Transgenic soybean overexpressing GmSAMT1 exhibits resistance to multiple-HG types of soybean cyst nematode Heterodera glycines.

Soybean (Glycine max (L.) Merr.) salicylic acid methyl transferase (GmSAMT1) catalyses the conversion of salicylic acid to methyl salicylate. Prior results showed that when GmSAMT1 was overexpressed in transgenic soybean hairy roots, resistance is conferred against soybean cyst nematode (SCN), Heterodera glycines Ichinohe. In this study, we produced transgenic soybean overexpressing GmSAMT1 and characterized their response to various SCN races. Transgenic plants conferred a significant reduction in the development of SCN HG type 1.2.5.7 (race 2), HG type 0 (race 3) and HG type 2.5.7 (race 5).

The plant apoplasm is an important recipient compartment for nematode secreted proteins.

Similarly to microbial pathogens, plant-parasitic nematodes secrete into their host plants proteins that are essential to establish a functional interaction. Identifying the destination of nematode secreted proteins within plant cell compartment(s) will provide compelling clues on their molecular functions. Here the fine localization of five nematode secreted proteins was analysed throughout parasitism in Arabidopsis thaliana. An immunocytochemical method was developed that preserves both the host and the pathogen tissues, allowing the localization of nematode secreted proteins within both org

Wimmera rye grass toxicity in Western Australia.

Outbreaks of Wimmera rye grass toxicity in the south-west of Western Australia, and the associated distribution of Wimmera rye grass parasitised by Anguina sp and Corynebacterium sp were investigated. Between 1970 and 1974, 58 outbreaks occurred on 26 farms in sheep and cattle grazing pasture, cereal crop, stubble, screenings, or hay containing parasitised Wimmera rye grass. Morbidity rates up to 77% were observed. Clinical signs were characterised by staggering, collapse, periods of convulsions and other nervous signs, often followed by death. A pale, friable liver and haemorrhages in differe

Organic acids induce plant defense responses and suppress root-knot nematodes in tomato via molecular mechanisms and molecular docking insights.

Root-knot nematodes (Meloidogyne spp.) are among the most destructive pathogens affecting tomato production worldwide, leading to severe yield losses and compromised plant health. This study evaluated the efficacy of six organic acids-benzoic, salicylic, citric, lactic, malic, and acetic acids-applied at three concentrations each, in managing Meloidogyne incognita infection in tomato cultivar "023."Among all treatments, salicylic acid (SA) exhibited the strongest nematicidal activity, achieving 100% juvenile mortality within 72 hours and the highest inhibition of egg hatchability, even at the

N6-Methyladenosine (m6A) Sequencing Reveals Heterodera glycines-Induced Dynamic Methylation Promoting Soybean Defense.

Unraveling the intricacies of soybean cyst nematode (Heterodera glycines) race 4 resistance and susceptibility in soybean breeding lines-11-452 (highly resistant) and Dongsheng1 (DS1, highly susceptible)-was the focal point of this study. Employing cutting-edge N6-methyladenosine (m6A) and RNA sequencing techniques, we delved into the impact of m6A modification on gene expression and plant defense responses. Through the evaluation of nematode development in both resistant and susceptible roots, a pivotal time point (3 days postinoculation) for m6A methylation sequencing was identified. Our seq

Genome Expression Dynamics Reveal the Parasitism Regulatory Landscape of the Root-Knot Nematode Meloidogyne incognita and a Promoter Motif Associated with Effector Genes.

Root-knot nematodes (genus Meloidogyne) are the major contributor to crop losses caused by nematodes. These nematodes secrete effector proteins into the plant, derived from two sets of pharyngeal gland cells, to manipulate host physiology and immunity. Successful completion of the life cycle, involving successive molts from egg to adult, covers morphologically and functionally distinct stages and will require precise control of gene expression, including effector genes. The details of how root-knot nematodes regulate transcription remain sparse. Here, we report a life stage-specific transcript

Plant Volatiles Reduce the Viability of the Root-Knot Nematode Meloidogyne incognita Either Directly or When Retained in Water.

Volatile organic compounds (VOC) produced by green residues for the management of plant-parasitic nematodes are poorly studied for oilseed plants and some Brassica spp. To investigate the activity of VOC in vitro and as biofumigants, dry and aqueous macerates of broccoli (Brassica oleracea var. italica) shoots and sunflower (Helianthus annuus) seed were used against the root-knot nematode Meloidogyne incognita. VOC produced by sunflower seed caused higher mortality of M. incognita second-stage juveniles (J2) than VOC produced by broccoli shoots but both plant species were equally effective in

Systemic changes in photosynthesis and reactive oxygen species homeostasis in shoots of Arabidopsis thaliana infected with the beet cyst nematode Heterodera schachtii.

Photosynthetic efficiency and redox homeostasis are important for plant physiological processes during regular development as well as defence responses. The second-stage juveniles of Heterodera schachtii induce syncytial feeding sites in host roots. To ascertain whether the development of syncytia alters photosynthesis and the metabolism of reactive oxygen species (ROS), chlorophyll a fluorescence measurements and antioxidant responses were studied in Arabidopsis thaliana shoots on the day of inoculation and at 3, 7 and 15 days post-inoculation (dpi). Nematode parasitism caused an accumulation

The novel GrCEP12 peptide from the plant-parasitic nematode Globodera rostochiensis suppresses flg22-mediated PTI.

