A Preliminary Analysis of the Influence of Elderberry Juice Consumption on Thyroid Metabolism in Mice and Humans Fed High-Fat Diets.

BACKGROUND/OBJECTIVES: Elderberry juice (EBJ) consumption prevents weight gain in mice fed a high-fat diet and increases fat oxidation in response to a meal challenge in overweight humans. Thyroid hormones influence metabolism and substrate oxidation, and the impact of EBJ consumption on thyroid homeostasis remains unexplored. Thus, the primary objective of this analysis was to investigate whether elderberry consumption in mice and humans affects serum thyroid biomarkers. METHODS: Serum samples from a previous trial incorporating an EBJ intervention were analyzed for thyroxine (T4), thyroid s

Avian influenza receptor expression in H5N1-infected and noninfected human tissues.

Avian and human influenza viruses preferentially bind to alpha-2,3-linked and alpha-2,6-linked sialic acids, respectively. Until today, the distributions of these two receptor types had never been investigated in H5N1-infected human tissue samples. Here, the expression of avian (AIV-Rs) and human influenza receptors (HuIV-Rs) is studied in various organs (upper and lower respiratory tracts, brain, placenta, liver, kidney, heart, intestines, and spleen) of two H5N1 cases and 14 control cases. Histochemical stains using biotinylated Maackia amurensis lectin II and Sambucus nigra agglutinin were

Identification of 9-O-acetyl-N-acetylneuraminic acid in normal canine pre-ocular tear film secreted mucins and its depletion in Keratoconjunctivitis sicca.

O-Acetylated sialic acids have been reported in many sialoglycoproteins where they mediate a variety of immune and other biological events. We have previously demonstrated that the protective mucus barrier on the surface of the canine eye contains sialoglycoproteins. We have also investigated the occurrence of O-acetylated sialic acids in these ocular mucins. Mucus aspirated from the surface of normal dog eyes and those with keratoconjunctivitis sicca (KCS) was fractionated into three pools by density gradient centrifugation. Sialic acids comprised 0.6-0.9% of the dry weight of the mucins isol

ST6GAL1-mediated sialyl linkage switching drives cancer-promoting α2,6-sialo-protrusions radiating from anti-inflammatory TAMs.

Tumor-associated macrophages (TAMs) are functionally diverse effector cells in the tumor microenvironment (TME). Pro- and anti-inflammatory TAMs are central to cancer progression by shaping inflammation and immune homeostasis, but it remains unknown if polarization-induced remodeling of the TAM glycocalyx critical for cellular communication and interactions occurs within the TME. Taking a systems glycobiology approach, we first used cell surface-focused glycomics and lectin flow cytometry of ex vivo polarized monocyte-derived macrophages to demonstrate profound sialyl linkage switching in proi

Structure of the N-linked oligosaccharides of the human transferrin receptor.

Human transferrin receptor was isolated from placenta and from the hepatocarcinoma cell line Hep G2. Asparagine-linked oligosaccharides were released by treatment of tryptic glycopeptides with endo-beta-N-acetylglucosaminidase H or peptide-N4-(N-acetyl-beta-glucosaminyl)asparagine amidase F. Oligosaccharide alditols were fractionated by anion-exchange high-performance liquid chromatography and by high-pH anion-exchange chromatography. Glycans from placental transferrin receptor were further characterized, after desialylation, by methylation analysis and, in part, by liquid secondary-ion mass s

Differences in IgG Sialylation Distinguish Asymptomatic From Symptomatic Antinuclear Antibody-Positive Individuals.

OBJECTIVE: The transition from asymptomatic antinuclear antibody (ANA) positivity to systemic autoimmune rheumatic disease (SARD) is associated with increased production of proinflammatory factors such as tumor necrosis factor α (TNFα). Here we investigate whether the relative absence of inflammation in asymptomatic ANA+ individuals (ANA+NS) results from a lack of circulating immune complexes (ICs) or from changes in the characteristics of the IgG autoantibodies produced. METHODS: Flow cytometry was used to characterize circulating microparticles (MPs) in 18 healthy controls (ANA-HC), 31 ANA+

Spatial mapping of influenza and coronavirus receptors in the respiratory and intestinal tract epithelium of beef cattle using advanced PixF image analysis.

