Pain and plasticity: is chronic pain always associated with somatosensory cortex activity and reorganization?

The somatosensory cortex remodels in response to sensory deprivation, with regions deprived of input invaded by neighboring representations. The degree of cortical reorganization is correlated with ongoing pain intensity, which has led to the assumption that chronic pain conditions are invariably associated with somatosensory cortex reorganization. Because the presentation and etiology of chronic pain vary, we determined whether cortical changes in human subjects are similar for differing pain types. Using functional and anatomical magnetic resonance imaging, we found that, while human patient

Altered expression of neuropeptides in the primary somatosensory cortex of the Down syndrome model Ts65Dn.

Down syndrome is the most common genetic disorder associated with mental retardation. Subjects and mice models for Down syndrome (such as Ts65Dn) show defects in the formation of neuronal networks in both the hippocampus and the cerebral cortex. The principal neurons display alterations in the morphology, density and distribution of dendritic spines in the cortex as well as in the hippocampus. Several evidences point to the possibility that the atrophy observed in principal neurons could be mediated by changes in their inhibitory inputs and, in fact, an imbalance between excitation and inhibit

Dynamic modulation of the primary somatosensory cortex during seeing and feeling a touched hand.

Previous work has demonstrated cross-modal links between vision and somatosensation at an early stage of sensory processing. Furthermore, recent behavioral studies have shown that viewing the stimulated body part can enhance tactile discrimination ability at the stimulated site. This study aims to investigate the role of the primary somatosensory cortex (SI) during visuotactile integration processes. Subjects looked at a hand in a video being touched on the first digit (D1) in synchrony with felt touches on their real hidden hand as compared with watching a video with asynchronous touches. Dur

Terbutaline is a developmental neurotoxicant: effects on neuroproteins and morphology in cerebellum, hippocampus, and somatosensory cortex.

Beta(2)-adrenoceptor agonists, especially terbutaline, are widely used to arrest preterm labor, but they also cross the placenta to stimulate fetal beta-adrenoceptors that control neural cell differentiation. We evaluated the effects of terbutaline administration in neonatal rats, a stage of neurodevelopment corresponding to human fetal development. Terbutaline administered on postnatal days PN2 to 5 elicited neurochemical changes indicative of neuronal injury and reactive gliosis: immediate increases in glial fibrillary acidic protein and subsequent induction of the 68-kDa neurofilament prote

Hand/face border as a limiting boundary in the body representation in monkey somatosensory cortex.

Horizontal intracortical connections may form one substrate for representational plasticity in somatosensory cortex. Electrophysiological mapping demonstrated the finer details of the representations of the hand, lower jaw, neck, and face in area 3b of normal macaque monkeys. Injections of two fluorescent tracers then defined the extent to which horizontal connections crossed from the face into the hand representations and vice versa in area 3b. Connections are widely distributed within cortical representations of skin areas innervated by cervical nerves or by the trigeminal nerve but do not c

Effects of prenatal exposure to ethanol on callosal projection neurons in rat somatosensory cortex.

The distribution and density of callosal projection neurons in the somatosensory cortex of mature rats was altered by prenatal exposure to ethanol. The density of callosal neurons was significantly greater in ethanol-treated rats than in controls. Ethanol exposure also altered the laminar distribution of callosal projection neurons. Whereas in control rats the cell bodies of callosal projection neurons were in layers II/III and V, in ethanol-treated rats most of these neurons were distributed in layers V and VI. Many of the ectopic neurons were generated toward the end of cortical neuronogenes

Benzodiazepines and synaptic processing in the spatial domain within the cat's primary somatosensory cortex.

In the primary somatosensory cortex of cats, the size of the receptive fields (RFs) of cutaneously responsive neurones is under the control of gamma-aminobutyric acid (GABA) mediated inhibition when the cells are situated in rapidly adapting (RA) background regions. Cells located in slowly adapting (SA) or low-velocity rapidly adapting (LVRA) background regions do not appear to be affected by GABA significantly in the spatial domain, although other response properties such as threshold and firing pattern are under the influence of bicuculline methiodide (BMI) sensitive processes. The GABA rece

Dynamic recruitment of PV interneurons in the somatosensory cortex induced by experience dependent plasticity.

Adaptive circuit plasticity plays crucial roles in the brain during development, learning, sensory experience, and after injury. During chronic whisker trimming, a well-studied paradigm for inducing experience dependent plasticity, whisker representations in the somatosensory barrel cortex (S1BF) undergo remapping, with expansion of maps for spared whiskers and contraction of maps for trimmed whiskers. At the cellular level, excitatory pyramidal cells in Layer 2/3 shift their whisker tuning, increasing selectivity to spared whiskers and away from deprived whiskers. While these changes are well

Learning induced neuronal identity switch in the superficial layers of the primary somatosensory cortex.

During learning, multi-dimensional inputs are integrated within the sensory cortices. However, the strategies by which the sensory cortex employs to achieve learning remains poorly understood. We studied the sensory cortical neuronal coding of trace eyeblink conditioning (TEC) in head-fixed, freely running mice, where whisker deflection was used as a conditioned stimulus (CS) and an air puff to the cornea delivered after an interval was used as unconditioned stimulus (US). After training, mice learned the task with a set of stereotypical behavioral changes, most prominent ones include prolonge

Pain-related neuronal ensembles in the primary somatosensory cortex contribute to hyperalgesia and anxiety.

