Catalytic activity of trypsin entrapped in electrospun poly(ϵ-caprolactone) nanofibers.

Trypsin was successfully entrapped in situ into nanofibers of poly(ϵ-caprolactone) (PCL) prepared by electrospinning. The spinning dope was an emulsion consisting of an aqueous phase with the solubilized enzyme in a pH buffer plus an oil phase of the polymer solubilized in chloroform (CF)/dimethylformamide (DMF). The optimized materials were composed by random arrays of bead-free fibers with outer diameters in the range 110-180 nm without showing core-shell structure. The fiber size and morphology, membrane porosity and surface properties were shown to be influenced by the polymer concentratio

Urinary trypsin inhibitor reduced inflammatory response after stent injury in minipig.

This study investigated whether urinary trypsin inhibitor (UTI) inhibits neointimal formation by reducing inflammatory response after stent injury. Twenty minipigs having undergone oversized bare material stent implantation in the left anterior descending artery were randomly subdivided into two groups: a UTI group (n=10) and a control group (n=10). Two systemic markers of inflammation (serum macrophage chemoattractant protein-1 and interleukin-6 levels measured by ELISA) were increased after stent implantation, and two days after stem implantation, their levels were positively correlated with

Trypsin-stabilized fluorescent gold nanocluster for sensitive and selective Hg2+ detection.

We report on trypsin-stabilized fluorescent gold nanoclusters (Au NCs) for the sensitive and selective detection of Hg(2+) ions. The Au NCs have an average size of 1 nm and show a red emission at 645 nm. The photostable properties of the trypsin-stabilized Au NCs were examined, and their photochemical stability was found to be similar to that of CdSe quantum dots. The fluorescence was particularly quenched by Hg(2+), and therefore the Au NCs can be used as fluorescent sensors for sensitive and selective Hg(2+) detection to a detection limit of 50 ± 10 nM and the quantitative detection of Hg(2+

Trypsin and thrombin accelerate aggregation of human endocrine pancreas precursor cells.

Human islet-derived precursor cells (hIPCs) and human pancreatic ductal carcinoma (PANC-1) cells can be induced to form aggregates that subsequently differentiate into hormone-expressing islet-like cell aggregates (ICAs). We show that challenge of hIPCs or PANC-1 cells with thrombin or trypsin resulted in stimulation of signaling via the inositol-tris-phosphate second messenger pathway leading to rapid, transient increases in cytosolic calcium ion concentration in the majority of the cells. Because we found that hIPCs, PANC-1 cells, human fetal pancreas, and human adult islets express two prot

Trypsin-like activity and thromboxane release in adult respiratory distress syndrome.

Plasmatic immunoreactive trypsin (IRT), thromboxane and trypsin-like enzymatic activity were measured in 117 patients at risk of developing adult respiratory distress syndrome (ARDS) (53 multiple injury, 30 abdominal surgery, 17 acute pancreatitis, 12 burnt and 5 disseminated intravascular coagulation patients). 69 of these patients developed ARDS. Immunoreactive trypsin and thromboxane were measured by radio-immuno-assay and trypsin-like enzymatic activity by spectrophotometry, using a specific chromogenic substrate. Mean IRT value was 675 ng/ml in ARDS and 265 ng/ml in non ARDS patients (p l

Riboflavin-induced photo-ATRP electrochemical strategy for detection of biomarker trypsin.

The detection of trypsin and its inhibitors is important for both clinical diagnosis and disease treatment. Abnormal trypsin activity affects pancreatic function and leads to corresponding pathological changes in the body. Therefore, the study presented a riboflavin-induced photo-ATRP electrochemical assay of trypsin activity and its inhibitor, including detection of trypsin activity in real urine samples. Experiments were performed on indium tin oxide (ITO) electrodes modified with sulfhydryl groups of 3-mercaptopropionic acid, and target trypsin-specific cleavage of BSA-Au nanocluster (BSA-A

Characterization of succinimide stability during trypsin digestion for LC-MS analysis.

