Transcriptome-guided development of a fibrosis-reversal compound reduces skin scarring and allows regeneration via mitochondrial uncoupling.

Skin scarring impairs function and aesthetics. Current therapies show limited efficacy and cause iatrogenic dermal disruption (e.g., triamcinolone acetonide [TA], a first-line corticosteroid for keloids), with topical medications demonstrating inferior outcomes. Through transcriptome-guided evaluation, we develop FR-1 (fibrosis-reversal compound 1), a small molecule that reverses fibrosis in vitro by inhibiting fibroblast proliferation, suppressing α-smooth muscle actin (α-SMA), and remodeling the extracellular matrix (ECM) via collagen downregulation and matrix metalloproteinase-1 (MMP1) indu

Randomized Controlled Trials for Oral Leukoplakia.

Oral leukoplakia is a white plaque with an increased risk of malignant transformation. Despite available treatment options such as active surveillance, surgical excision, laser ablation, and topical or systemic therapies, there is still a risk of recurrence and malignant transformation. We reviewed all randomized controlled trials on oral leukoplakia published in PubMed from 2014 to 2024 (n = 7) to identify emerging treatment strategies. These studies assessed the efficacy of topical Moringa oleifera, curcumin, beta-carotene with vitamin C, scalpel excision, carbon dioxide and diode laser abla

Engaging longitudinal cohort participants in a research priority setting process for the Raine Study.

BACKGROUND: The Raine Study, one of the world’s longest-running multigenerational cohort studies, has consistently engaged participants in shaping specific research projects within the Raine Study. While participant involvement has been integral to study operations, there has not previously been an opportunity for participants to actively help shape the overall research agenda. The Participant Priority Setting Project (the Project) was developed to address this gap, providing a structured process to establish participant-led research priorities that will guide future study directions. METHODS:

Rehabilitation of Peripheral Neuropathies: From Lexical Analysis of the Literature to Identification of Clinical Protocols.

Peripheral nervous system diseases are a complex and heterogenous group of diseases affecting the different nerves with various severity and impact on quality of life. The current literature does not provide a structured guide for the rehabilitation of these conditions. We performed a lexical literature evaluation based on graph theory to clarify this topic. We performed a search on PubMed and calculated the frequencies of the words indicating rehabilitation approaches, nerves, and diseases. We found the usefulness of exercises and different physical agents, like laser and ultrasound therapy a

What's new in atopic eczema? An analysis of systematic reviews published in 2018. Part 1: prevention and topical therapies.

This review is part of a series of annual updates that summarize the evidence base for atopic eczema (AE). The aim is to provide a succinct guide for clinicians on the key findings from 14 systematic reviews on the prevention and topical treatment of AE published or indexed in 2018. Various supplements, including long-chain polyunsaturated fatty acids, vitamin D and the probiotic Lactobacillus rhamnosus GG, given prenatally and postnatally, have not been shown to prevent AE in infants, although mixed strains of probiotics may decrease the risk of AE if given to the mother during pregnancy and

Community Guide Methods for Systematic Reviews of Economic Evidence.

INTRODUCTION: Community Guide systematic economic reviews provide information on the cost, economic benefit, cost-benefit, and cost-effectiveness of public health interventions recommended by the Community Preventive Services Task Force on the basis of evidence of effectiveness. The number and variety of economic evaluation studies in public health have grown substantially over time, contributing to methodologic challenges that required updates to the methods for Community Guide systematic economic reviews. This paper describes these updated methods. METHODS: The 9-step Community Guide econom

Teaching About Sex and Gender in Neuroscience: More Than Meets the "XY".

Offering courses on the neuroscience of sex and gender can help support an inclusive curriculum in neuroscience. At the same time, developing and teaching such courses can be daunting to even the most enthusiastic educators, given the subject's complexities, nuances, and the difficult conversations that it invites. The authors of this article have all developed and taught such courses from different perspectives. Our aim is to provide educators with an overview of important conceptual topics as well as a comprehensive, but non-exhaustive, guide to resources for teaching about sex/gender in neu

Understanding Delivery and Engagement With a Digital Self-Management Intervention for Chronic Obstructive Pulmonary Disease Across Two Clinical Settings: Qualitative Study.

