Leaf plasticity to light intensity in Italian cypress (Cupressus sempervirens L.): adaptability of a Mediterranean conifer cultivated in the Alps.

Italian cypress (Cupressus sempervirens L.) is native to the eastern Mediterranean, an area characterised by hot, dry summers and mild winters. Over the centuries, however, the species has been introduced into more northerly regions, a long way from its native range. The current, generally warmer climatic conditions brought about by global warming have favoured its cultivation in even more northerly areas in the Alps and other European alpine regions. Given that not only temperature, but also light availability are limiting factors for the spread of cypress in these environments, it is importa

Unique properties vs. common themes: the atypical cyanobacterium Gloeobacter violaceus PCC 7421 is capable of state transitions and blue-light-induced fluorescence quenching.

The atypical unicellular cyanobacterium Gloeobacter violaceus PCC 7421, which diverged very early during the evolution of cyanobacteria, can be regarded as a key organism for understanding many structural, functional, regulatory and evolutionary aspects of oxygenic photosynthesis. In the present work, the performance of two basic photosynthetic adaptation/protection mechanisms, common to all other oxygenic photoautrophs, had been challenged in this ancient cyanobacterium which lacks thylakoid membranes: state transitions and non-photochemical fluorescence quenching. Both low temperature fluore

Sulfoquinovosyldiacylglycerol (SQDG) in chloroplast-poor tissues: What can lipidomics tell us?

Sulfoquinovosyldiacylglycerol (SQDG) is an anionic sulfolipid of plastid membranes, classically associated with chloroplasts and thylakoids. Its detection by lipidomics in chloroplast-poor tissues raises a central problem: how can a reported SQDG signal be validated, and what evidence is required to determine its plastid context and biological meaning? In this critical narrative review, we reassess evidence beyond chloroplast-dominated tissues and evaluate both the strength of evidence for SQDG occurrence or function and the steps required to test lipidomics-derived hypotheses. The literature

Higher remediation efficiency of Cd and lower CO2 emissions in phytoremediation systems with biochar application.

Biochar has been considered a promising material for soil carbon sequestration. However, there are huge knowledge gaps regarding the carbon reduction effects of biochar-plant-polluted soil. Here, rice straw biochar (RB) was applied in ryegrass-cadmium (Cd)-contaminated soil to investigate the full-cycle carbon dioxide (CO2) emission and intrinsic mechanism. RB resulted in a 37.00 %-115.64 % reduction in accumulative CO2 emissions and a 31.61 %-45.80 % reduction in soil bioavailable Cd throughout the whole phytoremediation period. CO2 emission reduction triggered by RB can be attributed to the

Small heat-shock proteins regulate membrane lipid polymorphism.

Thermal stress in living cells produces multiple changes that ultimately affect membrane structure and function. We report that two members of the family of small heat-shock proteins (sHsp) (alpha-crystallin and Synechocystis HSP17) have stabilizing effects on model membranes formed of synthetic and cyanobacterial lipids. In anionic membranes of dimyristoylphosphatidylglycerol and dimyristoylphosphatidylserine, both HSP17 and alpha-crystallin strongly stabilize the liquid-crystalline state. Evidence from infrared spectroscopy indicates that lipid/sHsp interactions are mediated by the polar hea

One-Pot Biphasic Photocatalytic CO2-to-Carbonylation Cascades.

Photosynthesis compartmentalizes reactions across the thylakoid membrane in vivo to upcycle CO2 into complex chemicals that are fundamental to life. Herein, we demonstrate a step toward achieving a synthetic equivalent to the chloroplast with a compartmentalized cascade reaction in vitro that enables light driven CO2 utilization for organic synthesis in one-pot. This bioinspired compartmentalization enables the photocatalytic reduction of aqueous CO2 by water-soluble catalysts and dyes to produce CO, which then moves into the more buoyant organic layer and is consumed in aminocarbonylation or

A Light-Driven In Vitro Enzymatic Biosystem for the Synthesis of α-Farnesene from Methanol.

