Use of implantable intraperitoneal diffusion chambers to study Bordetella pertussis pathogenesis: growth and toxin production in vivo.

A new animal model for Bordetella pertussis infection is described. The system utilizes small chambers bounded by 0.22-micron filter membranes. The chambers are inoculated with B. pertussis, sealed, and surgically implanted into the peritoneal cavity of mice. The chamber system allows dynamic interchange of soluble host and bacterial factors, but not cells. We have used this system to study the growth of various clinical isolates and vaccine strains of B. pertussis, including an isogenic virulent/avirulent pair. All virulent strains tested grew well and had remarkably similar growth kinetics.

In Silico Analysis of Genetic VapC Profiles from the Toxin-Antitoxin Type II VapBC Modules among Pathogenic, Intermediate, and Non-Pathogenic Leptospira.

Pathogenic Leptospira spp. is the etiological agent of leptospirosis. The high diversity among Leptospira species provides an array to look for important mediators involved in pathogenesis. Toxin-antitoxin (TA) systems represent an important survival mechanism on stress conditions. vapBC modules have been found in nearly one thousand genomes corresponding to about 40% of known TAs. In the present study, we investigated TA profiles of some strains of Leptospira using a TA database and compared them through protein alignment of VapC toxin sequences among Leptospira spp. genomes. Our analysis ide

Toxic anti-phage defense proteins inhibited by intragenic antitoxin proteins.

UNLABELLED: Recombination-promoting nuclease (Rpn) proteins are broadly distributed across bacterial phyla, yet their functions remain unclear. Here we report these proteins are new toxin-antitoxin systems, comprised of genes-within-genes, that combat phage infection. We show the small, highly variable Rpn C -terminal domains (Rpn S ), which are translated separately from the full-length proteins (Rpn L ), directly block the activities of the toxic full-length proteins. The crystal structure of RpnA S revealed a dimerization interface encompassing a helix that can have four amino acid repeats

Current strategies for designing antidotes against botulinum neurotoxins.

INTRODUCTION: Botulinum neurotoxins (BoNTs) are proteins responsible for the deadly paralytic disease botulism. Extreme toxicity, ease of production and lack of antidotes against BoNT makes it a category A biothreat agent, according to the United States Center of Disease Control and Prevention. The only available therapy for BoNT is an equine antitoxin antibody or/and a protracted respiratory support system. Even then, antibody treatment can only prevent further exposure of the toxin and cannot rescue already intoxicated neurons. AREAS COVERED: In this article, the authors provide a summary o

Bacterial cell division protein FtsZ complexes with a phage protein to activate bacterial immunity.

Many antiphage defence systems directly bind a specific phage-encoded protein that acts similar to a pathogen-associated molecular pattern to activate an immune response. Such activation is often assumed to occur independent of host factors. Here we demonstrate that the antiphage defence protein CapRelEbc, a fused toxin-antitoxin system from Enterobacter chengduensis, senses the T7 phage-encoded protein Gp0.4 in complex with the host bacterial factor FtsZ, an essential cell division protein. During T7 infection, Gp0.4 sequesters monomeric FtsZ to block its polymerization and thereby inhibit ba

Enterotoxigenic Escherichia coli diarrhea in an endemic area prepares the intestine for an anamnestic immunoglobulin A antitoxin response to oral cholera B subunit vaccination.

We examined whether infection with enterotoxigenic Escherichia coli (ETEC) producing the heat-labile enterotoxin (LT) can prime the gut immune system to respond more efficiently to the immunologically related cholera B subunit component of a recently developed oral B subunit-whole-cell cholera vaccine (B-WCV). Nine Bangladeshi adults who had been hospitalized for watery diarrhea caused by LT-producing ETEC were given a single oral immunization with B-WCV on day 28 after hospital admission. The vaccine preparation used was adjusted to contain a lower-than-usual dose of B subunit, which had been

Structural Basis of Shiga Toxin Neutralization by Human Antibodies Targeting Canonical Receptor-Binding and Non-Canonical Assembly Sites.

Shiga toxin-producing Escherichia coli (STEC) infections cause severe systemic complications, yet targeted antitoxin therapeutics remain an unmet clinical need. Here, we isolated a panel of human monoclonal antibodies (mAbs) targeted at STEC toxin and characterized two potent neutralizing candidates, RDS045 and RDS059. The cryo-electron microscopy structure of the human mAb RDS059 in complex with the Shiga toxin 1a B subunit (Stx1aB) reveals that RDS059 recognizes the surface-exposed loop of Stx1aB, whereby it directly competes with the host glycolipid globotriaosylceramide (Gb3) to block toxi

Macrophages are sensitive to anthrax lethal toxin through an acid-dependent process.

Anthrax lethal toxin, which consists of two proteins, protective antigen and lethal factor, is lethal for experimental animals. This study describes the first in vitro system demonstrating lethality of the toxin. Mouse peritoneal macrophages are killed within 1 h of exposure to the toxin. Neither protein component alone shows any toxic activity. The minimal effective concentration of protective antigen and lethal factor was approximately equal to 10(-2) and approximately equal to 10(-3) micrograms/ml, respectively. None of the several established cell lines examined was killed. Cells could be

Drivers of bacterial genomes plasticity and roles they play in pathogen virulence, persistence and drug resistance.

Despite the advent of next-generation sequencing (NGS) technologies, sophisticated data analysis and drug development efforts, bacterial drug resistance persists and is escalating in magnitude. To better control the pathogens, a thorough understanding of their genomic architecture and dynamics is vital. Bacterial genome is extremely complex, a mosaic of numerous co-operating and antagonizing components, altruistic and self-interested entities, behavior of which are predictable and conserved to some extent, yet largely dictated by an array of variables. In this regard, mobile genetic elements (

Adoptive transfer of gut mucosal antitoxin memory by isolated B cells 1 year after oral immunization with cholera toxin.

