Extreme venom variation in Middle Eastern vipers: a proteomics comparison of Eristicophis macmahonii, Pseudocerastes fieldi and Pseudocerastes persicus.

UNLABELLED: Venoms of the viperid sister genera Eristicophis and Pseudocerastes are poorly studied despite their anecdotal reputation for producing severe or even lethal envenomations. This is due in part to the remote and politically unstable regions that they occupy. All species contained are sit and wait ambush feeders. Thus, this study examined their venoms through proteomics techniques in order to establish if this feeding ecology, and putatively low levels of gene flow, have resulted in significant variations in venom profile. The techniques indeed revealed extreme venom variation. This

Neonate-to-adult transition of snake venomics in the short-tailed pit viper, Gloydius brevicaudus.

UNLABELLED: Snake venoms undergo ontogenetic shifts in biochemical and pharmacological activities. This may be related to variation in venom components associated with the ontogenetic shift in diet. We used the short-tailed pit viper Gloydius brevicaudus that displays ontogenetic shifts in diet to examine whether the species displays a neonate-to-adult transition of snake venomics. Venoms from neonates and adults were pooled separately and then analyzed by 2-DE, MALDI-TOF-MS/MS and iTRAQ technologies. The 2-DE profiles showed that the main components in both types of venoms were acidic protein

[Adder bites in France].

Snake bites are a major public health problem in the tropics but they have a low incidence in Europe and are responsible for few deaths each year. The incidence is higher in children than in adults but no difference in severity seems to be observed between children and adults. In France, snake envenomations are due mainly to Vipera aspis and Vipera berus. The clinical presentation is usually limited to a local syndrome with pain and local inflammatory edema, but systemic signs occur in 17% of cases. Clinical grading published by the Institut Pasteur in Paris helps to assess the severity of env

Molecular phylogeny of Vipera Laurenti, 1768 and the related genera Macrovipera (Reuss, 1927) and Daboia (Gray, 1842), with comments about neurotoxic Vipera aspis aspis populations.

We used mtDNA sequences (cytochrome b and NADH dehydrogenase subunit 2) to reconstruct molecular phylogenies of Vipera sensu lato, Vipera sensu stricto, and Vipera aspis. Three major clades were identified within the Vipera s.l. group: (1) the European vipers, (2) the oriental vipers, consisting of Montivipera (Vipera 2) plus Macrovipera lebetina, and (3) a group of Asian and North African vipers consisting of Daboia russelii, V. palaestinae, and Macrovipera mauritanica. We also distinguished three clades within the monophyletic European Vipera group: V. ammodytes, V. aspis, and V. latastei, a

Antitumoral potential of Tunisian snake venoms secreted phospholipases A2.

Phospholipases type A2 (PLA2s) are the most abundant proteins found in Viperidae snake venom. They are quite fascinating from both a biological and structural point of view. Despite similarity in their structures and common catalytic properties, they exhibit a wide spectrum of pharmacological activities. Besides being hydrolases, secreted phospholipases A2 (sPLA2) are an important group of toxins, whose action at the molecular level is still a matter of debate. These proteins can display toxic effects by different mechanisms. In addition to neurotoxicity, myotoxicity, hemolytic activity, antib

Molecular cloning of serine proteases from elapid snake venoms.

Serine proteases are widely distributed in viperid snake venoms, but rare in elapid snake venoms. Previously, we have identified a fibrinogenolytic enzyme termed OhS1 from the venom of Ophiophagus hannah. The results indicated that OhS1 might be a serine protease, but there was no structural evidence previously. In the present study, the primary structure of OhS1 was determined by protein sequencing, in combination with RT-PCR and 5'-RACE methods. OhS1 precursor is composed of an 18-amino acid signal peptide, a 6-amino acid putative activation peptide and 236-amino acid mature protein. OhS1 ho

Diet and snake venom evolution.

