Characterization and annotation of Babesia orientalis apicoplast genome.

BACKGROUND: Babesia orientalis is an obligate intraerythrocytic protozoan parasite of the buffalo (Bubalus bubalis, Linnaeus, 1758) transmitted by the tick Rhipicephalus heamaphysaloides. It is the causative agent of water buffalo babesiosis, one of the most important pathogens of water buffalo in central and southern China. As a member of the phylum Apicomplexa, B. orientalis possesses a relatively independent and alga originated organelle the apicoplast. Apicoplasts in other apicomplexa parasites are involved in the biosynthesis of haem, fatty acids, iron-sulphur clusters and isoprenoids. So

Evidence for an increased geographical distribution of Dermacentor reticulatus in Germany and detection of Rickettsia sp. RpA4.

Two studies were performed to elucidate the current distribution of the tick Dermacentor reticulatus in Germany. In the first one in 2003, a total of 365 dogs from 171 sites in the states of Berlin and Brandenburg was screened for ticks, and the corresponding outdoor sites that the dogs usually visited were searched for host-seeking ticks by the flagging method. A total of 1155 ticks was removed from the dogs. The majority were Ixodes ricinus (88.5%), followed by D. reticulatus (9.1%) and I. hexagonus (2.4%). Altogether, 222 dogs carried I. ricinus (60.8%), 41 D. reticulatus (11.2%) and 15 I.

[Two cases of spotted fever group rickettsiosis contracted in southern parts of Africa].

A 40-year-old Japanese male stayed in Zimbabwe and developed a fever above 38 degrees C during which he noted a typical eschar in the lumbar region and also regional inguinal lymphadenopathy. Although not conspicuous, erythematous eruptions accompanied by itching were observed on the face, trunk and lower extremities. After returning to Japan and visiting our hospital, he was suspected of rickettsiosis and put on minocycline which gradually led to the improvement of the symptoms. Immunofluorescence antibody determinations disclosed rising titers against Rickettsia conorii (R. conorii) for both

The Namaqua rock mouse (Micaelamys namaquensis) as a potential reservoir and host of arthropod vectors of diseases of medical and veterinary importance in South Africa.

BACKGROUND: The role of endemic murid rodents as hosts of arthropod vectors of diseases of medical and veterinary significance is well established in the northern hemisphere. In contrast, endemic murids are comparatively understudied as vector hosts in Africa, particularly in South Africa. Considering the great rodent diversity in South Africa, many of which may occur as human commensals, this is unwarranted. METHODS: In the current study we assessed the ectoparasite community of a widespread southern African endemic, the Namaqua rock mouse (Micaelamys namaquensis), that is known to carry Bar

Hemorrhagic disease in dogs infected with an unclassified intraendothelial piroplasm in southern Brazil.

A hemorrhagic disease affecting dogs in Brazil, referred to popularly as "nambiuvú" (bloody ears) and believed to be transmitted by ticks, has been observed in animals infected with an organism described originally in 1910 as a piroplasm, and known locally as Rangelia vitalii. In this series of 10 cases, the disease was characterized by anaemia, jaundice, fever, spleno- and lymphadenomegaly, hemorrhage in the gastrointestinal tract, and persistent bleeding from the nose, oral cavity and tips, margins and outer surface of the pinnae. The ixodid ticks Rhipicephalus sanguineus and Amblyomma aureo

Hard tick infestation of birds of prey (Aves: Accipitriformes, Strigiformes, Falconiformes) in the Western Palearctic: New data and literature review.

Tick infestation of songbirds has been extensively studied at global and regional scales but relevant data on birds of prey remain scarce and fragmentary. Raptors are of particular epidemiological relevance because their large size, wide-ranging movements and nesting habits may facilitate long-distance dispersal of ticks, and endophilic tick species (e.g. from prey animals) may even persist in their re-used nests. In this study, we aimed to compensate for the lack of comprehensive data in this context. First, more than 1800 birds of prey were examined at 61 sites in Central and South-Eastern E

Potential spillover investigated by metagenome sequencing in Laikipia, Kenya reveals tick-borne pathogens and a novel bunyavirus.

Tick-borne infections continue to present a global public health threat, and require a One Health approach for successful mitigation. We conducted cross-sectional tick screening utilizing an agnostic metagenomic screening strategy based on nanopore sequencing (NS), in an area spanning a range of habitats with intensified human-livestock-wildlife interactions in central Kenya. We further used targeted amplification by polymerase chain reaction (PCR) and sequence independent single primer amplification (SISPA) for confirmation and genome characterization, as necessary. We initially screened 44 t

Predicting tick distributions in a changing climate: An ensemble approach for South Africa.

