Norovirus Detection at Oualidia Lagoon, a Moroccan Shellfish Harvesting Area, by Reverse Transcription PCR Analysis.

Norovirus (NoV) is the leading cause of acute viral gastroenteritis outbreaks in the world. These outbreaks are frequently associated with bivalve shellfish consumption, particularly because these products are often eaten raw or only slightly cooked. In Morocco, regulations concerning the acceptable levels of enteric bacteria indicator organisms in these products have been put in place. However, these regulations do not take into account the risk of viral contamination, and many gastroenteritis outbreaks have been linked to the ingestion of bivalve shellfish from areas that comply with the cur

Development of a F actin-based live-cell fluorimetric microplate assay for diarrhetic shellfish toxins.

A new cytotoxicity assay for detection and quantitation of diarrhetic shellfish toxins (DSP) is presented. This assay is based upon fluorimetric determination of F-actin depolymerization induced by okadaic acid (OA)-class compounds in the BE(2)-M17 neuroblastoma cell line. No interferences were observed with other marine toxins such as saxitoxin, domoic acid, or yessotoxin, thus indicating a good specificity of the assay as expected by the direct relationship between protein phosphatase inhibition and cytoskeletal changes. The proposed method is rapid (<2h) and shows a linear response in the r

Shellfish as a Potential Source of Hepatitis E Virus: Epidemiological Evidence, Biological Plausibility, and Research Gaps.

Hepatitis E virus (HEV) is an important cause of acute and chronic hepatitis worldwide, transmitted primarily through waterborne exposure and zoonotic foodborne pathways. In recent years, shellfish have attracted growing attention as a potential vehicle for HEV transmission. This interest is driven by epidemiological observations linking shellfish consumption to human HEV infection and by repeated detection of HEV RNA in bivalve mollusks across multiple geographic regions. This review critically evaluates the current evidence by integrating epidemiological data, environmental and food surveill

Use of capture based metagenomic to assess human enteric virus diversity in sewage and shellfish samples.

Human sewage is the main source of contamination of environmental waters with human enteric viruses, that can contaminate food such as shellfish. Metagenomic represents a new way of analyzing viral diversity through an a priori massive parallel sequencing approach. However, the precise identification of enteric viruses in sewage or shellfish matrices, is still challenging due to the low viral load, large diversity of viral genera and the large amounts of matrix masking viral sequences. This work compared three commercial kits using capture-based enrichment during the library preparation, for t

Survey, characterization and antimicrobial susceptibility of Clostridium difficile from marine bivalve shellfish of North Adriatic Sea.

Clostridium difficile is a major cause of infectious diarrhea associated to healthcare settings. Community-acquired infections are increasingly reported in the last decade and exposure other than to symptomatic patients rather to contaminated foods or animals is feasible. Occurrence of C. difficile in shellfish raises concern because spores can survive the cooking temperatures given that shellfish is often consumed poorly cooked or raw. Aim of our study was to investigate whether shellfish represents a reservoir of C. difficile human PCR-ribotypes (RTs). 702 shellfish samples of farmed and wil

Genomic Characterization of Arcobacter butzleri Isolated From Shellfish: Novel Insight Into Antibiotic Resistance and Virulence Determinants.

Arcobacter (A.) butzleri is an emerging pathogenic microorganism, whose taxonomy has been recently suggested to be emended to the Aliarcobacter (Al.) butzleri comb. nov. Despite extensive taxonomic analysis, only few fragmented studies have investigated the occurrence and the prevalence of virulence and antibiotic resistance determinants of this species in strains isolated from shellfish. Herein we report for the first time the whole genome sequencing and genomic characterization of two A. butzleri strains isolated from shellfish, with particular reference to the antibiotic, heavy metals and v

Occurrence of lipophilic marine toxins in shellfish from Galicia (NW of Spain) and synergies among them.

Lipophilic marine toxins pose a serious threat for consumers and an enormous economic problem for shellfish producers. Synergistic interaction among toxins may play an important role in the toxicity of shellfish and consequently in human intoxications. In order to study the toxic profile of molluscs, sampled during toxic episodes occurring in different locations in Galicia in 2014, shellfish were analyzed by liquid chromatography tandem mass spectrometry (LC-MS/MS), the official method for the detection of lipophilic toxins. The performance of this procedure was demonstrated to be fit for purp

Spatial distribution of lipophilic shellfish toxins in seawater and sediment in the Bohai Sea and the Yellow Sea, China.

Lipophilic shellfish toxins (LSTs) are widely distributed in marine environments worldwide, potentially threatening marine ecosystem health and aquaculture safety. In this study, two large-scale cruises were conducted in the Bohai Sea and the Yellow Sea, China, in spring and summer 2023 to clarify the composition, concentration, and spatial distribution of LSTs in the water columns and sediments. Results showed that okadaic acid (OA), dinophysistoxin-1 (DTX1) and/or pectenotoxin-2 (PTX2) were detected in 249 seawater samples collected in spring and summer. The concentrations of ∑LSTs in seawat

Prevalence, typing and antimicrobial resistance of Salmonella isolates from commercial shellfish in the North coast of Morocco.

Salmonellosis is one of the most common foodborne illnesses in the world. The irrational use of antibiotics in medicine and in animal nutrition has greatly favored the emergence and spread of resistant strains of non-typhoid Salmonella. This study aims the determination of the prevalence of Salmonella in bivalve mollusks in Northern Morocco, as well as the molecular typing and antibiotic susceptibility testing of the strains isolated from positive samples. In total, 150 samples from shellfish composed of mussels (Mytilus galloprovincialis), clams (Callista chione and Ruditapes descussatus) and

The effect of temperature on growth and production of paralytic shellfish poisoning toxins by the cyanobacterium Cylindrospermopsis raciborskii C10.

