Prevalence of chronic wasting disease and bovine tuberculosis in free-ranging deer and elk in South Dakota.

Heads of hunter-harvested deer and elk were collected throughout South Dakota (USA) and within established chronic wasting disease (CWD) surveillance areas from 1997-2002 to determine infection with CWD and bovine tuberculosis (TB). We used immunohistochemistry to detect CWD-infected individuals among 1,672 deer and elk sampled via geographically targeted surveillance. A total of 537 elk (Cervus elaphus nelsoni), 813 white-tailed deer (Odocoileus virginianus), and 322 mule deer (O. hemionus) was sampled for CWD. Estimated overall prevalence and associated confidence intervals (95%) in white-ta

Vaccinating wild badgers against bovine tuberculosis: an overview of 16 years of vaccine delivery in England.

INTRODUCTION: The European badger (Meles meles) is wildlife host for Mycobacterium bovis (bovine tuberculosis) in the UK and several European countries, complicating efforts to control the disease in livestock. Badger vaccination using the injectable BadgerBCG vaccine has been available in England since 2010 as a non-lethal tool to reduce TB transmission in badger populations and reduce risks to cattle. Despite long-standing interest, national deployment has historically been limited, and questions remain about operational feasibility. METHODS: Here we provide a comprehensive overview of 16 y

Strategy for Improving the Diagnostic Efficacy of Bovine Tuberculosis Using a Novel PCRBIO/DIG-ELISA Assay.

Bovine Tuberculosis (BTb) is an important zoonosis caused by Mycobacterium bovis. In Argentina, the Official National Control Program mandates cattle slaughter when a positive Tuberculin Skin Test Single (TSTS) is observed. Herds that test negative twice a year are considered infection-free; however, many false negatives have been observed in anergic cattle. In our previous study, 16% of herds with TSTS-positive animals were found to be negative by the Polymerase Chain Reaction Touch Down of Insertion Sequence 6110 (PCRTD-IS6110) for the detection of Mycobacterium bovis in dairy milk. To optim

Bovine tuberculosis in cattle slaughtered at Addis Ababa abattoir in Ethiopia and workforce awareness of zoonotic risk.

BACKGROUND: Bovine tuberculosis (bTB) is endemic and of zoonotic importance in Ethiopia. Despite this, there is limited recent information on the prevalence of bTB in cattle slaughtered at abattoirs. This study aimed to estimate the prevalence of bTB in cattle slaughtered at Addis Ababa municipality abattoir based on tuberculous lesions and region of difference (RD4) deletion typing and to assess the current practice and the awareness of occupational workers to zoonoses. METHODS: A total of 502 cattle slaughtered at the municipality abattoir (260 in the day shift and 242 in the night shift) w

Genotype Diversity of Mycobacterium bovis and Pathology of Bovine Tuberculosis in Selected Emerging Dairy Regions of Ethiopia.

Bovine tuberculosis (bTB) is endemic in Ethiopia with higher prevalence in cattle, particularly in the central parts. Spread of Mycobacterium bovis (M. bovis) to wider regions is inevitable in uncontrolled conditions. This study was conducted to explore the pathology, characterize M. bovis strains, and describe genotypic diversity to demonstrate possible epidemiological links in emerging dairy areas of Ethiopia, namely, Mekelle and Gondar. Twenty-seven bTB positive cattle identified by the Single Intradermal Comparative Cervical Tuberculin (SICCT) test were subjected to post-mortem inspection

Display of Antigens on Polyester Inclusions Lowers the Antigen Concentration Required for a Bovine Tuberculosis Skin Test.

The tuberculin skin test is the primary screening test for the diagnosis of bovine tuberculosis (TB), and use of this test has been very valuable in the control of this disease in many countries. However, the test lacks specificity when cattle have been exposed to environmental mycobacteria or vaccinated with Mycobacterium bovis bacille Calmette-Guérin (BCG). Recent studies showed that the use of three or four recombinant mycobacterial proteins, including 6-kDa early secretory antigenic target (ESAT6), 10-kDa culture filtrate protein (CFP10), Rv3615c, and Rv3020c, or a peptide cocktail derived

Farmers' views and attitudes towards bovine tuberculosis and bluetongue in Northern Ireland using semi-structured interviews.

