Prevalence of Intestinal Parasitic Infections and Associated Factors Among Food Handlers in Wolaita Sodo University Students Caterings, Wolaita Sodo, Southern Ethiopia: A Cross-Sectional Study.

Background: Intestinal Parasitic Infections (IPIs) are among the most prevalent diseases in the world, particularly in developing countries with low socio-economic and poor living conditions. From the estimated one third global population infected by intestinal parasitic infections; the majority lives in tropical and sub-tropical parts of the world. These diseases are highly prevalent in Ethiopia. However, its magnitude in the context of Wolaita Sodo University, where a large number of students reside in a campus is not studied. Therefore, the objective of this study was to assess the prevalen

Enzymatic chokepoints and synergistic drug targets in the sterol biosynthesis pathway of Naegleria fowleri.

Naegleria fowleri is a free-living amoeba that can also act as an opportunistic pathogen causing severe brain infection, primary amebic meningoencephalitis (PAM), in humans. The high mortality rate of PAM (exceeding 97%) is attributed to (i) delayed diagnosis, (ii) lack of safe and effective anti-N. fowleri drugs, and (iii) difficulty of delivering drugs to the brain. Our work addresses identification of new molecular targets that may link anti-Naegleria drug discovery to the existing pharmacopeia of brain-penetrant drugs. Using inhibitors with known mechanism of action as molecular probes, we

Differential Growth of Francisella tularensis, Which Alters Expression of Virulence Factors, Dominant Antigens, and Surface-Carbohydrate Synthases, Governs the Apparent Virulence of Ft SchuS4 to Immunized Animals.

The gram-negative bacterium Francisella tularensis (Ft) is both a potential biological weapon and a naturally occurring microbe that survives in arthropods, fresh water amoeba, and mammals with distinct phenotypes in various environments. Previously, we used a number of measurements to characterize Ft grown in Brain-Heart Infusion (BHI) broth as (1) more similar to infection-derived bacteria, and (2) slightly more virulent in naïve animals, compared to Ft grown in Mueller Hinton Broth (MHB). In these studies we observed that the free amino acids in MHB repress expression of select Ft virulence

Incidence and Risk Factors for Incident Syphilis among HIV-1-Infected Men Who Have Sex with Men in a Large Urban HIV Clinic in Tokyo, 2008-2015.

BACKGROUND: The epidemiology of incident syphilis infection among HIV-1-infected men who have sex with men (MSM) largely remains unknown. METHODS: The incidence and risk factors for incident syphilis (positive TPHA and RPR> = 1:8) among HIV-1-infected MSM who visited a large HIV clinic in Tokyo for the first time between 2008 and 2013 were determined, using clinical data and stored blood samples taken every three months for screening and determination of the date of incident syphilis. Poisson regression compared the incidence of syphilis at different observation periods. RESULTS: Of 885 HIV-

The role of Src kinase in the biology and pathogenesis of Acanthamoeba castellanii.

BACKGROUND: Acanthamoeba species are the causative agents of fatal granulomatous encephalitis in humans. Haematogenous spread is thought to be a primary step, followed by blood-brain barrier penetration, in the transmission of Acanthmaoeba into the central nervous system, but the associated molecular mechanisms remain unclear. Here, we evaluated the role of Src, a non-receptor protein tyrosine kinase in the biology and pathogenesis of Acanthamoeba. METHODS: Amoebistatic and amoebicidal assays were performed by incubating amoeba in the presence of Src kinase-selective inhibitor, PP2 (4-amino-5

Public health and clinical importance of amoebiasis in Malaysia: a review.

Entamoeba histolytica, the causative agent of human amoebiasis remains a significant cause of morbidity and mortality in developing countries and is responsible for up to 100,000 deaths worldwide each year. Entamoeba dispar, morphologically indistinguishable from E. histolytica is more common in humans in many parts of the world. Similarly Entamoeba moshkovskii, which was long considered to be a free-living amoeba is also morphologically identical to E. histolytica and E. dispar, and is highly prevalent in some E. histolytica endemic countries. Humans are the host of infection and there would

[Epidemic of autochthonous hepatic and intestinal amebiasis in a place near Grenoble].