Cylindrospermopsis raciborskii is a cyanobacterium which produces either cylindrospermopsine or paralytic shellfish poisoning (PSP) toxins. We studied the effect of temperature on growth and production of PSP toxins by C. raciborskii C10, isolated from a freshwater reservoir in Brazil. We analyzed the extracellular and intracellular content of PSP toxins at two different temperatures: 19 and 25 degrees C. C. raciborskii C10 produces STX, GTX2, and GTX3 at both temperatures. dcSTX was also detected at 25 degrees C in the intracellular extracts obtained at the end of the stationary phase. The gr

Epidemiology of Foodborne Diseases Transmitted by Fish, Shellfish and Marine Crustaceans in the United States, 1970-1978.

Fish, mollusks, marine crustaceans and marine mammals were implicated as vehicles in approximately 11% of foodborne disease outbreaks reported in the United States during the years 1970-1978. Tuna, clams, oysters, shrimp, and crab frequently were vehicles. These animals can acquire the dinoflagellates responsible for ciguatera, paralytic shellfish poisoning and neurotoxic shellfish poisoning from their marine habitats. Many common marine organisms found in and on fish can decarboxylate histidine to histamine, and do so during improper storage of fish. Vibrio parahaemolyticus and Clostridium bo

Meat, dairy, fish, and shellfish consumption and per- and polyfluoroalkyl substance (PFAS) exposure in the U.S. population.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants for which consumption of animal-derived foods represents an important exposure pathway. Using nationally representative data from the U.S. National Health and Nutrition Examination Survey (NHANES) 2003-2020, we examined associations between intake of meat, dairy, fish, and shellfish and serum PFAS concentrations. Total serum PFAS showed an approximately 75% decline over the study period, primarily reflecting reductions in perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA). Shellfish consumption

The occurrence of paralytic shellfish toxins in invertebrates, forage fishes and predatory fishes across southern Alaska.

In Alaska, paralytic shellfish poisoning (PSP) is associated with the toxic dinoflagellate Alexandrium catenella, which blooms annually along the coastline from the Canadian border to the Beaufort Sea. While it has long been recognized that these blooms cause annual outbreaks of shellfish toxicity each spring and summer, little is known about other pathways for PSP toxins in the marine food web. Here, we present data from a four-year study to characterize the occurrence of PSTs in marine non-bivalve invertebrates, forage fishes and predatory fishes across the breadth of southern Alaska. Time dependence of the pre-chromatographic oxidation method for bivalves contaminated with paralytic shellfish poisoning toxins.

The neurotoxins of the saxitoxin family can be the origin of the human neurological syndrome of paralytic shellfish poisoning via contaminated marine bivalve vectors. A pre-chromatographic oxidation method is the official testing method in the EU, also known as the 'Lawrence method'. It involves several steps, including toxin extraction, solid-phase clean-up, and oxidation to produce fluorescent derivatives. Despite the manual peroxide oxidation involving four pipetting steps, high coefficients of determination were commonly obtained for the calibration curves of the respective toxin oxidation

Development of enzyme-linked immunosorbent assay with nanogold-labeled probes for detection of diarrhetic shellfish toxins.

Diarrheal shellfish toxins (DSTs) are a type of liposoluble toxins produced by toxic algae in the ocean, with main components of okadaic acid (OA), dinotricin-1 (DTX-1), and dinotricin-2 (DTX-2). They can accumulate in shellfish through food chain. Human consumption of toxic shellfish can have adverse effects on health. Therefore, conducting DSTS detection can effectively ensure food safety. Base on the principle of competitive enzyme-linked immunosorbent assay (ELISA), referring to the seed growth method, we prepared goldnano rods using gold nanoparticles (AuNPs), and conjugated them with oka

A Generic LC-HRMS Screening Method for Marine and Freshwater Phycotoxins in Fish, Shellfish, Water, and Supplements.

Phycotoxins occur in various marine and freshwater environments, and can accumulate in edible species such as fish, crabs, and shellfish. Human exposure to these toxins can take place, for instance, through consumption of contaminated species or supplements and through the ingestion of contaminated water. Symptoms of phycotoxin intoxication include paralysis, diarrhea, and amnesia. When the cause of an intoxication cannot directly be found, a screening method is required to identify the causative toxin. In this work, such a screening method was developed and validated for marine and freshwater

First evidence that emerging pinnatoxin-G, a contaminant of shellfish, reaches the brain and crosses the placental barrier.

Massive proliferation of some toxic marine dinoflagellates is responsible for the occurrence of harmful algal blooms and the contamination of fish and shellfish worldwide. Pinnatoxins (PnTx) (A-H) comprise an emerging phycotoxin family belonging to the cyclic imine toxin group. Interest has been focused on these lipophilic, fast-acting and highly potent toxins because they are widely found in contaminated shellfish, and can represent a risk for seafood consumers. PnTx display a potent antagonist effect on nicotinic acetylcholine receptors (nAChR), and in this study we assessed in vivo the abil

A Mediterranean Alexandrium taylorii (Dinophyceae) Strain Produces Goniodomin A and Lytic Compounds but Not Paralytic Shellfish Toxins.

Species of the dinophyte genus Alexandrium are widely distributed and are notorious bloom formers and producers of various potent phycotoxins. The species Alexandrium taylorii is known to form recurrent and dense blooms in the Mediterranean, but its toxin production potential is poorly studied. Here we investigated toxin production potential of a Mediterranean A. taylorii clonal strain by combining state-of-the-art screening for various toxins known to be produced within Alexandrium with a sound morphological and molecular designation of the studied strain. As shown by a detailed thecal plate
