Characterization of Matrix-Adapted PEG Isolation of Exosome-Like Nanovesicles from Pandanus conoideus.

Plant-derived exosome-like nanovesicles (PDENs) have recently gained prominence as biocompatible, renewable nanocarriers, but lipid-dense tropical plants remain an underutilized vesicle reservoir and present substantial hurdles for efficient isolation. Here, we establish Pandanus conoideus (red fruit), a Papuan endemic with unusually high carotenoid, tocopherol, and unsaturated fatty acid content, as a new source of PDENs and develop a matrix-adapted polyethylene glycol (PEG 6000) workflow for their scalable isolation from fresh fruit (RFF), dried simplicia (RFS), and long-stored red fruit oil

Multi-omics data reveal causal associations of cellular senescence-related genes in rheumatoid arthritis: A summary-data-based Mendelian randomization and co-localization analysis.

Rheumatoid arthritis (RA) is a complex autoimmune disease. Recently, cell senescence has been identified as a key factor in its pathogenesis. This study integrated multi-omics summary data and applied Mendelian randomization (MR) and co-localization analysis to systematically evaluate the causal relationships between cell senescence-related genes and RA. We collected summary data on blood methylation quantitative trait loci (mQTL), expression quantitative trait loci, and protein quantitative trait loci. The FinnGen database was the primary discovery dataset, validated by the UK Biobank and GWA

Detailing long-term functional morbidity after resecting mobile spine chordomas across 35 patients.

OBJECTIVE: Surgery for mobile spine chordomas poses a challenge as en bloc resection often requires nerve root resection. Decision-making is challenging for patients because of this inherent high risk of postoperative deficits, and data demonstrating long-term postoperative functional outcomes are lacking. Here, the authors aimed to show the postoperative functional morbidity of a large series of patients with mobile spine chordomas and detail what was achieved intraoperatively and what nerve root(s) were deemed appropriate for sacrifice in each case. METHODS: A retrospective analysis was con

Decitabine Reprograms Temozolomide-Resistant Glioblastoma Through Epigenetic Reactivation and Mesenchymal Attenuation: A Multi-Omics Study.

Background/Objectives: Glioblastoma (GBM) is the most lethal primary brain malignancy in adults, with a median overall survival of approximately 15 months. Temozolomide (TMZ) resistance develops in virtually all patients, and no second-line regimen has improved outcomes over the past two decades. The DNA methyltransferase inhibitor decitabine (DAC) has attracted interest as a chemosensitizer, but whether it directly reverses the TMZ-resistance transcriptome or operates through distinct, complementary mechanisms has not been tested at multi-omics resolution. Methods: We performed an integrative

Exploring the potential causal relationship between lipid metabolism and oral carcinoma in situ: A 2-sample Mendelian randomization study.

Oral carcinoma in situ (OCIS) is a Stage 0 precancerous condition with a significant risk of developing into invasive oral squamous cell carcinoma. Despite this, the metabolic processes driving its onset and progression remain largely unknown. Lipid metabolism, a crucial factor in the development and behavior of various cancers, has been extensively studied. However, its exact role in OCIS has not been clearly defined, highlighting the need for further research in this area. The present study is designed to investigate the potential causal associations between specific lipid metabolic markers

Education Research: Educational Informatics: Characterizing Child Neurology Resident Clinical Experiences Using Electronic Health Record Data.

BACKGROUND AND OBJECTIVES: Gaps in preparedness for independent practice are common among recent graduates. Clinical experiences serve as the foundation for learning during residency, yet the breadth and variability of clinical exposure of child neurology residents remain poorly characterized. To inform programmatic curricular development and precision medical education (PME), we must better characterize learners' clinical exposure during training. We aimed to use electronic health record (EHR) data to catalog resident clinical experiences, linking diagnostic codes to educational benchmarks.

Genetic and epidemiological analyses of alcohol consumption patterns and age-related macular degeneration risk among current drinkers: a role for TNFRSF10A.

BACKGROUND: Alcohol intake, a major modifiable lifestyle factor, has been variably associated with age-related macular degeneration (AMD). We aim to investigate the association and potential causal relevance of alcohol consumption with AMD risk using complementary epidemiological and genetic approaches. METHODS: We used nonlinear Cox proportional hazards models to evaluate incident AMD associations among 440 052 participants of European ancestry with a mean age of 56.7 years and of whom 53.7% were female. We employed two-sample Mendelian randomisation (MR), integrating summary-level genetic a

CFD+CD14+ monocytes: potential pathogenic subset in myasthenia gravis uncovered by multi-omics integration and machine learning analysis.

