Fabrication and Surgical Application of Enriched Photo-crosslinked Gelatin-Riboflavin Hydrogels for Corneal Wound Repair in Rabbits.

This article presents a step‑by‑step protocol to fabricate, supplement, and surgically apply an in situ blue‑light-crosslinkable gelatin hydrogel for corneal stromal wound repair in rabbits. The hydrogel precursor comprises gelatin at 5% (w/v) and riboflavin phosphate at 0.01% (w/v), prepared under sterile and light‑protected conditions, sterile‑filtered, and warmed before use to achieve injectable viscosity. Optional incorporation of human amniotic membrane extract or rabbit autologous serum is detailed. On‑demand gelation is triggered on the ocular surface using blue light at λ = 420-480 nm

Biomechanical, Visual, and Refractive Outcomes of Total Corneal Wavefront-Guided Transepithelial PRK With Accelerated CXL in Keratoconus Using Objective Stability Markers.

PURPOSE: To evaluate the long-term safety and efficacy of simultaneous total corneal wavefront-guided transepithelial photorefractive keratectomy (TCWG-TransPRK) with accelerated corneal cross-linking (CXL) in mild to moderate keratoconus, incorporating corneal biomechanics as objective markers of structural stability. METHODS: This prospective, interventional, single-center study included patients with mild to moderate keratoconus. All underwent TCWG-TransPRK (maximum planned stromal ablation: 55 µm) followed by accelerated CXL (30 mW/cm2 for 3 minutes; total fluence: 5.4 J/cm2). Follow-up w

BACKGROUND: This study assessed patient-reported outcomes (PRO) of active-duty U.S. Military Servicemembers following refractive surgery. METHODS: We retrospectively reviewed the medical records of 375 U.S. Servicemembers who underwent LASIK (118 eyes), photorefractive keratectomy (PRK, 550 eyes), or small incision lenticule extraction (SMILE, 82 eyes). Surgeries occurred at the former Walter Reed Army Medical Centers Center for Refractive Surgery or the current FBCHs Warfighter Refractive Eye Surgery Program and Research Center (WRESP-RC) from 2004 to 2019. Preoperative and 6-month postopera

Genotype-Phenotype Correlations and Long-Term Surgical Outcomes in TGFBI-Linked Bowman's Layer Corneal Dystrophies.

PURPOSE: Reis-Bücklers corneal dystrophy (RBCD; Mendelian Inheritance in Man 602082) and Thiel-Behnke corneal dystrophy (TBCD; Mendelian Inheritance in Man 608470) are clinically similar corneal dystrophies affecting Bowman's layer, caused by the TGFBI variants p.(Arg124Leu) and p.(Arg555Gln), respectively. Yet their distinct clinical courses complicate therapeutic decisions. This study aimed to compare the clinical course and surgical outcomes of genetically confirmed RBCD and TBCD. METHODS: Medical records from 2016 to 2024 were reviewed for age at first surgical intervention, surgical proc

Dopamine Receptor 1 Treatment Promotes Epithelial Repair of Corneal Injury by Inhibiting NOD-Like Receptor Protein 3-Associated Inflammation.

PURPOSE: To elucidate the influence of dopamine receptor 1 (DRD1) on the proliferation of mouse corneal epithelial cells (MCECs) under inflammatory conditions. METHODS: In vitro, immortalized MCECs (iMCECs) were treated with IL-1β, with and without pcDNA3.1_DRD1. Primary MCECs (pMCECs) were exposed to IL-1β, with and without DRD1 agonist (A68930). Cell proliferation was quantified using the Cell Counting Kit-8 (CCK-8) assay and immunofluorescence staining for Ki-67 and p63. Expression levels of NOD-like receptor protein 3 (NLRP3), IL-1β, and IL-6 were assessed. To establish a corneal injury m

PURPOSE: Assessment of tomographic characteristics and interdevice comparability between Scheimpflug imaging (Pentacam HR, Oculus Optikgeräte GmbH, Wetzlar, Germany) and swept-source optical coherence tomography (CASIA2, Tomey Corp., Nagoya, Japan) in eyes with macular corneal dystrophy (MCD). METHODS: Eyes with MCD were examined by Pentacam HR and CASIA2. Interdevice comparison was performed using a Wilcoxon matched pairs test and Bland-Altman plots with 95% limit of agreement. A Spearman rank correlation coefficient was used for correlating indices of both devices. RESULTS: This retrospect

Mice Deficient in TAZ (Wwtr1) Demonstrate Clinical Features of Late-Onset Fuchs' Endothelial Corneal Dystrophy.

PURPOSE: We sought to define the role of Wwtr1 in murine ocular structure and function and determine the role of mechanotransduction in Fuchs' endothelial corneal dystrophy (FECD), with emphasis on interactions between corneal endothelial cells (CEnCs) and Descemet's membrane (DM). METHODS: A Wwtr1 deficient mouse colony was established, and advanced ocular imaging, atomic force microscope (AFM), and histology/immunofluorescence were performed. Corneal endothelial wound healing was assessed using cryoinjury and phototherapeutic keratectomy in Wwtr1 deficient mice. Expression of WWTR1/TAZ was

Vector Analysis of Changes in the Higher Order Ocular Aberrations and Central Corneal Thickness After T-PRK and Fs-LASIK.

INTRODUCTION: Refractive surgery procedures, transepithelial photorefractive keratectomy (T-PRK) and femtosecond laser in situ keratomileusis (Fs-LASIK) are regarded as safe and efficacious methods for correcting myopia and myopic astigmatism. These two methods do not have much differences in results when treating spherical myopia, while some differences does exist in treatment of myopic astigmatism. Vector analysis presents powerful tool to show the real differences between these two methods regarding higher order ocular aberrations and central corneal thickness of treated eyes. AIM: The aim
