BACKGROUND: Zearalenone (ZEA), a natural food contaminant, is reported to act as a mycoestrogen due to its estrogen-mimicking properties. According to studies, ZEA has a greater potential for estrogenic activity compared to any other naturally occurring non-steroidal estrogen. ZEA has been found in the endometrium of individuals with reproductive problems and the serum of children facing early puberty. These studies suggested a possible link between ZEA exposure and endometrial toxicity; nonetheless, no thorough research has been done. This study assessed the endometrium's response to chronic

Zearalenone (ZEN), a mycotoxin produced by Fusarium species, is known for its reproductive toxicity as an estrogen analogue. However, there are limited knowledge about its hepatointestinal toxicity, as well as the role that gut microbiota and metabolites play in this process. In this study, a total of 24 thirty-week-old hens were fed to investigate the hepatointestinal toxicity subjected to long-term ZEN consumption at 2.0 mg/kg for 90 d. And we employed uncultured 16S rRNA sequencing for gut microbiota and untargeted metabolomics for fecal metabolites assessment. Notably, ZEN induced signific

Zearalenone (ZEA) is widely present in feedstuffs and raw materials, causing reproductive disorders in animals. In this study, Bacillus licheniformis CK1 was used to detoxify ZEA in feed for alleviating its effect in Tibetan piglets. A total of 18 weaned female Tibetan piglets were randomly divided into 3 groups: control group (Control, ZEA-free basal diet); treatment group 1 (T1, ZEA-contaminated diet); and treatment group 2 (T2, ZEA-contaminated but pre-fermented by CK1 diet). There were no significant differences of average daily feed intake (ADFI), average daily gain (ADG), and feed effici

Zearalenone (ZEN) is an estrogen-like mycotoxin occurring in food and feeds, and it can cause oxidative damage and apoptosis in the testis, liver, and kidney. A current concern for researchers is how to reduce the harm it causes to humans and animals. In this study, our aim was to isolate and identify a novel and efficient ZEN-detoxifying strain of bacteria, and we aimed to assess the protective effect of the isolated strain on kidney damage caused by ZEN in mice. Our results indicated that a strain of Bacillus velezensis (B. velezensis), named A2, could completely degrade ZEN (7.45 μg/mL) aft

The present study was aimed at investigating the adverse effects of dietary zearalenone (ZEA) (1.1 to 3.2 mg/kg diet) on serum hormones, morphologic and apoptotic measurements of genital organs in post-weaning gilts. A total of twenty gilts (Landrace×Yorkshire×Duroc) weaned at 21 d with an average body weight of 10.36±1.21 kg were used in the study. Gilts were fed a basal diet with an addition of 0, 1.1, 2.0, or 3.2 mg/kg purified ZEA for 18 d ad libitum. Results showed that 3.2 mg/kg ZEA challenged gilts decreased (p<0.05) the serum levels of luteinizing hormone, however, serum levels of prol

Mycotoxins, secondary metabolites produced by moulds, have been shown to cause diverse toxic effects in animals and are also suspected of disease causation in humans. The present study compares the molecular mechanisms of the toxicity of zearalenone (ZEN), T-2 toxin and ochratoxin A (OTA) in human hepatoma cells HepG2. The three mycotoxins-induced a caspase-dependent mitochondrial apoptotic pathway. The mitochondrial alterations include: bax relocalisation into the mitochondrial outer membrane, loss of the mitochondrial transmembrane potential, PTPC opening, and cytochrome c (but not AIF) rele

This study was to investigate the effects of the combination of deoxynivalenol (DON) and zearalenone (ZON) on pigs. Twenty-four weaning piglets were divided into a control group fed a diet free of mycotoxins and a toxin group fed a diet containing 1 mg/kg DON and 250 microg/kg ZON. The results showed that supplementation of DON and ZON in diets had extensive effects on pigs. More specifically, DON and ZON caused levels of total protein, albumin, and globulin in sera to decrease (p < 0.05) by 14.5%, 6.5% and 11.3%, respectively, and at the same time increased (p < 0.05) the serum enzyme activit

The aim of the monitoring of zearalenone presence in the blood plasma of women with neoplastic lesions in the reproductive tract was to asses whether the phytosteride is noted in the patients blood and whether the correlation exists between its presence and the incidence of particular neoplasm. The presence of zearalenone or its metabolite--alpha-zearalenole, was noted in 13.51% of the examined women. In 60% of the patients with the confirmed presence of the xenobiotic it was noted in the lowest concentrations. These patients had neoplastic lesions of Carcinoma corpus uteri type.

Zearalenone (ZEN), a typical Fusarium-derived mycotoxin, accumulates widely in corn throughout harvesting, storage and processing. Upon entering organisms, ZEN is metabolized into α-zearalenol and β-zearalenol, which disrupt the endocrine systems of humans and animals, posing threats to food safety as well as human and livestock health. Efficient, eco-friendly and mild detoxification technologies for ZEN degradation remain limited. Herein, double dielectric barrier discharge (DDBD) cold plasma technology was adopted to degrade ZEN solution (1 μg/mL). Under fixed conditions (current 1 A, duty c

Zearalenone (ZEA) contaminates various food crops and water systems worldwide. We investigate whether gestational ZEA exposure disrupts the placental barrier and elucidates its underlying mechanisms. This study employed a mouse model and human trophoblast cells. We found that ZEA accumulated in mouse placental tissue. Compared with the well-defined tight junctions observed in controls, ZEA disrupted placental barrier integrity. Consistent with the morphological disruption, ZEA significantly downregulated the expression of placental barrier proteins. Furthermore, ZEA elevated fetal glucocortico