Terpenoids of substantial industrial interest are mainly obtained through direct extraction from plant sources. Recently, microbial cell factories or in vitro enzymatic biosystems have emerged as promising alternatives for terpenoid production. Here, we report a route for the synthesis of α-farnesene based on an in vitro enzyme cascade reaction using methanol as an inexpensive and renewable C1 substrate. Thirteen biocatalytic reactions divided into 2 modules were optimized and coupled to achieve methanol-to-α-farnesene conversion via integration with natural thylakoid membranes as a green ener

Effect of moderate and high light on photosystem II function in Arabidopsis thaliana depleted in digalactosyl-diacylglycerol.

The response of the heat-sensitive dgd1-2 and dgd1-3 Arabidopsis mutants depleted in the galactolipid DGDG to photoinhibition of chloroplasts photosystem II was studied to verify if there is a relationship between heat stress vulnerability due to depletion in DGDG and the susceptibility to photoinhibitory damage. Non-photochemical quenching (NPQ) is known to dissipate excessive absorbed light energy as heat to protect plants against photodamage. The main component of NPQ is dependent of the transthylakoid pH gradient and is modulated by zeaxanthin (Zx) synthesis. These processes together with

Acclimation of tobacco leaves to high light intensity drives the plastoquinone oxidation system--relationship among the fraction of open PSII centers, non-photochemical quenching of Chl fluorescence and the maximum quantum yield of PSII in the dark.

Responses of the reduction-oxidation level of plastoquinone (PQ) in the photosynthetic electron transport (PET) system of chloroplasts to growth light intensity were evaluated in tobacco plants. Plants grown in low light (150 micromol photons m-2 s-1) (LL plants) were exposed to a high light intensity (1,100 micromol photons m-2 s-1) for 1 d. Subsequently, the plants exposed to high light (LH plants) were returned back again to the low light condition: these plants were designated as LHL plants. Both LH and LHL plants showed higher values of non-photochemical quenching of Chl fluorescence (NPQ

Dissection of photodamage at low temperature and repair in darkness suggests the existence of an intermediate form of photodamaged photosystem II.

Irradiation of the photosynthetic machinery with strong light induces damage to the photosystem II complex (PSII), and this phenomenon is referred to as photodamage. In an attempt to characterize the mechanism of photodamage to PSII, we examined the events associated with photodamage by monitoring the phenomenon in Synechocystis sp. PCC 6803 at a low temperature. After the activity of PSII had been reduced to 10% of the original activity by exposure of Synechocystis cells to strong light at 10 degrees C, recovery was allowed to proceed at 34 degrees C in darkness. Under these conditions, appro

[Attachment of horse cecum Ciliata to plant fragments. Degradation of chloroplasts. Attachment of bacteria to cecal Ciliata].

Cecum microfauna association with different plant tissues was examined by scanning and transmission electron microscopy. The ciliates were attached to the damaged areas of the leaves and their highest concentrations were found on the epidermis and mesophyll tissues. The degradation of plant tissue was due to protozoal ingestion of the plant fragments. The morphology of ingested envacuolized chloroplasts changed rapidly, showing different stages of digestion inside the ciliate endoplasm. Intact chloroplasts were rarely observed but the grana of fragmented thylakoid membranes was often seen. The

Differential chlorophyll and carotenoid degradation underpin poor curing colour of low-quality flue-cured tobacco leaves.

This study investigates the physiological basis for the inferior curing quality of low-quality flue-cured tobacco leaves. Using 'Yunyan 300' tobacco, we compared normal (CK), multi-fertilizer (LH), and reviving (FQ) leaves. Post-curing, LH and FQ leaves exhibited uneven, discoloured appearances versus the uniform golden-yellow CK. Physiologically, LH showed delayed water loss and electrolyte leakage, while FQ exhibited premature stress responses. Chemically, low-quality leaves maintained significantly higher post-curing nitrogenous compounds (total nitrogen, alkaloids, protein) and aberrant st

Host-Dependent Cytopathological Responses to Beet Curly Top Virus Infection in Four Plant Species.

Beet curly top viruses (BCTVs) are highly devastating viral pathogens affecting a wide range of plant species worldwide and are transmitted only by leafhoppers in the genus Circulifer. During viral infection, host plant cells undergo morphological changes known as cytopathic effects (CPEs). This study investigated CPEs induced by BCTVs infecting four plant hosts: tomato, pepper, sugar beet, and hemp. BCTV infection resulted in severe stunting, leaf curling, and yellowing, leading to reduced photosynthetic capacity and yield losses. Transmission electron microscopy of ultrathin leaf sections re

Molecular underpinnings of hornwort CO2 concentrating mechanisms: subcellular localization of putative key molecular components in the model hornwort Anthoceros agrestis.

