Transporter-Driven Glycerophosphocholine (GPC) Toxicity Is Conserved from Fission Yeast to Budding Yeast: Roles for Inositol Pyrophosphates and Gde1 Regulation in Fission Yeast.

Glycerophosphocholine (GPC) and glycerophosphoinositol (GPI) are phospholipid metabolites generated by phospholipase-mediated deacylation. In budding yeast, they enter cells via the Git1 permease; in fission yeast, the homolog is Tgp1. This study investigates why GPC is toxic to asp1-STF mutants, where Tgp1 is upregulated due to loss of Asp1 pyrophosphatase, resulting in elevated inositol pyrophosphate 1,5-IP8. We show that S. pombe Tgp1 specifically transports GPC, explaining why GPC, but not GPI, impairs growth. Increased GPC uptake slows doubling time but does not reduce viability. Toxicity

Baker's Yeast Clinical Isolates Provide a Model for How Pathogenic Yeasts Adapt to Stress.

Global outbreaks of drug-resistant fungi such as Candida auris are thought to be due at least in part to excessive use of antifungal drugs. Baker's yeast Saccharomyces cerevisiae has gained importance as an emerging opportunistic fungal pathogen that can cause infections in immunocompromised patients. Analyses of over 1000 S. cerevisiae isolates are providing rich resources to better understand how fungi can grow in human environments. A large percentage of clinical S. cerevisiae isolates are heterozygous across many nucleotide sites, and a significant proportion are of mixed ancestry and/or a

Expression of AtMed15 of Arabidopsis in yeast causes flocculation and increases ethanol production in yeast culture.

Mediator, a multiprotein complex involved in transcription of class II genes, was first discovered in yeast and then characterized in many metazoans revealing a striking structural conservation of the complex. However, sequences of Mediator subunits are not well conserved raising a question on the functional conservation of these individual subunits. In this study, expression of Med15 of Arabidopsis (AtMed15) in gal11∆ yeast could not complement the function of ScGal11 in galactose metabolism and resistance against cycloheximide. Surprisingly, AtMed15 changed the morphology of the yeast cells.

Azole-Driven Cross-Resistance and Transporter Gene Expression in Malassezia Yeasts.

Malassezia are commensal lipid dependent yeasts which can cause opportunistic skin infection. Topical imidazole antifungals such as clotrimazole and ketoconazole are the frontline treatment. However, the tendency of fungal infections to recur, combined with the emergence of multi-azole-resistant Malassezia isolates means that many patients have used these antifungal treatments repeatedly or for extended durations with limited efficacy. While the impact of single azole treatments has been studied, the ability of specific azoles to induce cross-resistance is unclear. Understanding the effect of

Mitochondrial NAD kinase Pos5 is required for CoQ biosynthesis in yeasts.

Coenzyme Q (CoQ) is an essential component of the electron transport chain, and ten genes involved in CoQ biosynthesis have been identified in Schizosaccharomyces pombe. To gain further insight into CoQ biosynthesis, we screened the Bioneer gene-deletion library and found that the Δpos5 strain produced only 0.2-fold of the wild-type CoQ10 level. Pos5 shares homology with Saccharomyces cerevisiae Pos5 (ScPos5), a mitochondrial NADH (or NAD+) kinase that generates NADPH (or NADP+). Heterologous expression of ScPOS5 in the S. pombe Δpos5 strain recovered CoQ content to 0.9-fold of the wild-type l

De novo L-(+)-tartaric acid biosynthesis in multi-modular engineered yeasts.

L-(+)-tartaric acid (L-TA) is a high-value chiral organic acid essential for food and pharmaceuticals. Despite its industrial importance, sustainable green production is constrained by the lack of a fully defined biosynthetic pathway. Here, we report the de novo biosynthesis of L-TA in Saccharomyces cerevisiae through reaction-guided enzyme mining, experimental validation, and Enzyme Commission-specific Catalytic Hybrid Optimizer (ECHO)-assisted enzyme prioritization. We first elucidate the elusive two-step conversion from precursor 5-keto-D-gluconic acid (5-KGA) to L-TA, catalyzed by transket

Biotechnological applications of D-xylose-fermenting autochthonous yeasts in lignocellulosic biorefineries.

