MecE, MecB, and MecC proteins orchestrate methyl group transfer during dichloromethane fermentation.

Dichloromethane (DCM), a common hazardous industrial chemical, is anaerobically metabolized by four bacterial genera: Dehalobacter, Dehalobacterium, Ca. Dichloromethanomonas, and Ca. Formimonas. However, the pivotal methyltransferases responsible for DCM transformation have remained elusive. In this study, we investigated the DCM catabolism of Dehalobacterium formicoaceticum strain EZ94, contained in an enriched culture, using a combination of biochemical approaches. Initially, enzymatic assays were conducted with cell-free protein extracts, after protein separation by blue native polyacrylami

Transformation of kidney epithelial cells by a quinol thioether via inactivation of the tuberous sclerosis-2 tumor suppressor gene.

Although hydroquinone (HQ) is a rodent carcinogen, because of its lack of mutagenicity in standard bacterial mutagenicity assays it is generally considered a nongenotoxic carcinogen. 2,3,5-Tris-(glutathion-S-yl)HQ (TGHQ) is a potent nephrotoxic metabolite of HQ that may play an important role in HQ-mediated nephrocarcinogenicity. TGHQ mediates cell injury by generating reactive oxygen species and covalently binding to tissue macromolecules. We determined the ability of HQ and TGHQ to induce cell transformation in primary renal epithelial cells derived from the Eker rat. Eker rats possess a ger

Assessing the impact of lime on chromium migration in soil caused by basic chromium sulfate in tannery.

Chromium (Cr) pollution is the primary pollution problem of the soil in tannery. However, the effect of tanning chemicals on Cr migration in soil has not been clearly elucidated. Column leaching tests were designed in this study to reveal the transport and transformation of Cr from basic chromium sulfate (BCS) into soil and the effects of lime on Cr migration and transformation. The results showed that BCS was mainly leached out in the state of Cr(VI) after entering the soil, and the Cr concentration in leachate decreased with the increase of the bulking thickness of the BCS. Compared with the

Harnessing bacterial symbionts of beetles for the valorization of lignin into value-added byproducts: Recent advances and future perspectives.

Lignin is the most abundant renewable aromatic heteropolymer in nature and is highly recalcitrant due to its complex structure. The gut systems of beetles function as an evolutionarily optimized microreactor wherein mechanical pretreatment, physicochemical gradients, and selectively maintained microbial consortia act synergistically to transform, mineralize, and metabolize lignin as an energy source. Beetle-associated bacterial symbionts represent a promising yet underexploited resource for lignin valorization into value-added products. This review provides emerging evidence of lignin degradat

Establishment of a High-Efficiency In Vitro Regeneration and Agrobacterium-Mediated Genetic Transformation System for Ajania achilleoides (Turcz.) Poljakov.

Ajania achilleoides, a wild species with strong environmental adaptability, is a valuable germplasm resource for genetic improvement and functional gene studies. However, efficient in vitro regeneration and genetic transformation systems for this species remain undeveloped, limiting functional gene characterization and genetic improvement of this valuable germplasm resource. In this study, leaf explants were used to establish an efficient regeneration system by evaluating different combinations of plant growth regulators and to develop an Agrobacterium tumefaciens-mediated genetic transformati

Biochar as an enhancer of the stability, mesoporous structure and oxytetracycline adsorption capacity of ferrihydrite: Role of the silicon component.

The char component of biochar can act as an electron shuttle and redox agent to accelerate the transformation of ferrihydrite, but how the silicon component of biochar affects ferrihydrite transformation and pollutant removal remains unclear. In this paper, infrared spectroscopy, electron microscopy, transformation experiments and batch sorption experiments were conducted to examine a 2-line ferrihydrite formed by alkaline precipitation of Fe3+ on a rice straw-derived biochar. Fe-O-Si bonds were developed between the precipitated ferrihydrite particles and biochar silicon component, increasing

Competence pili in Streptococcus pneumoniae are highly dynamic structures that retract to promote DNA uptake.

