DPV UL46 activates EGFR-PI3K-AKT pathway to antagonize innate immunity by interfering with cGAS-STING-TBK-IRF7 Signalsome formation.

Duck plague virus (DPV), a representative species of the Alphaherpesvirinae subfamily, poses significant economic and biosecurity threats to poultry production. Although the EGFR-PI3K-AKT signaling pathway is known to regulate diverse cellular processes-including cell growth, protein synthesis and immune regulation-and plays a significant role in viral infections, its interplay with the cGAS-STING pathway during DPV infection remains unclear. In this study, we demonstrate that DPV infection induces AKT1 phosphorylation in duck embryo fibroblast (DEF) cells. Overexpression of AKT1 significantly

Message in a bottle: lessons learned from antagonism of STING signalling during RNA virus infection.

STING has emerged in recent years as an important signalling adaptor in the activation of type I interferon responses during infection with DNA viruses and bacteria. An increasing body of evidence suggests that STING also modulates responses to RNA viruses, though the mechanisms remain less clear. In this review, we give a brief overview of the ways in which STING facilitates sensing of RNA viruses. These include modulation of RIG-I-dependent responses through STING's interaction with MAVS, and more speculative mechanisms involving the DNA sensor cGAS and sensing of membrane remodelling events

STING deficiency alleviates scar formation after glaucoma filtration surgery by suppressing p38 MAPK-induced inflammation in mice.

BACKGROUND: Glaucoma filtration surgery (GFS) often fails because of excessive scar formation driven by inflammation and fibroblast activation. Although the stimulator of interferon genes (STING) pathway is involved in inflammatory responses, its role in post-surgical fibrosis remains unclear. METHODS: A mouse GFS model was established in wild-type (WT) and STING-knockout (STING-KO) animals. A parallel cohort of WT mice received a single intraoperative subconjunctival injection of the STING inhibitor H151. Bleb survival, intraocular pressure, histopathology, collagen deposition, and inflammat

STING signaling sensing of DRP1-dependent mtDNA release in kupffer cells contributes to lipopolysaccharide-induced liver injury in mice.

Aberrant pro-inflammatory activation of Kupffer cells (KCs) is strongly involved in the pathogenesis of septic liver injury. Recent evidence indicates the crucial roles of excessive stimulator of interferon genes (STING) signaling activation during sepsis. However, the role of STING signaling in septic liver injury remains unclear. In this study, we demonstrated that STING signaling was markedly activated in KCs isolated from wild type mice after lipopolysaccharide (LPS) treatment. STING deficiency effectively protected liver function, attenuated systemic inflammatory response and decreased mo

A cell-based high throughput screening assay for the discovery of cGAS-STING pathway agonists.

Stimulator of interferon genes (STING) is an endoplasmic reticulum transmembrane protein that serves as a molecular hub for activation of interferon and inflammatory cytokine response by multiple cellular DNA sensors. Not surprisingly, STING has been demonstrated to play an important role in host defense against microorganisms and pharmacologic activation of STING is considered as an attractive strategy to treat viral diseases and boost antitumor immunity. In light of this we established a HepAD38-derived reporter cell line that expresses firefly luciferase in response to the activation of cyc

cGAS-STING Targeting Offers Novel Therapeutic Opportunities in Liver Diseases.

Cyclic GMP/AMP (cGAMP) synthase (cGAS), coupled with the endoplasmic reticulum (ER)-anchored adaptor protein stimulator of interferon genes (STING), constitute key components of the type 1 interferon signaling network. cGAS detects both pathogen-derived DNA and aberrant cytosolic self-DNA, establishing the cGAS-STING pathway as a central player in autoimmune disorders, sterile inflammation, and senescence-related processes. However, sustained abnormal activation of this signaling axis is implicated in the pathogenesis of chronic inflammatory and autoimmune conditions. Recent studies have uncov

Myeloid-Specific STING-YBX1 Interaction Alleviates Coagulation in Deep Vein Thrombosis.

BACKGROUND: Deep vein thrombosis (DVT), the third most common cause of cardiovascular deaths, is characterized by intravascular clot formation, often accompanied by inflammation. Although the STING (stimulator of interferon genes) signaling pathway is well recognized for its role in mediating inflammation in the context of infection, cellular stress, and tissue, its intricate involvement in DVT remains enigmatic. METHODS: In this study, we investigated the role of myeloid cell-intrinsic STING signaling in DVT progression using murine models. Both STING inhibitors and genetic depletion of myel

Nanoadjuvant-based membrane poration boosts antitumor immunity via modulating mitochondrial metabolism to downregulate PD-L1 and upregulate STING.

Activation of the stimulator of interferon genes (STING) pathway represents a promising strategy for cancer immunotherapy. However, clinical translation of STING agonists has been hampered by inefficient intracellular delivery and compensatory upregulation of the negative feedback immune checkpoint molecule of programmed death-ligand 1 (PD-L1). In addition, nuclear PD-L1 induces resistance by transcriptionally repressing STING, but clinically approved anti-PD-L1 antibodies cannot target nuclear PD-L1, limiting their efficacy. Here, we developed a nanoadjuvant (cGAMP + Halicin@PFDBA) to potenti

STING regulates porphyromonas gingivalis lipopolysaccharide-induced pyroptosis and inflammatory response through the NF-κB/NLRP3 signaling pathway in human gingival fibroblasts.

