Neuropathology of blepharospasm.

BACKGROUND: The dystonias are a group of disorders characterized by excessive muscle contractions leading to abnormal repetitive movements or postures. In blepharospasm, the face is affected, leading to excessive eye blinking and spasms of muscles around the eyes. The pathogenesis of blepharospasm is not well understood, but several imaging studies have implied subtle structural defects in several brain regions, including the cerebellum. OBJECTIVE: To delineate cerebellar pathology in brains collected at autopsy from 7 human subjects with blepharospasm and 9 matched controls. METHODS: Sectio

Diseases of the myoneural junction.

LEMS appears to associate specifically with SCLC but not with other lung tumors. Experimental and clinical evidence indicates that the autoantibody response is triggered by tumor VGCC determinants, cross-reactivity with determinants at motor nerve terminals leading to the neurological disorder. LEMS currently provides the only example of a paraneoplastic autoantibody-mediated neurological disorder that meets the critical criterion of passive transfer to experimental animals by systemic IgG injections. In young-onset MG cases, the rate of anti-AChR antibody synthesis by cultured thymic cells co

Evidence for in situ and in vitro association between beta-dystroglycan and the subsynaptic 43K rapsyn protein. Consequence for acetylcholine receptor clustering at the synapse.

The accumulation of dystrophin and associated proteins at the postsynaptic membrane of the neuromuscular junction and their co-distribution with nicotinic acetylcholine receptor (AChR) clusters in vitro suggested a role for the dystrophin complex in synaptogenesis. Co-transfection experiments in which alpha- and beta-dystroglycan form a complex with AChR and rapsyn, a peripheral protein required for AChR clustering (Apel, D. A., Roberds, S. L., Campbell, K. P., and Merlie, J. P. (1995) Neuron 15, 115-126), suggested that rapsyn functions as a link between AChR and the dystrophin complex. We ha

Quantification of cerebellar hemispheric purkinje cell linear density: 32 ET cases versus 16 controls.

Although essential tremor (ET) is among the most prevalent neurological diseases, its precise pathogenesis is not understood. Purkinje cell loss has been observed in some studies and is the focus of interest and debate. Expressing these data as Purkinje cells/layer length allows one to adjust for the inherent curved nature of the cerebellar folia. Capitalizing on the Essential Tremor Centralized Brain Repository, we quantified Purkinje cell linear density in cases versus controls. Free-floating 100-μm parasagittal cerebellar hemispheric sections were subjected to rabbit polyclonal anti-Calbind

Structural mechanism of muscle nicotinic receptor desensitization and block by curare.

Binding of the neurotransmitter acetylcholine to its receptors on muscle fibers depolarizes the membrane and thereby triggers muscle contraction. We sought to understand at the level of three-dimensional structure how agonists and antagonists alter nicotinic acetylcholine receptor conformation. We used the muscle-type receptor from the Torpedo ray to first define the structure of the receptor in a resting, activatable state. We then determined the receptor structure bound to the agonist carbachol, which stabilizes an asymmetric, closed channel desensitized state. We find conformational changes

Barbiturate action is dependent on the conformational state of the acetylcholine receptor.

BACKGROUND: Barbiturates act on many neuronal ion channels by poorly understood mechanisms. The authors investigated the hypothesis that barbiturates inhibit the transient open-channel conformation of the nicotinic acetylcholine receptor (nAchR) by binding to a discrete site. METHODS: Inhibition curves of the agonist-stimulated efflux of 8bRb+ from nAchR-rich membrane vesicles prepared from the electric tissue of Torpedo nobiliana were obtained for 14 barbiturates using a filter assay. RESULTS: When added simultaneously with agonist, all agents inhibited the ion efflux with half-inhibitory c

Histopathology of the cerebellar cortex in essential tremor and other neurodegenerative motor disorders: comparative analysis of 320 brains.

In recent years, numerous morphologic changes have been identified in the essential tremor (ET) cerebellar cortex, distinguishing ET from control brains. These findings have not been fully contextualized within a broader degenerative disease spectrum, thus limiting their interpretability. Building off our prior study and now doubling the sample size, we conducted comparative analyses in a postmortem series of 320 brains on the severity and patterning of cerebellar cortex degenerative changes in ET (n = 100), other neurodegenerative disorders of the cerebellum [spinocerebellar ataxias (SCAs, n

A juxta-membrane epitope on the human acetylcholine receptor recognized by T cells in myasthenia gravis.

T cell proliferative responses to synthetic peptides taken from the human nicotinic acetylcholine receptor (AChR) alpha-chain sequence, or to whole AChR purified from electric fish (Torpedo marmorata), have been studied, using blood, thymus, and lymph node cells, from 34 patients with myasthenia gravis (MG) and 17 controls mostly with other neurological diseases. Peptides were selected because they contained amino acid motifs that recur in most defined T cell epitopes. Peptide 257-269 (from the extracellular loop of the AChR alpha-chain between the second and third trans-membrane domains) stim

Barbiturates bind to an allosteric regulatory site on nicotinic acetylcholine receptor-rich membranes.