BACKGROUND: Understanding the attitude of farmers towards infectious diseases and government interventions is essential for understanding behavioural responses during outbreaks, compliance with control measures, and preventing and eradicating diseases. OBJECTIVES: This study aimed to improve the understanding of farmers’ attitudes towards bluetongue (BT) and bovine tuberculosis (bTB), which are key epizootic and endemic diseases in Northern Ireland, and provide insight into how this may affect disease control and eradication. METHODOLOGY: Short answer question slips, each containing a single q

Mycobacterium bovis infected domestic cats in an officially bovine tuberculosis free country resulting in human infection.

Although the Netherlands is an officially bovine tuberculosis (bTB) free (OTF) country, sporadic infections with Mycobacterium bovis still cause tuberculosis (TB) in (non-bovine) mammals, including humans. We describe for the first time cases of M. bovis infection in domestic cats in the Netherlands with transmission between companion animals and humans. In January 2023, a domestic cat, euthanized due to severe respiratory clinical signs, was diagnosed with M. bovis. Subsequently, three other cats from the household were euthanized and also diagnosed with M. bovis. The remaining kitten and dog

A DIVA-compatible Mycobacterium bovis triple mutant vaccine confers protection against bovine tuberculosis in mouse model.

Bovine tuberculosis (bTB) is a pulmonary infectious disease caused by Mycobacterium bovis, affecting cattle and a wide range of mammals, including humans. Despite its significant impact on global livestock production, no commercial vaccine is currently available, partly due to potential interference with standard diagnostic tests. In this study, we evaluated the protective efficacy of a triple M. bovis mutant lacking the immunodominant antigens ESAT-6 and CFP-10, as well as the virulence factor Ag85A. This mutant is compatible with DIVA (Differentiation of Infected from Vaccinated Animals) dia

Abattoir surveillance: Identifying risk factors associated with bovine tuberculosis lesion detection in a low prevalence region.

Abattoir surveillance is a key component in control and eradication programs against bovine tuberculosis (bTB). In low-prevalence or officially tuberculosis-free (OTF) regions in which active surveillance is typically limited or non-existent, postmortem inspection at the abattoir constitutes one of the main diagnostic tool to guarantee the absence of disease transmission. Here, we evaluated the performance of abattoir postmortem inspection in Galicia, a low-prevalence region in Spain (now OTF). Between 2014 and 2019, 1,784,261 animals were culled in 41 abattoirs, of which a small proportion (0

Once bitten twice shy: Risk factors associated with bovine tuberculosis recurrence in Castilla y Leon, Spain.

Persistence of bovine tuberculosis (bTB) in cattle herd remains a major challenge in disease elimination due to the ineffectual removal of all infected animals in a bTB breakdown. Characterization of herds with a higher probability of experiencing further bTB breakdowns can help to implement specific risk-based policies for disease control and eradication. Here, our aim was to identify herd- and breakdown-level risk factors in bTB infected herds in Castilla y Leon, Spain, associated with a decreased time to recurrence and an increased risk of recurrence using a mixed effects Cox proportional h

Bovine Tuberculosis Case Intervention Using the T.SPOT.TB Assay to Screen Dairy Workers in Bailey County, Texas.

Background: One potential exposure on a dairy farm is Mycobacterium bovis or bovine tuberculosis (bTB)-an infectious zoonotic pathogen. The prevalence of tuberculosis among dairy workers in the U.S. is unknown largely due to insufficient surveillance and testing practices. Our objective was to determine the prevalence and risk factors of LTBI among dairy workers potentially exposed to cattle infected with bTB in two Bailey County, Texas dairy farms in 2016. Methods: This study involved a secondary analysis of data that were collected by Texas Department of State Health Services (DSHS) Public H

A DNA prime-Mycobacterium bovis BCG boost vaccination strategy for cattle induces protection against bovine tuberculosis.