The efficient fermentation of D-xylose by yeasts remains a major bottleneck for the sustainable production of second-generation bioethanol, as this pentose represents a substantial fraction of hemicellulosic sugars in lignocellulosic biomass. While conventional industrial yeasts show limited native capacity for D-xylose utilization, numerous autochthonous (native or wild-derived) yeasts isolated from natural environments harbor metabolic traits that have been naturally selected and enable effective pentose assimilation under challenging conditions. Understanding and harnessing this diversity i

Chemical Fingerprints of Honey Fermented by Conventional and Non-Conventional Yeasts.

Previous studies have shown the positive effects of non-conventional Metschnikowia spp. yeasts in mixed cultures with Saccharomyces cerevisiae on the properties of fruit wines. In this study, we investigated the effects of using conventional S. cerevisiae and non-conventional Metschnikowia pulcherrima yeasts as starter cultures in controlled mixed fermentations of honey wort. Other non-conventional yeasts were also tested for comparison, including Wickerhamomyces anomalus, Dekkera/Bretannomyces bruxellensis, and Wickerhamomyces anomalus. We evaluated the tolerance of the tested yeasts to high

Basidiomycete Yeasts of Wine Grapes and Their Potential Applications in Winemaking.

Vineyards support highly diverse communities of native yeasts, but only a small proportion are fermentative ascomycetes capable of alcoholic fermentation. Many non-fermentative species are also present, including a range of metabolically active basidiomycete yeasts that can influence wine aromatic profiles, especially in the early stages of fermentation. In some cases, basidiomycete yeasts, such as Filobasidium, Rhodotorula, Sporobolomyces, and Vishniacozyma, are more abundant and diverse than ascomycete yeasts in grape musts, with some persisting throughout fermentation. As the existing infor

Secondary Metabolites from Food-Derived Yeasts Inhibit Virulence of Candida albicans.

A sparse number of available antifungal drugs, therapeutic side effects, and drug resistance are major challenges in current antifungal therapy to treat Candida albicans-associated infections. Here, we describe two food-derived yeasts, Saccharomyces cerevisiae and Issatchenkia occidentalis, that inhibit virulence traits of C. albicans, including hyphal morphogenesis, biofilm formation, and adhesion to intestinal epithelial cells. These yeasts also protect the model host Caenorhabditis elegans from C. albicans infection. We demonstrate that the protective activity is primarily retained in the s

Molecular identification of yeasts from the order Trichosporonales causing superficial infections.

BACKGROUND: The molecular reclassification of the order Trichosporonales placed the medically relevant Trichosporon species into three genera of the family Trichosporonaceae: Cutaneotrichosporon, Trichosporon, and Apiotrichum. From the clinical and epidemiological standpoint, it is important to identify any species of the family Trichosporonaceae because they present different antifungal susceptibility profiles. In Mexico, little is known about trichosporonosis etiology because the fungi are identified through phenotypic methods. AIMS: To identify at a molecular level 12 yeast isolates morfol

Fermentation characteristics of four non-Saccharomyces yeasts in green tea slurry.

The fermentation characteristics of non-Saccharomyces yeasts (Pichia kluyveri FrootZen, Torulaspora delbrueckii Prelude, Williopsis saturnus var. mrakii NCYC2251 and Torulaspora delbrueckii Biodiva) were evaluated in green tea slurry fermentation. Each yeast showed different fermentation performances: strains Prelude and Biodiva utilized sucrose faster than the other two yeasts; strain NCYC2251 was the only species that metabolized xylose. Strain FrootZen increased the caffeine content significantly and strain Prelude showed the opposite trend, both at a statistical level, while theanine conte

Miltefosine Has a Postantifungal Effect and Induces Apoptosis in Cryptococcus Yeasts.

Cryptococcus spp. are common opportunistic fungal pathogens, particularly in HIV patients. The approved drug miltefosine (MFS) has potential as an alternative antifungal against cryptococcosis; however, the mechanism of action of MFS in Cryptococcus is poorly understood. Here, we examined the effects of MFS on C. neoformans and C. gattii yeasts (planktonic and biofilm lifestyles) to clarify its mechanism of action. MFS presented inhibitory and fungicidal effects against planktonic Cryptococcus cells, with similar activities against dispersion biofilm cells, while sessile biofilm cells were les