BACKGROUND: Adherence to chronic obstructive pulmonary disease (COPD) self-management plans can improve health outcomes, yet access to pulmonary rehabilitation remains limited. Digital self-management interventions, such as myCOPD, offer a potential means to extend support across care pathways. However, little is known about how these tools are delivered in routine practice or how delivery influences patient engagement across different clinical settings. OBJECTIVE: This study explored perceived barriers and facilitators to the delivery of and engagement with myCOPD by examining the perspectiv

Radiofrequency Ablation of Pancreatic Solid Tumors.

Today, endoscopic ultrasound-guided radiofrequency ablation has become increasingly accepted for the treatment of different precancerous and neoplastic lesions of the pancreas, particularly in patients who are unfit for surgery. However, thermal ablation has long been suspected to induce pancreatitis or to injure adjacent structures. Published case reports and case series on this topic are of limited size and are often based on a heterogeneous study population, reporting on functional and nonfunctional pancreatic neuroendocrine tumors. Nevertheless, the overall results are promising, with high

Why patients self-refer to the Emergency Department: A qualitative interview study.

RATIONALE, AIMS, AND OBJECTIVES: There have been multiple studies investigating reasons for patients to self-refer to the Emergency Department (ED). The majority made use of questionnaires and excluded patients with urgent conditions. The goal of this qualitative study is to explore what motives patients have to self-refer to an ED, also including patients in urgent triage categories. METHODS: In a large teaching hospital in the Netherlands, a qualitative interview study focusing on reasons for self-referring to the ED was performed. Self-referred patients were included until no new reasons f

Topical application of nitrates onto the papilla of Vater: manometric and clinical results.

BACKGROUND AND STUDY AIMS: Systemic administration of nitrates inhibits motility of the sphincter of Oddi (SO) but is hampered by a decrease of the arterial blood pressure. We evaluate whether topical application of glyceril trinitrate (GTN) onto the papilla can facilitate cannulation of the common bile duct during routine endoscopic retrograde cholangiopancreatography (ERCP) and compare the effect of topical GTN with that of isosorbide dinitrate (ISDN) on SO motility. PATIENTS AND METHODS: A total of 80 patients undergoing routine ERCP for suspected biliary obstruction received topical appli

Insights into the nutritional prevention of macular degeneration based on a comparative topic modeling approach.

Topic modeling and text mining are subsets of natural language processing (NLP) with relevance for conducting meta-analysis (MA) and systematic review (SR). For evidence synthesis, the above NLP methods are conventionally used for topic-specific literature searches or extracting values from reports to automate essential phases of SR and MA. Instead, this work proposes a comparative topic modeling approach to analyze reports of contradictory results on the same general research question. Specifically, the objective is to identify topics exhibiting distinct associations with significant results

Comparative experimental anesthesia efficacy study of epidural injection at the sacrococcygeal space using ultrasound guidance versus blindness technique in Egyptian donkeys (Equus asinus).

The study investigates the effectiveness of blind and ultrasound-guided epidural injections in Egyptian donkeys in the sacrococcygeal region, a topic of limited research, by comparing and assessing their onset in 20 healthy adult donkeys of both sexes. In the first group, we used ten cadaver donkeys that were humanely euthanized. In the second group, we used 10 live, healthy adult donkeys to assess the efficacy and time of analgesia onset for blind and ultrasound-guided epidural injections at the sacrococcygeal region. Cadavers were randomly designated to blind (n = 5) or US-guided (n = 5) epi

Transient Quadriparesis and Cervical Neuropraxia in Elite Athletes.

Elite athletes are often faced with difficult decisions when faced with a cervical spinal disorder. There are many aspects to consider such as the risk of further injury, short- and long-term effects on an athlete's life both during and after his/her career, and the options for treatment. Although there have been some recent contributions to this topic, the evidence-based literature is generally devoid of high-level clinical studies to help guide the decision-making process. This article reviews the pertinent available data/criteria and offer an algorithm for return-to-play considerations.