Biophysical CO2 concentrating mechanisms (CCMs) operating at the single-cell level have evolved independently in eukaryotic algae and a single land plant lineage, hornworts. An important component for an efficient eukaryotic CCM is a pyrenoid whose biology is well-characterized in the unicellular green alga, Chlamydomonas reinhardtii. By contrast, pyrenoids and CCM are little understood in hornworts. Here, we investigate the molecular underpinnings and dynamics of hornwort pyrenoids. We do so by studying the subcellular localization of candidate proteins homologous to essential CCM genes in C.

Drought stress and rehydration affect the balance between MGDG and DGDG synthesis in cowpea leaves.

Membranes are main targets of drought, and there is growing evidence for the involvement of membrane lipid in plant adaptation to such an environmental stress. Biosynthesis of the galactosylglycerolipids, monogalactosyl-diacylglycerol (MGDG) and digalactosyl-diacylglycerol (DGDG), which are the main components of chloroplast envelope and thylakoid membranes, could be important for plant tolerance to water deficit and for recovery after rehydration. In this study, galactolipid (GL) biosynthesis in cowpea (Vigna unguiculata L. Walp) leaves was analysed during drought stress and subsequent rewate

Inhibition of chloroplast ATPase by the K+ channel blocker alpha-dendrotoxin.

Possible structural and functional similarities between the channel part, CF0, of chloroplast ATPase (CF0CF1) and ion channels permeable to monovalent cations were investigated using high-affinity toxins mainly targeted against voltage-sensitive K+ channels. In particular, the effect of the K(+)-channel blocker alpha-dendrotoxin and the crude scorpion venom of Leiurus quinquestriatus hebraeus (LQ venom) on ATP synthesis in thylakoid membranes and in CF0CF1-containing liposomes was characterised. Alpha-dendrotoxin (K(i) approximately 5.05 microM) and the LQ venom (K(i) approximately 1.55 microg

Photo-Powered Artificial Organelles for ATP Generation and Life-Sustainment.

Adenosine triphosphate (ATP) is the most important immediate energy source for driving intracellular biochemical reactions in nearly all life forms. Controllable generation of ATP in life is still an unrealized goal. Here, thylakoid fragments are recombined with lipid molecules to synthesize a synthetic/biological hybrid proteoliposome, named highly efficient life-support intracellular opto-driven system (HELIOS) for the generation of ATP. With red light irradiation, HELIOS can improve the intracellular ATP concentration to 1.38-2.45 times in various cell lines. Moreover, it is noticed that HE

Effects of manganese deficiency and added cerium on photochemical efficiency of maize chloroplasts.

The mechanism of the fact that manganese deprivation and cerium addition affect the photochemical efficiency of plants is unclear. In this study, we investigated the improvement by cerium of the damage of the photochemical function of maize chloroplasts under manganese-deprived stress. Chlorophyll fluorescence induction measurements showed that the ratio of variable to maximum fluorescence (Fv/Fm) underwent great decreases under manganese deficiency, which was attributed to the reduction of intrinsic quantum efficiency of the photosystem II units. The electron flow between the two photosystems

Two types of FtsH protease subunits are required for chloroplast biogenesis and Photosystem II repair in Arabidopsis.

FtsH protease is important in chloroplast biogenesis and thylakoid maintenance. Although bacteria contain only one essential FTSH gene, multiple genes exist in cyanobacteria and higher plants. However, the functional significance of FTSH multiplication in plants is unclear. We hypothesized that some FTSH genes may be redundant. To test this hypothesis, we generated double mutant combinations among the different FTSH genes in Arabidopsis thaliana. A double mutant of ftsh1 and ftsh8 showed no obvious phenotypic alterations, and disruption of either FTSH1 or FTSH5 enhanced the phenotype of the ft

Electron paramagnetic resonance (EPR) spectroscopy has been used to determine this physiological response quantitatively in intact cells having different amounts of plastocyanin. The results obtained by the EPR technique were compared with data on isolated plastocyanin determined either by spectrophotometry or immunoelectrophoresis.
