Interest in the identification of cognitive decline in its earliest manifestations and the heterogeneity of clinically diagnosed Alzheimer's disease (AD) explain the growing number of neuroimaging studies of AD. Alzheimer-type lesions are associated with loss of neurons, and magnetic resonance imaging (MRI) can detect predominantly left atrophic changes in the entorhinal cortex, amygdala and anterior hippocampus several years before the onset of clinical symptoms. Cerebrovascular disease can mimic AD in the elderly whereas MR markers of subcortical vascular disease-leukoaraiosis, lacunar infar

Anticonvulsant properties of furosemide have been suggested to reduce neuronal synchronization via its inhibitory effect on the Na+/K+/2Cl- co-transport system. We have studied effects of furosemide on spontaneous epileptiform activity and analysed effects of furosemide on amplitudes of stimulus-induced population-spikes, on stimulus-induced K+ changes, on extracellular pH changes at rest and during stimulation, and on changes in the extracellular space-volume. We used three different in vitro models of epilepsy in the combined hippocampal-entorhinal cortex slice preparation. Furosemide revers

In the CNS the cell adhesion molecule L1 plays a role in axonal growth and fasciculation. Since its roles in synapse formation and CNS regeneration are unknown, we followed the staining of L1 through the sequence of degeneration and reactive axon sprouting in the denervated outer molecular layer (ML) of the hippocampal dentate gyrus following ipsilateral entorhinal cortex (ERC) lesion. We compared immunohistological and ultrastructural localization of L1 and employed image analysis to evaluate lamina-specific changes over time. L1 staining was uniformly distributed over the ML in unlesioned an

Despite the need for successful navigation, humans vary greatly in their ability to navigate, and these individual differences may relate to variation in brain structure. While prior research provides support for a correlation between hippocampal volume and navigation ability in both navigation experts and in older individuals, this relationship is under scrutiny for healthy, young adults. We assessed 99 healthy young adults' ability to navigate in a virtual, desktop maze and correlated their performance with total hippocampal gray matter volume. For a subset of these individuals, we further s

Bridging integrator 1 (BIN1) is the second most prevalent genetic risk factor identified by genome-wide association studies (GWAS) for late-onset Alzheimer's disease. BIN1 encodes an adaptor protein that regulates membrane dynamics in the context of endocytosis and neurotransmitter vesicle release. In vitro evidence suggests that BIN1 can directly bind to tau in the cytosol. In addition, BIN1's function limits extracellular tau seed uptake by endocytosis and subsequent propagation as well as influences tau release through exosomes. However, the in vivo roles of BIN1 in tau pathogenesis and tau

Loneliness is associated with cognitive decline and dementia risk, but the role of Alzheimer's disease (AD) pathology in its associations with medial temporal lobe (MTL) morphology remains unclear. We examined whether loneliness was related to total hippocampal volume, hippocampal subfields, and entorhinal cortex (ERC) volumes, and whether plasma phosphorylated tau 217 (pTau217) moderated these relationships. We analyzed baseline data from cognitively unimpaired older adults aged 65-80 years enrolled in IGNITE. Loneliness was assessed using the revised UCLA Loneliness Scale. Structural 3 T MRI

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is associated with neurological complications, yet the underlying mechanisms remain poorly understood. Here, we used an integrated spatial multiomics approach to investigate the effects of SARS-CoV-2 on human hippocampus–entorhinal cortex (HP–EC) tissues from autopsies of individuals with severe COVID-19. Our analysis revealed a cascade of pathological events initiated by viral infection. First, we found that SARS-CoV-2 preferentially infects neurons, with the highest viral loads in the dentate gyrus. Second, this infection

Normal explicit/declarative memory requires a system of anatomically related structures in the medial temporal lobe that includes the hippocampus and the entorhinal, perirhinal, and parahippocampal cortex. Although extensive medial temporal lobe damage in primates causes robust anterograde amnesia, defining the contribution of the hippocampus proper has proved challenging. We revisited that enduring puzzle here, directly addressing a number of factors suspected to contribute to conflicting results across earlier studies. Among them, we explored the effects of selective hippocampus lesions prod

Abnormal tau proteins are characterized by accumulation of abnormally phosphorylated tau followed by irreversible loss of neurons. Progranulin (PGRN) is a secreted pleiotropic glycoprotein against the development of multiple neurodegenerative diseases. However, it remains unclear whether PGRN could modulate the relationships of abnormal tau proteins with cognition and neurodegeneration. A total of 606 non-demented participants were followed for 8 years and were annually assessed for cognition and brain volumes. Abnormal tau protein (TN) status was determined by cerebrospinal fluid (CSF) levels

