Mitochondrial dysfunction and chronic disease: treatment with natural supplements.

Loss of function in mitochondria, the key organelle responsible for cellular energy production, can result in the excess fatigue and other symptoms that are common complaints in almost every chronic disease. At the molecular level, a reduction in mitochondrial function occurs as a result of the following changes: (1) a loss of maintenance of the electrical and chemical transmembrane potential of the inner mitochondrial membrane, (2) alterations in the function of the electron transport chain, or (3) a reduction in the transport of critical metabolites into mitochondria. In turn, these changes

Inhibition of lindane-induced toxicity using alpha-lipoic acid and vitamin E in the brain of Mus musculus.

In the present investigation, we have used adenosine triphosphatase (ATPase) activity as biochemical test of toxic action of lindane that was explained by lipid peroxidation model. Study was also undertaken to ascertain the potential protective role of alpha-lipoic acid (ALA) and vitamin E on the same parameters. Highly acute dose of lindane, i.e., 40 mg/kg bw for 18 h exposure, was used for creating lesions in brain. Lipid peroxidation was measured in terms of glutathione peroxidase and thio barbituric acid-reacting substances (TBARS). Various brain regions under investigation were cerebellum

Engineered Biomimetic Nanomicelles Target Inflammation in Sepsis-Associated Acute Lung Injury by Scavenging ROS and Reprogramming Macrophages.

BACKGROUND: Sepsis-associated acute lung injury (SALI) has high mortality, largely driven by a damaging cycle of oxidative stress and inflammation, with a lack of effective treatments. To address this, a novel biomimetic nanodrug was developed. It uses an amphiphilic copolymer (PT) to encapsulate the antioxidant/anti-inflammatory agent carnosic acid (CA), forming PT@CA micelles. These micelles are then coated with M2 macrophage membranes (MM) to create MM@PT@CA. Compared to traditional liposomes, the macrophage membrane has better inflammatory targeting and biological safety. METHODS: The PT

A Comprehensive Review of Safety, Efficacy, and Indications for the Use of Alpha-Lipoic Acid and Acetyl-L-Carnitine in Neuropathic Pain.

The debilitating, chronic symptoms of neuropathic pain result in decreased quality of life, depressed mood, and anxiety in patients suffering from neuropathic pain. Despite hundreds of dollars in monthly treatment-related costs, more than half of the patients report inadequate pain relief. Traditional first-line agents are expensive and may have disruptive side effects. Given the disease burden of neuropathic pain, many patients turn to over-the-counter supplements. Here we review two supplements, alpha-lipoic acid (ALA), also known as thioctic acid, and acetyl-L-carnitine (ALC), and data of t

Unlocking New Therapeutic Options for Vincristine-Induced Neuropathic Pain: The Impact of Preclinical Research.

Vincristine, a vinca alkaloid, is used in chemotherapy protocols for cancers such as acute leukemia, Hodgkin's disease, neuroblastoma, cervical carcinoma, lymphomas, breast cancer, and melanoma. Among the common adverse effects of vincristine is peripheral neuropathy, with most patients receiving a cumulative dose over 4 mg/m2 who develop varying degrees of sensory neuropathy. The onset of vincristine-induced peripheral neuropathy can greatly affect patients' quality of life, often requiring dose adjustments or the discontinuation of treatment. Moreover, managing vincristine-induced peripheral

Randomized, double-blind, controlled trial of a combination of alpha-lipoic acid and pregabalin for neuropathic pain: the PAIN-CARE trial.

We compared a combination of the nonsedating antioxidant, alpha-lipoic acid (ALA), with the sedating anticonvulsant, pregabalin, vs each monotherapy to treat neuropathic pain due to peripheral neuropathies. In this randomized, double-blind, 3-period crossover trial, participants received oral ALA, pregabalin, and their combination-each for 6 weeks. The primary outcome was mean daily pain intensity at maximal tolerated doses (MTD); secondary outcomes included quality of life (SF-36), sleep (Medical Outcomes Study-Sleep Scale), adverse effects, drug doses, and other measures. Of 55 participants

Alpha-lipoic acid reduces oxidative damage and ameliorates follicular abnormalities in vitrified cat ovarian tissue.