The ability of barbiturates to bind to acetylcholine receptor-rich membranes purified from the electroplaques of Torpedo nobiliana was examined by centrifugation assay. [14C]Amobarbital both partitioned into the membrane and bound displaceably to a site with an equilibrium dissociation constant of 12 microM. This low affinity made the stoichiometry difficult to obtain despite the high specific activity of acetylcholine receptors in this membrane preparation. However, the data are not inconsistent with a stoichiometry of one barbiturate-binding site per acetylcholine-binding site. Displaceable

"Hairy baskets" associated with degenerative Purkinje cell changes in essential tremor.

Essential tremor (ET) is one of the most common neurologic diseases. Increased numbers of torpedoes and Purkinje cell (PC) loss have been documented in the brains of patients with ET. We recently observed a dense and tangled appearance ("hairiness") of the basket cell axonal plexuses that surround PC soma in Bielschowsky preparations of cerebellar cortex in ET brains. Here, we assessed basket cell "hairiness" in 37 ET (32 cerebellar ET; 5 Lewy body variant ET), 21 nondisease control, and 48 disease control brains using a semiquantitative scale. In 8 cerebellar ET cases (25%), there were high b

A non-radioactive receptor assay for snake venom postsynaptic neurotoxins.

A non-radioactive assay was developed for detecting the binding of postsynaptic neurotoxins to acetylcholine receptor (AchR) from Torpedo californica. Enzyme linked immunosorbent assay (ELISA) wells coated with long or short chain neurotoxins specifically bound to purified AchR while crotamine or two different cardiotoxins did not. Bound receptor was detected by antibody against AchR. Specificity was determined by dose-response experiments and competition studies using carbamylcholine chloride, acetylcholine chloride, or Naja naja atra cobrotoxin mixed with receptor.

Diversity and community structure of ectoparasites infecting some elasmobranch species off the Tunisian coast.

With more than 62 reported species, Tunisia has a rich diversity of elasmobranchs. However, investigations of their parasites in Tunisian waters remain rare and fragmented. With the global biodiversity crisis that most living species are facing, the study of parasite diversity is crucial for assessing ecosystem health and host-parasite interactions. In this study, 2,092 specimens of cartilaginous fishes (Chondrichthyes: Elasmobranchii) belonging to eight species, six genera and five families were sampled along the Tunisian coast and examined for their ectoparasites. The different host species

Tooth reorientation affects tooth function during prey processing and tooth ontogeny in the lesser electric ray, Narcine brasiliensis.

The dental anatomy of elasmobranch fishes (sharks, rays and relatives) creates a functional system that is more dynamic than that of mammalian dentition. Continuous dental replacement (where new teeth are moved rostrally to replace older ones) and indirect fibrous attachment of the dentition to the jaw allow teeth to reorient relative to the jaw over both long- and short-term scales, respectively. In this study, we examine the processing behavior and dental anatomy of the lesser electric ray Narcine brasiliensis (Olfers, 1831) to illustrate that the freedom of movement of elasmobranch dentitio

An anatomical study of an electric organ and its nerve supply in the electric ray (Torpedinidae Narke japonica).

Despite anatomist's great interest in the electric organ (EO) of the electric ray, its detailed morphology remains unclear. In order to understand more completely the architecture of the EO and the branchial organ, it is necessary to examine detailed relationships regarding the origin, course and distribution of nerves innervating the EO. We thus carried out a macroscopic and microscopic anatomical study, focusing on issues, using the 18 sides of nine electric rays. The following results were obtained: (i) the EO was innervated exclusively by the facial, glossopharyngeal and vagus nerves; (ii)

Neuropathologic findings in essential tremor.

Pathologic findings, including cerebellar changes and brainstem Lewy bodies, distinguished 10 essential tremor (ET) cases from 12 controls. Numbers of torpedoes (p = 0.009) and Bergmann glia (p = 0.046) were increased in cases. Six cases (60%) had Lewy bodies vs 2 controls (16.7%) (odds ratio 7.5, 95% CI 1.04 to 54.1; p = 0.035). Four of these six had an atypical distribution of brainstem Lewy bodies. ET may be pathologically heterogeneous.

Characterisation of alpha-dystrobrevin in muscle.

Dystrophin-related and associated proteins are important for the formation and maintenance of the mammalian neuromuscular junction. Initial studies in the electric organ of Torpedo californica showed that the dystrophin-related protein dystrobrevin (87K) co-purifies with the acetylcholine receptors and other postsynaptic proteins. Dystrobrevin is also a major phosphotyrosine-containing protein in the postsynaptic membrane. Since inhibitors of tyrosine protein phosphorylation block acetylcholine receptor clustering in cultured muscle cells, we examined the role of alpha-dystrobrevin during syna

Climbing fiber-Purkinje cell synaptic pathology across essential tremor subtypes.

BACKGROUND: Essential tremor (ET) is heterogeneous in nature and cases may be subdivided based on clinical features. ET patients may thus be subdivided by age of onset, family history of tremor, and presence of head tremor. We recently described climbing fiber-Purkinje cell (CF-PC) synaptic abnormalities in ET; however, these CF pathological features have not been studied across different ET subtypes. OBJECTIVES: To explore whether these CF-PC synaptic abnormalities differ across ET subtypes. METHODS: We studied two climbing fiber (CF-PC) synaptic pathologies (CF synaptic density and percent
