Attraction, Feeding Preference, and Performance of Spodoptera frugiperda Larvae (Lepidoptera: Noctuidae) Reared on Two Varieties of Maize.

The fall armyworm, Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), is an economically important pest of maize and other crops in the Americas. Studies suggest that modern varieties of maize lost some of their natural defense mechanisms against herbivores during domestication and agricultural selection. In the present study, we evaluated the attraction, feeding preference (host fidelity and consumption rate), and performance of S. frugiperda larvae reared on hybrid (Pioneer P4063W) and landrace (Tuxpeño) varieties of maize. We also evaluated the damage caused by S. frugiperda to P

The Arabidopsis immune regulator SRFR1 dampens defences against herbivory by Spodoptera exigua and parasitism by Heterodera schachtii.

Plants have developed diverse mechanisms to fine tune defence responses to different types of enemy. Cross-regulation between signalling pathways may allow the prioritization of one response over another. Previously, we identified SUPPRESSOR OF rps4-RLD1 (SRFR1) as a negative regulator of ENHANCED DISEASE SUSCEPTIBILITY1 (EDS1)-dependent effector-triggered immunity against the bacterial pathogen Pseudomonas syringae pv. tomato strain DC3000 expressing avrRps4. The use of multiple stresses is a powerful tool to further define gene function. Here, we examined whether SRFR1 also impacts resistanc

Synergistic effects of botanical curcumin-induced programmed cell death on the management of Spodoptera litura Fabricius with avermectin.

Chemical pesticides and adjuvants have caused many negative effects. Botanical compounds provide solutions for the development of environment friendly pesticides and the management of increasing pest resistance. Curcumin, a natural polyphenol, showed synergistic effects on avermectin upon the destructive agricultural pest, Spodoptera litura. However, the botanical synergist and its relevant mechanisms remain unclear. In the article, curcumin significantly enhanced the growth inhibition and midgut structural damage of avermectin on the larvae of S. litura, and the synergistic effects were confi

Changes in the haemolymph proteome of Spodoptera littoralis induced by the parasitoid Chelonus inanitus or its polydnavirus and physiological implications.

The egg-larval parasitoid Chelonus inanitus induces in its host Spodoptera littoralis two major developmental effects, namely a precocious onset of metamorphosis followed by a developmental arrest in the prepupal stage. Along with each egg, the wasp injects polydnavirus and venom into the host egg. The polydnavirus has been shown to play a major role in inducing the developmental arrest while the parasitoid larva is instrumental in inducing the precocious onset of metamorphosis. Here we report that experimental dilution of haemolymph of polydnavirus-containing larvae can partially prevent the

Biochemical and molecular responses of Spodoptera frugiperda to insecticide exposure: detoxification enzymes, gene expression, and genotoxic effects.

Spodoptera frugiperda (fall armyworm) is a highly destructive agricultural pest with a remarkable ability to develop resistance to insecticides, posing a severe threat to global food security. Understanding the physiological and molecular responses of this species to insecticide exposure is crucial for improving resistance monitoring and developing effective management strategies. In this study, the toxicity and the associated biochemical, molecular, and genotoxic responses of third-instar S. frugiperda larvae were investigated following exposure to four commonly used insecticides: emamectin b

UDP-glycosyltransferases-mediated vitellogenin protein biogenesis reveals juvenile hormone I-specific dominance in Spodoptera frugiperda reproductive programming.

Juvenile hormone (JH) critically regulates reproduction in Spodoptera frugiperda (FAW) via its signaling cascade. We dissected isoform-specific roles of JH I, II, and III by using hormone injection, RNAi, and RNA-seq. Systematic modulation of JH variants revealed distinct roles in ovarian development, egg production, and vitellogenin (Vg) dynamics. Transcriptomic profiling identified JH I as the most potent inducer of reproductive pathways, specifically enriching the steroid hormone biosynthesis pathway. Within this pathway, two UDP-glycosyltransferases (UGT), SfUGT2 and SfUGT2-like, were iden

Depleting SfCarE: Innovative strategies for enhancing chlorantraniliprole toxicity through mixing with carbaryl in Spodoptera frugiperda (Lepidoptera: Noctuidae).

The sustained application of insecticides has led to the development of resistance. Carboxylesterases (CarEs) play important roles in the resistance and synergism of chlorantraniliprole and carbaryl. In this study, the key Spodoptera frugiperda CarEs gene, SfCarE, was identified. SfCarE was highly expressed at the 4th instar and in the midgut of the 6th instar larvae. Knockdown of SfCarE resulted in significant decrease in CarE activity and an increase in susceptibility to chlorantraniliprole, as well as to the mixture of chlorantraniliprole and carbaryl. Metabolic assays demonstrated that the

Synergistic enhancement of emamectin benzoate and Metarhizium anisopliae against late-instar Spodoptera frugiperda by promoting fungal pathogenicity.

Spodoptera frugiperda, a globally invasive pest, has developed severe resistance to chemical insecticides, necessitating sustainable control strategies. This study investigated the synergistic interaction between emamectin benzoate (EB) and the entomopathogenic fungus Metarhizium anisopliae CQMa421 against fourth-instar larvae. Bioassays revealed a synergistic ratio (SR) of 4.19 when EB was combined with CQMa421 spores (2 × 107 spores/mL), reducing EB usage by 76.1 %. CQMa421 did not cause significant effects on S. frugiperda, whereas emamectin benzoate (EB) significantly disrupted feeding, de

Comparative analysis of testicular fusion in Spodoptera litura (cutworm) and Bombyx mori (silkworm): Histological and transcriptomic insights.

