Sodium-Taste Cells Require Skn-1a for Generation and Share Molecular Features with Sweet, Umami, and Bitter Taste Cells.

Taste buds are maintained via continuous turnover of taste bud cells derived from local epithelial stem cells. A transcription factor Skn-1a (also known as Pou2f3) is required for the generation of sweet, umami (savory), and bitter taste cells that commonly express TRPM5 and CALHM ion channels. Here, we demonstrate that sodium-taste cells distributed only in the anterior oral epithelia and involved in evoking salty taste also require Skn-1a for their generation. We discovered taste cells in fungiform papillae and soft palate that show similar but not identical molecular feature with sweet, uma

Histological and immunohistochemical studies of the fungiform and the circumvallate papillae through the life stages from 6- to 72-week-old Sprague-Dawley male rats.

Taste sensitivity decreases with age. Therefore, we investigated the histological and immunohistochemical changes in the receptive fields circumvallate papilla (CvP) and fungiform papilla (FfP) to explore the mechanism underlying age-related changes in taste sensitivity in 6- to 72-week-old rats. We analyzed papilla size, the thickness of the keratin layer of the papilla and stratified squamous epithelium, taste bud size, the keratin layer around the taste pores in the CvP and FfP, and the number and distribution of taste buds in the CvP coronal section. We further assessed the expression of m

Role of saliva in the maintenance of taste sensitivity.

Saliva is the principal fluid component of the external environment of the taste receptor cells and, as such, could play a role in taste sensitivity. Its main role includes transport of taste substances to and protection of the taste receptor. In the initial process of taste perception, saliva acts as a solvent for taste substances; salivary water dissolves taste substances, and the latter diffuse to the taste receptor sites. During this process, some salivary constituents chemically interact with taste substances. For example, salivary buffers (e.g., bicarbonate ions) decrease the concentrati

Recent advancements in the taste transduction mechanism, identification, and characterization of taste components.

In the realm of human nutrition, the phenomenon known as taste refers to a distinctive sensation elicited by the consumption of food and various compounds within the oral cavity and on the tongue. Moreover, taste affects the overall comfort in the oral cavity, and is a fundamental attribute for the assessment of food items. Accordingly, clarifying the material basis of taste would be conducive to deepening the cognition of taste, investigating the mechanism of taste presentation, and accurately covering up unpleasant taste. In this paper, the basic biology and physiology of transduction of bit

Wnt-beta-catenin signaling initiates taste papilla development.

Fungiform taste papillae form a regular array on the dorsal tongue. Taste buds arise from papilla epithelium and, unusually for epithelial derivatives, synapse with neurons, release neurotransmitters and generate receptor and action potentials. Despite the importance of taste as one of our five senses, genetic analyses of taste papilla and bud development are lacking. We demonstrate that Wnt-beta-catenin signaling is activated in developing fungiform placodes and taste bud cells. A dominant stabilizing mutation of epithelial beta-catenin causes massive overproduction of enlarged fungiform papi

Learning-based recovery from perceptual impairment in salt discrimination after permanently altered peripheral gustatory input.

Rats lacking input to the chorda tympani (CT) nerve, a facial nerve branch innervating anterior tongue taste buds, show robust impairments in salt discrimination demonstrating its necessity. We tested the sufficiency of the CT for salt taste discrimination and whether the remaining input provided by the greater superficial petrosal (GSP) nerve, a facial nerve branch innervating palatal taste buds, or by the glossopharyngeal (GL) nerve, innervating posterior tongue taste buds, could support performance after extended postsurgical testing. Rats presurgically trained and tested in a two-response

Epithelial plasticity enhances regeneration of committed taste receptor cells following nerve injury.

Taste receptor cells are taste bud epithelial cells that are dependent upon the innervating nerve for continuous renewal and are maintained by resident tissue stem/progenitor cells. Transection of the innervating nerve causes degeneration of taste buds and taste receptor cells. However, a subset of the taste receptor cells is maintained without nerve contact after glossopharyngeal nerve transection in the circumvallate papilla in adult mice. Here, we revealed that injury caused by glossopharyngeal nerve transection triggers the remaining differentiated K8-positive taste receptor cells to dedif

Gustatory trophic action of arterial chemosensory neurones in the cat.

The proximal stump of the carotid sinus nerve (c.s.n.) branch of the IXth cranial nerve was surgically cross-anastomosed to the distal stump of the lingual nerve (l.n.) branch of the same cranial nerve to test the ability of regenerating c.s.n. axons to reinnervate and induce taste buds on cat circumvallate papillae. The l.n. branch of the IXth nerve, which normally innervates circumvallate papillae, was directed away from the tongue by suturing it to the distal stump of the c.s.n. Animals with normal and re-anastomosed (cut, then resutured) l.n. served as control preparations. 2-19 months fol

Severe acute respiratory syndrome coronavirus 2 pathology and cell tropism in tongue tissues of COVID-19 autopsies.

Since 2019, Coronavirus Disease 2019(COVID-19) has affected millions of people worldwide. Except for acute respiratory distress syndrome, dysgeusis is also a common symptom of COVID-19 that burdens patients for weeks or permanently. However, the mechanisms underlying taste dysfunctions remain unclear. Here, we performed complete autopsies of five patients who died of COVID-19. Integrated tongue samples, including numerous taste buds, salivary glands, vessels, and nerves were collected to map the pathology, distribution, cell tropism, and receptor distribution of severe acute respiratory syndro

Anatomy and development of the human taste system.

