Transgenerational effects of in ovo stimulation with synbiotic and choline on gonadal tissue across three generations.

Epigenetic mechanisms play a pivotal role in hereditary processes, shaping phenotypic outcomes across generations. This study investigates the transgenerational impacts of in ovo injection of bioactive substances on gene expression and DNA methylation in the male gonads using the Green-legged Partridgelike chickens as a model organism. Synbiotic PoultryStar® (Biomin; PS) and choline were injected in ovo on the 12th day of egg incubation. In the F1 generation, three groups were established: (1) control (C, 0.9% physiological saline); (2) PS synbiotic (SYN, 2 mg/embryo); and (3) PS synbiotic com

Randomized controlled trial evaluating synbiotic supplementation as an adjuvant therapy in the treatment of Parkinson's disease.

BACKGROUND AND AIM: Neuroinflammatory mechanisms have been closely related to the microbiota-gut-brain axis and could lead to degeneration of dopaminergic neurons with subsequent development and progression of Parkinson's disease (PD). Targeting this pathway for the treatment of PD has sparked a lot of interest. Hence, this study investigates the therapeutic potential of a synbiotic supplement, in conjunction with L-dopa for the management of PD. METHODS: This randomized controlled trial enrolled 66 Parkinson's disease patients, who were randomly assigned to two groups: a control group (n = 3

Dietary Synbiotic Attenuated the Intestinal Inflammation in Weaned Piglets Challenged with Escherichia coli Lipopolysaccharide.

At weaning, piglets deal with numerous changes and stressors, which can lead to reduced feed intake, digestive disturbances, and gut inflammation. In this context, there is a compelling need to develop new and innovative nutritional strategies aimed at restoring intestinal balance in piglets after weaning and controlling the weaning-associated intestinal inflammation. This study investigated the effect of a diet, including a synbiotic additive (a mix of grape seed and camelina meals as the prebiotic and a lactobacilli mixture as the probiotic) on intestinal inflammation in piglets after weanin

Dose-dependent impact of enrofloxacin on broiler chicken gut resistome is mitigated by synbiotic application.

Fluoroquinolone agents are considered critical for human medicine by the World Health Organization (WHO). However, they are often used for the treatment of avian colibacillosis in poultry production, creating considerable concern regarding the potential spread of fluoroquinolone resistance genes from commensals to pathogens. Therefore, there is a need to understand the impact of fluoroquinolone application on the reservoir of ARGs in poultry gut and devise means to circumvent potential resistome expansion. Building upon a recent dose optimization effort, we used shotgun metagenomics to investi

A Designer Synbiotic Attenuates Chronic-Binge Ethanol-Induced Gut-Liver Injury in Mice.

Gut dysbiosis and altered short-chain fatty acids are associated with ethanol-induced liver injury. SCFA are fermentation byproducts of the gut microbiota known to have many beneficial biological effects. We tested if a designer synbiotic could protect against ethanol-induced gut-liver injury. C57BL/6 female mice were exposed to chronic-binge ethanol feeding consisting of ethanol (5% vol/vol) for 10 days, followed by a single gavage (5 g/kg body weight) 6 h before euthanasia. A group of mice also received oral supplementation daily with a designer synbiotic, and another group received fecal sl

Synbiotics, a promising approach for alleviating exacerbated allergic airway immune responses in offspring of a preclinical murine pollution model.

Exposure to pollutants like environmental cigarette smoke (CS) poses a major global health risk, affecting individuals from an early age. Therefore, this study explores how postnatal synbiotic supplementation affects allergic asthma symptoms in house-dust-mite (HDM)- challenged offspring maternally exposed to CS. In HDM-allergic offspring of CS-exposed dams, lung resistance was elevated, but synbiotic supplementation effectively reduced this resistance. Elevated eosinophil BALF counts following HDM challenge were intensified in pups maternally exposed to CS. Similarly, Th2 cell activation and

Genome-scale metabolic modeling-driven design of synbiotics for methane mitigation and quality improvement in fermented total mixed ration.

Methane is a major greenhouse gas with a global warming potential 28 times greater than that of carbon dioxide, making reduction of livestock emissions a critical challenge. Current mitigation strategies, such as feed management, chemical additives, and algae supplementation, face limitations related to safety, supply instability, and economic feasibility. To address these issues, we investigated the development of methane-reducing feeds utilizing synbiotics. In a preliminary study, we evaluated the methane reduction potential of Komagataeibacter intermedius and Zygosaccharomyces parabailii (K

In vitro screening of synbiotics based on a four-strain probiotic blend and their therapeutic potential for ulcerative colitis.

This study explores the therapeutic effects of synbiotics on ulcerative colitis (UC) using an in vitro fermentation model and a Dextran Sulfate Sodium (DSS)-induced UC mouse model. We assessed the impact of synbiotics on probiotic proliferation, short-chain fatty acid (SCFA) production, metabolic regulation, and intestinal barrier function. Fructooligosaccharide (FOS) and Inulin (INU) significantly promoted probiotic growth and increased SCFA production, especially acetate, propionate, butyrate, and isobutyrate (p < 0.01). Metabolomics analysis revealed compound probiotics (CP)+FOS and CP+INU

Dual-responsive microgel beads prepared from pectin-stabilized emulsions for the controlled release of synbiotics to ameliorate ulcerative colitis.

The benefits of synbiotics in ulcerative colitis are now supported by a growing amount of evidence. However, achieving its targeted delivery in vivo and reaching effective doses for therapeutic effects remains to be explored. To further explore the potential role of synbiotics in alleviating ulcerative colitis, this study designed a dual-response "egg-box" shaped microgel microsphere for targeted delivery and precise release of synbiotics. The internal "egg" structure was constructed using an apple pectin-zein hybrid particles (AZHPs) stabilized W1/O/W2 double emulsions system, while a dense n

The Effects of Pro-, Pre-, and Synbiotics on Muscle Wasting, a Systematic Review-Gut Permeability as Potential Treatment Target.

Muscle wasting is a frequently observed, inflammation-driven condition in aging and disease, known as sarcopenia and cachexia. Current treatment strategies target the muscle directly and are often not able to reverse the process. Because a reduced gut function is related to systemic inflammation, this might be an indirect target to ameliorate muscle wasting, by administering pro-, pre-, and synbiotics. Therefore, this review aimed to study the potential of pro-, pre-, and synbiotics to treat muscle wasting and to elucidate which metabolites and mechanisms affect the organ crosstalk in cachexia