A protocol was elaborated for the adoptive transfer of lymphocytes from mice which were orally immunized with cholera toxin (CT) to enable the study of long-term gut mucosal immunological memory at the single-cell level. Mesenteric lymph node (MLN) cells were transferred 1 year after priming immunizations, and recipient animals were challenged perorally on days 1 and 2 with CT before sacrifice on day 6 to 7 following transfer of cells. Strong antitoxin ELISPOT spot-forming cell (SFC) responses were recorded in spleens, MLN, and laminae propriae (LP) of recipient mice. In contrast, no SFC were

Genomic characterization of a pathogenic Bacillus licheniformis strain LSDY01: deciphering its genetic diversity and virulence-associated traits.

BACKGROUND: Bacillus licheniformis is an opportunistic pathogen in clinical settings. However, the emergence of clinical strains carrying horizontally acquired virulence determinants, including chromosomal genomic islands harboring yopX, a putative type IV secretion system (T4SS), and plasmids bearing toxin-antitoxin systems and additional virulence factors, poses a significant challenge to diagnosis and treatment. Moreover, the genetic basis of the pathogenicity of clinical isolates has not been comprehensively studied. METHODS: A pathogenic B. licheniformis strain (LSDY01) isolated from a s

Proteomic insight into the primycin fermentation process of Saccharomonospora azurea.

Saccharomonospora azurea SZMC 14600 is a member of the family Pseudonocardiaceae exclusively used for industrial scale production of primycin a large 36-membered non-polyene macrolide lactone antibiotic belonging to the polyketide class of natural products. Even though maximum antibiotic yield has been achieved by empirically optimized two-step fermentation process, little is known about the molecular components and mechanisms underlying the efficient antibiotic production. In order to identify differentially expressed proteins (DEPs) between the pre- and main-fermentation stages of primycin,

Clarifying the Link between Toxin-Antitoxin Modules and Bacterial Persistence.

While most of a bacterial population is killed upon antibiotic exposure, a fraction transiently exhibits a multidrug-tolerant phenotype termed antibiotic persistence. This phenomenon enables the bacteria to escape killing by drugs and is presumed to be, at least partly, responsible for the recalcitrance of many bacterial infections. For this reason, understanding mechanisms allowing a fraction of a bacterial population to become transiently multidrug-tolerant represents an essential step to eradicate these persisting subpopulations. Toxin-antitoxin (TA) systems were proposed as perfect candida

Type I toxin-dependent generation of superoxide affects the persister life cycle of Escherichia coli.

Induction of growth stasis by bacterial toxins from chromosomal toxin-antitoxin systems is suspected to favor formation of multidrug-tolerant cells, named persisters. Recurrent infections are often attributed to resuscitation and regrowth of persisters upon termination of antibiotic therapy. Several lines of evidence point to oxidative stress as a crucial factor during the persister life cycle. Here, we demonstrate that the membrane-depolarizing type I toxins TisB, DinQ, and HokB have the potential to provoke reactive oxygen species formation in Escherichia coli. More detailed work with TisB r

Rare human skin infection with Corynebacterium ulcerans: transmission by a domestic cat.

Corynebacterium ulcerans is mainly known for its ability to cause animal infections. Some strains of C. ulcerans produce diphtheria toxin, which can cause life-threatening cardiopathies and neuropathies in humans. Human cutaneous C. ulcerans infection is a very rare disease that mimics classical cutaneous diphtheria. We present a very rare case of a C. ulcerans skin infection caused by a non-diphtheria toxin-producing strain of C. ulcerans that resolved after 3 weeks of therapy with amoxicillin-clavulanate. A pet cat was the probable source of infection. The presence of C. ulcerans in the mout

Characterization of Clostridioides difficile Persister Cells and Their Role in Antibiotic Tolerance.

Clostridioides difficile is a Gram-positive pathogen known for its toxin production and spore formation. It is primarily responsible for most cases of antibiotic-associated diarrhea. Bacterial persisters are a small subset of the population that exhibits transient tolerance to bactericidal substances, and they are of significant medical concern due to their association with the emergence of antibiotic resistance and difficult-to-treat chronic or recurrent infections. Vancomycin, the predominant antibiotic utilized in the management of C. difficile infection, is extensively applied in the realm

Toxic shock syndrome toxin-1, not α-toxin, mediated Bundaberg fatalities.

The 1928 Bundaberg disaster is one of the greatest vaccine tragedies in history. Of 21 children immunized with a diphtheria toxin-antitoxin preparation contaminated with Staphylococcus aureus, 18 developed life-threatening disease and 12 died within 48  h. Historically, the deaths have been attributed to α-toxin, a secreted cytotoxin produced by most S. aureus strains, yet the ability of the Bundaberg contaminant microbe to produce the toxin has never been verified. For the first time, the ability of the original strain to produce α-toxin and other virulence factors is investigated. The study

Coccoid H. pylori: A key driver of recurrent infections and a target for novel therapies.

Helicobacter pylori is a globally prevalent pathogen linked to gastritis, peptic ulcers, and gastric cancer, with over half the world's population infected as of recent estimates. While its helical form is well characterized for colonization and virulence, the bacterium's ability to transform into a coccoid morphology under stress conditions remains enigmatic. This review explores the role of the coccoid form in the recurrence of H. pylori infection, emphasizing its potential as a viable but non-culturable (VBNC) state that facilitates persistence, immune evasion, and reinfection. We highlight
