Distinct Roles of Transketolase (TktA) and Transaldolase (talB) in Metabolism, Biofilm Formation, and Flea Colonization in Yersinia pestis.

The flea-borne transmission of Yersinia pestis relies on biofilm formation and metabolic adaptation within the insect gut. The pentose phosphate pathway (PPP) is central to these processes, yet the contribution of its non-oxidative branch remains poorly defined. Here, we investigated the roles of transketolase (TktA) and transaldolase (TalB) in plague bacillus physiology, metabolism, and flea colonization. TktA was essential for growth, preventing assessment of its role in biofilm formation and in vivo colonization. In contrast, TalB was dispensable for growth but required for optimal biofilm

In situ localization of transketolase activity in epithelial cells of different rat tissues and subcellularly in liver parenchymal cells.

Metabolic mapping of enzyme activities (enzyme histochemistry) is an important tool to understand (patho)physiological functions of enzymes. A new enzyme histochemical method has been developed to detect transketolase activity in situ in various rat tissues and its ultrastructural localization in individual cells. In situ detection of transketolase is important because this multifunctional enzyme has been related with diseases such as cancer, diabetes, Alzheimer's disease, and Wernicke-Korsakoff's syndrome. The proposed method is based on the tetrazolium salt method applied to unfixed cryostat

The relationship between erythrocyte transketolase activity and the 'TPP effect' in Wernicke's encephalopathy and other thiamine deficiency states.

Patients (n = 104) were judged to be thiamine deficient by the criteria of erythrocyte transketolase activity (ETK) less than 0.6 U/g of hemoglobin, or greater than 17% increase in this activity on addition of thiamine pyrophosphate in vitro (TPP effect). ETK activated by TPP in vitro (AETK) was related to ETK by a linear regression of slope greater than or equal to 1, implying that transketolose apoenzyme (apoTK) was constant or decreased as ETK decreased. For most patient groups the value of apoTK was 0.1 U/g and the slope 1.033 to 1.050. In the subgroup of non-vomiting drinkers with Wernick

Expression of transketolase-like gene 1 (TKTL1) depends on disease phase in patients with chronic myeloid leukaemia (CML).

PURPOSE: Overexpression of transketolase-like gene 1 (TKTL1) on RNA and protein level has been linked to tumour progression, metastasis and unfavourable patient outcome in many solid tumours. Chronic myeloid leukaemia (CML) cells show metabolic characteristics resembling deviations observed in TKTL1 overexpressing solid tumour cells. We therefore sought to evaluate TKTL1 gene expression in different phases of CML. METHODS: A total of 120 peripheral blood samples from 69 patients in various phases of CML and 21 healthy individuals were investigated. TKTL1 expression levels were determined by r

Thiamine pyrophosphate effect and normalized erythrocyte transketolase activity ratio in Wernicke-Korsakoff patients and acute alcoholics undergoing detoxification.

Thiamine deficiency may be assessed clinically by an abnormally low specific erythrocyte transketolase activity and/or by abnormally large activation by thiamine diphosphate in vitro (or 'TPP effect'). In the present investigation, we report erythrocyte transketolase activation by TPP in acute alcoholics and Wernicke-Korsakoff patients undergoing detoxification. A new age-dependent parameter was used to improve the reliability of transketolase activity as an indicator of marginal thiamine deficiency. Thus normalized transketolase activity ratio (NTKZ), primary activation ratio (PAR) and furthe

Pentose-phosphate pathway in Saccharomyces cerevisiae: analysis of deletion mutants for transketolase, transaldolase, and glucose 6-phosphate dehydrogenase.

Deletion mutants for the yeast transketolase gene TKL1 were constructed by gene replacement. Transketolase activity was below the level of detection in mutant crude extracts. Transketolase protein could be detected as a single protein band of the expected size by Western-blot analysis in wild-type strains but not in the deletion mutant. Deletion of TKL1 led to a reduced but distinct growth in synthetic medium without an aromatic amino-acid supplement. We also isolated double and triple mutants for transketolase (tkl1), transaldolase (tal1), and glucose 6-phosphate dehydrogenase (zwf1) by cross

Discovery of Pyrazole-Fusaric Acid Derivatives as the Potential Herbicidal Lead Compounds Targeting Both Transketolase and Protoporphyrinogen Oxidase.

Transketolase (TKL, EC 2.2.1.1) was confirmed as a potential new target for herbicides. In this study, a series of pyrazole-fusaric acid derivatives 9a-9o were designed and synthesized via structural optimization of compound 7r. Among them, compound 9f exhibited good inhibition on Digitaria sanguinalis and Amaranthus retroflexus (inhibition >95%) even at 10 mg/L via the small cup method. It exhibited excellent inhibition against D. sanguinalis and A. retroflexus (inhibition >90%, 150 g ai/ha) via the pot bioassay superior to the positive control. Significantly, it showed good crop safety to ma

Transketolase, pyruvate and oxoglutarate dehydrogenase activities and [14C]thiamin turnover in tissues of mice fed thiamin-deficient diet.

A study was made of turnover of [14C]thiamin (5 or 2 micrograms/mouse) in mice fed a thiamin-deficient diet. Simultaneously the activities of the thiamin-dependent enzymes (transketolase, pyruvate and oxoglutarate dehydrogenases) were measured as an index of efficiency of fulfilling the coenzyme function of the vitamin under conditions of different thiamin status. After [14C]thiamin injections of 5 micrograms/mouse, kidney, spleen, stomach and pancreas tissue stores turned over completely on day 9, whereas by day 13 this process had not yet been finished in liver, heart and brain. [Alkaline phosphatase and transketolase activity during immunogenesis in the lymphoid organs of rats with vitamin B1 deficiency caused by oxythiamine].