INTRODUCTION: Vitrification may induce oxidative damage, mitigating cryodamage is crucial for preserving follicular development and ovarian function. Alpha lipoic acid (α-ALA) is an antioxidant which exerts its antioxidant activity directly or indirectly. METHODS: In this study, we established a α-ALA concentration gradients (0, 100, and 150 μM) in the vitrification solution, assessed follicular abnormalities through hematoxylin and eosin (HE), analyzed cell apoptosis via TUNEL staining analysis, immunohistochemistry (IHC) and Masson staining, explored differential genes by transcriptome sequ

Dihydrolipoic acid improves diabetic endothelial cell function via the H2S-producing enzyme 3-mercaptopyruvate sulfurtransferase.

High glucose-induced endothelial dysfunction is a hallmark of diabetic microvascular complications. Reduced hydrogen sulphide (H2S) levels and dysregulation of the H2S-producing enzyme 3-mercaptopyruvate sulfurtransferase (3-MST) are linked to this dysfunction. Dihydrolipoic acid (DHLA), formed from alpha-lipoic acid in mitochondria, has been proposed to enhance H2S release from 3-MST; however, this has not been empirically tested in diabetes. Using a cell culture model, this study demonstrates that DHLA supplementation enhances H2S production via 3-MST and improves endothelial function under

Double-blind, randomized, placebo-controlled crossover trial of alpha-lipoic acid for the treatment of fibromyalgia pain: the IMPALA trial.

Fibromyalgia is a common and challenging chronic pain disorder with few, if any, highly effective and well-tolerated treatments. Alpha-lipoic acid (ALA) is a nonsedating antioxidant with evidence of efficacy in the treatment of symptomatic diabetic neuropathy that has not been evaluated in the setting of fibromyalgia treatment. Thus, we conducted a single-centre, proof-of-concept, randomized, placebo-controlled, crossover trial of ALA for the treatment of fibromyalgia. Twenty-seven participants were recruited, and 24 participants completed both treatment periods of the trial. The median maxima

Robust photogeneration of H2 in water using semiconductor nanocrystals and a nickel catalyst.

Homogeneous systems for light-driven reduction of protons to H(2) typically suffer from short lifetimes because of decomposition of the light-absorbing molecule. We report a robust and highly active system for solar hydrogen generation in water that uses CdSe nanocrystals capped with dihydrolipoic acid (DHLA) as the light absorber and a soluble Ni(2+)-DHLA catalyst for proton reduction with ascorbic acid as an electron donor at pH = 4.5, which gives >600,000 turnovers. Under appropriate conditions, the precious-metal-free system has undiminished activity for at least 360 hours under illuminati

[Pyruvate oxidation and S35-lipoic acid fixation by breast tumor homogenates].

The rate of pyruvic acid oxidation by human mammary gland tumor homogenates is several times lower compared with that by homogenates of the adjacent non-involved tissues. This dependence is especially manifest with malignant tumors, although in this case both in the involved and non-involved tissues the pyruvate oxidation is slower than in benign tumors. In the in vitro experiments the rate of S35 lipoic acid binding by the precipitated in 10000 g fractions of the homogenates in question was the highest for malignant tumors and 3.36 times lower for the adjacent tissues. In benign tumors these

Natural Bone Regeneration-Inspired Poly(Lipoic Acid)-Based Organic-Inorganic Bone Adhesive for Osteoporotic Bone Defect Repair.

Osteoporosis, featured by imbalanced bone remodeling, faces clinical challenges including microenvironment dysregulation, insufficient mineralization, and poor material fixation. Inspired by natural bone regeneration-where ossification centers and surrounding soft tissues orchestrate mineralization and tissue integration-we herein develop a biomimetic adhesive in which strontium titanate (SrTiO3) serves as the mineralization core to mimic ossification centers, boosting biomineralization and osteoblast differentiation. Meanwhile, poly(lipoic acid) (PolyLA), synthesized via one-step polymerizati

Renal-Protective Roles of Lipoic Acid in Kidney Disease.

The kidney is a crucial organ that eliminates metabolic waste and reabsorbs nutritious elements. It also participates in the regulation of blood pressure, maintenance of electrolyte balance and blood pH homeostasis, as well as erythropoiesis and vitamin D maturation. Due to such a heavy workload, the kidney is an energy-demanding organ and is constantly exposed to endogenous and exogenous insults, leading to the development of either acute kidney injury (AKI) or chronic kidney disease (CKD). Nevertheless, there are no therapeutic managements to treat AKI or CKD effectively. Therefore, novel th