The competence pili of transformable Gram-positive species are phylogenetically related to the diverse and widespread class of extracellular filamentous organelles known as type IV pili. In Gram-negative bacteria, type IV pili act through dynamic cycles of extension and retraction to carry out diverse activities including attachment, motility, protein secretion, and DNA uptake. It remains unclear whether competence pili in Gram-positive species exhibit similar dynamic activity, and their mechanism of action for DNA uptake remains unclear. They are hypothesized to either (1) leave transient cav

Secretion of a pneumococcal type II secretion system pilus correlates with DNA uptake during transformation.

Streptococcus pneumoniae is a major human pathogen that successfully adapts to the host environment via an efficient uptake system for free DNA liberated from other organisms in the upper respiratory tract, facilitating immune evasion and drug resistance. Although the initial signaling events leading to pneumococcal competence for DNA transformation and the fate of DNA when it has been taken up have been extensively studied, the actual mechanism by which DNA in the environment may traverse the thick capsular and cell wall layers remains unknown. Here we visualize that induction of competence r

Natural competence is a major mechanism for horizontal DNA transfer in the oral pathogen Porphyromonas gingivalis.

UNLABELLED: Porphyromonas gingivalis is a Gram-negative anaerobe that resides exclusively in the human oral cavity. Long-term colonization by P. gingivalis requires the bacteria to evade host immune responses while adapting to the changing host physiology and alterations in the composition of the oral microflora. The genetic diversity of P. gingivalis appears to reflect the variability of its habitat; however, little is known about the molecular mechanisms generating this diversity. Previously, our research group established that chromosomal DNA transfer occurs between P. gingivalis strains. I

Collateral damage: insights into bacterial mechanisms that predispose host cells to cancer.

Infections are estimated to contribute to 20% of all human tumours. Viruses are known to induce cell transformation, but evidence has also linked bacteria, such as Helicobacter pylori and Salmonella enterica subsp. enterica serovar Typhi, to different cancer types. In addition, Chlamydia trachomatis, Fusobacterium nucleatum and Bacteroides fragilis are associated with the development of cancer, although a causal relationship has not yet been established. Bacterial effectors such as colibactin and the virulence factor cytotoxin-associated gene A (CagA) can promote cancer directly by influencing

Colonic bacterial metabolites and human health.

The influence of the microbial-mammalian metabolic axis is becoming increasingly important for human health. Bacterial fermentation of carbohydrates (CHOs) and proteins produces short-chain fatty acids (SCFA) and a range of other metabolites including those from aromatic amino acid (AAA) fermentation. SCFA influence host health as energy sources and via multiple signalling mechanisms. Bacterial transformation of fibre-related phytochemicals is associated with a reduced incidence of several chronic diseases. The 'gut-liver axis' is an emerging area of study. Microbial deconjugation of xenobioti

Insight into the mechanisms of insoluble phosphate transformation driven by the interactions of compound microbes during composting.

Phosphate-solubilizing (PS) microbes are important to improve phosphorus availability and transformation of insoluble phosphate, e.g., rock phosphate (RP). The use of phosphate solubilizing bacteria (PSB) as inoculants have been proposed as an alternative to increase phosphate availability in RP and composting fertilizers. In this study, the effect of compound PSB coinoculation and single-strain inoculation on the transformation of insoluble phosphate were compared in a liquid medium incubation and RP-enriched composting. The goal of this study was to understand the possible mechanisms of inso

Transition Metal Ion-Mediated Tyrosine-Based Short-Peptide Amphiphile Nanostructures Inhibit Bacterial Growth.

We report the design and synthesis of a biocompatible small-peptide-based compound for the controlled and targeted delivery of encapsulated bioactive metal ions through transformation of the internal nanostructures of its complexes. A tyrosine-based short-peptide amphiphile (sPA) was synthesized and observed to self-assemble into β-sheet-like secondary structures. The self-assembly of the designed sPA was modulated by application of different bioactive transition-metal ions, as was confirmed by spectroscopic and microscopic techniques. These bioactive metal-ion-conjugated sPA hybrid structures

Expression and characterization of an iron-containing superoxide dismutase from Burkholderia pseudomallei.

