By using a new member of the neurotropic equine herpesviruses, EHV-9, which induced encephalitis in various species via various routes, an ocular infection model was developed in suckling hamsters. The suckling hamsters were inoculated with EHV-9 via the conjunctival route and were sacrificed after 6, 12, 24, 36, 48, 72, 96, 120, and 144 hours (h) post inoculation (PI). Three horizontal sections of the brains, including the eyes and cranial cavity, were examined histologically to assess the viral kinetics and time-course neuropathological alterations using a panoramic view. At 6 to 24 h PI, th

Horses are hosts to 2 types of gammaherpesviruses, Equid herpesvirus 2 and 5 (EHV-2 and EHV-5, respectively). Both EHV-2 and EHV-5 are common in horses in Iceland. An Icelandic EHV-5 isolate was recovered by sequential culture in primary fetal horse kidney and rabbit kidney cells. Glycoprotein B, glycoprotein H, and DNA terminase genes of the isolate were fully sequenced, and the DNA polymerase gene was partly sequenced. To date, the glycoprotein B gene of EHV-5 was the only gene that has been reported to be completely sequenced in addition to small parts of the glycoprotein H, DNA polymerase,

INTRODUCTION: Felid herpesvirus 1 (FeHV-1) is widespread worldwide and is sometimes associated with chronic diseases in cats; therefore, it is clinically important. Vaccination can be used to control FeHV-1 infection. We previously isolated a strain identical to the live attenuated vaccine (LAV) strain F2 from a feline eye with a dendritic ulcer, indicating that the F2 strain may be pathogenic. The origin of the F2 strain that we previously isolated is unknown, and we surmised that the F2 strain may exist in the field. To determine whether the LAV strain F2 exists in the field, we, therefore,

Effective vaccines against sexually transmitted pathogens must elicit protective immunity at mucosal surfaces, particularly within the female reproductive tract. Caprine herpesvirus 1 (CpHV-1) causes genital disease, reproductive failure, and neonatal mortality in goats and provides a biologically relevant large-animal model for human genital herpesvirus infection. Here, we evaluated a mucosal vaccination strategy based on a bovine herpesvirus 4 (BoHV-4) vector expressing the CpHV-1 immunodominant glycoprotein D (BoHV4-A-gD(cp)gD(106)ΔTK). Goats were immunized via the intravaginal or intranasa

This study investigated the prevalence of upper respiratory tract disease (URTD) pathogens in domestic cats during the SARS-CoV-2 pandemic in southern Brazil, focusing on the detection of feline herpesvirus-1 (FHV-1), feline calicivirus (FCV), Mycoplasma felis, Chlamydia felis, and SARS-CoV-2. A total of 120 cats presenting URTD symptoms and with recent contact (within 15 days) with SARS-CoV-2-positive tutors were analyzed. The detection rates were 38.3% for FHV-1, 34.1% for FCV, 43.3% for M. felis, 19.16% for C. felis, and 1.2% for SARS-CoV-2. Co-infections were common, affecting 64.45% of ca

BACKGROUND: The feline viral rhinotracheitis, calicivirus, and panleukopenia (FVRCP) vaccine, prepared from viruses grown in the Crandell-Rees feline kidney cell line, can induce antibodies to cross-react with feline kidney tissues.
OBJECTIVES: This study surveyed the prevalence of autoantibodies to feline kidney tissues and their association with the frequency of FVRCP vaccination.
METHODS: Serum samples and kidneys were collected from 156 live and 26 cadaveric cats. Antibodies that bind to kidney tissues and antibodies to the FVRCP antigen were determined by enzyme-linked immunosorbent ass

Feline herpesvirus-1 (FHV-1) infection occurs worldwide and is a leading cause of respiratory and ocular diseases in cats. Current vaccines reduce the severity of symptoms but do not prevent infection and, therefore, do not provide defense against an establishment of latency and reactivation. We hypothesize that immunomodulation of FHV-1 is the cause of lack in protection and that deletion of virulence/immune modulatory genes of FHV-1 will enhance safety and immunogenicity. Our objective was to use feline respiratory epithelial cell (FREC) cultures to define in vitro growth characteristics and

Although famciclovir is efficacious in feline herpesvirus type 1 (FHV-1)-infected cats, effects of a single dose early in disease course have not been reported. In this two part, randomized, masked, placebo controlled study, cats received a single dose of 125 mg famciclovir (n = 43) or placebo (n = 43; pilot study), or 500 mg famciclovir (n = 41) or placebo (n = 40; clinical trial) on entering a shelter. FHV-1 PCR testing was performed, bodyweight and food intake were recorded, and signs of respiratory disease were scored prior to and 7 days following treatment. FHV-1 DNA was detected in 40% o

