Biological Control of Chili Damping-Off Disease, Caused by Pythium myriotylum.

Pythium myriotylum is a notorious soil-borne oomycete that causes post-emergence damping-off in chili pepper. Of various disease management strategies, utilization of plant growth promoting rhizobacteria (PGPR) in disease suppression and plant growth promotion is an interesting strategy. The present study was performed to isolate and characterize PGPR indigenous to the chili rhizosphere in Pakistan, and to test the potential to suppress the damping-off and plant growth promotion in chili. Out of a total of 28 antagonists, eight bacterial isolates (4a2, JHL-8, JHL-12, 1C2, RH-24, 1D, 5C, and RH

Fate of nitrogen-fixing bacteria in crude oil contaminated wetland ultisol.

The effect of crude oil on the growth of legumes (Calopogonium muconoides and Centrosema pubescens) and fate of nitrogen-fixing bacteria in wetland ultisol was investigated using standard cultural techniques. The results revealed observable effects of oil on soil physico-chemistry, plant growth and nodulation as well as on densities of heterotrophic, hydrocarbonoclastic and nitrogen fixing bacteria. The effects however varied with different levels (0.5%, 1%, 5%, 10%, 15% and 20%) of pollution. Ammonium and nitrate levels were high in the unpolluted soil but decreased with increase in pollution

The preprotein translocase YajC mediates biofilm formation and symbiotic nitrogen-fixation in Mesorhizobium huakuii.

UNLABELLED: In bacteria, the auxiliary complex SecDF-YajC participates in the Sec system, facilitating post-translational translocation of preproteins across the membrane through the SecYEG channel. Although YajC homologs are strongly conserved in rhizobial strains and may play important roles in many processes, the functions and mechanisms by which they are involved in the interaction between rhizobia and host legumes are unknown. By constructing Mesorhizobium huakuii yajC mutant, we observed an increase in the strain growth rate, but a reduction in both the biomass and average thickness of t

Protocol for Quantitative Estimation of Hydrogen Cyanide Production from Bacteria.

Hydrogen cyanide (HCN) is a volatile, nitrogen-containing secondary metabolite produced by various bacterial species, primarily during the idiophase of growth under nutrient-limiting or competitive conditions. It plays a significant ecological role as a biocontrol agent by inhibiting the respiratory enzymes of plant pathogens and modulating microbial competition in the rhizosphere. Although protocols for detecting HCN production have existed for over a century, they have largely remained qualitative and are rarely optimized for quantitative assessment. This is mainly due to the volatile nature

Herbicide-treated soil as a reservoir of beneficial bacteria: microbiome analysis and PGP bioinoculants in maize.

BACKGROUND: Herbicides are integral to agricultural weed management but can adversely affect non-target organisms, soil health, and microbiome. We investigated the effects of herbicides on the total soil bacterial community composition using 16S rRNA gene amplicon community profiling. Further, we aimed to identify herbicide-tolerant bacteria with plant growth-promoting (PGP) capabilities as a mitigative strategy for these negative effects, thereby promoting sustainable agricultural practices. RESULTS: A bacterial community analysis explored the effects of long-term S-metolachlor application o

Kribbellichelins A and B, Two New Antibiotics from Kribbella sp. CA-293567 with Activity against Several Human Pathogens.

Current needs in finding new antibiotics against emerging multidrug-resistant superbugs are pushing the scientific community into coming back to Nature for the discovery of novel active structures. Recently, a survey of halophilic actinomyectes from saline substrates of El Saladar del Margen, in the Cúllar-Baza depression (Granada, Spain), led us to the isolation and identification of 108 strains from the rhizosphere of the endemic plant Limonium majus. Evaluation of the potential of these strains to produce new anti-infective agents against superbug pathogens was performed through fermentatio

Enhancing Phytate Availability in Soils and Phytate-P Acquisition by Plants: A Review.

Phytate (myo-inositol hexakisphosphate salts) can constitute a large fraction of the organic P in soils. As a more recalcitrant form of soil organic P, up to 51 million metric tons of phytate accumulate in soils annually, corresponding to ∼65% of the P fertilizer application. However, the availability of phytate is limited due to its strong binding to soils via its highly-phosphorylated inositol structure, with sorption capacity being ∼4 times that of orthophosphate in soils. Phosphorus (P) is one of the most limiting macronutrients for agricultural productivity. Given that phosphate rock is a

Polyhydroxyalkanoate (PHA) Polymer Accumulation and pha Gene Expression in Phenazine (phz⁻) and Pyrrolnitrin (prn⁻) Defective Mutants of Pseudomonas chlororaphis PA23.

