The bloodstream represents a hostile environment that bacteria must overcome to cause bacteraemia. To understand how the major human pathogen Staphylococcus aureus manages this we have utilised a functional genomics approach to identify a number of new loci that affect the ability of the bacteria to survive exposure to serum, the critical first step in the development of bacteraemia. The expression of one of these genes, tcaA, was found to be induced upon exposure to serum, and we show that it is involved in the elaboration of a critical virulence factor, the wall teichoic acids (WTA), within

Out of the 2,527 strains typed in 1976, 1977, 1978 and 1979, 226 strains (8.94%) were lysed by phages 94, 96 or 94 + 96. The two phages lyse together 83.17% of strains, whilst phage 94 alone lyses only 3.96% and phage 96 alone 12.87%. 94/96 strains have the S. aureus species antigens, particularly that of biotype A (human) at levels similar to those previously described: Protein A 96%, beta Ribitol teichoic acid (93%). Very characteristic is the presence in these strains of type antigens c1 (89%) and sigma (92%), which are significantly unusual or absent in the strains of other phages types. 9

A cell wall-associated polysaccharide has been isolated from a noncapsulated mutant, 89R50, of the alpha-hemolytic filamentous streptococcus of provisional capsular type 89. Chemical analysis of the polysaccharide indicates that it consists of galactosamine, glucosamine, glucose, and phosphorus. Immunological cross-reactivity was observed between the streptococcal cell wall antigen and antibody to the cell wall-like capsular polysaccharide of pneumococcus. This cross-reactivity appears attributable to similarities in the teichoic acid moieties rather than in the mucopeptide moieties of the two

The unique cell wall of mycobacteria is essential to their viability and the target of many clinically used anti-tuberculosis drugs and inhibitors under development. Despite intensive efforts to identify the ligase(s) responsible for the covalent attachment of the two major heteropolysaccharides of the mycobacterial cell wall, arabinogalactan (AG) and peptidoglycan (PG), the enzyme or enzymes responsible have remained elusive. We here report on the identification of the two enzymes of Mycobacterium tuberculosis, CpsA1 (Rv3267) and CpsA2 (Rv3484), responsible for this function. CpsA1 and CpsA2

Cell surface carbohydrates have been proven optimal targets for vaccine development. Conjugation of polysaccharides to a carrier protein triggers a T-cell-dependent immune response to the glycan moiety. Licensed glycoconjugate vaccines are produced by chemical conjugation of capsular polysaccharides to prevent meningitis caused by meningococcus, pneumococcus and Haemophilus influenzae type b. However, other classes of carbohydrates (O-antigens, exopolysaccharides, wall/teichoic acids) represent attractive targets for developing vaccines. Recent analysis from WHO/CHO underpins alarming concern

Staphylococcus aureus is a leading cause of hospital-associated and, more recently, community-associated infections caused by highly virulent methicillin-resistant strains (CA-MRSA). S. aureus survival in the human host is largely defined by the ability to evade attacks by antimicrobial peptides (AMPs) and other mechanisms of innate host defence. Here we show that AMPs induce resistance mechanisms in CA-MRSA via the aps AMP sensor/regulator system, including (i) the d-alanylation of teichoic acids, (ii) the incorporation of lysyl-phosphatidylglycerol in the bacterial membrane and a concomitant

Staphylococcus epidermidis is a major cause of late onset sepsis in extremely preterm neonates. Antibody therapies are considered as an interesting strategy to prevent sepsis. However, previous clinical trials with intravenous immunoglobulin (IVIG) and a monoclonal antibody (mAb) targeting staphylococcal lipoteichoic acid (LTA) (Pagibaximab) failed to show significant protection from invasive infections in extremely preterm neonates. Here we use an age-specific in vitro platform to compare immune protection by Pagibaximab with two other mAbs recognizing invasive S. epidermidis in the context o

The gut microbiota represents a promising target to enhance cancer immunotherapy, yet the microbial molecules mediating anti-tumor immunity remain poorly defined. Here, we identify a human-derived strain, Lactobacillus fermentum MI37, that augments anti-tumor immune responses and synergizes with anti-PD-1 therapy. MI37 induces robust IFN-γ production with minimal IL-10, increases tumor-infiltrating cytotoxic T lymphocytes, and upregulates inflammatory gene programs in a syngeneic mouse model. Notably, heat-killed MI37 retains its immunostimulatory and anti-tumor effects, indicating a structura

The use of gel immunodiffusion for the determination of antibodies to nontype-specific cell-wall protein antigens of group A streptococcus allowed to eliminate reactions caused by antibodies to type-specific antigens, group polysaccharide and teichoic acid. Antibodies to nontype-specific antigens, and particularly to two such antigens, occurred more frequently in the blood sera of patients with rheumatism in the active phase than in patients with rheumatism in the inactive phase, erysipelas and with non-rheumatic cardiac diseases, and in donors. Antibodies to nontype-specific antigens occurred

