Metallomics and other omics approaches in antiparasitic metal-based drug research.

Potential use of metal complexes in medicine is an area of bioinorganic chemistry that has gained much interest. High-throughput omics approaches can provide in-depth insights into the mechanism of action of new metal-based compounds. Discovering new metallodrugs against Trypanosoma cruzi is an emerging field. Combining metallomics, proteomics, and transcriptomics allows the identification of multiple molecular targets and several parasitic metabolic pathways affected by V(IV), Pt(II), and Pd(II) potential antiparasitic drugs. Specifically, metallomics studies with Pd(II) and Pt(II) analogous

Isolation and Total Synthesis of Kinenzoline, an Antitrypanosomal Linear Depsipeptide Isolated from a Marine Salileptolyngbya sp. Cyanobacterium.

Kinenzoline (1), a new linear depsipeptide, was isolated from a marine Salileptolyngbya sp. cyanobacterium. Its structure was elucidated by spectroscopic analyses and degradation reactions. In addition, we achieved a total synthesis of 1 and confirmed its structure. Kinenzoline (1) showed highly selective antiproliferative activity against the causative organism of sleeping sickness, Trypanosoma brucei rhodesiense (IC50 4.5 μM), compared to normal human cells (WI-38, IC50 > 100 μM). Kinenzoline (1) is a promising lead compound for the development of new antitrypanosomal drugs.

Ethyl Pyruvate Emerges as a Safe and Fast Acting Agent against Trypanosoma brucei by Targeting Pyruvate Kinase Activity.

BACKGROUND: Human African Trypanosomiasis (HAT) also called sleeping sickness is an infectious disease in humans caused by an extracellular protozoan parasite. The disease, if left untreated, results in 100% mortality. Currently available drugs are full of severe drawbacks and fail to escape the fast development of trypanosoma resistance. Due to similarities in cell metabolism between cancerous tumors and trypanosoma cells, some of the current registered drugs against HAT have also been tested in cancer chemotherapy. Here we demonstrate for the first time that the simple ester, ethyl pyruvate,

Assessing the efficacy of three trypanocidal drugs in controlling Trypanosoma evansi.

Trypanosoma evansi is an emerging zoonotic threat causing significant economic and health issues, especially in tropical and subtropical regions. Due to limited effective trypanocidal agents and increasing drug resistance, this investigation sought to evaluate the therapeutic efficacy of quinapyramine sulfate and chloride, melarsamine hydrochloride, and diminazene aceturate in 72 female Swiss albino mice. Each drug was administered at two dosages: quinapyramine (3 and 6 mg/kg), melarsamine hydrochloride (0.25 and 0.5 mg/kg), and diminazene aceturate (3.5 and 7 mg/kg). The efficacy was evaluate

N6-modification of 7-Deazapurine nucleoside analogues as Anti-Trypanosoma cruzi and anti-Leishmania agents: Structure-activity relationship exploration and In vivo evaluation.

Chagas disease and leishmaniasis are two poverty-related neglected tropical diseases that cause high mortality and morbidity. Current treatments suffer from severe limitations and novel, safer and more effective drugs are urgently needed. Both Trypanosoma cruzi and Leishmania are auxotrophic for purines and absolutely depend on uptake and assimilation of host purines. This led us to successfully explore purine nucleoside analogues as chemotherapeutic agents against these and other kinetoplastid infections. This study extensively explored the modification of the 6-amino group of tubercidin, a n

Advances and challenges in the search for new treatments for Chagas disease.

The treatment of Chagas disease (CD) has relied for more than five decades on two drugs, benznidazole (BZ) and nifurtimox (NTX), both with significant limitations and severe adverse effects. Their limited efficacy during the chronic phase underscores the urgent need for new chemotherapeutic strategies. The pursuit of new therapeutic agents for CD is focused on identifying molecular targets essential for Trypanosoma cruzi survival that are either absent or highly divergent in human, thereby enhancing selectivity and minimising off-target toxicity. These targets exploit the parasite's unique bio

Trypanosoma evansi: Molecular Characterization and Validation of a Cathepsin-L-like Peptidase as a Potential Therapeutic Target.

C1A cysteine peptidases (CPs) are major contributors to host-parasite crosstalk and have been proposed as anti-trypanosomal chemotherapeutic targets. Trypanosoma evansi is the most widespread animal trypanosome of African origin causing the trypanosomosis known as "Surra" or "derrengadera". Gelatin zymography combined with peptidase class-specific inhibitors identified four CPs in T. evansi lysates with relative masses of 28, 31, 36 and 40 kDa (p28, p31, p36 and p40). The CPs present in the T. evansi lysate preferentially hydrolyzed the Z-Phe-Arg-AMC fluorogenic substrate over Z-Gly-Arg-AMC an

Macrocyclic skeletal modification approach to the anti-trypanosomal macrolides, actinoallolides.