Multiple myeloma has a continued need for more effective and durable therapies. B cell maturation antigen (BCMA), a plasma cell surface antigen and member of the tumor necrosis factor (TNF) receptor superfamily, is an attractive target for immunotherapy of multiple myeloma due to its high prevalence on malignant plasma cells. The current work details the pre-clinical evaluation of BCMA expression and development of a chimeric antigen receptor (CAR) targeting this antigen using a fully human single chain variable fragment (scFv). We demonstrate that BCMA is prevalently, but variably expressed b

Essential Oil of Angelica archangelica Roots as a Sustainable Alternative for Management of Meloidogyne incognita.

Bioactive essential oil (EO) based nanoemulsions (NEs) exhibit superior stability and bioactivity over conventional formulations, thereby emerging as a promising strategy for efficient management of phyto-parasitic nematodes. The present investigation emphasized the development of eco-benign botanical nematicide from EO of Angelica archangelica using nanotechnological interventions. GC-MS analysis of the EO revealed abundance of limonene (18.39%), vinylguaiacol (14.10%), and citronellol acetate (15.91%). Nanoemulsification of the volatile oil was performed using ultrasonication, followed by ch

Root-knot-nematode-derived mimics of RGF peptides hijack host signalling to orchestrate feeding site formation.

Root-knot nematodes establish long-term parasitic relationships with diverse hosts by inducing specialized feeding cells. However, the molecular mechanisms by which nematodes manipulate this developmental reprogramming process remain largely unknown. Here we identify a class of ROOT MERISTEM GROWTH FACTOR (RGF)-like peptide effectors conserved in root-knot nematodes. MgRGF from Meloidogyne graminicola and MiRGF1 from M. incognita are expressed in subventral gland cells during early infection and secreted into the host apoplast. Functional analysis reveals that nematode RGFs are critical for fe

The root-knot nematode effector Mi1D08B targets a NINJA-family co-repressor to suppress jasmonate production and promote infection in soybean.

Plant-parasitic nematodes deploy secreted effector proteins that reprogram host cellular processes to establish parasitism. Here, we characterized Mi1D08B, a putative dorsal-gland effector from the root-knot nematode Meloidogyne incognita and defined its role in subverting soybean jasmonate-mediated immunity. Mi1D08B is conserved across several Meloidogyne species and localizes to the plant nucleus and cytoplasm. Overexpression of Mi1D08B significantly increased galling and egg production, demonstrating a strong virulence role. A gall-specific yeast two-hybrid screen identified the NINJA-famil

RNA tomography reveals spatial gene expression maps of Arabidopsis thaliana roots infected with Heterodera schachtii.

Plant-parasitic cyst nematodes, such as Heterodera schachtii, cause substantial crop losses world-wide and induce specialized feeding structures in host roots, yet the molecular mechanisms underlying feeding structure initiation and development remain poorly understood. We introduce RNA tomography for plants, a powerful untargeted spatial transcriptomics technology that allows studying gene expression at high spatial resolution. We applied RNA tomography to Arabidopsis (Arabidopsis thaliana) roots infected with H. schachtii, capturing 96 consecutive cross sections of 20 micrometers at 1- and 2

Aboveground herbivory induced jasmonates disproportionately reduce plant reproductive potential by facilitating root nematode infestation.

Different plant feeders, including insects and parasitic nematodes, can influence each other by triggering systemic changes in their shared host plants. In most cases, however, the underlying mechanisms are unclear, and the consequences for plant fitness are not well understood. We studied the interaction between leaf feeding Manduca sexta caterpillars and root parasitic nematodes in Nicotiana attenuata. Simulated M. sexta attack increased the abundance of root parasitic nematodes in the field and facilitated Meloidogyne incognita reproduction in the glasshouse. Intact jasmonate biosynthesis w

Yellow Pigment Aurovertins Mediate Interactions between the Pathogenic Fungus Pochonia chlamydosporia and Its Nematode Host.

Nematophagous fungi are globally distributed soil fungi and well-known natural predators of soil-dwelling nematodes. Pochonia chlamydosporia can be found in diverse nematode-suppressive soils as a parasite of nematode eggs and is one of the most studied potential biological control agents of nematodes. However, little is known about the functions of small molecules in the process of infection of nematodes by this parasitic fungus or about small-molecule-mediated interactions between the pathogenic fungus and its host. Our recent study demonstrated that a P. chlamydosporia strain isolated from

Identification of proteins expressing differences among isolates of Meloidogyne spp. (Nematoda: Meloidogynidae) by nano-liquid chromatography coupled to ion-trap mass spectrometry.

Total protein variation (up to ninety-five different positions) was revealed by two-dimensional electrophoresis (2-DE) in 18 isolates from populations of M. arenaria (6 isolates), M. incognita (10), M. javanica (1) plus an unclassified isolate in a previously reported study. Isolates of M. arenaria, M. javanica, Meloidogyne sp., and M. incognita formed two separate groups defined on the basis of two sets of protein positions that could be considered as diagnostic characters, but we could not identify these proteins by MALDI-TOF. To identify these marker positions, nano-liquid chromatography as

Enhancing Plant Immunity and Mitigating Environmental Risk: A Chitosan Oligosaccharide-Functionalized Nanoplatform for Targeted Delivery of Fluopyram.

To balance pesticide efficacy and ecological safety, a synergistic delivery system (FLU@MSNs/CMCO) was constructed by grafting chitosan oligosaccharide (COS) onto mesoporous silica nanoparticles (MSNs) to encapsulate the hydrophobic pesticide fluopyram (FLU). This cationic nanosystem (131 nm) achieved a 35.0% loading capacity and sustained release. It orchestrates a "dual-attack" mechanism, the cationic surface promotes electrostatic targeting on nematode cuticles and fungal cell walls, while the COS shell acts as an immune elicitor, enhancing plant root chitinase activity and vigor.
