Combined treatment of xyloglucan derivative hydrogel and anti-C5a receptor antibody in preventing peritoneal adhesion.

Postoperative peritoneal adhesion is a common complication after surgery with high morbidity. In addition to improving surgical operations, medical therapy and physical barriers are the two main ways to prevent postoperative peritoneal adhesion. Satisfactory efficacy is not often obtained by the single antiadhesion method, and the combination of barrier therapy and antiadhesion drugs has attracted more attention. In this study, we first demonstrated that aberrant complement activation was associated with peritoneal injury and inflammatory responses. Correspondingly, blocking the C5a-C5aR axis

Relevance, structure and analysis of ferulic acid in maize cell walls.

Phenolic compounds in foods have been widely studied due to their health benefits. In cereals, phenolic compounds are extensively linked to cell wall polysaccharides, mainly arabinoxylans, which cross-link with each other and with other cell wall components. In maize, ferulic acid is the phenolic acid present in the highest concentration, forming ferulic acid dehydrodimers, trimers and tetramers. The cross-linking of polysaccharides is important for the cell wall structure and growth, and may protect against pathogen invasion. In addition to the importance for maize physiology, ferulic acid ha

Advanced structural profiling of arabinoxylans from cereal grains by high performance anion exchange chromatography with parallel pulsed amperometric and mass spectrometric detection.

Arabinoxylans (AX) are major hemicellulosic polysaccharides of cell walls of cereal grains. Potential health benefits and techno-functional properties of AX are determined, among others, by the extent and distribution of arabinofuranose (Araf) units as substituents along the β-(1 → 4)-linked d-xylopyranose (Xylp) backbone. Conventional carbohydrate analytical methods, such as methylation analysis, are time-consuming and often insufficient to capture structural diversity. Therefore, a semi-quantitative profiling method based on enzymatic AX degradation and on high-performance anion-exchange chr

Selective aqueous hemicellulose extraction from hazelnut shells through planetary ball milling.

Hazelnuts are among the most widely consumed nuts, but their shells are notoriously difficult to valorise beyond the generation of electricity and heat. Here, we cracked the hazelnut shell valorisation through planetary ball milling, allowing effective aqueous extraction of hemicellulose. Optimized planetary ball milling showed that 4 h milling allowed 50% w/w hemicellulose extraction (80 °C; 24 h), leaving all cellulose and lignin unextracted. The hemicellulose recovered through ethanol precipitation was characterised as 4-O-methylglucuronoxylan (GX; 96% Xyl; 4% MeGlcA) of high DP (Mw 25 kDa)

Preparation of xyloglucan-grafted poly(N-hydroxyethyl acrylamide) copolymer by free-radical polymerization for in vitro evaluation of human dermal fibroblasts.

Xyloglucan is a rigid polysaccharide that belongs to the carbohydrate family. This hemicellulose compound has been widely used in biomedical research because of its pseudoplastic, mucoadhesive, mucomimetic, and biocompatibility properties. Xyloglucan is a polyose with no amino groups in its structure, which also limits its range of applications. It is still unknown whether grafting hydrophilic monomers onto xyloglucan can produce derivatives that overcome these shortcomings. This work aimed to prepare the first copolymers in which N-hydroxyethyl acrylamide is grafted onto tamarind xyloglucan b

Molecular modification, structure-activity relationship and practical application of wheat bran arabinoxylan: A review.

Wheat bran arabinoxylan (WBAX), a major dietary fiber component in wheat bran, has garnered increasing attention due to its versatile physicochemical properties and potential health benefits, including immunomodulatory, prebiotic, and hypoglycemic effects. Despite its abundance as a by-product of wheat milling, WBAX remains underutilized in food systems. This review systematically summarizes recent progress in the extraction, purification, molecular modification, and structural characterization of WBAX. It further explores the structure-activity relationships underlying its bioactivities, such

Integration of subcritical water extraction and treatment with xylanases and feruloyl esterases maximises release of feruloylated arabinoxylans from wheat bran.

Wheat bran is an abundant and low valued agricultural feedstock rich in valuable biomolecules as arabinoxylans (AX) and ferulic acid with important functional and biological properties. An integrated bioprocess combining subcritical water extraction (SWE) and enzymatic treatments has been developed for maximised recovery of feruloylated arabinoxylans and oligosaccharides from wheat bran. A minimal enzymatic cocktail was developed combining one xylanase from different glycosyl hydrolase families and a feruloyl esterase. The incorporation of xylanolytic enzymes in the integrated SWE bioprocess i

Arabinoxylan from Corn Fiber Obtained through Alkaline Extraction and Membrane Purification: Relating Bioactivities with the Phenolic Compounds.

Arabinoxylan has prebiotic properties, as it is able to resist digestion in the small intestine and undergoes fermentation in the large intestine. In this work, arabinoxylan was extracted from corn fiber using an alkaline solution and further purified with membrane processing. It was found that the extracts were mainly composed of xylose (50-52%), arabinose (37-39%), galactose (9%) and glucose (1-4%), with an A/X ratio of 0.72-0.77. All the extracts were composed of phenolic compounds, including ferulic acid derivatives such as dimers, trimers and tetramers. The purified extract had a lower co

Optimization of the Cellulase Free Xylanase Production by Immobilized Bacillus Pumilus.

