Microinjections of leupeptin in the frontal cortex or dorsal hippocampus block spatial learning in the rat.

Adult male Long-Evans hooded rats were given bilateral microinjections of 18 mM leupeptin or saline through cannulae implanted with tips aimed at the frontal cortex or CA1 or CA3 hippocampal cell fields. Five minutes following injections animals were allowed to complete an eight-arm radial maze. The acquisition criterion required that the animal make 7 correct choices of the first 8, and 8 correct choices of the first 10, for five consecutive sessions. Leupeptin slowed acquisition of the eight-arm radial maze task when injected into the CA1 and CA3 hippocampal fields or the frontal cortex, but

Intact spatial learning in Fmr1-/y , Syngap1+/-, and Grin2b+/- rat models of neurodevelopmental disorders.

INTRODUCTION: FMR1, SYNGAP1, and GRIN2B gene mutations Q8 are known causes of mild to severe intellectual disability (ID) in humans, with affected individuals frequently co-diagnosed with autism spectrum disorder (ASD). These three genes influence molecular mechanisms underlying synaptic plasticity, and thus spatial memory behaviors have been of interest in studying rodent models of these mutations. However, evidence of spatial memory impairment across these models is inconsistent, with some studies reporting deficits and others finding none. METHOD: In the present study, we used a variant of

HisToSpatialCNV: an interpretable deep learning method predicting spatial copy number variations from histopathology images.

Copy number variations (CNV) are key drivers of cancer progression, yet methods for predicting spatial CNVs directly from haematoxylin and eosin (H&E) images are currently lacking. We introduce HisToSpatialCNV, a multiscale deep-learning framework that integrates histopathological features from H&E-stained images to infer spatial CNV patterns. Combining interpretable feature extraction with graph neural networks and multihead self-attention, our approach captures both local and global tissue contexts. Applied to HER2+ breast cancer, skin cancer and brain cancer datasets, HisToSpatialCNV outper

A Learning-based Framework for Spatial Impulse Response Compensation in 3D Photoacoustic Computed Tomography.

Photoacoustic computed tomography (PACT) is a promising imaging modality that combines the advantages of optical contrast with ultrasound detection. Utilizing ultrasound transducers with larger surface areas can improve detection sensitivity. However, when computationally efficient analytic reconstruction methods that neglect the spatial impulse responses (SIRs) of the transducer are employed, the spatial resolution of the reconstructed images will be compromised. Although optimization-based reconstruction methods can explicitly account for SIR effects, their computational cost is generally hi

Dual-value reinforcement learning with delayed local welfare feedback in spatial public goods games.

Cooperation is difficult to sustain in public-goods dilemmas because contributing can reduce an individual's immediate payoff, while benefits to the surrounding group may emerge only after several rounds. We introduce a dual-value reinforcement-learning model for a spatial public goods game that evaluates these two consequences separately. One value system learns from the individual payoff obtained after each action, whereas the other learns from local welfare accumulated over multiple rounds. The two evaluations are combined only when an action is selected. Within the tested settings, numeric

SPADE: A Deep Learning Framework for Spatial Mapping and Quantitative Cell-Cell Interaction Inference.

Spatial transcriptomics (ST) enables the study of tissue architecture by resolving gene expression in space, but current ST platforms are constrained by limited sequencing depth and indirect single-cell identification. Existing deconvolution methods that integrate single-cell RNA sequencing (scRNA-seq) data with ST often overlook the biological principle that cells in communication with each other tend to be closer spatially. Here we introduce SPADE, a deep learning framework that aligns scRNA-seq data to spatial locations by jointly modeling expression similarity between scRNA-seq and ST data

Cascaded deep learning enables multimodal brain PET spatial normalization and quantification for Alzheimer's disease.

Semi-quantitative positron emission tomography (PET) analysis, particularly Centiloid and CenTauRz scaling, is essential for Alzheimer's disease (AD) research and diagnosis. However, standard quantification workflows often depend on structural MRI for spatial normalization (SN) or rely on computationally intensive software, limiting clinical accessibility. In this retrospective, multi-center study (3539 patients; 6535 scans; 2005-2025), we compiled data across 7 modalities and 13 tracers to develop and validate the Deep Cascaded Cerebral Calculator (DCCC). This fully automated, PET-only framew

Pseudogene Lamr1-ps1 Aggravates Early Spatial Learning Memory Deficits in Alzheimer's Disease Model Mice.

Alzheimer's disease (AD), a neurodegenerative disorder with complex etiologies, manifests through a cascade of pathological changes before clinical symptoms become apparent. Among these early changes, alterations in the expression of non-coding RNAs (ncRNAs) have emerged as pivotal events. In this study, we focused on the aberrant expression of ncRNAs and revealed that Lamr1-ps1, a pseudogene of the laminin receptor, significantly exacerbates early spatial learning and memory deficits in APP/PS1 mice. Through a combination of bioinformatics prediction and experimental validation, we identified

EEG Temporal-Spatial Feature Learning for Automated Selection of Stimulus Parameters in Electroconvulsive Therapy.

The risk of adverse effects in Electroconvulsive Therapy (ECT), such as cognitive impairment, can be high if an excessive stimulus is applied to induce the necessary generalized seizure (GS); Conversely, inadequate stimulus results in failure. Recent efforts to automate this task can facilitate statistical analyses on individual parameters or qualitative predictions. However, this automation still significantly lags behind the requirements in clinical practices. This study addresses this issue by predicting the probability of GS induction under the joint restriction of a patient's EEG (electro

Integrating machine learning and spatial clustering for malaria case prediction in Brazil's Legal Amazon.