Spodoptera litura commonly known as the cutworm, is among the most destructive lepidopteran pests affecting over 120 plants species. The powerful destructive nature of this lepidopteran is attributable to its high reproductive capacity. The testicular fusion that occurs during metamorphosis from larvae to pupa in S.litura positively influences the reproductive success of the offspring. In contrast, Bombyx mori, the silkworm, retains separate testes throughout its life and does not undergo this fusion process. Microscopic examination reveals that during testicular fusion in S.litura, the perito

Differential pathogenicity of heterologous nucleopolyhedroviruses on Spodoptera frugiperda corresponds to modulated expression of immune-response-related genes.

The fall armyworm Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) is a notorious invasive pest wreaking havoc on various crops globally. Nucleopolyhedroviruses (NPVs) are viral pathogens that specially target lepidopteran pests. However, the homologous virus, Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV), has not been commercialized in China. Therefore, understanding the molecular mechanisms underlying heterologous virus-host interactions can inform the design of virus-based insecticides for controlling S. frugiperda. The pathogenicity of the four heterologous NPVs o

Spinosad induces programmed cell death involves mitochondrial dysfunction and cytochrome C release in Spodoptera frugiperda Sf9 cells.

Spinosad, a reduced-risk insecticide, acts on the nicotinic acetylcholine receptors and the gamma-aminobutyric acid receptor in the nervous system of target insects. However, its mechanism of action in non-neural insect cells is unclear. This study aimed to evaluate mitochondrial functional changes associated with spinosad in Spodoptera frugiperda (Sf9) insect cells. Our results indicate that in Sf9 cells, spinosad induces programmed cell death and mitochondrial dysfunction through enhanced reactive oxygen species production, mitochondrial permeability transition pore (mPTP) opening, and mitoc

The F116V mutation and overexpression of CYP9A186 confer high-level resistance to emamectin benzoate and abamectin in Spodoptera exigua.

The beet armyworm, Spodoptera exigua, is a polyphagous, migratory pest of worldwide importance. It has developed serious resistance to emamectin benzoate and abamectin due to the widespread use of these insecticides in pest control. Previous research demonstrated that the F116V mutation in CYP9A186 confers 265-fold resistance to emamectin benzoate (EB) and 45-fold resistance to abamectin in the WH-EB strain of S. exigua, a near-isogenic strain obtained by introgression of field resistance into the susceptible WH-S line via repeated backcrossing. Here, we examine the contribution of CYP9A186 in

Post-transcriptional regulation of ABCB1 by miR-34-5p is associated with emamectin benzoate susceptibility in Spodoptera frugiperda.

Emamectin benzoate serves as a critical insecticide for controlling Spodoptera frugiperda. However, the emergence of resistance to emamectin benzoate poses a significant challenge to effective pest management, and the underlying genetic mechanisms remain poorly elucidated. This study demonstrates that resistance levels in field populations of S. frugiperda have progressively increased, with ABCB1 expression was positively associated with resistant phenotypes. Although microRNAs (miRNAs) are well-established as crucial regulators of gene expression at the post-transcriptional level, their speci

Toward Sustainable Management of Spodoptera frugiperda (Lepidoptera: Noctuidae): A Review on the Role of Endophytic Fungi in Crop Protection.

The fall armyworm (FAW), Spodoptera frugiperda, is a highly polyphagous pest that has rapidly expanded across Africa, Asia, and Oceania, threatening food security and agricultural productivity. Reliance on synthetic insecticides for FAW management is increasingly challenged by insecticide resistance, environmental contamination, and adverse effects on non-target organisms and human health. This review synthesizes current knowledge on the potential of endophytic fungi as a sustainable strategy for FAW management. Following a systematic literature search, 59 original research articles published

Metabolomics of susceptible and insecticide-resistant strains of Spodoptera frugiperda (Lepidoptera: Noctuidae) and metatranscriptomics of their midgut-associated microbiota.

The gut microbiota of insects can influence host physiology and, like the host, can be affected by stressors. In this study, we investigated the effects of insecticide exposure on the metabolome and gut microbial metatranscriptome of insecticide resistant and susceptible Spodoptera frugiperda larvae. Metabolomic analysis revealed distinct metabolic profiles influenced by host phenotype and treatment with three insecticides: flubendiamide, spinosad and teflubenzuron. Amino acids, monosaccharides and fatty acids were prominent among the differentially abundant metabolites. Insecticide exposure a

Baculovirus entry into the central nervous system of Spodoptera exigua caterpillars is independent of the viral protein tyrosine phosphatase.

Neuroparasitism concerns the hostile take-over of a host's nervous system by a foreign invader, in order to alter the behaviour of the host in favour of the parasite. One of the most remarkable cases of parasite-induced host behavioural manipulation comprises the changes baculoviruses induce in their caterpillar hosts. Baculoviruses may manipulate caterpillar behaviour in two ways: hyperactivity (increased movement in the horizontal plane) and/or tree-top disease (movement to elevated levels in the vertical plane). Those behavioural changes are followed by liquefaction and death of the caterpi

Constitutive expression of Arabidopsis NPR1 confers enhanced resistance to the early instars of Spodoptera litura in transgenic tobacco.

In Arabidopsis, NPR1 (AtNPR1) regulates salicylic acid (SA)-mediated activation of PR genes at the onset of systemic acquired resistance. AtNPR1 also modulates SA-induced suppression of jasmonic acid-responsive gene expression, and npr1 mutants manifest enhanced herbivore resistance. We have raised stable transgenic tobacco lines, expressing AtNPR1 constitutively, which showed elevated expression of PR1 and PR2 genes upon SA treatment. Herbivore bioassays with a generalist polyphagous pest, Spodoptera litura, revealed that the transgenic lines exhibited enhanced resistance compared to the wild