Venom composition within snake species can show considerable geographical variation, an important consideration because bites by conspecific populations may differ in symptomatology and require different treatments. The underlying causes of this phenomenon have never been explained. Here we present evidence that the variation in the venom of the pitviper Calloselasma rhodostoma (Serpentes: Viperidae) is closely associated with its diet. We also evaluated other possible causes of geographic variation in venom using partial Mantel tests and independent contrasts, but rejected both contemporary g

A new species of Hepatozoon in the common lancehead snake (Bothrops atrox) from the Eastern Amazonia region.

Although venomous snakes from the family Viperidae, such as Bothrops atrox, are recognized for their medical importance due to snakebite accidents, few studies on parasitological aspects have been carried out with them, especially in the Amazonia region. Using morphological and molecular tools, we described a novel haemogregarine species infecting the common lancehead snake B. atrox from Eastern Amazonia, Brazil. Hepatozoon atrocis sp. nov. has mature gamonts that are morphologically distinct from those reported in the literature, which are often compact, with dispersed or encapsulated cytopla

Snake antivenom production in Ecuador: Poor implementation, and an unplanned cessation leads to a call for a renaissance.

Snakebite envenomation is a global health problem. This health problem asymmetrically affects rural populations in developing countries to such an extent that it recently has been listed as a priority neglected tropical disease (NTD). It is estimated that 5.4 million individuals are bitten by snakes each year, causing at least 2.7 million envenomations and more than 100,000 deaths each year. Ecuador has one of the highest snakebite envenomation incidence rates in Latin America, mostly in the coastal and Amazonian provinces. Envenomations in these regions are the result of bites primarily by sp

Local envenoming by the Schokari sand racer, Psammophis schokari Forskål, 1775 (Serpentes, Psammophiidae) and a brief review of reported bites by sand racers (Psammophis spp.).

A recent case of a bite by a psammophiid snake, Psammophis schokari, is described and analyzed. This is the first report of local envenoming by this species. The 1 m long P. schokari inflicted a protracted bite on the third digit, right hand of the male 59 year-old victim who developed mild, but locally progressive edema and persistent pain; full resolution required almost three months. All documented cases of bites by snakes of the genus Psammophis are briefly reviewed and discussed. Finally, we encourage the use of a standardized method to describe the observed symptoms of bites by non-front

Amino acid sequence and homology modeling of obtustatin, a novel non-RGD-containing short disintegrin isolated from the venom of Vipera lebetina obtusa.

Disintegrins represent a group of cysteine-rich peptides occurring in Crotalidae and Viperidae snake venoms, and are potent antagonists of several integrin receptors. A novel disintegrin, obtustatin, was isolated from the venom of the Vipera lebetina obtusa viper, and represents the first potent and selective inhibitor of the binding of integrin alpha(1)beta(1) to collagen IV. The primary structure of obtustatin contains 41 amino acids and is the shortest disintegrin described to date. Obtustatin shares the pattern of cysteines of other short disintegrins. However, in contrast to known short d

Venomous bites and stings in the tropical world.

Snakes of the families Viperidae and Elapidae are responsible for the high incidence of morbidity and mortality after snake bites in countries of West Africa, the Indian subcontinent, South-East Asia, New Guinea and Latin America. Envenoming can cause local effects, notably tissue necrosis; and systemic effects, including paralysis, haemostatic disturbances, shock, increased capillary permeability, myocardial damage, rhabdomyolysis and acute renal failure. Specific hyperimmune serum (antivenom) is the mainstay of medical treatment for severe envenoming. Ancillary treatments such as assisted ve

Characteristics and significance of "green snake" bites in Myanmar, especially by the pit vipers Trimeresurus albolabris and Trimeresurus erythrurus.

Snakebite is an important problem in Myanmar. Regionally, bites by Eastern Russell's vipers, Daboia siamensis (Viperidae, Viperinae), and monocled cobras, Naja kaouthia are considered medically important, but those categorised as "green snake" bites are not. However, these may include bites by green pit vipers, Trimeresurus spp. (Viperidae, Crotalinae) for which no antivenom is available in Myanmar. Elsewhere in Southeast Asia, these snakes are reported to cause local and systemic envenoming. As part of the Myanmar Snakebite Project, prospective case data were collected over 3 years from five

Comparative Proteomic and Peptidomic Analysis of Venom From Three Viper Taxa: Evaluating Software-Specific Protein and Peptide Profiles.