The mechanism by which acute pain or itch information at the periphery is processed in the primary somatosensory cortex (S1) remains unclear. To elucidate this, we used a viral-mediated targeted-recombination-in-active population system to target S1 neuronal ensembles that are active during pain or itch sensations. We induced the expression of excitatory or inhibitory designer receptors exclusively activated by designer drugs in pain- or itch-related S1 neurons. We identified neuronal populations in mice that regulate the sensory components of pain and itch in the S1 hind paw region. Notably,

Responses of neurons in the primary somatosensory cortex to itch- and pain-producing stimuli in rats.

Understanding of cortical encoding of itch is limited. Injection of pruritogens and algogens into the skin of the cheek produces distinct behaviors, making the rodent cheek a useful model for understanding mechanisms of itch and pain. We examined responses of neurons in the primary somatosensory cortex by application of mechanical stimuli (brush, pressure, and pinch) and stimulations with intradermal injections of pruritic and algesic chemical of receptive fields located on the skin of the cheek in urethane-anesthetized rats. Stimuli included chloroquine, serotonin, β-alanine, histamine, capsa

Reorganization of the somatosensory cortex in hemiplegic cerebral palsy associated with impaired sensory tracts.

Functional neuroimaging studies argue that sensory deficits in hemiplegic cerebral palsy (HCP) are related to deviant somatosensory processing in the ipsilesional primary somatosensory cortex (S1). A separate body of structural neuroimaging literature argues that these deficits are due to structural damage of the ascending sensory tracts (AST). The relationship between the functional and structural integrity of the somatosensory system and the sensory performance is largely unknown in HCP. To address this relationship, we combined findings from magnetoencephalography (MEG) and probabilistic di

Seeing is not feeling: posterior parietal but not somatosensory cortex engagement during touch observation.

Observing touch has been reported to elicit activation in human primary and secondary somatosensory cortices and is suggested to underlie our ability to interpret other's behavior and potentially empathy. However, despite these reports, there are a large number of inconsistencies in terms of the precise topography of activation, the extent of hemispheric lateralization, and what aspects of the stimulus are necessary to drive responses. To address these issues, we investigated the localization and functional properties of regions responsive to observed touch in a large group of participants (n

Illusory and veridical mapping of tactile objects in the primary somatosensory and posterior parietal cortex.

While several behavioral and neuroscience studies have explored visual, auditory, and cross-modal illusions, information about the phenomenology and neural correlates of somatosensory illusions is meager. By combining psychophysics and somatosensory evoked potentials, we explored in healthy humans the neural correlates of 2 compelling tactuo-proprioceptive illusions, namely Aristotle (1 object touching the contact area between 2 crossed fingers is perceived as 2 lateral objects) and Reverse illusions (2 lateral objects are perceived as 1 between crossed-fingers object). These illusions likely

Does very premature birth affect the functioning of the somatosensory cortex?--A magnetoencephalography study.

Increased survival of extremely low birth weight infants has led to a need for new prognostic methods to predict possible future neurological impairment. We investigated the early development of the somatosensory system by recording the somatosensory evoked magnetic fields (SEFs) during natural sleep at fullterm age in 16 very prematurely born infants and 16 healthy newborns born at term. The purpose was to determine possible changes in the function of the somatosensory cortex in the prematurely born infants by comparing the latency, strength, location and morphology of the SEFs with those of

Oxygen and glucose deprivation induces major dysfunction in the somatosensory cortex of the newborn rat.

The mechanisms and functional consequences of ischemia-induced injury during perinatal development are poorly understood. Subplate neurons (SPn) play a central role in early cortical development and a pathophysiological impairment of these neurons may have long-term detrimental effects on cortical function. The acute and long-term consequences of combined oxygen and glucose deprivation (OGD) were investigated in SPn and compared with OGD-induced dysfunction of immature layer V pyramidal cortical neurons (PCn) in somatosensory cortical slices from postnatal day (P)0-4 rats. OGD for 50 min follo

Disinhibition of the somatosensory cortex in cervical dystonia-decreased amplitudes of high-frequency oscillations.

OBJECTIVE: To determine whether patients with cervical dystonia have electrophysiological signs of disinhibition in the somatosensory cortex by recording high-frequency oscillations (HFOs) in somatosensory evoked potentials (SEPs). METHODS: HFOs were recorded in 13 patients and 10 age-matched control subjects, and the data were analyzed statistically by paired comparison and by Pearson's correlation. RESULTS: In patients with cervical dystonia, the early part of HFOs showed a significant decrease in amplitude, and the amplitude ratios of both early and late parts of HFOs/N20 potential were a

Correlating cytoarchitecture and function in cat primary somatosensory cortex: the challenge of individual differences.

Principles of organization for the primary somatosensory cortex are generalizations derived by examining data obtained in different individuals. The manner in which these data are combined influences the conclusions derived. We found the line representing the widest anteroposterior distance across the sigmoid gyrus to be a useful reference in the cat somatosensory cortex for combining and comparing electrophysiological and cytoarchitectonic data from different individuals when we constructed cytoarchitectonic and functional maps of the bank of the medial ansate sulcus; maps prepared from combi

Uneven densities of corticopontine neurons in the somatosensory cortex: a quantitative experimental study in the cat.

By use of large injections in the pontine nuclei of wheat germ agglutinin-horseradish peroxidase conjugate, the distribution of corticopontine cells in the primary somatosensory cortex (SI) was mapped quantitatively. The borders of the cytoarchitectonic areas 3a, 3b, 1 and 2, together constituting SI, were determined cyto- and myeloarchitectonically. Flat maps showing the distribution of labelled cells in SI were constructed. All labelled cells were confined to lamina V, and present in deep as well as superficial parts of this lamina. The size spectrum of the corticopontine cells seemed to enc