Pseudomonas chlororaphis PA23 was isolated from the rhizosphere of soybeans and identified as a biocontrol bacterium against Sclerotinia sclerotiorum, a fungal plant pathogen. This bacterium produces a number of secondary metabolites, including phenazine-1-carboxylic acid, 2-hydroxyphenazine, pyrrolnitrin (PRN), hydrogen cyanide, proteases, lipases and siderophores. It also synthesizes and accumulates polyhydroxyalkanoate (PHA) polymers as carbon and energy storage compounds under nutrient-limited conditions. Pseudomonads like P. chlororaphis metabolize glucose via the Entner-Doudoroff and Pen

Spores and soil from six sides: interdisciplinarity and the environmental biology of anthrax (Bacillus anthracis).

Environmentally transmitted diseases are comparatively poorly understood and managed, and their ecology is particularly understudied. Here we identify challenges of studying environmental transmission and persistence with a six-sided interdisciplinary review of the biology of anthrax (Bacillus anthracis). Anthrax is a zoonotic disease capable of maintaining infectious spore banks in soil for decades (or even potentially centuries), and the mechanisms of its environmental persistence have been the topic of significant research and controversy. Where anthrax is endemic, it plays an important eco

Transporter promiscuity and redox-driven metal partitioning in plant responses to chemically analogous metals.

Chemically similar metal(loid)s exploit nutrient transport systems and destabilize integrated metal-homeostasis networks, triggering redox imbalance, transcriptional reprogramming, and multiscale regulatory responses that ultimately determine plant adaptation or toxicity. Plant growth depends on the homeostasis of mineral nutrients, but it is short in heterogeneous soils where required elements are found together with chemically similar harmful metal(loid)s. The divalent ionic and coordination properties of Cd-Zn and Ni-Fe are similar, while the arsenate-phosphate interactions are structurally

Plant-growth promotion by proteobacterial strains depends on the availability of phosphorus and iron in Arabidopsis thaliana plants.

Phosphorus (as phosphate, Pi) and iron (Fe) are critical nutrients in plants that are often poorly available in the soil and can be microbially affected. This work aimed to evaluate how plant-rhizobacteria interaction changes due to different Pi or Fe nutritional scenarios and to study the underlying molecular mechanisms of the microbial modulation of these nutrients in plants. Thus, three proteobacteria (Paraburkholderia phytofirmans PsJN, Azospirillum brasilense Sp7, and Pseudomonas putida KT2440) were used to inoculate Arabidopsis seeds. Additionally, the seeds were exposed to a nutritional

Bacteria associated with oak and ash on a TCE-contaminated site: characterization of isolates with potential to avoid evapotranspiration of TCE.

BACKGROUND, AIM, AND SCOPE: Along transects under a mixed woodland of English Oak (Quercus robur) and Common Ash (Fraxinus excelsior) growing on a trichloroethylene (TCE)-contaminated groundwater plume, sharp decreases in TCE concentrations were observed, while transects outside the planted area did not show this remarkable decrease. This suggested a possibly active role of the trees and their associated bacteria in the remediation process. Therefore, the cultivable bacterial communities associated with both tree species growing on this TCE-contaminated groundwater plume were investigated in o

Qualitative and quantitative evaluation of phosphate-solubilizing ability of Lysinibacillus macroides: a climate-resilient biofertilizer candidate for sustainable crop nutrition.

Phosphorus (P) is a critical macronutrient governing plant productivity, however its bioavailability in agricultural soils remains severely constrained due to fixation in insoluble mineral complexes, particularly calcium-bound phosphates in alkaline systems. The low use efficiency of chemical fertilizers, coupled with escalating environmental concerns, necessitates biologically driven strategies for sustainable phosphorus management. In this study, rhizospheric bacteria associated with chili (Capsicum annuum L.) were systematically investigated to elucidate the phosphate-solubilizing potential

Genomic characterization and niche adaptive analysis of Pseudomonas promysalinigenes W2469: the first clinical isolate from a human bile specimen.

BACKGROUND: Pseudomonas promysalinigenes is a newly described bacterial species renowned for producing promysalin, a species-selective lipopeptide antibiotic. All previously reported strains of this species are derived from environmental niches such as plant rhizospheres, and no clinical infection cases associated with this bacterium have been documented to date. Thus, the clinical microbiology relevance, genomic features, and adaptive potential of P. promysalinigenes remain largely unexplored, and current reference databases have limited coverage of this rare species. Enterococcus faecalis OG1RF Evolution at Low pH Selects Fusidate-Sensitive Mutants in Elongation Factor G and at High pH Selects Defects in Phosphate Transport.

Enterococcus bacteria inhabit human and soil environments that show a wide range of pH values. Strains include commensals as well as antibiotic-resistant pathogens. We investigated the adaptation to pH stress in E. faecalis OG1RF by conducting experimental evolution under acidic (pH 4.8), neutral pH (pH 7.0), and basic (pH 9.0) conditions. A serial planktonic culture was performed for 500 generations and in a high-pH biofilm culture for 4 serial bead transfers. Nearly all of the mutations led to nonsynonomous codons, indicating adaptive selection. All of the acid-adapted clones from the plankt