UNLABELLED: Like varicella-zoster virus (VZV), simian varicella virus (SVV) reactivates to produce zoster. In the present study, 5 rhesus macaques were inoculated intrabronchially with SVV, and 5 months later, 4 monkeys were immunosuppressed; 1 monkey was not immunosuppressed but was subjected to the stress of transportation. In 4 monkeys, a zoster rash developed 7 to 12 weeks after immunosuppression, and a rash also developed in the monkey that was not immunosuppressed. Analysis at 24 to 48 h after zoster revealed SVV antigen in the lung alveolar wall, in ganglionic neurons and nonneuronal ce

Alphaherpesvirus reactivation from thoracic sympathetic ganglia (TSG) and transaxonal spread to target organs cause human visceral disease. Yet alphaherpesvirus latency in TSG has not been well characterized. In this study, quantitative PCR detected varicella-zoster virus (VZV), herpes simplex virus 1 (HSV-1), and HSV-2 DNA in 117 fresh TSG obtained postmortem from 15 subjects. VZV DNA was found in 76 (65%) ganglia from all subjects, HSV-1 DNA was found in 5 (4%) ganglia from 3 subjects, and no HSV-2 was found.

Ganglia of monkeys with reactivated simian varicella virus (SVV) contained more CD8 than CD4 T cells around neurons. The abundance of CD8 T cells was greater less than 2 months after reactivation than that at later times and correlated with that of CXCL10 RNA but not with those of SVV protein or open reading frame 61 (ORF61) antisense RNA. CXCL10 RNA colocalized with T-cell clusters. After SVV reactivation, transient T-cell infiltration, possibly mediated by CXCL10, parallels varicella zoster virus (VZV) reactivation in humans.

Primary infection with varicella zoster virus (VZV) results in varicella (more commonly known as chickenpox) after which VZV establishes latency in sensory ganglia. VZV can reactivate to cause herpes zoster (shingles), a debilitating disease that affects one million individuals in the US alone annually. Current vaccines against varicella (Varivax) and herpes zoster (Zostavax) are not 100% efficacious. Specifically, studies have shown that 1 dose of varivax can lead to breakthrough varicella, albeit rarely, in children and a 2-dose regimen is now recommended. Similarly, although Zostavax result

Varicellovirus bovinealpha 1 (BoAHV-1) is a significant pathogen responsible for respiratory disease in cattle, capable of inducing lung damage independently or co-infection with bacteria. The widespread spread of BoAHV-1 in cattle herds has caused substantial economic losses to the cattle industry. The pathogenic mechanisms of BoAHV-1 are often relevant to robust inflammatory responses, increased oxidative burden, and the initiation of apoptosis. Glycyrrhizin (GLY) is a small-molecule triterpenoid saponin compound obtained from the herb liquorice, which has a broad spectrum of pharmacological

Caprine herpesvirus 1 (CpHV-1) infection in goats induces genital vesicular-ulcerative lesions that strictly resemble those produced by human herpesvirus 2 in humans. In previous studies, the potent inhibition of CpHV-1 by cidofovir was demonstrated. Cidofovir antiherpetic activity was evaluated in goats infected experimentally by the vaginal route with CpHV-1 and then treated locally at different times after infection. The administration of 1% cidofovir cream onto vaginal mucosa was able to prevent the onset of genital lesions and to decrease significantly the titers of the virus shed by the

BACKGROUND: Feline panleukopenia, feline calicivirus infection, feline viral rhinotracheitis, and feline infectious peritonitis are significant diseases that threaten feline health. The trend of mixed infections is increasing, and current diagnostic methods are limited in scope and unable to provide rapid, simultaneous detection of these diseases.
METHODS: Four groups of primers and probes targeting the VP2 gene of Feline Panleukopenia virus (FPV), the TK gene of Feline Herpesvirus (FHV-1), the ORF2 gene of Feline Calicivirus (FCV), and the N gene of Feline Infectious Peritonitis Virus (FIPV)

In this study we examined the effects of non-myeloablative total body irradiation (TBI) in combination with immunosuppressive chemotherapy on immune homeostasis in rhesus macaques. Our results show that the administration of cyclosporin A or tacrolimus without radiotherapy did not result in lymphopenia. The addition of TBI to the regimen resulted in lymphopenia as well as alterations in the memory/naive ratio following reconstitution of lymphocyte populations. Dendritic cell (DC) numbers in whole blood were largely unaffected, while the monocyte population was altered by immunosuppressive trea

Feline calicivirus (FCV) and feline herpesvirus 1 (FHV-1) are major pathogens threatening feline health, and prophylactic immunity remains the primary preventive approach. To develop a safe and effective multivalent vaccine against the two viruses, two recombinant rabies virus (RABV) vaccines expressing the FCV VP1 and FHV-1 gB/gD genes were constructed. The exogenous genes showed stable inheritance following serial viral passages, and the recombinant RABVs exhibited growth kinetics comparable to the parental strain, with a delayed peak titer. After verifying the viral pathogenicity, the recom