Snake venom is increasingly recognized in biomedical research as a potential source of relevant proteins that are still relatively unknown in various species. In this experiment, we performed proteomic and peptidomic quantification and identification of the venomic profile of three viper taxa: Montivipera blugardaghica subsp. bulgardaghica (MB), Vipera ammodytes subsp. montandoni (VA), and Vipera kaznakovi (VK); and compared the performance of three peptide identification software: PEAKS, MaxQuant, and Proteome Discoverer. Overall, PEAKS identified 19 unique proteins (19 in MB, 11 in VA, and 1

Proteomic and functional analyses of Lachesis acrochorda snake venom from the Valle del Cauca Department of Colombia.

Lachesis acrochorda envenomation has a lethality rate of approximately 90%. Despite its high lethality, little is known about its local and systemic effects and its relationship with its protein content. Thus, to increase knowledge of L. acrochorda snake venom from the Southwestern ecoregion of Colombia, we developed a proteomic analysis using a "bottom-up shotgun proteomic profiling" approach. Besides, we evaluated toxinological properties and compared the effects with the Bothrops asper snake venom activities. The RP-HPLC profile showed similarities with the L. acrochorda snake venom from th

Biochemistry and pharmacology of proteins and peptides purified from the venoms of the snakes Macrovipera lebetina subspecies.

The isolation and characterization of individual snake venom components is important for a deeper understanding of the pathophysiology of envenomations, for improving the therapeutic procedures of patients, and it also opens possibilities for the discovery of novel toxins that might be useful as tools for understanding cellular and molecular processes. This review provides a summary of the different toxins that have been isolated and characterized from the venoms of Vipera lebetina (Macrovipera lebetina) subspecies Macrovipera lebetina cernovi, Macrovipera lebetina lebetina, Macrovipera lebeti

Cardiotoxic effects of the Vipera ammodytes ammodytes venom fractions in the isolated perfused rat heart.

The nose-horned viper (Vipera ammodytes ammodytes) is the most venomous European snake. Its venom is known as haematotoxic, myotoxic and neurotoxic but it exerts also cardiotoxic effects. To further explore the cardiotoxicity of the venom we separated it into four fractions by gel filtration chromatography. Three fractions that contain polypeptides (A, B, and C) were tested for their effects on isolated rat heart. Heart rate (HR), incidence of arrhythmias (atrioventricular (AV) blocks, ventricular tachycardia, ventricular fibrillation, and asystolia), coronary flow (CF), systolic, developed an

Mechanism of Vipera berus venom activity and the principles of antivenom administration in treatment.

In the present paper, the actual knowledge on the composition and toxic properties of the European common viper venom was analyzed. The organism response to the particular components and the methods of neutralization of venom toxin in bitten person were presented. On the basis of literature data, the clinical course of envenomation with its classification according to the severity of symptoms was characterized. In the paper the situations in which administration of antivenom is required to neutralize toxic properties of venom and its possible adverse reactions were also described.

Envenoming following bites by the Balkan adder Vipera berus bosniensis - first documented case series from Bulgaria.

We report the first detailed accounts of bites by the Balkan adder, Vipera berus bosniensis from Bulgaria. Documentation of bites by this subspecies is very rare in the literature and most available accounts are from the northern limit of its distribution. V. berus bosniensis is considered to possess neurotoxic venom but little evidence has hitherto been available to support this supposition. In this case series symptoms typical of adder bites developed including oedema, nausea, dizziness, lymphangitis, vomiting, and diarrhoea together with aberrant symptoms such as diplopia and ptosis that co

In Slovak Republic and the region of Central and Eastern Europe the viper is the only species of venomous snake living in wild. During the last nine observed years (1993-2002) the Toxicological Information Center (TIC) was consulted in 81 cases of individuals attacked by snakes. 51 of these cases were by viper, rattlesnake 6, and green mamba once. 23 cases were by non-venomous snakes; adder 22 times and royal snake once. The viper and adder attacks occurred in the wild and the other cases with snake keepers. None of affected individuals died. Our contribution contains the new recommended thera
