Cross Kingdom Metabolic Engineering Paradigm Elevating Sustainable Protein Production.

Confronting the dual crisis of escalating global protein demand and unsustainable agriculture necessitates transformative solutions. Here, we pioneer evolutionary insights from maize nitrogen optimization via asparagine synthetase (ASNS) to rewire metabolism in Pichia pastoris. Empirically, the tri-copy ASNS strain achieved superior protein titers: 62.48% crude protein, 47.86% total amino acids, and 8.05% branched-chain amino acids, nutritionally surpassing conventional protein sources. Genome-scale modeling and transcriptomic studies provided convergent evidence that ASNS overexpression drove

Homopolymerization and copolymerization of ε-caprolactone and l-lactide organocatalyzed by carboxylic acids with mono-, di-, and tri-functionality.

A challenge in the design of biocompatible polymers involves the use of non-cytotoxic catalysts in the ring-opening polymerization (ROP) of lactones. In this study, a family of seven mono-, di-, and tricarboxylic acids was used as organocatalysts for the ROP of ε-caprolactone (CL) and l-lactide (l-LA). The use of a long-chain aliphatic alcohol such as 1-docosanol (C22OH) has been demonstrated to accelerate the polymerization rate. For the ROP of CL, carboxylic acids with pk a values less than 4 and an increase in functional groups exhibited the highest catalytic efficiency. In contrast, the RO

Integrated landscape of cardiac metabolism in end-stage human nonischemic dilated cardiomyopathy.

Heart failure (HF) is a leading cause of mortality. Failing hearts undergo profound metabolic changes, but a comprehensive evaluation in humans is lacking. We integrate plasma and cardiac tissue metabolomics of 678 metabolites, genome-wide RNA-sequencing, and proteomic studies to examine metabolic status in 87 explanted human hearts from 39 patients with end-stage HF compared with 48 nonfailing donors. We confirm bioenergetic defects in human HF and reveal selective depletion of adenylate purines required for maintaining ATP levels. We observe substantial reductions in fatty acids and acylcarn

Significance of phototherapy-induced riboflavin deficiency in the full-term neonate.

As a result of impaired fatty acid oxidation, a characteristic urinary dicarboxylic aciduria occurs in the riboflavin deficient animal. We compared the occurrence of riboflavin deficiency induced by phototherapy with changes in urinary organic acid profiles in 8 full-term, breast-fed neonates who received phototherapy for hyperbilirubinemia, and in 10 full-term, breastfed controls. Riboflavin status was assessed by measuring flavin adenine dinucleotide saturation of erythrocyte glutathione reductase. All 8 neonates exposed to phototherapy developed riboflavin deficiency (p less than 0.001). Ri

Testosterone regulates thymic remodeling by altering metabolic reprogramming in male rats.

The thymus is an energy-consuming organ, and its metabolism changes with atrophy. Testosterone regulates thymus remodeling (atrophy and regeneration). However, the characteristics of the energy metabolism during testosterone-mediated thymic atrophy and regeneration remain unclear. In this study, we demonstrated that testosterone ablation (implemented by immunocastration and surgical castration) induced global metabolic changes in the thymus. Efficient modulation of a barium metal-organic framework using amino acids.

In recent years, significant advances have been made in the precise control of the physical properties of metal-organic frameworks (MOFs) via the linker-modulated method in which modulators compete with linkers and impose kinetic limitations through crystal growth. In this regard, the structure of a new barium-organic framework [Ba(H2BTC)2(H2O)4]n, BaBTC (BTC = 1,3,5-benzene tricarboxylic acid) is introduced, which allows the competitive coordination strategy and growth orientation of an alkaline-earth metal-organic framework (AEMOF) to be probed without sacrificing phase purity, porosity and

Pyruvate carboxylase and cancer progression.

Pyruvate carboxylase (PC) is a mitochondrial enzyme that catalyzes the ATP-dependent carboxylation of pyruvate to oxaloacetate (OAA), serving to replenish the tricarboxylic acid (TCA) cycle. In nonmalignant tissue, PC plays an essential role in controlling whole-body energetics through regulation of gluconeogenesis in the liver, synthesis of fatty acids in adipocytes, and insulin secretion in pancreatic β cells. In breast cancer, PC activity is linked to pulmonary metastasis, potentially by providing the ability to utilize glucose, fatty acids, and glutamine metabolism as needed under varying

Comprehensive quantification of fuel use by the failing and nonfailing human heart.

The heart consumes circulating nutrients to fuel lifelong contraction, but a comprehensive mapping of human cardiac fuel use is lacking. We used metabolomics on blood from artery, coronary sinus, and femoral vein in 110 patients with or without heart failure to quantify the uptake and release of 277 metabolites, including all major nutrients, by the human heart and leg. The heart primarily consumed fatty acids and, unexpectedly, little glucose; secreted glutamine and other nitrogen-rich amino acids, indicating active protein breakdown, at a rate ~10 times that of the leg; and released intermed

Revealing Hidden Variables in DESI-Based Spatial Metabolomics: Solvent Composition and Tissue Type as Critical Drivers.

In the development of a desorption electrospray ionization (DESI) workflow for spatial metabolomics, we investigated the impact of two commonly used solvent systems, 90% acetonitrile (ACN) and 90% methanol (MeOH), on the spatial metabolomic profiling of various murine tissues. The performance of both solvents was evaluated across several metabolite classes (central carbon metabolites, amino acids, and fatty acids). Although the ACN-based solvent system led to higher signal intensities for small polar metabolites involved in glycolysis, the tricarboxylic acid (TCA) cycle, and amino acid metabol

Glucose and fatty acids catabolism during in vitro decidualization of human endometrial stromal cells.