The variable efficacy of bacillus Calmette-Guérin (Mycobacterium bovis BCG) in protecting humans and cattle against tuberculosis has prompted a search for a more effective vaccination regimen. A prime-boost strategy was investigated in cattle naturally sensitized to environmental mycobacteria by using a combination of three DNA vaccines coding for Hsp 65, Hsp 70, and Apa for priming, followed by a boost with BCG prior to experimental challenge with virulent M. bovis. Controls were vaccinated with DNA or BCG alone or were not vaccinated. The immune responses were monitored throughout the study,

Field Trial with Vaccine Candidates Against Bovine Tuberculosis Among Likely Infected Cattle in a Natural Transmission Setting.

Background/Objectives: Vaccines may improve the control and eradication of bovine tuberculosis. However, the evaluation of experimental candidates requires the assessment of the protection, excretion, transmission and biosafety. A natural transmission trial among likely infected animals was conducted. Methods: Seventy-four male heifers were randomly distributed (five groups) and vaccinated subcutaneously with attenuated strains (M. bovis Δmce2 or M. bovis Δmce2-phoP), a recombinant M. bovis BCG Pasteur (BCGr) or M. bovis BCG Pasteur. Then, they cohoused with a naturally infected bTB cohort und

Evaluation of five serologic assays for bovine tuberculosis surveillance in domestic free-range pigs from southern Spain.

In countries where bovine tuberculosis (bTB) is still prevalent the contact among different animal species in extensive systems contributes to the circulation of Mycobacterium bovis (M. bovis) and other members of the Mycobacterium tuberculosis complex (MTC). Thus, free-range pigs can develop subclinical infections and may contribute to disease spread to bovine and wildlife. Serodiagnosis has been proposed as a screening tool for detecting infected pig herds; however, the value of this method to obtain an accurate diagnosis in this species is still not clear. In this study, sensitivity (Se) an

Bovine tuberculosis in South Darfur State, Sudan: an abattoir study based on microscopy and molecular detection methods.

Bovine tuberculosis (BTB) is a widespread zoonosis in developing countries but has received little attention in many sub-Saharan African countries including Sudan and particularly in some parts such as Darfur states. This study aimed to detect bovine tuberculosis among caseous materials of cattle slaughtered in abattoirs in South Darfur State, Sudan by using microscopic and PCR-based methods. The study was a cross-sectional abattoir-based study which examined a total of 6,680 bovine carcasses for caseous lesions in South Darfur State between 2007 and 2009. Collected specimens were examined for

Vaccination of cattle with Mycobacterium bovis culture filtrate proteins and interleukin-2 for protection against bovine tuberculosis.

In this study vaccines prepared from culture filtrate proteins (CFP) of Mycobacterium bovis and interleukin-2 (IL-2) were tested in cattle for their capacity to stimulate immune responses and to protect against an intratracheal challenge with virulent M. bovis. Nine groups of cattle were vaccinated with combinations of different doses of CFP and bovine IL-2 mixed with a monophosphoryl lipid A (MPL) adjuvant. An additional group was vaccinated with M. bovis BCG. Immune responses in b1P-IL-2-vaccinated animals differed from those seen in BCG-vaccinated animals by inducing high antigen-specific a

Incidence and nature of human tuberculosis due to bovine tubercle bacilli in South-East England: 1977-1987.

A total of 201 new cases of tuberculosis due to bovine tubercle bacilli was confirmed in South-East England between 1977 and 1987 inclusive. This represents about 1% of all cases of tuberculosis in this region. Most cases occurred amongst older individuals of indigenous white British origin, although some younger patients of Southern European and Indian subcontinent ethnic origin were also diagnosed. The lung was the most frequent site of disease, followed by the genito-urinary tract. In view of the known risk of transmission of disease from man to cattle via the respiratory and urinary tracts