Zearalenone-14-glucoside (Z14G) is a widely contaminating modified mycotoxin in the global food and feed. For the first time, we report that Z14G formed a self-assembled supramolecular gel. Results from our morphological and chemical characterizations, rheological testing, and molecular simulation dynamics demonstrated that Z14G is a type of thermal-responsive, self-assembled supramolecular gel. Both the microbial key metabolic enzymes (MKME)-Caco2/HepG2 and rat metabolic models were adopted to explore the release of Z14G. These in vivo and in vitro results suggested that Z14G slowly released

Zearalenone (ZEN), a mycotoxin commonly found in animal feed, impairs the female reproductive function owing to its estrogen-mimicking ability. ZEN exposure induces endoplasmic reticulum (ER) stress, oxidative stress, and autophagy activation. However, the involvement of inositol-requiring enzyme 1 (IRE1)-mediated ER-phagy in ZEN-induced ER stress and embryonic competence during in vitro culture (IVC) remains unclear. Porcine embryos were cultured with 2.5, 5, and 10 μM ZEN for two days after in vitro fertilization (IVF) to assess blastocyst development. ZEN exposure caused a significant dose-

A novel fluorescent lateral flow assay (FLFA) based on a zirconium-based metal aggregation-induced emission luminogen framework (Zr-MAF) is reported for the highly sensitive detection of zearalenone (ZEN) in cereals. A carambola-like Zr-MAF with a high fluorescence quantum yield of 36.9% was synthesized via a hydrothermal method using Zr4+ and 1,1,2,2-tetra(4-carboxylphenyl) ethylene (TCPE). The prepared Zr-MAF-labeled monoclonal antibody probe exhibited intense luminescence, robust stability, and high affinity toward ZEN, collectively contributing to a significant enhancement in detection sen

Based on the strategy of rare-earth MOF coated with molecular imprinted polymer (MIP), a high-performance fluorescent colorimetric sensor was constructed for the detection of Zearalenone (Zen). Bisquinoline acid was selected as the organic ligand to construct the Eu-BCA MOF with europium nitrate, then the quinoline-N in the ligand of MOFs was used to participate in the construction of the SiO2 molecularly imprinted layer to form the Eu-BCA@SiO2-MIP, which can respond quickly to trace Zen in the range of 0-1.56 × 10-6 mol L-1, and the detection limit is as low as 1.66 ng mL-1. BACKGROUND: Mycotoxin-producing fungal species and their mycotoxins pose a global threat for crop production and for human and animal health. Given the increasing demand for healthier food and feed, alternative non-pesticide approaches for reducing fungal infections in crops and eliminating mycotoxin contamination in feedstock are becoming more prevalent. For such purposes, yeast species can be good candidates. Thus, the present study examined the mycotoxin tolerance and mycotoxin elimination ability of several yeast strains belonging to the Trichomonascaceae family.
RESULTS: While none of th

Zearalenone (ZEA) is commonly found in crops and feed, with a high detection rate and concentration. This substance adversely affects reproduction and development, with particularly noticeable effects on pigs. Given these detrimental effects, there is an urgent need to identify effective protective agents against ZEA toxicity. Sulforaphane (SFN) has emerged as a promising candidate owing to its widely acknowledged antioxidant, anti-inflammatory, and detoxifying properties. This study elucidates ZEA's mechanism of oxidative injury in porcine endometrial stromal cells (ESCs) via mitochondrial-as

Zearalenone (ZEA) is an environmentally widespread mycotoxin capable of posing a serious threat to food safety and public health, and porcine endometrial stromal cells (ESCs) are particularly sensitive to the toxic effects of ZEA. We hypothesized that Rutin, a flavonoid antioxidant, could significantly alleviate ZEA-induced ferroptosis through the p53 signaling pathway. In this study, we used porcine ESCs as a research model. When porcine ESCs were co-cultured with the addition of Rutin and ZEA following p53 gene silencing via siRNA transfection, Rutin significantly mitigated ZEA-induced mitoc

In order to alleviate toxic effects of aflatoxins B1 (AFB1) and zearalenone (ZEA) on broiler production performance and gut microbiota, three kinds of compound probiotics (CP) were selected. The optimal ratios of Bacillus subtilis, Lactobacillus casei and Candida utilis in broiler diets were 7, 5 and 6 log CFU/g for ZEA biodegradation (CP1); 6, 7 and 7 log CFU/g for AFB1 biodegradation (CP2); 7, 6 and 7 log CFU/g for ZEA + AFB1 biodegradation (CP3). A total of 350 1-day-old Ross broilers were randomly divided into 7 groups. Group A was the basal diet, group B-G contained ZEA, AFB1, ZEA + AFB1,

The worldwide contamination of grains designated to human and animal feeding with Fusarium mycotoxins is a significant problem. Among Fusarium mycotoxins, deoxynivalenol (DON) and zearalenone (ZEA) are the most prevalent mycotoxins found in cereals. Co-occurrence of DON and ZEA is also very frequent and indicates that these mycotoxins might be involved in a wide range of synergistic or additive interactions. Both mycotoxins have been linked to various male reproduction problems including downregulation of steroidogenesis. In this study, the impact of DON and ZEA alone or in combination on the

Both earlier investigations and complete formulas of pet food for dogs show that professionally prepared pet food is based in 30% on the material of the plant origin. Our own investigations prove that animal feeding stuffs of the plant origin are very often vectors of many undesirable substances (including micotoxins) that are unfavourable for carnivores. Zearalenone is a micotoxin, which frequently occurs among them. It was revealed in pet food in quantities from 5.0-299.5 microg/kg. It is a very dangerous xenobiotic because of its quick and easy absorption in the organism after per os applic
