The woodchuck as an animal model for pathogenesis and therapy of chronic hepatitis B virus infection.

This review describes the woodchuck and the woodchuck hepatitis virus (WHV) as an animal model for pathogenesis and therapy of chronic hepatitis B virus (HBV) infection and disease in humans. The establishment of woodchuck breeding colonies, and use of laboratory-reared woodchucks infected with defined WHV inocula, have enhanced our understanding of the virology and immunology of HBV infection and disease pathogenesis, including major sequelae like chronic hepatitis and hepatocellular carcinoma. The role of persistent WHV infection and of viral load on the natural history of infection and dise

Animal models of Alport syndrome: advancing the prospects for effective human gene therapy.

Several animal models for Alport syndrome have been described. These are available for studies on the pathogenetic mechanisms of the disease, as well as for the development of new technologies for gene therapy in this progressive hereditary kidney disease. This review summarizes current knowledge on the molecular basis of Alport syndrome, and on the animal models which all remarkably well resemble the human disease. Recent work aimed at the development of gene therapy, including hurdles and progress are discussed.

Anti-inflammatory therapy for acute lung injury. A review of animal and clinical studies.

The adult respiratory distress syndrome (ARDS) continues to demonstrate high mortality. This syndrome is frequently observed as a remote complication of another disease process and is characterized by a significant inflammatory component. The purpose of this review is to compare and contrast published research on the use of anti-inflammatory agents, steroidal and nonsteroidal, in animal models of acute lung injury. Emphasis is given to the nature of the experimental pulmonary injury, infusion (ie, oleic acid and zymosan-activated plasma) or bacteriologically (ie, endotoxin and live bacteria) i

[Lung lavage and artificial surfactant replacement therapy--animal experimentation and clinical application].

Lung lavage may cause a surfactant deficient condition. We examined the effect of artificial surfactant replacement on the post-lavage condition in rabbits and in a patient with alveolar pulmonary proteinosis. Adult rabbits (n = 21) were subjected to whole-lung lavage, and the PaO2 was made to decrease to less than 70 mmHg (FIO2 = 1.0). In the replaced group (n = 11), an artificial surfactant which was a saline suspension of lipids (mixture of dipalmitoylphosphatidylcholine, phosphatidylglycerol and tripalmitin) was administered into the airway. The PaO2 was elevated to a level higher than 400

[Calcium blockers in the therapy of vertebrobasilar insufficiency. Results of animal experiments and clinical studies].

Vertebrobasilar insufficiency (VBI) is a clinical syndrome which includes different transient brain stem symptoms. In addition to anamnesis and clinical study electronystagmography (ENG) is regarded as the most important method for testing the labyrinthine and brain stem system. Typical findings, not only in patients with VBI, but also in animal experiments (occlusion of one vertebral artery) are an increase in nystagmus frequency and decrease in nystagmus amplitude. The present study concerns two calcium antagonists: flunarizine and nimodipine. During tests on patients the caloric and the rot

Neural Stem/Progenitor Cell Therapy in Patients and Animals with Amyotrophic Lateral Sclerosis: A Systematic Review and Meta-analysis.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative malady that causes progressive degeneration and loss of motor neuron function in the brain and spinal cord, eventually resulting in muscular atrophy, paralysis, and death. Neural stem/progenitor cell (NSPC) transplantation can improve bodily function in animals and delay disease progression in patients with ALS. This paper summarizes and analyzes the efficacy and safety of neural stem/progenitor cell (NSPC) transplantation as a treatment for ALS, aiming to improve function and delay disease progression in patients. We present a summary

Severe burn injury, burn shock, and smoke inhalation injury in small animals. Part 2: diagnosis, therapy, complications, and prognosis.

OBJECTIVE: To review the evaluation and treatment of patients suffering from severe burn injury (SBI), burn shock, and smoke inhalation injury. Potential complications and prognosis associated with SBI are also discussed. DIAGNOSIS: Diagnosis of burn injury and burn shock is based on patient history and clinical presentation. Superficial burn wounds may not be readily apparent for the first 48 h whereas more severe wounds will be evident at presentation. Patients are diagnosed with local or SBI by estimating total body surface area involved using the 'Rule of Nines' or the Lund-Browder chart

Assessing the translational value of animal models in developing pain-targeting therapies for clinical osteoarthritis.

Osteoarthritis (OA) is a highly prevalent and disabling disease, with pain as its most debilitating symptom. Despite extensive preclinical research, no therapy has successfully translated from animal models into routine clinical use. Commonly used animal models (e.g., acute, surgically, or chemically induced OA in young, single-sex animals) often fail to reflect the chronic and multifactorial human condition, which is often seen in older, postmenopausal women with comorbidities and mixed nociceptive-neuropathic pain. This review aims to identify animal models and pain assessment methods for pr

Gene editing therapy as a therapeutic approach for cardiovascular diseases in animal models: A scoping review.

BACKGROUND: Cardiovascular diseases (CVDs) are the leading cause of mortality worldwide, with hereditary genetic factors contributing substantially to disease burden. Current treatments, including lifestyle modifications, pharmacotherapy, and surgical interventions, focus primarily on symptom management but fail to address underlying genetic causes, often resulting in disease progression or recurrence. Gene therapy has emerged as a transformative approach, offering a potential treatment. This review explores its efficacy and safety in animal models, identifying opportunities for future advance

Does extracorporeal shockwave therapy treat leg length discrepancy? an experimental animal study.

BACKGROUND: Extracorporeal shockwave therapy (ESWT) is widely used to treat musculoskeletal diseases, but its impact on adolescents with unhealed epiphyseal plates is concerning. It remains unclear whether ESWT applied to growth plates promotes or inhibits bone growth. Low energy ESWT does not cause damage of articular cartilage and promotes the growth of articular cartilage. Therefore, the application of ESWT to treat the leg length discrepancy is a possibleoption. METHODS: Here, the 96 adolescent rats were used to demonstrate that different levels of ESWT developed different effects on the

Photobiomodulation therapy on skeletal muscles exposed to diabetes mellitus: a systematic review of animal studies.

Diabetes-related muscle damage has been overlooked despite its known association with increased morbidity and mortality in DM individuals. PBMT is a recognized alternative to improve skeletal muscle health in other populations, but its effectiveness in DM is still unclear. To address this issue, we reviewed preclinical studies, available in any language and period, in ten sources of information. The methods were previously registered at PROSPERO (CRD42021271041), based on PRISMA recommendations. Studies in murine models of T1DM or T2DM that reported quantitative analyses of skeletal muscles tr
