Wuchereria bancrofti: detection of microfilariae in asymptomatic microfilaremic individuals with Setaria digitata antigens.

A dot-ELISA for detection of microfilariae of Wuchereria bancrofti in an endemic area was developed. This test can differentiate the endemic normals from the microfilaraemic asymptomatic individuals. Antigens of molecular weight 130 and 52 kDa of the cattle filaria worm Setaria digitata were used for this test. It was observed that these two antigens were also present in the serum of asymptomatic microfilaraemic individuals.

Experimental transmission of nocturnally periodic Wuchereria bancrofti to Indian leaf monkey (Presbytis entellus).

Successful experimental transmission of the human lymphatic dwelling nocturnal periodic strain of Wuchereria bancrofti has been achieved from man to Indian leaf monkey (Presbytis entellus) through a susceptible strain of Aedes aegypti. The prepatent period varied between 195 and 240 days. Microfilaria (mf) levels were in general low and the peak count was attained at 1800 hr. Of the adult worms 5-12.5% were recovered from male langur while females revealed poor (2.54%) recovery. This host--parasite model thus makes available the target parasite material in substantial quantity to work on chemo

Factors affecting transmission of Wuchereria bancrofti by anopheline mosquitoes. 1. Uptake of microfilariae.

Ingestion of Wuchereria bancrofti microfilariae (mf) from humans by 639 Anopheles gambiae, 557 An. arabiensis, 117 An. melas and 9 An. funestus was investigated. The mf densities in blood fell into 3 groups; 0-32mf/ml, 107-122 mf/ml and 421-1140 mf/ml. In An. gambiae and An. arabiensis percentage of mosquitoes ingesting mf was strongly associated with mf density in host blood; in An. melas the association was much weaker. Mean number of mf ingested per mosquito was also strongly correlated to mf density in An. gambiae and An. arabiensis but not in An. melas. At low mf densities both An. gambia

Ultrastructure of the anterior alimentary tract of infective-stage Wuchereria bancrofti (Nematoda: Filarioidea).

The anterior alimentary tract of infective-stage Wuchereria bancrofti is divided into the following segments: stoma or buccal capsule, muscular esophagus, glandular esophagus, esophageal-intestinal valve, and intestine. Invaginated external cuticle lines only the anterior stoma. External cuticle and esophageal lining are not continuous and are ultrastructurally distinct; the latter is compared morphologically to the amorphous component of elastin. The glandular esophagus is a composite structure of a stellate contractile epithelial core, surrounded by a sleeve of secretory epithelium. The glan

Sex ratio of Wuchereria bancrofti in surgical specimens from an endemic area of Brazil.

The sex ratio of adult Wuchereria bancrofti from 172 histologically diagnosed cases of lymphatic filariasis, all from an endemic area of Recife, in north-eastern Brazil, was investigated. Of 172 tissue specimens examined, 74 (43%) were lymph nodes and 98 (57%) lymphatic-vessel segments. The morphology of the worms was generally disrupted, in some cases to the point where the worms were almost completely absorbed by the granulomatous inflammatory response. Parasite gender was clearly determined in only 110 (64%) specimens, 61 (55.5 %) of which were lymph nodes and 49 (44.5%) lymphatic vessels.

Setaria cervi: in vitro released collagenases and their inhibition by Wuchereria bancrofti infected sera.

In vitro released products of adult Setaria cervi females, microfilariae and extracts showed considerable amounts of collagenase activity. On the basis of per mg protein released in vitro, the products of both microfilariae and adult females exhibited comparable activity but this was much higher than that of extract of microfilariae and adult females. Two collagenase enzymes with molecular masses of 50 kDa and 70 kDa were separated using DEAE-sepharose CL6B and Sephadex G-100 column chromatography. The 50 kDa and 70 kDa collagenase exhibited pH optima of 5.2 and 7.0, respectively. Considering

Control of the Aedes vectors of the dengue viruses and Wuchereria bancrofti: the French Polynesian experience.

In most of the 130 islands of French Polynesia, the stenotopic mosquitoes Aedes aegypti (the main local vector for the viruses causing dengue) and Aedes polynesiensis (the main local vector of Wuchereria bancrofti) share many breeding sites in water containers such as discarded cans, coconut shells, buckets and water-storage pots and drums. In addition to selective application of insecticides, non-polluting methods of controlling these mosquitoes have been evaluated during the last decade in two main ecological situations: (1) villages, where Aedes breeding sites are typically peridomestic; an

On the uptake of Wuchereria bancrofti microfilariae in vector mosquitoes of different susceptibility to filarial infections.

The quantitative uptake of Wuchereria bancrofti microfilariae by mosquito species of different susceptibility to filarial infection was studied. There was no relationship between the degree of susceptibility and the number of ingested microfilariae. However, in all tested mosquito strains the females ingested as a mean 1.2-1.9 times more microfilariae of W. bancrofti than expected.

The prevalence of naturally acquired multiple infections of Wuchereria bancrofti and human malarias in anophelines.

Malaria and filaria infection rates were determined for anopheline mosquitoes collected whilst biting and resting in village houses in Papua New Guinea. The number of anophelines infected with both parasites was greater than expected from the infection rates of each parasite and this difference was significant in resting collections. The excess of multiply infected mosquitoes is probably a result of a vector population composed of individuals with differing numbers of opportunities to become infected. Malaria-positive Anopheles punctulatus from resting catches had a significantly greater numbe

The susceptibility of five laboratory colonies of mosquitoes to the human nematode Wuchereria bancrofti (Cobbold).