BACKGROUND: Transcriptome-wide association study (TWAS) contributes to discovering novel susceptibility genes related to diseases. Single-cell RNA sequencing (scRNA-seq) has been extensively applied to characterize cellular heterogeneity in various biological and pathological conditions. However, few studies have integrated TWAS and scRNA-seq to decode the pathogenesis of myasthenia gravis (MG) at cellular resolution. METHODS: We integrated multi-omics data from GEO, genome-wide association study (GWAS) catalog, and Genotype-Tissue Expression (GTEx) databases, including scRNA-seq data, GWAS s

Evolution-guided prioritization identifies a tissue-specific phosphorylation switch on herpes simplex virus 1 UL7 regulating viral replication and pathogenicity.

UNLABELLED: Although numerous phosphorylation sites on herpes simplex virus 1 (HSV-1) proteins have been identified through phosphoproteomic analyses, their virological significance remains largely unclear. Here, we developed an evolution-guided prioritization strategy that integrates phosphoproteomic data with residue-level conservation across the genus Simplexvirus to identify phosphorylation sites as candidates for functional relevance. Applying this strategy to a previously reported phosphoproteomic data set, we identified 46 phosphorylation sites with a conservation level of 88% or higher

Limited contribution of compensatory mutations to MDR/RR-TB clustering in Hunan Province, China: a population-based whole-genome sequencing study.

Transmission of rifampicin-resistant and multidrug-resistant tuberculosis (MDR/RR-TB) is a major driver of the global drug-resistant TB epidemic, yet the contribution of compensatory mutations to its spread remains uncertain. We analyzed 206 non-duplicate MDR/RR-TB isolates collected from five surveillance sites in Hunan Province, China, between 2013 and 2020 using whole-genome sequencing. Transmission clusters were defined by ≤12 single-nucleotide polymorphism (SNP) differences, and compensatory mutations in rpoA, rpoB, and rpoC were identified through a curated mutation catalog and phylogene

Understanding the ancient classic and famous prescriptions via the property of Chinese materia medica.

BACKGROUND: Ancient classic and famous prescriptions (ACFPs), derived from traditional Chinese medicine (TCM) classics, are widely utilized due to their precise therapeutic effects and distinctive clinical advantages. Existing research predominantly focuses on individual prescriptions, and there is lack of systematic exploration of medication patterns within the official ACFPs catalog. The property of Chinese materia medica (PCMM), a multidimensional representation of medicinal properties, offers a novel perspective for systematically analyzing TCM formulas. OBJECTIVE: In this study, we aim t

Immune Cells as Mediators of Lipidome Influence on Osteoporosis: Evidence from a Mediation Analysis.

Background: Although clinical studies have indicated a possible association between dyslipidemia and osteoporosis, the underlying genetic basis and mechanistic pathways remain insufficiently defined. Most prior research has concentrated on conventional lipid markers, which are prone to confounding and limit causal inference. Exploring lipidomic profiles offers a more comprehensive view of lipid metabolism and may reveal novel genetic links beyond traditional lipid traits. Additionally, alterations in immune cell function, often triggered by metabolic disturbances, may contribute to osteoporosi

The role of N6-methyladenosine (m6A) RNA methylation modification in kidney diseases: from mechanism to therapeutic potential.

BACKGROUND: Kidney disease is a major global health issue, causing numerous deaths and a loss of life years. This prompts us to explore potential targets or mechanisms that may increase the likelihood of diagnosing and treating kidney diseases. N6-methyladenosine (m6A) modifications dynamically regulate RNA through "writer" enzymes, "eraser" enzymes, and "reader" proteins, influencing its processing, stability, and translation efficiency. In cases of kidney disease, there is a likelihood that m6A methylation is a significant contributor to the pathological mechanisms of acute kidney injury (AK

Genetically predicted plasma metabolites mediate the causal relationship between gut microbiota and primary immune thrombocytopenia (ITP).

BACKGROUND: Primary immune thrombocytopenia (ITP) is an immune-mediated hematologic disorder characterized by a reduction in platelet count, increasing the risk of bleeding. Recent studies have indicated a close association between alterations in gut microbiota and the development of ITP. However, the mechanisms by which gut microbiota influence the occurrence and progression of ITP through plasma metabolites remain poorly understood. Evidence suggests extensive interactions between gut microbiota and plasma metabolites, implying a potential role for gut microbiota in influencing ITP through a