Malaria remains a major global health challenge, particularly in Brazil's Legal Amazon region, where environmental and socioeconomic conditions foster favorable conditions for disease transmission. Traditional control measures have shown limited effectiveness, emphasizing the need for better predictive approaches to support timely and targeted public health interventions. This study evaluates the performance of six computational models-Long Short-Term Memory (LSTM), Gated Recurrent Units (GRU), Support Vector Regression (SVR), Random Forest (RF), eXtreme Gradient Boosting (XGBoost), and Autore

Deep Learning-Enhanced Hand-Driven Spatial Encoding Microfluidics for Multiplexed Molecular Testing at Home.

The frequent global outbreaks of viral infectious diseases have significantly heightened the urgent demand for molecular testing at home. However, the labor-intensive sample preparation and nucleic acid amplification steps, along with the complexity and bulkiness of detection equipment, have limited the large-scale application of molecular testing at home. Here, we propose artificial intelligence-enhanced hand-driven microfluidic system (MACRO) based on RPA and CRISPR technologies for home diagnosis of multiple types of infectious diseases. Improved pharmacokinetic parameter estimation from DCE-MRI via spatial-temporal information-driven unsupervised learning.

Objective.Pharmacokinetic (PK) parameters derived from dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) provide quantitative characterization of tissue perfusion and permeability. However, existing deep learning methods for PK parameter estimation rely on either temporal or spatial features alone, overlooking the integrated spatial-temporal characteristics of DCE-MRI data. This study aims to remove this barrier by fully leveraging the spatial and temporal information to improve parameter estimation.Approach.A spatial-temporal information-driven unsupervised deep learning method (

Spatial patterns of rural opioid-related hospital emergency department visits: A machine learning analysis.

As opioid-related overdose emergency department visits continue to rise in the United States, there is a need to understand the location and magnitude of the crisis, especially in at-risk rural areas. We analyzed sets of ZIP code level electronic health records for emergency department visits from 6 hospitals for two rural regions of Maryland with higher opioid-related overdose rates. Analysis of the demographics of visits found Black or African American emergency department visits in both rural regions were higher than the proportion of their population per region. We trained random forest mo

Dynamics of Awake Hippocampal-Prefrontal Replay for Spatial Learning and Memory-Guided Decision Making.

Spatial learning requires remembering and choosing paths to goals. Hippocampal place cells replay spatial paths during immobility in reverse and forward order, offering a potential mechanism. However, how replay supports both goal-directed learning and memory-guided decision making is unclear. We therefore continuously tracked awake replay in the same hippocampal-prefrontal ensembles throughout learning of a spatial alternation task. We found that, during pauses between behavioral trajectories, reverse and forward hippocampal replay supports an internal cognitive search of available past and f

The chemotherapeutic agent paclitaxel selectively impairs learning while sparing source memory and spatial memory.

Chemotherapeutic agents are widely used to treat patients with systemic cancer. The efficacy of these therapies is undermined by their adverse side-effect profiles such as cognitive deficits that have a negative impact on the quality of life of cancer survivors. Cognitive side effects occur across a variety of domains, including memory, executive function, and processing speed. Such impairments are exacerbated under cognitive challenges and a subgroup of patients experience long-term impairments. Episodic memory in rats can be examined using a source memory task. In the current study, rats rec

Long-lasting effects of neonatal pentobarbital administration on spatial learning and hippocampal synaptic plasticity.

Exposure of newborn rats to antiepileptics such as barbiturates has long-lasting detrimental effects on the hippocampus and hippocampus-dependent behavior. However, the long-term consequences of neonatal administration with barbiturates on the hippocampal synaptic plasticity remain unresolved. In this study, we investigated the long-lasting effects of a neonatal administration of pentobarbital on spatial memory, paired-pulse plasticity in the population spikes, and long-term potentiation (LTP) in the hippocampal CA1 region of rats in vivo. Eight weeks after administration of pentobarbital (10

Overcoming missing data in spatial metabolomics with machine learning imputation to accelerate downstream discovery.

Mass spectrometry imaging (MSI)-based spatial metabolomics exhibits extensive missing values; yet, practical guidance on how imputation choices affect both imputation accuracy and downstream spatial analyses remains limited. In this study, we evaluated eight imputation methods, including both existing approaches and a graph convolutional network (GCN)-based method specifically designed for spatial metabolomics data, to identify suitable approaches for spatial metabolomics. To enable comprehensive assessment, we developed an evaluation framework focusing on two objective criteria: (a) imputatio

Deciphering spatial-temporal mechanisms of PD-1 blockade resistance via biologically informed machine learning.

Immune checkpoint inhibitors (ICIs), especially PD-1/PD-L1 blockade, have transformed cancer therapy; yet objective response rates to anti-PD-(L)1 monotherapy remain ∼20-30% and resistance is common. Meanwhile, multi-omics, spatial profiling, imaging, and clinical datasets are expanding faster than our ability to extract mechanistic insight, creating a "data-rich but mechanism-poor" bottleneck in immuno-oncology. Conventional biomarkers such as tumor mutational burden and PD-L1 expression lack spatiotemporal resolution, while purely data-driven artificial intelligence models often suffer from

AugGCL: Multimodal graph learning for spatial transcriptomics analysis with enhanced gene and morphological data.

Spatial transcriptomics enables the measurement of gene expression in intact tissues. Despite this, reconstructing anatomically accurate spatial domains remains challenging, primarily due to expression sparsity, complex tissue architecture that is characterized by sharp boundaries and long-range continuity, and weak spatial signals. Traditional pipelines typically rely on expression-driven clustering and spatial smoothing, which underperform at boundaries and in sparse regions while neglecting morphological information. To address these challenges, AugGCL is proposed, an augmented graph-convol