Laboratory colonies of Anopheles gambiae, An. arabiensis, An. merus, An. quadriannulatus and Aedes aegypti formosus were artificially fed on blood containing microfilariae of Wuchereria bancrofti. The anopheline colonies all supported parasite development to the infective stage, with An. quadriannulatus being the most heavily infected. The parasite did not develop at all in the Ae. aegypti formosus colony.

A quantitative analysis of the diagnostic value of diethylcarbamazine provocation in endemic Wuchereria bancrofti infection.

The efficiency (delta) of the day-time provocative effect of diethylcarbamazine (DEC) on the circulation of microfilariae (mf) of nocturnally periodic Wuchereria bancrofti compared with the night mf count was evaluated in terms of sensitivity and specificity. In one test, the mean mf count for finger-blood samples from the same person on 3 successive nights was compared with the day-time post-DEC count; another test compared the mf count in one night-time finger-blood sample with the following day-time post-DEC count. Both tests were efficient (delta = 0.74 and 0.77, S.E. = 0.07 and 0.07, resp

Comparative densities of Wuchereria bancrofti microfilaria in paired samples of capillary and venous blood.

Concurrent finger-prick and venous blood samples were obtained from 43 patients infected with Wuchereria bancrofti. Microfilariae were counted in blood smears and on nuclepore filters. Based on the numbers of microfilariae in 20-microliters finger-prick samples, an expected (theoretical) number of microfilariae in venous samples was calculated. This expected value was compared to the actual number of microfilariae present in venous blood. The actual number of microfilariae present ranged from 0.7 to 30 times (median 3) less than expected. The exponential function y = 16.74e.17x, r2 = .69, wher

[Wuchereria bancrofti: value of the diethylcarbamazine provocation test for the diurnal detection of microfilaria carriers].

Sixty-nine of 83 (83%) patients with circulating microfilariae of nocturnally periodic Wuchereria bancrofti were detected in daytime blood films following the one-time administration of diethylcarbamazine at a dose per patient of 1 +/- .25 mg/kg of body weight. This method of detecting microfilaria carriers in Haiti is recommended as a reasonably sensitive alternative when circumstances make it impossible to conduct a nighttime blood survey.

Immunodiagnostic potential of Wuchereria bancrofti L1 antigen-based filarial immunoglobulin G4 detection assay.

BACKGROUND: After mass drug administration to eliminate human lymphatic filariasis, there is a need for surveillance to detect the measurable endpoint of the program. METHODS: An immunodominant seroreactive clone, WbL1, was identified through immunoscreening of a Wuchereria bancrofti L3 complementary DNA expression library. Recombinant WbL1 (rWbL1) was analysed with sera from W. bancrofti patients. Diagnostic evaluation was carried out by developing an enzyme-linked immunosorbent assay (ELISA) to detect the filarial-specific antibodies in various categories of filarial sera samples against re

Recombinant Wolbachia surface protein (WSP)-induced T cell responses in Wuchereria bancrofti infections.

Human lymphatic filariasis is a debilitating parasitic disease characterized by downregulation of the host's immune response in asymptomatic carriers along with profound hyperreactivity in chronic patients apart from putatively immune endemic normals. The endosymbiont Wolbachia, a bacterium of filarial nematodes has received much attention as possible chemotherapeutic target and its involvement in disease pathogenesis. The role of recombinant Wolbachia surface protein (rWSP), one of the most abundantly expressed proteins of the endosymbiont, in modulating cell-mediated immune responses in pati

Field application of PCR-based assays for monitoring Wuchereria bancrofti infection in Africa.

Approximately 50 million people in Egypt and sub-Saharan Africa have bancroftian filariasis and together they represent about a third of all cases of lymphatic filariasis (LF) world-wide. Currently, the Global Programme to Eliminate Lymphatic Filariasis, which was launched by the World Health Organization (WHO) in 1998, is largely based on repeated annual cycles of mass drug administration (MDA) to endemic populations. Also, some countries, including Egypt, are taking steps to improve LF vector-control interventions, to break the transmission cycle more effectively than is achievable with MDA

Diagnosis of Wuchereria bancrofti infection by the polymerase chain reaction employing patients' sputum.

A preliminary evaluation of the diagnostic potential of a polymerase chain reaction (PCR) assay using diurnally collected sputum from bancroftian filariasis patients is described. A new set of PCR primers amplifying a 254-bp-long sequence termed AccI, derived from a long dispersed repeated sequence and SspI primers previously employed for PCR-based diagnosis were employed in this study with similar results. Of the 34 sputum samples from patients, 32 (94%) were PCR positive. Of the 18 patients with low to high microfilaremia (21-1560 microfilariae/ml), 16 (88.8%) were PCR positive. Of the remai

Acute disease episodes in a Wuchereria bancrofti-endemic area of Papua New Guinea.

Acute disease episodes of Bancroftian filariasis were monitored prospectively in a rural area of Papua New Guinea. The frequency and duration of episodes were recorded for the leg, arm, scrotum, and breast. A very high incidence of acute disease was observed; 0.31 episodes per person-year in the leg alone. Incidence generally increased with age, except in the breast, where episodes were concentrated in the reproductive age range. Males had slightly higher incidence than females in the leg and arm. Chronic disease was strongly associated with acute disease incidence in all locations. Microfilar
