Genome Mining of Deep-Sea Cold Seep-Derived Fungus Reveals a Laccase-Fasciclin System Modulating Regioselective Naphthopyranone Dimerization.

Naphthopyranones represent a structurally diverse family of fungal polyketides exhibiting a broad range of biological activities, including antibacterial, antifungal, and cytotoxic properties. Despite extensive investigations of terrestrial-derived naphthopyranones, the biosynthetic machinery responsible for their production in marine fungi has remained unexplored. Here, we report the first characterization of naphthopyranone biosynthetic gene clusters (BGCs) from a deep-sea-derived fungus. Genome mining of the cold seep-associated Penicillium javanicum OUCF108 revealed two highly homologous p

N-terminal lipidation enhances membrane interaction and antifungal activity of a Trematocine-derived decapeptide.

Invasive fungal infections are a growing global health threat, driven by the limited availability of antifungal therapies and the rapid spread of multidrug-resistant yeasts. This study explored the effects of N-terminal lipidation on Trem-4, a decapeptide derived from the antimicrobial peptide Trematocine-HSK, to enhance its antifungal potential. Trem-4 was modified with caprylic (C8) and myristic (C14) fatty acid chains, generating two lipopeptides: Cap-T4 and Myr-T4. Biophysical analyses, including fluorescence-based assays and circular dichroism, showed that lipidation significantly improve

The Upcoming Antifungal Drugs in Clinical Development for the Treatment of Invasive Candidiasis.

BACKGROUND: Invasive candidiasis (IC) is a major cause of morbidity and mortality among hospitalized patients, and treatment success relies on early diagnosis and prompt initiation of an appropriate antifungal agent. The increasing rates of resistance to first-line antifungal agents (azoles, echinocandins, and polyenes), along with changes in the epidemiology of IC and the spread of multidrug-resistant C. auris, pose further challenges for the management of IC. Limited therapeutic options and routes of administration, drug-drug interactions, and drug toxicities of commonly utilized antifungals

Bioactive Lyocell Fibers with Inherent Antibacterial, Antiviral and Antifungal Properties.

Functional Lyocell fibers gain interest in garments and technical textiles, especially when equipped with inherently bioactive features. In this study, Lyocell fibers are modified with an ion exchange resin and subsequently loaded with copper (Cu) ions. The modified Lyocell process enables high amounts of the resin additive (>10%) through intensive dispersion and subsequently, high uptake of 2.7% Cu throughout the whole cross-section of the fiber. Fixation by Na2CO3 increases the washing and dyeing resistance considerably. Cu content after dyeing compared to the original fiber value amounts to

Persistence, Source Control, and Secondary Bacterial Bloodstream Infection in Catheter-Related Candidemia: A Retrospective Cohort Study.

BACKGROUND: In catheter-related candidemia, a true catheter-source infection is often difficult to distinguish from a catheter that merely marks critical illness. Our primary aim was to identify predictors of 30-day mortality; secondary aims were to characterize species-specific patterns of persistent candidemia, its prognostic meaning within the host immune-inflammatory context, and the relationship between echinocandin therapy and secondary bacterial bloodstream infection (BSI). METHODS: We retrospectively reviewed 89 consecutive episodes of catheter-related candidemia at Bursa City Hospita

Insights into invasive fungal infection diagnostic and treatment capacities in tertiary care centres of Germany.

INTRODUCTION: In Germany, the growing incidence of invasive fungal infections (IFIs) is a significant health concern, particularly impacting individuals with compromised immune systems due to factors like increasing transplant recipients, an ageing population, and heightened use of immunosuppressive medications. Diagnosing IFI remains challenging, and the integration of biomarker assays into clinical practice is difficult. Antifungal resistance, exemplified by pan-antifungal-resistant Candida auris cases, adds complexity to treatment. This study aims to provide a concise overview of the diagno

Discovery of the antifungal compound ilicicolin K through genetic activation of the ilicicolin biosynthetic pathway in Trichoderma reesei.

BACKGROUND: Given the global rise in antimicrobial resistance, the discovery of novel antimicrobial agents and production processes thereof are of utmost importance. To this end we have activated the gene cluster encoding for the biosynthesis of the potent antifungal compound ilicicolin H in the fungus Trichoderma reesei. While the biosynthetic gene cluster (BGC) is silent under standard cultivation conditions, we achieved BGC activation by genetically overexpressing the transcription factor TriliR. RESULTS: Successful activation was confirmed by RT-qPCR, proteomic and metabolomic analyses. M

Fungal extracellular vesicles mediate conserved cross-species communication and immunomodulation.

Extracellular vesicles (EVs) play crucial roles in fungal communication and host immune modulation, representing potential therapeutic targets for fungal infections. This study investigated the role of fungal EVs in both intra- and interspecies communication, focusing on their effects on virulence and immune responses. Co-incubation experiments were performed using EVs derived from Candida albicans and Candidozyma auris to assess interactions with C. albicans planktonic cells and biofilms, as well as Cryptococcus neoformans and Cryptococcus gattii EVs interacting with C. neoformans cultures. Bioactive glass S53P4 cream kills ESKAPE panel multidrug resistant pathogens and Staphylococcus aureus biofilms.

INTRODUCTION: Orthopedic implant-associated infections, predominantly caused by S. aureus, pose significant challenges due to biofilm formation and antibiotic resistance. Bioactive Glass (BAG) S53P4 is a unique material with antimicrobial and bone regenerative properties. We aimed to characterize a novel BAG S53P4 cream, consisting of BAG powder and a binder, for its capacity to kill Staphylococcus aureus in suspension and biofilms in the absence or presence of titanium implant material. Since the BAG antimicrobial activity depends on ions eluted, we also analyzed the eluates of the cream and

Candidaemia and Central Line-Associated Candidaemia in a Network of Indian ICUs: Impact of COVID-19 Pandemic.

BACKGROUND AND OBJECTIVES: Candidaemia is a potentially life-threatening emergency in the intensive care units (ICUs). Surveillance using common protocols in a large network of hospitals would give meaningful estimates of the burden of candidaemia and central line associated candidaemia in low resource settings. We undertook this study to understand the burden and epidemiology of candidaemia in multiple ICUs of India, leveraging the previously established healthcare-associated infections (HAI) surveillance network. Our aim was also to assess the impact that the pandemic of COVID-19 had on the