Pharyngitis is an inflammation of the pharynx caused by viral, bacterial, or non-infectious factors. In the present study, the anti-inflammatory efficacy of carvacrol was assessed using an in vitro model of streptococcal pharyngitis using human tonsil epithelial cells (HTonEpiCs) induced with Streptococcus pyogenes cell wall antigens. HTonEpiCs were stimulated by a mixture of lipoteichoic acid (LTA) and peptidoglycan (PGN) for 4 h followed by exposure to carvacrol for 20 h. Following exposure, interleukin (IL)-6, IL-8, human beta defensin-2 (HBD-2), epithelial-derived neutrophil-activating pro

Lipoteichoic acid from Staphylococcus aureus was a potent inducer of procoagulant activity in isolated mononuclear cells but not in whole blood. In contrast, staphylococcal peptidoglycan showed equal levels of potency in isolated mononuclear cells and whole blood, suggesting that peptidoglycan is an important inducer of procoagulant activity in severe sepsis involving gram-positive bacteria.

BACKGROUND: Periodontal disease and diabetes mellitus (DM) are highly prevalent and interrelated diseases, resulting in altered host response microbiota. Thus, this study aimed to evaluate the impact of DM on local levels of lipopolysaccharide (LPS) and lipoteichoic acid (LTA) and their relationship with cytokines and matrix metalloproteinases' (MMPs) profile.
METHODS: This case-control study included diabetic (n = 15) and non-diabetic (n = 15) subjects presenting Stage 3-4, Grade C, Periodontitis. Gingival crevicular fluid (GCF) was collected, and LPS and LTA levels were analyzed by enzyme-l

Clostridiodes (Clostridium) difficile is an anaerobic Gram-positive, spore-forming nosocomial, gastrointestinal pathogen causing C. difficile-associated disease with symptoms ranging from mild cases of antibiotic-associated diarrhea to fatal pseudomembranous colitis. We developed murine monoclonal antibodies (mAbs) specific for a conserved cell surface antigen, lipoteichoic acid (LTA)of C. difficile. The mAbs were characterized in terms of their thermal stability, solubility, and their binding to LTA by surface plasmon resonance and competitive ELISA. Synthetic LTA molecules were prepared in o

Central metabolic pathways control virulence and antibiotic resistance, and constitute potential targets for antibacterial drugs. In Staphylococcus aureus the role of the pentose phosphate pathway (PPP) remains largely unexplored. Mutation of the 6-phosphogluconolactonase gene pgl, which encodes the only non-essential enzyme in the oxidative phase of the PPP, significantly increased MRSA resistance to β-lactam antibiotics, particularly in chemically defined media with physiologically-relevant concentrations of glucose, and reduced oxacillin (OX)-induced lysis. Expression of the methicillin-res

Listeria monocytogenes causes listeriosis, an infectious and potentially fatal disease of animals and humans. A diverse network of transcriptional regulators, including LysR-type catabolite control protein C (CcpC), is critical for the survival of L. monocytogenes and its ability to transition into the host environment. In this study, we explored the physiological and genetic consequences of deleting ccpC and the effects of such deletion on the ability of L. monocytogenes to cause disease. We found that ccpC deletion did not impact hemolytic activity, whereas it resulted in significant reducti

Staphylococcus aureus is a major opportunistic pathogen whose ability to form biofilms significantly enhances its persistence and antibiotic resistance. We previously found that human neutrophil peptide 1 (HNP1), a member of the α-defensin family, promotes biofilm formation in Acinetobacter baumannii. Here, we demonstrate that HNP1 enhances biofilm formation across a range of Gram-positive and Gram-negative bacteria, with particularly strong effects in S. aureus, which increases its tolerance to multiple antibiotics. Mechanistically, HNP1 promotes bacterial adhesion and self-aggregation, facil

A study was undertaken to test the hypothesis that Staphylococcus aureus colonization of the lid margins could determine the expression of allergic eye disease in atopic patients. The authors compared lid isolates of S. aureus from 23 adults who had both atopic dermatitis and chronic conjunctivitis and isolates from 14 patients who had atopic dermatitis but who lacked ocular disease. No significant difference was found in either the staphylococcal protein A or hemolytic toxin production by isolates from the two disease groups, and there was no difference between groups in the quantity of serum

Humoral reactions to native culture filtrates of Streptococcus bovis were studied in patients with endocarditis by means of two-dimensional crossed immunoelectrophoresis. Serum from patients with S bovis endocarditis produced eight different precipitin arcs. Lex antigen cross-reacted with sera from patients with viridans streptococcal endocarditis, and TA antigen cross-reacted with ribitol teichoic acid-positive sera from patients with Staphylococcus aureus bacteremia. One antigen, common (c), was immunoreactive with all 10 sera from S bovis-infected patients. Antibody to this antigen was not

The use of β-lactam antibiotics to treat infections caused by Staphylococcus aureus has been severely compromised by the acquisition by horizontal gene transfer of a gene that encodes the β-lactam-insensitive penicillin-binding protein PBP2a. This allows methicillin-resistant S. aureus (MRSA) to proliferate in the presence of β-lactam antibiotics. Paradoxically the dependence on PBP2a for the essential transpeptidase activity in cell wall peptidoglycan biosynthesis is the 'Achilles heel' of MRSA. Compounds that disrupt the divisome, wall teichoic acid, and functional membrane microdomains act
