Histochemical study of the effects on abomasal mucins of Haemonchus contortus or Teladorsagia circumcincta infection in lambs.

Previously, chemical analysis of gastric fundic mucin showed that infection of sheep with Haemonchus contortus or Teladorsagia circumcincta changed the proportions of monosaccharides and decreased terminal mucin fucosylation and sialylation. To identify the effects of these parasites on the two mucin-secreting cell lineages, fundic and antral tissues were collected for histochemistry from 69 lambs aged from 3-4 to 9-10 months-of-age which had received a single infection of either H. contortus or T. circumcincta and euthanased at Day 21 or 28 post- infection respectively. All fundic tissues wer

Bioflocculation: chemical free, pre-treatment technology for the desalination industry.

Rapid sand filtration (RSF), proceeded by chemical coagulation and flocculation, is a commonly used, effective pretreatment in the desalination industry. We designed and tested a novel, large pilot-scale, two-stage granular Rapid Bioflocculation Filter (RBF) based on a first-stage Bioflocculator (BF) unit followed by a mixed-media bed filter (MBF). The BF filter bed consisted of an extremely porous volcanic Tuff granular medium which provided an enlarged surface area for microbial development and biofilm proliferation. We compared the efficiency of the pilot RBF to that of a full-scale RSF, op

Detection of cardia intestinal metaplasia: do the biopsy number and location matter?

BACKGROUND AND AIM: Presence of intestinal metaplasia in the gastric cardia (cardia intestinal metaplasia, CIM) has been reported in 5-34% of patients undergoing upper endoscopy and is a topic of interest given the rising incidence of cancer in this location. The aim of this article is to determine the prevalence of CIM in biopsies obtained from two separate locations within the gastric cardia. METHODS: Patients presenting to the endoscopy unit for upper endoscopy for any symptoms were invited to participate in the study. The biopsy protocol included: eight biopsies from the gastric cardia, f

Allantoin Enhances Proliferation, Wound Closure, and Nrf2-Associated Antioxidant Defense in Mouse Embryonic Palatal Mesenchymal Cells:A Dose-Response Study.

BACKGROUND: Cleft palate is a congenital craniofacial anomaly that originates from disrupted palatogenesis. During palatal shelf morphogenesis, palatal mesenchyme must expand, remodel, and withstand environmental stress, and oxidative imbalance is considered a convergent cellular risk that can compromise developmental robustness. Allantoin is a clinic-friendly small molecule widely used in topical formulations and reported to modulate cell behavior, but its dose-dependent effects and antioxidant mechanism in embryonic palatal mesenchyme remain unclear. Here, we investigated the concentration w

Quercetin protected the gut barrier in ulcerative colitis by activating aryl hydrocarbon receptor.

BACKGROUND: Ulcerative colitis (UC) is characterized by abdominal pain and bloody diarrhoea and restoring the gut barrier is the core goal of UC treatment. Activation of aryl hydrocarbon receptor (Ahr) was reported to effectively alleviate symptoms and repair the gut barrier damage. Neutrophil extracellular traps (NETs) have been recognized as potential targets in the treatment of UC. Ahr activation has been found to be capable of upregulating Nqo1, thereby reducing the production of reactive oxygen species (ROS), which is important in the formation of NETs. Quercetin (QUE), which is derived f

Exploration of the distribution of intestinal bacteria in mice under normal and intestinal leakage (IBD) conditions.

Introduction. The distribution of micro-organisms in healthy organisms remains a subject of debate. Emerging evidence revealed the colonization of microbial communities in multiple anatomical sites previously considered sterile under homeostatic conditions. However, the mechanistic relationship between compromised intestinal epithelial barrier integrity and subsequent translocation of gut-resident bacteria into systemic circulation has yet to be comprehensively elucidated.Hypothesis/Gap Statement. Under intestinal leakage, gut micro-organisms can break through the intestinal barrier and then t

Microdistribution of Magnetic Resonance Imaging Contrast Agents in Atherosclerotic Plaques Determined by LA-ICP-MS and SR-μXRF Imaging.

PURPOSE: Contrast-enhanced magnetic resonance imaging (MRI) has the potential to replace angiographic evaluation of atherosclerosis. While studies have investigated contrast agent (CA) uptake in atherosclerotic plaques, exact CA spatial distribution on a microscale is elusive. The purpose of this study was to investigate the microdistribution of gadolinium (Gd)- and iron (Fe) oxide-based CA in atherosclerotic plaques of New Zealand White rabbits. PROCEDURES: The study was performed as a post hoc analysis of archived tissue specimens obtained in a previous in vivo MRI study conducted to invest

Perforated caecal diverticulum as a content of inguinal hernia - report of a rare case.

FUN14 domain‑containing 1 (FUNDC1) is a receptor that has been previously reported to activate hypoxia‑induced mitophagy. However, the potential role of FUNDC1 in the pathophysiology of dental pulp diseases remains unknown. Therefore, present study first collected tissue specimens from patients with pulpitis and from healthy individuals. The results of reverse transcription‑quantitative PCR and immunohistochemical staining revealed markedly increased FUNDC1 and hypoxia‑inducible factor‑1α expression in pulpitis tissue specimens compared with those from healthy individuals. Aldehyde fuchsin staining, direct or after oxidation: problems and remedies, with special reference to human pancreatic B cells, pituitaries, and elastic fibers.

Successful production of aldehyde fuchsin (AF) having the unique properties described by Gomori depends on each of many critical variables. AF made from basic fuchsins which contain mainly rosanilin (C.I. 42510) do not stain properly-fixed pancreatic B cells, pituitary basophils, or elastic fibers in unoxidized sections. AF made from basic fuchsins containing mainly pararosanilin (C.I. 42500) stains these entities strongly. Substances stained by AF without oxidation fall into two classes: 1) nonacidic peptides and proteins, most of which contain half-cystines, and 2) polyanions, particularly w

Zanthoxylum nitidum-derived active alkaloid nitidine chloride mitigates ulcerative colitis by remodeling gut microbiota and activating SIRT1 to restore intestinal barrier integrity.

BACKGROUND: Ulcerative colitis (UC) is a recurrent inflammatory bowel disorder with rising prevalence and limited efficacy of current treatments. Nitidine chloride (NC), a characteristic bioactive constituent of Zanthoxylum nitidum (Liang-Mian-Zhen), possesses diverse pharmacological properties, including anti-inflammatory, antitumor, and antioxidant activities. Nevertheless, its role in UC and the associated regulatory mechanisms remain incompletely defined. PURPOSE: This study investigated whether NC protects against experimental colitis and clarified its effects on epithelial barrier integ

High-fat diet exacerbates experimental colitis by inhibiting lysosomal function via the STAT3-TFEB Axis.

An elevated risk for inflammatory bowel disease (IBD) has been linked to the intake of high-fat diet (HFD), yet the underlying molecular mechanisms remain unclear. The lysosome and the macroautophagy/autophagy-lysosome pathway (ALP) are critical for maintaining the intestinal epithelial barrier. By employing both an in vivo model of dextran sulfate sodium (DSS)-induced colitis in mice and an in vitro model using lipopolysaccharide (LPS)-treated NCM460 cells, we established that HFD in vivo and palmitic acid (PA) in vitro profoundly impair epithelial barrier function and amplify inflammation, w
