Gellan gum formulations containing natural polyphenolic compounds to treat oral candidiasis.

Oral candidiasis (OC) is caused by Candida albicans, targeting immunocompromised individuals and developing drug resistance, highlighting the need for advanced therapeutics. Polyphenols such as caffeic acid phenethyl ester (CAPE) and ellagic acid (EA) display antifungal and immune-modulating properties. Incorporating CAPE and EA in gellan gum (GG) formulation enhances their applicability and effectiveness against OC. We developed GG-based formulations loaded with CAPE (1,000 µg/mL), EA (1,000 µg/mL), or CAPE + EA (1,000 µg/mL each) against C. albicans. GG formulations containing gellan (0.6% a

Salvianolic acids from antithrombotic Traditional Chinese Medicine Danshen are antagonists of human P2Y1 and P2Y12 receptors.

Many hemorheologic Traditional Chinese Medicines (TCMs) that are widely-used clinically lack molecular mechanisms of action. We hypothesized that some of the active components of hemorheologic TCMs may function through targeting prothrombotic P2Y1 and/or P2Y12 receptors. The interactions between 253 antithrombotic compounds from TCM and these two G protein-coupled P2Y receptors were evaluated using virtual screening. Eleven highly ranked hits were further tested in radioligand binding and functional assays. Among these compounds, salvianolic acid A and C antagonized the activity of both P2Y1 a

Epigenetic modulation of inflammation and synaptic plasticity promotes resilience against stress in mice.

Major depressive disorder is associated with abnormalities in the brain and the immune system. Chronic stress in animals showed that epigenetic and inflammatory mechanisms play important roles in mediating resilience and susceptibility to depression. Here, through a high-throughput screening, we identify two phytochemicals, dihydrocaffeic acid (DHCA) and malvidin-3'-O-glucoside (Mal-gluc) that are effective in promoting resilience against stress by modulating brain synaptic plasticity and peripheral inflammation. DHCA/Mal-gluc also significantly reduces depression-like phenotypes in a mouse mo

Emerging roles of ferroptosis in liver pathophysiology.

Ferroptosis is a widely recognized process of regulated cell death linking redox state, metabolism, and human health. It is considered a defense mechanism against extensive lipid peroxidation, a complex process that may disrupt the membrane integrity, eventually leading to toxic cellular injury. Ferroptosis is controlled by iron, reactive oxygen species, and polyunsaturated fatty acids. Accumulating evidence has addressed that ferroptosis plays an unneglectable role in regulating the development and progression of multiple pathologies of the liver, including hepatocellular carcinoma, liver fib

Salvianolic acid A prevents UV-induced skin damage by inhibiting the cGAS-STING pathway.

DNA damage resulting from UV irradiation on the skin has been extensively documented in numerous studies. In our prior investigations, we demonstrated that UVB-induced DNA breakage from keratinocytes can activate the cGAS-STING pathway in macrophages. The cGAS-STING signaling pathway serves as the principal effector for detecting and responding to abnormal double-stranded DNA in the cytoplasm. Expanding on our previous findings, we have further validated that STING knockout significantly diminishes UVB-induced skin damage, emphasizing the critical role of cGAS-STING activation in this context.

Novel antioxidants are not toxic to normal tissues but effectively kill cancer cells.

Pancreatic ductal adenocarcinoma (PDAC) represents a challenge for a multidisciplinary oncology team. Diagnosis of PDAC remains challenging due to overlapping imaging features with benign lesions, notwithstanding great advances with computed tomography (CT) and magnetic resonance imaging (MRI). The term "Radiomics" has recently been introduced to define a mathematical process to extract countless quantitative features from medical images (including each diagnostic technique) with high throughput computing for diagnosis and prediction. This article is an updated overview of the imaging techniqu

Future perspectives of propolis-chitosan biomembranes in wound healing: molecular mechanism, formulation and therapeutic properties.

Wound management remains a persistent global health burden because conventional dressings largely address the symptoms of a wound rather than the disrupted molecular signalling that stalls its healing. Rather than cataloguing the properties and applications of propolis and chitosan in isolation, this review interrogates the mechanistic basis by which their combination as a biomembrane redirects wound biology. Although attention to propolis-chitosan systems is increasing, the physicochemical basis of their interaction remains poorly understood. Specifically, how chitosan's cationic amine groups

Salvianolic Acid A Protects Against Angiotensin II-Induced Hypertensive Kidney Disease by Targeting TLR4 and Inhibiting TLR4/Myd88/PI3K/AKT/NF-κB Pathway.