OBJECTIVE: The production of reactive oxygen species caused by antimicrobial response during periodontitis leads to the activation of NOD-like receptor protein 3 (NLRP3) inflammasome and pyroptosis. Stimulator of interferon genes (STING) has been found to be involved in regulating pyroptosis and inflammation in a variety of diseases. The present study aimed to investigate whether STING is involved in Porphyromonas gingivalis lipopolysaccharide (P.g LPS)-stimulated human gingival fibroblasts (HGFs) by regulating pyroptosis and inflammation. DESIGN: After culturing and identifying HGFs, HGFs we

Molecular Characterization, Expression and Functional Analysis of Chicken STING.

Innate immunity is an essential line of defense against pathogen invasion which is gained at birth, and the mechanism involved is mainly to identify pathogen-associated molecular patterns through pattern recognition receptors. STING (stimulator of interferon genes) is a signal junction molecule that hosts the perception of viral nucleic acids and produces type I interferon response, which plays a crucial role in innate immunity. However, relatively few studies have investigated the molecular characterization, tissue distribution, and potential function of STING in chickens. In this study, we c

The ubiquitin E3 ligase TRIM21 suppresses type I interferon signaling via STING degradation and ameliorates systemic autoimmunity.

Tripartite motif-containing 21 (TRIM21) is a cytoplasmic protein with E3 ubiquitin ligase activity. Although autoantibodies against TRIM21 are frequently detected in patients with systemic lupus erythematosus (SLE), its role in disease pathogenesis remains unclear. Here we demonstrate that TRIM21 directly interacts with the stimulator of interferon genes (STING) to regulate type I interferon (IFN) production. In both induced and spontaneous murine models of lupus, TRIM21 deficiency exacerbated lupus-like pathology and heightened IFN production after STING activation. By contrast, TRIM21 overex

Expanding roles of cGAS-STING signaling in neuroinflammation.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a central mediator of cytosolic DNA-induced innate immune responses, driving the production of type I IFNs and pro-inflammatory cytokines. Beyond its canonical role in cytosolic DNA sensing, increasing attention has been directed toward the noncanonical functions of cGAS and STING, particularly within the nucleus. Recent studies implicate dysregulated cGAS-STING signaling in neurodegenerative diseases and brain aging, with a prominent contribution to glial activation-associated neuroinflammation, a hallmark of m

Deubiquitinase OTUD6a drives cardiac inflammation and hypertrophy by deubiquitination of STING.

BACKGROUND: Cardiac hypertrophy is a crucial pathological characteristic of hypertensive heart disease and subsequent heart failure. Deubiquitinating enzymes (DUBs) have been found to be involved in the regulation of myocardial hypertrophy. OTU Domain-Containing Protein 6a (OTUD6a) is a recently identified DUB. To date, the potential role of OTUD6a in myocardial hypertrophy has not yet been revealed. METHODS AND RESULTS: We examined the up-regulated level of OTUD6a in mouse or human hypertrophic heart tissues. Then, transverse aortic constriction (TAC)- or angiotensin II (Ang II)- induced ven

Puerarin from Pueraria montana var. lobata (Willd.) alleviates hepatic steatosis and inflammation in MAFLD through suppressing STING-IRF3/NF-κB signaling in macrophages.

ETHNOPHARMACOLOGICAL RELEVANCE: Metabolic-associated fatty liver disease (MAFLD) has emerged as the most prevalent chronic liver disease globally. Accumulating evidence indicates that aberrant activation of the stimulator of interferon genes (STING) signaling pathway in hepatic macrophages plays a pivotal role in driving hepatic inflammation and steatosis. Puerarin, a bioactive isoflavone derived from Pueraria montana var. lobata (Willd.), has been reported to exert anti-inflammatory and lipid-lowering effects. However, the detailed mechanism of its actions in treating MAFLD remains to be full

The cGAS-STING Pathway in Bacterial Infection and Bacterial Immunity.

Cyclic guanosine monophosphate (GMP)-adenosine monophosphate (AMP) (cGAMP) synthase (cGAS), along with the adaptor stimulator of interferon genes (STING), are crucial components of the innate immune system, and their study has become a research hotspot in recent years. Many biochemical and structural studies that have collectively elucidated the mechanism of activation of the cGAS-STING pathway with atomic resolution have provided insights into the roles of the cGAS-STING pathway in innate immunity and clues to the origin and evolution of the modern cGAS-STING signaling pathway. The cGAS-STING

STING expression in colorectal cancer: prognostic role and immune microenvironment link via mIHC.

Colorectal cancer (CRC) is a malignant tumor originating in the mucosal epithelium of the colon or rectum and is among the most common cancers of the digestive system. Now it is the fourth deadliest cancer in the world. Stimulator of Interferon Genes (STING) is a dimeric transmembrane protein on the endoplasmic reticulum membrane that induces the secretion of type I interferons and pro-inflammatory cytokines and is triggered by the cell membrane DNA of pathogens and hosts. It is widely expressed in immune cells (such as dendritic cells, macrophages) and some tumor cells, and has been shown to

Alpha-lipoic acid (ALA) ameliorates early brain injury after subarachnoid hemorrhage in Sprague-Dawley (SD) rats via inhibiting STING-NLRP3 inflammatory signaling.

Neuroinflammation is intimately associated with poor prognosis in patients with subarachnoid hemorrhage (SAH). Alpha-lipoic acid (ALA), a disulfide antioxidant, has been shown to be neuroprotective in an in vivo model of neurological injury; however, the role of ALA in SAH has never been evaluated. In this study, the Sprague-Dawley rats SAH model was induced by endovascular perforation method. ALA was transplanted intravenously into rats, and SR-717, a stimulator of interferon genes (STING) agonist, was injected intraperitoneally. The effects of ALA on early brain injury were assayed by neurol