LC-MS peptide mapping is the most commonly used method to analyze protein modifications. The proteins are generally digested using trypsin at a slightly basic pH at 37 °C from several hours to overnight. Assay-induced artifacts can be generated during this procedure, potentially causing false-positive or false-negative results for a given modification. Unfortunately, for the analysis of succinimide, both false-negative and false-positive results can be generated within the same procedure. This study evaluates the stability of succinimide during the peptide mapping procedure and has demonstrate

Artificial insemination using trypsin-treated sperm in the silkworm, Bombyx mori.

A new method was developed for artificial insemination of the silkworm. Virgin females artificially inseminated by this method oviposited fertilized eggs at a rate almost similar to that of females mated with males. Using inactive sperm collected from the seminal vesicles, we confirmed the previous finding by Omura, S., 1936a, Artificial insemination of Bombyx mori, Journal of Faculty of Agriculture, Hokkaido Imperial University 38, 135-150, and Omura, S., 1938, Studies of the reproductive system of the male of Bombyx mori II, Post-testicular organs and post-testicular behaviour of the spermat

Gastrin and cholecystokinin levels in rats fed soya bean trypsin inhibitor.

The effect of dietary soya bean trypsin inhibitor (SBTI) on rat pancreas, stomach, duodenum and two gastrointestinal hormones (gastrin and cholecystokinin (CCK] levels was investigated over a 21 day period. Rats were fed either a complete diet containing 17% protein as casein (control animals), or the same diet supplemented with 1.1% SBTI (test animals). After 21 days the weight of the pancreas in the test group was 90% higher than that in the controls. The stomach antrum, and duodenum showed no change. The gastrin content in the antrum in fasted rats increased after SBTI feeding whereas plasm

Immediate prediction of post-ERCP pancreatitis: a novel approach using serum trypsin.

BACKGROUND: As trypsin activation occurs early in the pathogenesis of pancreatitis, evaluating serum trypsin (TRY) may allow immediate prediction of post-ERCP pancreatitis (PEP). This study aims to evaluate the diagnostic performance of TRY measurement for predicting PEP. METHODS: This multicenter prospective observational study was conducted at 12 tertiary centers in Japan. Serum TRY, amylase (AMY), pancreatic-type AMY (P-AMY), and lipase (LIP) were measured before ERCP, immediately post-procedure (0 h), and 2 h post-procedure. Predictive performances of pancreatic enzymes for PEP were evalu

Trypsin Activates Human Platelets via Proteolytic Cleavage of Protease-Activated Receptor 4.

Human platelets express protease-activated receptor (PAR) 1 and 4 on the platelet membrane. PAR1 activation is crucial for hemostasis, while PAR4 activation produces a prolonged pro-inflammatory platelet response. Once activated, the PAR receptors cause platelet activation and aggregation. The PARs are activated by proteolytic cleavage of their N-terminus, revealing a new N-terminus called the tethered ligand. This tethered ligand folds back and activates the receptor. PAR1 and PAR4 differ in their tethered ligand sequence as well as their signaling cascades; however, they are both activated b

Trypsin-induced aggregation of transthyretin Valine 30 variants associated with hereditary amyloidosis.

Amyloid fibrils of transthyretin (TTR) consist of full-length TTR and C-terminal fragments starting near residue 50. However, the molecular mechanism underlying the production of the C-terminal fragment remains unclear. Here, we investigated trypsin-induced aggregation and urea-induced unfolding of TTR variants associated with hereditary amyloidosis. Trypsin strongly induced aggregation of variants V30G and V30A, in each of which Val30 in the hydrophobic core of the monomer was mutated to less-bulky amino acids. Variants V30L and V30M, in each of which Val30 was mutated to bulky amino acids, a

Dietary wheat amylase trypsin inhibitors exacerbate CNS inflammation in experimental multiple sclerosis.