Herein, we report the construction of 16-membered macrocycles that were designed as intermediates toward the unified synthesis of actinoallolides. Key to our synthesis was the use of Mitsunobu macrocyclization, followed by a sequence of Birch reduction and oxidative C-C cleavage to edit the macrocycle.

Polyamine Metabolism for Drug Intervention in Trypanosomatids.

Neglected tropical diseases transmitted by trypanosomatids include three major human scourges that globally affect the world's poorest people: African trypanosomiasis or sleeping sickness, American trypanosomiasis or Chagas disease and different types of leishmaniasis. Different metabolic pathways have been targeted to find antitrypanosomatid drugs, including polyamine metabolism. Since their discovery, the naturally occurring polyamines, putrescine, spermidine and spermine, have been considered important metabolites involved in cell growth. With a complex metabolism involving biosynthesis, ca

Evolution, function and roles in drug sensitivity of trypanosome aquaglyceroporins.

Aquaglyceroporins (AQPs) are membrane proteins that function in osmoregulation and the uptake of low molecular weight solutes, in particular glycerol and urea. The AQP family is highly conserved, with two major subfamilies having arisen very early in prokaryote evolution and retained by eukaryotes. A complex evolutionary history indicates multiple lineage-specific expansions, losses and not uncommonly a complete loss. Consequently, the AQP family is highly evolvable and has been associated with significant events in life on Earth. In the African trypanosomes, a role for the AQP2 paralogue, in

Screening and Identification of Pathogen Box® Compounds with anti-Trypanosoma evansi Activity.

The development of new drugs targeting neglected animal diseases is imperative. In Asia and South America, Trypanosoma evansi is a pathogen that affects horses and other species, causing economic losses associated with reduced animal productivity and death. In order to accelerate the identification of drugs with activity against neglected diseases, Medicines for Malaria Venture has developed Pathogen Box®, a library of 400 different molecules. The present work aimed to identify compounds present in the Pathogen Box® library, measuring in vitro activity against T. evansi. Among the 400 compound

Water-soluble ruthenium complexes bearing activity against protozoan parasites.

Parasitic illnesses are major causes of human disease and misery worldwide. Among them, both amebiasis and Chagas disease, caused by the protozoan parasites, Entamoeba histolytica and Trypanosoma cruzi, are responsible for thousands of annual deaths. The lack of safe and effective chemotherapy and/or the appearance of current drug resistance make the development of novel pharmacological tools for their treatment relevant. In this sense, within the framework of the medicinal inorganic chemistry, metal-based drugs appear to be a good alternative to find a pharmacological answer to parasitic dise

[Control of human African trypanosomiasis: back to square one].

The natural history of sleeping sickness is cyclic. The first epidemic outbreak in the 19th century devastated the population and resolved spontaneously for lack of victims. Intensive development during the colonial period and the movement of population that it spawned led to another epidemic in the early 1920s that reached such severe proportions that drastic steps had to be taken. At that time, Jamot was given complete political, administrative, and financial freedom to combat the disease. This program led to the development of the mobile team concept and so-called vertically structured vect

Immunomodulatory potential of Sarcophaga argyostoma larval hemolymph as a natural alternative to berenil in treating Trypanosoma evansi in vivo.

This study compared effects of diminazene aceturate (berenil), commonly used to treat domestic animals infected with Trypanosoma evansi, with the hemolymph of Sarcophaga argyostoma larva. The hemolymph may be acting as a possible natural alternative to berenil, based on immunomodulation mediated inflammatory response. Inflammatory mediators and histopathological changes in liver, kidney, and spleen of albino mice experimentally infected with T. evansi were studied. Mice were divided into five groups: G1, uninfected, untreated (negative control); G2, T. evansi infected (positive control); G3, i

Single-dose pentamidine substantially reduces viability of trypanosomes in human East African trypanosomiasis.

Examination of viability of trypomastigotes before and after single-dose pentamidine treatment demonstrated that single-dose pentamidine substantially affected motility of trypomastigotes, a proxy of drug efficacy. This suggests that single-dose pentamidine may be of added value to bridge time until suramin will be available for treatment of human East Africa trypanosomiasis.

Trypanosoma cruzi Malic Enzyme Is the Target for Sulfonamide Hits from the GSK Chagas Box.

Chagas disease, an infectious condition caused by Trypanosoma cruzi, lacks treatment with drugs with desired efficacy and safety profiles. To address this unmet medical need, a set of trypanocidal compounds were identified through a large multicenter phenotypic-screening initiative and assembled in the GSK Chagas Box. In the present work, we report the screening of the Chagas Box against T. cruzi malic enzymes (MEs) and the identification of three potent inhibitors of its cytosolic isoform (TcMEc). One of these compounds, TCMDC-143108 (1), came out as a nanomolar inhibitor of TcMEc, and 14 new