Predicting the potential distribution of disease vectors is crucial for vector management and disease transmission surveillance. This study aims to assess changes in the geographic projection of the ecological niche of ticks of veterinary, public health, and economic importance in South Africa, and to predict areas suitable for their establishment under current and future climate scenarios. We used a suite of six algorithms within the ensemble modelling framework of the biomod2 package in R version 4.4.2 to produce species distribution models for current (2021-2040) and future (2041-2060) clim

Prevalence of Borrelia burgdorferi in Ixodidae Tick around Asia: A Systematic Review and Meta-Analysis.

Lyme disease (LD) is a common arthropod-borne inflammatory disorder prevalent in the northern hemisphere. LD is caused by a spirochete named Borrelia burgdorferi s.l., which is transmitted to humans by ticks. Climate, environment, and other factors affect land use; recreational-behavior changes affect human contact with infected ticks. Studies in Europe and North America have looked at these aspects, but studies in Asia have not. We searched databases to identify all relevant abstracts published until March 2021. A meta-analysis was undertaken using the standard methods and procedures establis

Illuminating the bacterial microbiome of Australian ticks with 16S and Rickettsia-specific next-generation sequencing.

Next-generation sequencing (NGS) studies show that mosquito and tick microbiomes influence the transmission of pathogens, opening new avenues for vector-borne pathogen control. Recent microbiological studies of Australian ticks highlight fundamental knowledge gaps of tick-borne agents. This investigation explored the composition, diversity and prevalence of bacteria in Australian ticks (n = 655) from companion animals (dogs, cats and horses). Bacterial 16S NGS was used to identify most bacterial taxa and a Rickettsia-specific NGS assay was developed to identify Rickettsia species that were ind

Clinicopathological and molecular profiles of Babesia vogeli infection and Ehrlichia canis coinfection.

BACKGROUND AND AIM: Canine babesiosis, a tick-borne parasitic disease, is caused by the hemoprotozoa, Babesia vogeli, and Babesia gibsoni. Infection with these parasites, which is endemic globally, leads to life-threatening immunosuppression in dogs. The merozoites invade the red blood cells (RBCs) of infected dogs. Ehrlichia canis, an intracellular bacterium that infects monocytes, is transmitted by the same tick species (Rhipicephalus sanguineus) during blood consumption and coinfection with B. vogeli and E. canis has been reported. Although the hematology and biochemistry of canine babesios

Molecular detection of Rickettsia spp., Borrelia spp., Bartonella spp. and Yersinia pestis in ectoparasites of endemic and domestic animals in southwest Madagascar.

Little is known about the presence of vector-borne bacteria in southwest Madagascar. Anthropogenic alteration of natural habitats represents an important driver for the emergence of new diseases. Especially the involvement of livestock and the involuntary maintaining of invasive synanthropic animals (particularly rats) facilitate disease transmission from wildlife to humans and associated animals and vice versa. The dissemination or acquisition of ectoparasites is most likely in regions where human/wildlife contact is increasing. Little is known about the presence of vector-borne bacteria in s

Transgenic Babesia bovis lacking 6-Cys sexual-stage genes as the foundation for non-transmissible live vaccines against bovine babesiosis.

Babesia bovis, a tick-borne apicomplexan parasite responsible for bovine babesiosis has a complex life cycle including sexual development in its Rhipicephalus microplus vector. Understanding the molecular mechanisms involved in sexual development is essential for developing future-generation transmission blocking vaccines (TBVs) and/or non-transmissible attenuated live vaccines. The widely conserved members of the 6-Cys gene family likely play roles in the development of sexual stages of B. bovis, and are candidates for developing novel TBV. The recently defined sexual markers 6-CysA and 6-Cys

Dog survey in Russian veterinary hospitals: tick identification and molecular detection of tick-borne pathogens.

BACKGROUND: Species of Canidae in Russia can be infested with up to 24 different tick species; however, the frequency of different tick species infesting domestic dogs across Russia is not known. In addition, tick-borne disease risks for domestic dogs in Russia are not well quantified. The goal of this study was to conduct a nationwide survey of ticks collected from infested dogs admitted to veterinary clinics in Russian cities and to identify pathogens found in these ticks. METHODS: Ticks feeding on dogs admitted to 32 veterinary clinics in 27 major cities across Russia were preserved in eth

Endemic, exotic and novel apicomplexan parasites detected during a national study of ticks from companion animals in Australia.

BACKGROUND: Apicomplexan tick-borne pathogens that cause disease in companion animals include species of Babesia Starcovici, 1893, Cytauxzoon Neitz & Thomas, 1948, Hepatozoon Miller, 1908 and Theileria Bettencourt, Franca & Borges, 1907. The only apicomplexan tick-borne disease of companion animals that is known to occur in Australia is babesiosis, caused by Babesia canis vogeli Reichenow, 1937 and Babesia gibsoni Patton, 1910. However, no molecular investigations have widely investigated members of Apicomplexa Levine, 1980 in Australian ticks that parasitise dogs, cats or horses, until this p