The differentiation of endometrial stromal cells, named decidualization, is essential for the proper formation of the materno-fetal interphase. One important feature of decidualization is the increased glucose consumption and its utilization by endometrial cells to produce energy. Besides glucose, fatty acids are another important energy source for living cells and it has been described that endometrial stromal cells rely on the proper function of the oxidation of fatty acids for the correct decidualization. It is, however, unknown whether the turn-over of fatty acid degradation is modified du

Toxoplasma gondii DNA methyltransferases regulate parasitic energy metabolism.

The intracellular protozoan Toxoplasma gondii results in serious diseases such as encephalitis, and retinochoroiditis in immunocompromised patients. The interconversion between tachyzoites and bradyzoites under the host's immune pressure results in the interchange of acute infection and chronic infection. We previously reported two functional DNA methyltransferases (DNMT) in Toxoplasma gondii named TgDNMTa and TgDNMTb. In this research, proteomics analysis for T. gondii tachyzoites of ME49 WT, dnmta knockout (ME49-∆Tgdnmta), and dnmtb knockout (ME49-∆Tgdnmtb) strains, revealed 362 significantl

Gene expression analysis in mitochondria from chagasic mice: alterations in specific metabolic pathways.

Cardiac hypertrophy and remodelling in chagasic disease might be associated with mitochondrial dysfunction. In the present study, we characterized the cardiac metabolic responses to Trypanosoma cruzi infection and progressive disease severity using a custom-designed mitoarray (mitochondrial function-related gene array). Mitoarrays consisting of known, well-characterized mitochondrial function-related cDNAs were hybridized with 32P-labelled cDNA probes generated from the myocardium of mice during immediate early, acute and chronic phases of infection and disease development. The mitoarray succe

BACKGROUND: Heroin addiction and withdrawal have been associated with an increased risk for infectious diseases and psychological complications. However, the changes of metabolites in heroin addicts during withdrawal remain largely unknown. METHODS: A total of 50 participants including 20 heroin addicts with acute abstinence stage, 15 with protracted abstinence stage and 15 healthy controls, were recruited. We performed metabolic profiling of plasma samples based on ultraperformance liquid chromatography coupled to tandem mass spectrometry to explore the potential biomarkers and mechanisms of

Metabolic studies of the edematous cerebral cortex of the pyrithiamine-treated thiamine-deficient rat.

The pyrithiamine-treated rats produced by our method histopathologically exhibited the edematous lesion not only around the third ventricle and at the tegmentum of pons, as expected, but also in the cerebral cortex. These animals were, therefore, used in a biochemical study of the cerebral cortex, in particular, glucose metabolism. The results indicate an accumulation of lactate and severe impairment of the lipid metabolism in the edematous lesion of the cerebral cortex among the pyrithiamine-treated rats. The fact that oxygen consumption was hardly diminished, however, strongly suggested that

Modulating tris-acid integrated proton-confined motifs for efficient uranium extraction from natural seawater.

Uranium predominantly exists as the anionic [UO2(CO3)3]4- complex in seawater; conventional uranium adsorbents cannot displace carbonate ligands from the uranyl core via competitive coordination, resulting in inherently low uranium extraction efficiency. Herein, we report a class of unprecedented porous adsorbents featuring tris-acid (triphosphonate/trisulfonate/tricarboxylate) integrated proton-confined motifs. In contrast to traditional sorbents, the incorporated tris-acid moieties suppress proton release through extensive hydrogen-bonding networks, enabling the formation of protonated urany

Metabolomic landscape of macrophage discloses an anabolic signature of dengue virus infection and antibody-dependent enhancement of viral infection.

Dengue virus (DENV) infection causes dengue fever, the most prevalent arthropod-transmitted viral disease worldwide. Viruses are acellular parasites and obligately rely on host cell machinery for reproduction. Previous studies have indicated metabolomic changes in endothelial cell models and sera of animal models and patients with dengue fever. To probe the immunometabolic mechanism of DENV infection, here, we report the metabolomic landscape of a human macrophage cell model of DENV infection and its antibody-dependent enhancement. DENV infection of THP-1-derived macrophages caused 202 metabol

In vitro synthesis of the iron-molybdenum cofactor of nitrogenase from iron, sulfur, molybdenum, and homocitrate using purified proteins.

Biological nitrogen fixation, the conversion of atmospheric N2 to NH3, is an essential process in the global biogeochemical cycle of nitrogen that supports life on Earth. Most of the biological nitrogen fixation is catalyzed by the molybdenum nitrogenase, which contains at its active site one of the most complex metal cofactors known to date, the iron-molybdenum cofactor (FeMo-co). FeMo-co is composed of 7Fe, 9S, Mo, R-homocitrate, and one unidentified light atom. Here we demonstrate the complete in vitro synthesis of FeMo-co from Fe(2+), S(2-), MoO4(2-), and R-homocitrate using only purified

Glycine Betaine-Induced Metabolic Responses Under Heat and Cold Stress in Passiflora edulis f. flavicarpa.

Temperature extremes represent a major constraint for the cultivation of yellow passion fruit (Passiflora edulis Sims f. flavicarpa), a tropical crop increasingly exposed to heat waves and chilling events under climate change. Glycine betaine (GB) is a widely studied osmoprotectant in plants, yet its influence on metabolic responses of passion fruit under contrasting temperature stresses remains poorly characterized. This study investigated the effects of exogenous GB on primary metabolite profiles of passion fruit seedlings subjected to heat (25, 35, and 45 °C) and cold (25, 15, and 5 °C) con