The sense of taste relies on well-defined neuroanatomical structures, namely, the taste buds and afferent nerve fibers. Taste buds are clusters of 50-100 neuroepithelial cells located throughout the oral cavity, including the epiglottis and larynx. They are responsible for the initial transduction process that ultimately results in the perception of bitter, sour, salty, sweet, and umami (savory) sensations. They service as the initial sentinel for a sensory system critical in evolution for distinguishing "dangerous" food components, often perceived as bitter or unpleasant, from "useful" ones,

Developing a sense of taste.

Taste buds are found in a distributed array on the tongue surface, and are innervated by cranial nerves that convey taste information to the brain. For nearly a century, taste buds were thought to be induced by nerves late in embryonic development. However, this view has shifted dramatically. A host of studies now indicate that taste bud development is initiated and proceeds via processes that are nerve-independent, occur long before birth, and governed by cellular and molecular mechanisms intrinsic to the developing tongue. Here we review the state of our understanding of the molecular and ce

Taste receptor cells arise from local epithelium, not neurogenic ectoderm.

Except for taste bud cells, all sensory receptor cells and neurons have been shown to originate from neurogenic ectoderm (i.e., neural tube, neural crest, or ectodermal placodes). Descriptive studies on taste buds indicate that they, however, may arise from local epithelium. To determine whether taste receptor cells originate from neurogenic ectoderm or from local epithelium, the tongues of X chromosome-inactivation mosaic mice were examined. Results of this analysis show that taste bud cells and their surrounding epithelium always match in terms of the mosaic marker. This suggests that taste

Optogenetics in taste research: A decade of enlightenment.

The electrophysiological function of the tongue involves complicated activities in taste sense, producing the perceptions of salty, sweet, bitter, and sour. However, therapies and prevention of taste loss arising from dysfunction in electrophysiological activity require further fundamental research. Optogenetics has revolutionized neuroscience and brought the study of sensory system to a higher level in taste. The year 2022 marks a decade of developments of optogenetics in taste since this technology was adopted from neuroscience and applied to the taste research. This review summarizes a deca

Tongue papillae density and fat taster status- a cardinal role on sweet and bitter taste perception among Indian population.

Obesity is a complex nutritional disorder that may be influenced by calorie intake and eating behaviours. Aside from many studies, the influence of papillae count on obesity is still debated. Despite the multiple variables connected to weight gain and altered taste perception, determining the association between papillae count and taste sensitivity to fat, sweet and bitter tastes, in particular, has recently become a focus of attention. This study aimed to rule out the relationship between the number of papillae on different areas of the tongue and taste sensitivity in people (n = 150) among t

Substance-P-containing fibers in the incisive papillae of the rat hard palate. Light- and electron-microscopic immunohistochemical study.

Substance P (SP)-containing fibers in the incisive papillae of rat hard palates, which include various components of sensory receptors, i.e. mechanoreceptors, free nerve endings and chemosensory corpuscles (taste buds), were examined using immunoperoxidase techniques and light and electron microscopes. Immunolabeled fibers were consistently distributed in the medial part of the orifice of the incisive canals, i.e. in the taste-bud-enriched region. Dense immunolabeled fibers were found in subgemmal regions and in the lamina propria papillae. Some fine fibers entered and ascended the taste buds

Defects in the peripheral taste structure and function in the MRL/lpr mouse model of autoimmune disease.

While our understanding of the molecular and cellular aspects of taste reception and signaling continues to improve, the aberrations in these processes that lead to taste dysfunction remain largely unexplored. Abnormalities in taste can develop in a variety of diseases, including infections and autoimmune disorders. In this study, we used a mouse model of autoimmune disease to investigate the underlying mechanisms of taste disorders. MRL/MpJ-Fas(lpr)/J (MRL/lpr) mice develop a systemic autoimmunity with phenotypic similarities to human systemic lupus erythematosus and Sjögren's syndrome. Our r

Developing and regenerating a sense of taste.

Gustation, or the sense of taste, is essential for distinguishing harmful and nutritious substances, and therefore crucial for health and survival. Taste buds (TBs) located in specialized gustatory papillae on the dorsal surface of the tongue are assemblages of specialized epithelial cells called taste receptor cells (TRCs). With the help of saliva, TRCs transduce sweet, sour, salt, bitter and umami stimuli into electrochemical signals that are transmitted to the brain via gustatory sensory neurons of the VIIth and IXth cranial ganglia. TBs in the anterior tongue are derived from embryonic ect

Novel Insights Into Egyptian Tortoise (Testudo kleinmanni) Tongue Morphology Using Immunofluorescence Analysis.

The Egyptian tortoise (Testudo kleinmanni) exhibits remarkable adaptations; yet, significant gaps remain in the understanding of taste bud distribution and the molecular characteristics of its tongue cells. The unique adaptations were investigated using immunofluorescence with six specific antibodies, light microscopy, and scanning electron microscopy. Vimentin and nestin identified superficial taste buds in the lingual body, notably larger than those in the lingual root. The lingual root's fungiform papillae contained deep taste buds. Vimentin showed broad expression across all taste bud cell

Variability in P2X receptor composition in human taste nerves: implications for treatment of chronic cough.

BACKGROUND: Antagonists to the P2X purinergic receptors on airway sensory nerves relieve refractory or unexplained chronic cough (RCC/UCC) but can evoke unwanted dysgeusias because the gustatory nerves innervating taste buds express this same family of receptors. However, the subunit composition of the P2X receptors in these systems may differ, with implications for pharmacological intervention of RCC/UCC. In most species, the extrapulmonary airway nerves involved in cough predominantly express P2X3 subunits that form homotrimeric P2X3 receptors. In contrast, most sensory nerves innervating ta