Hypertensive nephropathy (HN), a significant consequence of chronic hypertension, remains a leading contributor to end-stage renal disease. Excessive Angiotensin II (Ang II)-driven inflammation, apoptosis, and fibrosis are central to its pathogenesis. Salvianolic acid A (SAA), a polyphenolic compound from Salvia miltiorrhiza, has anti-inflammatory properties, but its therapeutic potential in HN is unclear. HN was induced in mice by continuous subcutaneous infusion of Ang II (1.44 mg/kg/day) for 28 days. SAA (10 or 20 mg/kg/day) was administered orally every day during the final 14 days. Networ

Salvianolic acid A ameliorates sepsis through inhibiting inflammation via binding STING and modulating TBK1/IRF3 signaling pathway.

Sepsis is a condition characterized by a systemic inflammatory response due to infection, resulting in numerous organ dysfunction. Salvianolic acid A (SAA) is a phenolic acid substance extracted from the plant Salvia miltiorrhiza Bunge, possessing antioxidant and anti-platelet aggregation properties. Although aberrant stimulator of interferon genes (STING) signaling is associated with sepsis, it is uncertain if SAA can influence this pathway to avert sepsis-induced organ injury. This study examined the antiseptic efficacy and biological mechanisms of SAA. The pharmacodynamics and mechanism of

Salvianolic Acid A Mitigates Osteoporotic Bone Loss by Repressing Reactive Oxygen Species via the Nrf2-HO-1 Pathway.

Osteoporosis, characterized by osteoclastic bone resorption, has been the focus of research. Studies implicate that reactive oxygen species (ROS) accumulate intracellularly during osteoclastogenesis. Salvianolic acid A (SAA), a compound derived from Salvia miltiorrhiza, has been widely used to treat cardiovascular and cerebrovascular disorders, owing to its antioxidant and anti-inflammatory properties. In this study, we investigated the therapeutic effects of SAA on osteoporotic bone loss in vitro and in osteoporotic mice induced by ovariectomy (OVX) and explored the underpinning mechanisms. I

Dietary phytochemicals modulate experience-dependent changes in Neurexin gene expression and alternative splicing in mice after chronic variable stress exposure.

Neurexins (NRXNs) are cell-adhesion molecules important in the formation and remodeling of neural circuits. It has been shown that aversive environmental stimuli can affect the expression pattern of Neurexin genes (Nrxns) impacting the regulation of synaptic strength. Accumulated evidence suggests that, after chronic exposure to psychological stress, the triggered changes in gene expression and splicing patterns of Nrxns may be involved in aversive conditioning. Previously, we have demonstrated that a novel treatment using dietary phytochemicals can modulate the response to chronic variable st

Microfluidic cell sorting by stiffness to examine heterogenic responses of cancer cells to chemotherapy.

Cancers consist of a heterogeneous populations of cells that may respond differently to treatment through drug-resistant sub-populations. The scarcity of these resistant sub-populations makes it challenging to understand how to counter their resistance. We report a label-free microfluidic approach to separate cancer cells treated with chemotherapy into sub-populations enriched in chemoresistant and chemosensitive cells based on the differences in cellular stiffness. The sorting approach enabled analysis of the molecular distinctions between resistant and sensitive cells. Consequently, the role

Effects of salvianolic acid a on oxidative stress and liver injury induced by carbon tetrachloride in rats.

In the present study, we investigated the hepatoprotective effects of salvianolic acid A, a novel antioxidant, against oxidative stress and acute liver injury induced by carbon tetrachloride (CCl(4)) in rats, and the mechanisms underlying its protective effects. Administration of CCl(4) to rats caused severe hepatic damage, as demonstrated by the significant increase in the levels of serum alanine aminotransferase, aspartate aminotransferase and classic histological changes including hepatocyte necrosis or apoptosis, haemorrhage, fatty degeneration, etc. Co-treatment with salvianolic acid A (2

Antioxidant Potential of Ethiopian Medicinal Plants and Their Phytochemicals: A Review of Pharmacological Evaluation.

BACKGROUND: Free radicals are very reactive molecules produced during oxidation events that in turn initiate a chain reaction resulting in cellular damage. Many degenerative diseases in humans, including cancer and central nervous system damage, are caused by free radicals. Scientific evidence indicates that active compounds from natural products can protect cells from free radical damage. As a result, the aim of this review is to provide evidence of the use of diverse Ethiopian medicinal plants with antioxidant properties that have been scientifically validated in order to draw attention and