OBJECTIVE: Wheat has become a main staple globally. We studied the effect of defined pro-inflammatory dietary proteins, wheat amylase trypsin inhibitors (ATI), activating intestinal myeloid cells via toll-like receptor 4, in experimental autoimmune encephalitis (EAE), a model of multiple sclerosis (MS). DESIGN: EAE was induced in C57BL/6J mice on standardised dietary regimes with defined content of gluten/ATI. Mice received a gluten and ATI-free diet with defined carbohydrate and protein (casein/zein) content, supplemented with: (a) 25% of gluten and 0.75% ATI; (b) 25% gluten and 0.19% ATI or

The Inter-α-Trypsin Inhibitor Family: Versatile Molecules in Biology and Pathology.

Inter-α-trypsin inhibitor (IαI) family members are ancient and unique molecules that have evolved over several hundred million years of vertebrate evolution. IαI is a complex containing the proteoglycan bikunin to which heavy chain proteins are covalently attached to the chondroitin sulfate chain. Besides its matrix protective activity through protease inhibitory action, IαI family members interact with extracellular matrix molecules and most notably hyaluronan, inhibit complement, and provide cell regulatory functions. Recent evidence for the diverse roles of the IαI family in both biology an

Juvenile hormone affects the splicing of Culex quinquefasciatus early trypsin messenger RNA.

The full length of Culex quiquefasciatus early trypsin has been cloned and sequenced and a three-dimensional (3D) model of the enzyme was built showing that the enzyme has the canonical trypsin's active pocket containing H78, D123, S129, and D128. The biosynthesis of juvenile hormone (JH) III by the corpora allata (CA) in female Cx. quiquefasciatus is sugar-dependent. Females that were maintained on water after emergence synthesize very little JH III, JH III bisepoxide, and methyl farnesoate (MF) (3.8, 1.1, and 0.8 fmol/4 hr/CA, respectively). One hour after sugar feeding, the synthesis of JH

Human airway trypsin-like protease exerts potent, antifibrotic action in pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is characterized by the deposition of excessive extracellular matrix and the destruction of lung parenchyma, resulting from an aberrant wound-healing response. Although IPF is often associated with an imbalance in protease activity, the mechanisms underlying the sustained repair mechanisms are not fully understood. Here, we addressed the role of the recently identified, membrane-anchored serine protease human airway trypsin-like protease (HAT). In the present study, we show that both HAT expression and activity were up-regulated in human IPF specimens. Next,

SdPI, the first functionally characterized Kunitz-type trypsin inhibitor from scorpion venom.

BACKGROUND: Kunitz-type venom peptides have been isolated from a wide variety of venomous animals. They usually have protease inhibitory activity or potassium channel blocking activity, which by virtue of the effects on predator animals are essential for the survival of venomous animals. However, no Kunitz-type peptides from scorpion venom have been functionally characterized. PRINCIPAL FINDINGS: A new Kunitz-type venom peptide gene precursor, SdPI, was cloned and characterized from a venom gland cDNA library of the scorpion Lychas mucronatus. It codes for a signal peptide of 21 residues and

Bauhinia variegata var. variegata trypsin inhibitor: from isolation to potential medicinal applications.

Here we report for the first time of a new Kunitz-type trypsin inhibitor (termed BvvTI) from seeds of the Camel's foot tree, Bauhinia variegata var. variegata. BvvTI shares the same reactive site residues (Arg, Ser) and exhibits a homology of N-terminal amino acid sequence to other Bauhinia protease inhibitors. The trypsin inhibitory activity (K(i), 0.1 x 10(-9)M) of BvvTI ranks the highest among them. Besides anti-HIV-1 reverse transcriptase activity, BvvTI could significantly inhibit the proliferation of nasopharyngeal cancer CNE-1 cells in a selective way. This may partially be contributed