Influenza A viruses (IAV) and coronaviruses (CoV) pose significant threats to various animal species, including cattle. Reports of SARS-CoV-2 infections and recent outbreaks of highly pathogenic avian influenza (HPAI H5N1) in dairy cattle highlight the need to understand receptor distributions critical for viral entry. This study investigates the spatial distribution of IAV and CoV receptors in bovine tissues using PixF, a novel and newly developed web-based image analysis tool. Respiratory (trachea and lung) and intestinal (small and large intestine) tissues from crossbred Holstein-Angus stee

A glutaminyl cyclase-catalyzed α-synuclein modification identified in human synucleinopathies.

Parkinson's disease (PD) is a progressive neurodegenerative disorder that is neuropathologically characterized by degeneration of dopaminergic neurons of the substantia nigra (SN) and formation of Lewy bodies and Lewy neurites composed of aggregated α-synuclein. Proteolysis of α-synuclein by matrix metalloproteinases was shown to facilitate its aggregation and to affect cell viability. One of the proteolysed fragments, Gln79-α-synuclein, possesses a glutamine residue at its N-terminus. We argue that glutaminyl cyclase (QC) may catalyze the pyroglutamate (pGlu)79-α-synuclein formation and, ther

Association of the Sialylation of Antibodies Specific to the HCV E2 Envelope Glycoprotein with Hepatic Fibrosis Progression and Antiviral Therapy Efficacy.

The E2 envelope glycoprotein of the hepatitis C virus (HCV) is a major target of broadly neutralizing antibodies that are closely related to a spontaneous cure of HCV infection. There is still no data about the diversity of E2-specific antibodies (Abs) glycosylation. The aim of this study was to analyze the level and sialylation of E2 IgG Abs, the relation of the respective changes to hepatic fibrosis (F) progression and their possible association with the efficacy of interferon-α-2a plus ribavirin (IFN-RBV) antiviral therapy. One hundred three HCV infected treatment-naive patients were examin

The role of integrin glycosylation in galectin-8-mediated trabecular meshwork cell adhesion and spreading.

Primary open angle glaucoma (POAG) is a major blindness-causing disease, characterized by elevated intraocular pressure due to an insufficient outflow of aqueous humor. The trabecular meshwork (TM) lining the aqueous outflow pathway modulates the aqueous outflow facility. TM cell adhesion, cell-matrix interactions, and factors that influence Rho signaling in TM cells are thought to play a pivotal role in the regulation of aqueous outflow. In a recent study, we demonstrated that galectin-8 (Gal8) modulates the adhesion and cytoskeletal arrangement of TM cells and that it does so through binding

Alpha-2,3 sialylation differentiate the limbal and corneal epithelial cell phenotypes.

PURPOSE: The initial differentiation event for the corneal epithelial cell lineage occurs as the limbally localized stem cells yield, through mitosis, the highly proliferative, transiently amplifying corneal peripheral cells. This differentiation is characterized by the expression of tissue-specific cytokeratins, as well as the loss of alpha-enolase and pigmentation. All these are intracellular events. The aim of this study was to identify and characterize, through lectin analysis, changes in cell surface properties associated with differentiation. METHODS: Cryostat sections of the limbo-corn

Divergence of Ethnobotanical Knowledge of Slovenians on the Edge of the Mediterranean as a Result of Historical, Geographical and Cultural Drivers.

State boundaries limit human contacts in a homogenous context of a landscape and its natural features, including plants. After nine centuries of separation, finally the two territories in Slovenia share the same political history. In this paper we tried to answer the question to which extent the past political borders, geographical and cultural drivers affect today's traditional knowledge on wild plants use of Slovenians, living unified in the same political entity. Data were collected using 60 in-depth semi-structured interviews, from March to August 2019, in two municipalities: Komen at Kars