BACKGROUND: The extracellular xylanase secreted by microorganisms is a hydrolytic enzyme, which arbitrarily cleaves the β-1, 4 backbone of the polysaccharide xylan; an enzyme used in the food processing, bio-pulping and bio-bleaching. The commercial production of the xylanase is limited because of a higher cost involvement, which can be overcome by the cost-effective production of the xylanase through immobilization of the microbial cell by the non-toxic substances. OBJECTIVES: In this work, the optimization of the extra-cellular cellulase free xylanase production by the immobilized cell of t

Valorization of Lignocellulose with One-Step Acidified Monophasic Phenoxyethanol Fractionation.

Effective fractionation of lignocelluosic biomass and subsequent valorization of all three major components under mild conditions were achieved. Pretreatment with acidified monophasic phenoxyethanol (EPH) efficiently removed 92.6 % lignin and 80 % xylan from poplar at 110 °C in 60 min, yielding high-value EPH-xyloside, EPH-modified lignin (EPHL), and a solid residue nearly purely composed of carbohydrates. After removing the grafted acetyl groups using 1 % NaOH at 50 °C, the highest enzymatic digestibility reached 92.3 %. EPHL could be recovered in high yield and purity with an uncondensed str

Metabolic engineering of Caldicellulosiruptor bescii for 2,3-butanediol production from unpretreated lignocellulosic biomass and metabolic strategies for improving yields and titers.

The platform chemical 2,3-butanediol (2,3-BDO) is used to derive products, such as 1,3-butadiene and methyl ethyl ketone, for the chemical and fuel production industries. Efficient microbial 2,3-BDO production at industrial scales has not been achieved yet for various reasons, including product inhibition to host organisms, mixed stereospecificity in product formation, and dependence on expensive substrates (i.e., glucose). In this study, we explore engineering of a 2,3-BDO pathway in Caldicellulosiruptor bescii, an extremely thermophilic (optimal growth temperature = 78°C) and anaerobic bacte

Enzymatic Hydrolysis of Soybean Hull Without Pretreatment and Its Enhancement of Bioethanol Production Using Xylose-Fermenting Escherichia coli (FBR5).

BACKGROUND: Lignocellulosic materials, such as soybean hulls, possess a complex and recalcitrant structure that requires efficient pretreatment or enzymatic processing for effective conversion into valuable products. However, pretreatment processes often generate inhibitory byproducts (e.g., furfural, hydroxymethyl furfural (HMF), phenols, and lignin degradation products), which can impede enzymatic activity and increase overall production costs. This study explores soybean hulls, a byproduct of oil and meal production, as a potential high-carbohydrate biorefinery resource, assessing their che

Cellulase adsorption and relationship to features of corn stover solids produced by leading pretreatments.

Although essential to enzymatic hydrolysis of cellulosic biomass to sugars for fermentation to ethanol or other products, enzyme adsorption and its relationship to substrate features has received limited attention, and little data and insight have been developed on cellulase adsorption for promising pretreatment options, with almost no data available to facilitate comparisons. Therefore, adsorption of cellulase on Avicel, and of cellulase and xylanase on corn stover solids resulting from ammonia fiber expansion (AFEX), ammonia recycled percolation (ARP), controlled pH, dilute acid, lime, and s

Comparative genomic analysis of hemicellulose-degrading potential in bacterial isolates from the anterior intestine of Eisenia andrei (Bouché, 1972).

Earthworms and their associated microbiota can degrade various types of lignocellulosic residues, but the enzymes, carbohydrate-binding modules, and sugar transporters involved in this process remain imperfectly understood. The present study aimed to identify genes and characterize hemicellulose degradation strategies of bacterial strains with high xylanolytic activity from the earthworm gut. The strains, originally isolated from the anterior intestine of earthworms fed on a diet of palm fiber (Streptomyces thermophilus PF5-2S and Niallia circulans PF7-2S) and coffee husk (Cellulosimicrobium c

Enzymatic Production of Prebiotic Xylooligosaccharides Using a Bacillus pumilus GH30_8 Glucuronoxylanase: Structural Basis of Glucuronoxylan Recognition and Hydrolysis.

Transformation of agro-industrial products into value-added products, such as prebiotic oligosaccharides, is a key element of the emerging bioeconomy. Here, we characterized a new GH30_8 glucuronoxylanase from Bacillus pumilus (BpXyn30_8A) for its potential in producing xylooligosaccharides (XOS). BpXyn30_8A showed tolerance to ethanol and NaCl and released both linear and branched XOS containing MeGlcA at the penultimate nonreducing end residue. Its X-ray structure, determined at 2.16 Å resolution, revealed high similarity to other glucuronoxylanases. Furthermore, BpXyn30_8A achieved higher x