We had the opportunity of studying an epidemic of autochthonous amoebiasis occurring in the autumn of 1974 in a small town of 4000 inhabitants 30 km from Grenoble. Attention was originally attracted by the occurrence in this town of two cases of hepatic amoebiasis and one of intestinal amoebiasis identified by rectoscopy. Systematic investigations (coproctic examinations and serological tests for amoebiasis by indirect antibody fluorescence) were then carried out on everyone in the locality with digestive disorders which were possibly referable to amoebiasis, and on the other members of their

Comparative morphology and phagocytic capacity of primary human adult microglia with time-lapse imaging.

Microglia provide immune surveillance within the brain and spinal cord. Various microglial morphologies include ramified, amoeboid, and pseudopodic. The link between form and function is not clear, especially for human adult microglia which are limited in availability for study. Here, we examined primary human microglia isolated from normal-appearing white matter. Pseudopodic and amoeboid microglia were effective phagocytes, taking up E. coli bioparticles using ruffled cell membrane sheets and retrograde transport. Pseudopodic and amoeboid microglia were more effective phagocytes as compared t

Phage/nanoparticle cocktails for a biocompatible and environmentally friendly antibacterial therapy.

Antibiotic resistance continues to rise, necessitating alternative strategies. Bacteriophages have emerged as promising natural antibacterial agents, offering a targeted approach to combating bacterial infections. Combining bacteriophages with nanoparticles presents a novel approach that could enhance antibacterial potency while reducing the risk of resistance. While phage/antibiotic cocktails are widely explored to enhance antibacterial efficacy and prevent resistance, research on phage/nanoparticle combinations remains limited. We explore the synergy between green tea extract-capped silver n

Indigenous knowledge and nutritional and morphological characterization of okra (Abelmoschus esculentus (L.) Moecnh) varieties in Western Ethiopia.

Okra (Abelmoschus esculentus (L.) Moecnh) is a versatile crop that is widely grown in western Ethiopia, despite that it is less researched. This study, therefore, investigates the traditional knowledge, nutritional, and morphological characteristics of two commonly grown okra varieties (Kenketse and Sharma) in west Ethiopia. A semi-structured questionnaire, checklist, and observations were used to collect primary data from households, key informants, and farms. In addition, proximate composition parameters like moisture content, total ash, crude fat, crude protein, crude fiber, utilizable carb

Evaluation of the C-Terminal Fragment of Entamoeba histolytica Gal/GalNAc Lectin Intermediate Subunit as a Vaccine Candidate against Amebic Liver Abscess.

BACKGROUND: Entamoeba histolytica is an intestinal protozoan parasite that causes amoebiasis, including amebic dysentery and liver abscesses. E. histolytica invades host tissues by adhering onto cells and phagocytosing them depending on the adaptation and expression of pathogenic factors, including Gal/GalNAc lectin. We have previously reported that E. histolytica possesses multiple CXXC sequence motifs, with the intermediate subunit of Gal/GalNAc lectin (i.e., Igl) as a key factor affecting the amoeba's pathogenicity. The present work showed the effect of immunization with recombinant Igl on

Designing Synthetic Microcapsules That Undergo Biomimetic Communication and Autonomous Motion.

Inspired by the collective behavior of slime molds and amoebas, we designed synthetic cell-like objects that move and self-organize in response to self-generated chemical gradients, thereby exhibiting autochemotaxis. Using computational modeling, we specifically focused on microcapsules that encompass a permeable shell and are localized on an adhesive surface in solution. Lacking any internal machinery, these spherical, fluid-filled shells might resemble the earliest protocells. Our microcapsules do, however, encase particles that can diffuse through the outer shell and into the surrounding fl

In vitro inhibition of protease-activated receptors 1, 2 and 4 demonstrates that these receptors are not involved in an Acanthamoeba castellanii keratitis isolate-mediated disruption of the human brain microvascular endothelial cells.

Granulomatous amoebic encephalitis is a rare but serious human disease leading almost always to death. The pathophysiology of amoebic encephalitis is better understood, while events leading to the constitution of brain infection are largely unknown. Traversal of the blood-brain barrier is a key step in amoebae invasion of the central nervous system and facilitated by amoebic extracellular proteases. By using specific inhibitors of protease-activated receptors 1, 2 and 4, here we studied the role of these host receptors in Acanthamoeba castellanii-mediated damage to human brain microvasculature