A superoxide dismutase (SOD) gene from Burkholderia pseudomallei, the causative agent of melioidosis, was cloned and expressed in Escherichia coli, and its product was functionally and physically characterized. The gene has an open-reading frame of 579 bp. The deduced amino acid sequence has 192 residues with a calculated molecular mass of ~22 kDa. Sequence comparison with other bacterial SODs showed that the protein contains typical metal-binding motifs and other Fe-SOD-conserved residues. The sequence has substantial similarity with other bacterial Fe-SOD sequences. The enzymatic activity of

Nonthermal plasma processing catalyzed by CuFe2O4 for organic pollutants remediation and bacterial inactivation with density functional theory.

The suggested nonthermal plasma has been employed for organic pollutants remediation and bacterial inactivation with catalyst (CuFe2O4) via reactive oxygen and nitrogen species, along with catalytic density functional theory processing. The plasma generated species O2- (g.), OH• (g.), H2O2 (aq.), and NOx (aq.) are used for the remediation of organic pollutants, such as reactive black5 and bromocresol green with catalytic oxidative and reductive transformation, like as from Fe2+ (aq.) to Fe3+ (aq.) and from Cu2+ (aq.) to Cu1+ (aq.), respectively. In the presence of plasma with CuFe2O4, the poll

Two D-loop resolution systems enable natural genetic transformation in bacteria.

Natural transformation is a widespread mechanism driving genetic exchanges in bacteria. It proceeds from the capture of extracellular DNA released by other cells. Internalized in linear single strands, this exogenous DNA is ultimately integrated into the genome by homologous recombination. It is unknown how the RecA-directed D-loop intermediate of this dedicated recombination pathway is processed. We report that resolution of the transformation D-loop depends on two endonucleases of opposing phylogenetic distribution in bacteria. One is YraN, which has coevolved and interacts with the ComM hel

[Pathogenic neisseriae--model of bacterial virulence and genetic flexibility].

The early stages of an infection with pathogenic Neisseriae are determined by receptor-mediated events that finally lead to the attachment and invasion of human mucous tissues. The factors participating in this process, the pili, OPA proteins, and perhaps lipopolysaccharide (LPS), are subject to complex genetic changes that enable the pathogens to produce multiple variant forms of these factors. Antigenic variation of the pathogenic Neisseriae permits both, the escape from the human immune response and the interaction with different cells and tissues of the human host. One of the intrinsic mec

Effects of straw return combined with different amendments on soil enzyme activities and bacterial community structure in acidic purple soil.

Soil microorganisms are fundamental drivers of nutrient cycling and ecosystem functioning in agricultural soils, but their responses to management practices in acidic agroecosystems remain insufficiently understood. Acidification-induced changes in soil environmental conditions can constrain bacterial activity and disrupt carbon and nutrient transformation processes. This study investigated how straw return combined with different amendments affects soil enzyme activities and bacterial community structure in acidic purple soil under a 2-year rice-rapeseed rotation system. The Mr 90,000 heat shock protein-free androgen receptor has a high affinity for steroid, in contrast to the glucocorticoid receptor.

Transformed and bacterially expressed glucocorticoid receptors free from Mr 90,000 heat shock protein (hsp90) have a 100-fold lower steroid-binding affinity than the hsp90-bound nontransformed receptor, suggesting that hsp90 is needed for high-affinity steroid binding [Nemoto, T., Ohara-Nemoto, Y., Denis, M., & Gustafsson, J.-A. (1990) Biochemistry 29, 1880-1886]. To investigate whether or not this phenomenon is common to all steroid receptors, we investigated the steroid-binding affinities of bacterially expressed and transformed androgen receptors.