OBJECTIVE: Limbic Predominant Age-Related TDP43 Encephalopathy (LATE-NC) is a recently defined (2019) degenerative transactive response DNA binding TDP43 proteinopathy whose prevalence increases after age 80, marked by an amnestic presentation. Anatomically, atrophy predominantly occurs in the hippocampal region, in a unilateral or bilateral manner, yet workup is negative for amyloid and phosphorylated tau (e.g. PET scan; cerebrospinal fluid). Postmortem immunohistochemical data revealing neurofibrillary tangles accumulation initially occurring in the limbic (e.g. amygdala) region with subsequ

Temporal lobe epilepsy (TLE) is the most common type of acquired epilepsy characterized by recurrent spontaneous seizures. The entorhinal cortex (EC) lies in the medial temporal lobe and serves as a relay center between the hippocampus and neocortex, and contains cholecystokinin-positive (CCK+) neurons. Increased CCK expression has been reported in patients with TLE suggesting its role in epilepsy development. Here, we sought to understand whether CCK+ neurons in EC are involved in TLE development and also to evaluate the therapeutic efficacy of a CCK-2 receptor antagonist in ameliorating seiz

One of the pathologic hallmarks of Alzheimer's disease (AD) is neurofibrillary tau tangles. Despite our knowledge that tau typically initiates in the medial temporal lobe (MTL), the mechanisms driving tau to spread beyond MTL remain unclear. Emerging evidence reveals distinct patterns of functional connectivity change during aging and preclinical AD: while connectivity within-network decreases, connectivity between-network increases. Building upon increased between-network connectivity, our study hypothesizes that this increase may play a critical role in facilitating tau spread in early stage

INTRODUCTION: Alzheimer's disease (AD), primary age-related tauopathy (PART), and chronic traumatic encephalopathy (CTE) all feature hyperphosphorylated tau (p-tau)-immunoreactive neurofibrillary degeneration, but differ in neuroanatomical distribution and progression of neurofibrillary degeneration and amyloid beta (Aβ) deposition.
METHODS: We used Nanostring GeoMx Digital Spatial Profiling to compare the expression of 70 proteins in neurofibrillary tangle (NFT)-bearing and non-NFT-bearing neurons in hippocampal CA1, CA2, and CA4 subregions and entorhinal cortex of cases with autopsy-confirm

Alzheimer's disease (AD) pathology and amyloid-beta (Aβ) plaque deposition progress slowly in the cerebellum compared to other brain regions, while the entorhinal cortex (EC) is one of the most vulnerable regions. Using a knock-in AD mouse model (App KI), we show that within the cerebellum, the deep cerebellar nuclei (DCN) has particularly low accumulation of Aβ plaques. To identify factors that might underlie differences in the progression of AD-associated neuropathology across regions, we profiled gene expression in single nuclei (snRNAseq) across all cell types in the DCN and EC of wild-typ

BACKGROUND: Western studies indicate potential associations between hippocampal volume and memory in the trajectory of Alzheimer's disease (AD). However, limited availability of neuroimaging technology and neuropsychological tests appropriate for sub-Saharan African (SSA) countries makes it difficult to establish neuroanatomical associations of hippocampus and memory in this locale.
OBJECTIVE: This study examined hippocampal volumes and memory in healthy control (HC) and probable AD groups in the Democratic Republic of Congo (DRC).
METHODS: Forty-six subjects with probable AD and 29 HC subje

INTRODUCTION: Progressive supranuclear palsy (PSP) is an atypical parkinsonism caused by the intracerebral aggregation of the microtubule-associated protein tau (MAPT) which is encoded by MAPT gene. Although PSP is a sporadic disease, MAPT mutations have been reported in rare cases.
METHODS: Among 190 patients with PSP who were recruited by the Neurodegenerative Research Group at Mayo Clinic during 2009-2023, we identified two patients who fulfilled diagnostic criteria for PSP-Richardson's syndrome (PSP-RS) and harbor novel MAPT mutations. To better investigate the potential effects of these

OBJECTIVE: Subjective cognitive decline (SCD) is a potential early risk marker for Alzheimer's disease (AD), but its utility may vary across individuals. We investigated the relationship of SCD severity with memory function and cerebral blood flow (CBF) in areas of the middle temporal lobe (MTL) in a cognitively normal and overall healthy sample of older adults. Exploratory analyses examined if the association of SCD severity with memory and MTL CBF was different in those with lower and higher cardiovascular disease (CVD) risk status.
METHODS: Fifty-two community-dwelling older adults underwe

This work aimed to investigate potential pathways linking age and imaging measures to early age- and pathology-related changes in cognition. We used [18F]-Flutemetamol (amyloid) and [18F]-Flortaucipir (tau) positron emission tomography (PET), structural MRI, and neuropsychological assessment from 232 elderly individuals aged 50-89 years (46.1% women, 23% APOE-ε4 carrier, 23.3% MCI). Tau-PET was available for a subsample of 93 individuals. Structural equation models were used to evaluate cross-sectional pathways between age, amyloid and tau burden, grey matter thickness and volumes, white matte