A study was made of the effect of different doses of hydroxythiamine on activity of alkaline phosphatase and transketolase in rat spleen and thymus during primary and secondary administration of human gamma-globulin. Immunization of rats significantly raised alkaline phosphatase activity in both organs. Administration of a large single dose of hydroxythiamine (400 mg/kg bw) reduced transketolase activity in the thymus and spleen in both immunized and non-immunized animals and did not change alkaline phosphatase activity in these organs.

[Study of the thiamine supply of healthy school-age children and adolescents by examining transketolase activity and the TDP effect in the blood].

A modified method is described for determination of transketolase (TK) activity in the blood and the degree of its stimulation under the effect of exogenous thiamine diphosphate (TDP-effect). The method allows for an appreciable reduction of the time required for an analysis and to decrease 2-fold the blood volume necessary for the determination. With the use of the modification described 70 schoolchildren of three age groups (7-10, 11-13 and 14-15 years old) attending one of the Moscow non-specialized prolonged-day schools were examined. It was found that the magnitude of TDP-effect in the sc

Role of thiamine status and genetic variability in transketolase and other pentose phosphate cycle enzymes in the progression of diabetic nephropathy.

BACKGROUND: Pentose phosphate pathway (PPP) represents a potentially 'protective' mechanism in hyperglycaemia due to shunting of glycolytic intermediates into PPP reactions. We hypothesized that thiamine status (plasma and erythrocyte levels of thiamine and its esters) together with genetic variability in key PPP enzymes-transketolase (TKT), transaldolase and TKT-like-might contribute to the progression of diabetic nephropathy (DN) and mortality of diabetics. METHODS: A total of 240 diabetic subjects with variable degree of kidney disease were included at baseline and were followed up for a m

[Transketolase activity in thiamine deficiency of the kidney in rainbow trout (Salmo gairdneri) following continous feeding with raw hearring].

The feeding of crude, thiaminase-containing herring to rainbow trout produced external symptoms of thiamine deficiency after a period of 150 days. The level of transketolase activity in the kidney decreased continuously until the first appearance of deficiency symptoms, while the TPP effect showed an upward trend. After prolonged feeding of crude herring the decline of transketolase activity is accompanied by a deficiency of the apoenzyme of transketolase. The administration of thiamine-containing food to the thiamine-deficient animals induced an immediate increase n transketolase activity. On

Randomised trial of intravenous thiamine and/or magnesium sulphate administration on erythrocyte transketolase activity, lactate concentrations and alcohol withdrawal scores.

Alcohol withdrawal syndrome (AWS) occurs in 2% of patients admitted to U.K. hospitals. Routine treatment includes thiamine and benzodiazepines. Laboratory studies indicate that thiamine requires magnesium for optimal activity, however this has not translated into clinical practice. Patients experiencing AWS were randomized to three groups: (group 1) thiamine, (group 2) thiamine plus MgSO4 or (group 3) MgSO4. Pre- and 2-h post-treatment blood samples were taken. AWS severity was recorded using the Glasgow Modified Alcohol Withdrawal Score (GMAWS). The primary outcome measure was 15% change in e

[The effect of cocarboxylase treatment on erythrocyte transketolase and blood thiamine in patients with end stage renal disease undergoing maintenance hemodialysis].

The depressed ETKA in ESRD patients is supposed to be caused and/or aggravated by several factors among which the diminished content of thiamine in blood and/or disturbances of thiamine utilization seem to play the major role. This role stems from the fact that thiamine acts as the cofactor of transketolase. In order to check the therapeutic significance of this relationship we introduced the thiamine pyrophosphoric acid ester chloride (Cocarboxylasum-CC) administration in 25 patients (mHD + CC). Immediately after each HD performance CC was given i.v. during 12 weeks in a doses of 5 mg/kg b.w.

Cloning of human transketolase cDNAs and comparison of the nucleotide sequence of the coding region in Wernicke-Korsakoff and non-Wernicke-Korsakoff individuals.

Variants of the enzyme transketolase which possess reduced affinity for its cofactor thiamine pyrophosphate (high apparent Km) have been described in chronic alcoholic patients with Wernicke-Korsakoff syndrome. Since the syndrome has been shown to be directly related to thiamine deficiency, it has been hypothesized that such transketolase variants may represent a genetic predisposition to the development of this syndrome. To test this hypothesis, human transketolase cDNA clones were isolated, and their nucleotide and predicted amino acid sequence were determined. Transketolase was found to be

Comparison of transketolase activity and thiamin pyrophosphate levels in erythrocytes and liver of rainbow trout (Salmo gairdneri) as indicators of thiamin status.

Yearling rainbow trout (Salmo gairdneri) were fed a purified diet with and without thiamin supplementation for 30 wk, at which time overt signs of thiamin deficiency appeared in the deficient group. Overt signs of thiamin deficiency were anorexia, darkening and ataxia. Death rapidly followed the development of overt thiamin deficiency. Transketolase activity and thiamin pyrophosphate levels were measured monthly in erythrocyte and liver samples. Significant differences in erythrocyte transketolase activity between fish fed the thiamin-deficient and control diets were measured after 24 wk of fe
