Using phase-resolved vibrational sum-frequency imaging to probe the impact of head-group functionality on hierarchical domain structure in lipid membranes.

The substantial diversity in phospholipids within a plasma membrane, varying in tail length, degree of saturation, and head-group functionality, generates widespread structural heterogeneity. This exists both laterally across the membrane through the spontaneous formation of condensed domains that differ from their surrounding expanded phase in density, composition, and molecular packing order, as well as between its two leaflets, which normally maintain significant compositional asymmetry. Of particular importance is the exposure of phosphatidylserine (PS) lipids which is a marker for importa

Analysis of phytosterols encapsulated in pegylated liposomes.

Dipalmitoylphosphatidylcholine (DPPC) lipids were encapsulated in PEGylated liposomes with free stigmasterol (ST), stigmasterol myristate (ME), and stigmasterol oleate (OE). Their quality was assessed using TEM, zeta potential, and hydrodynamic diameter measurements. The liposomes were heated to 60 °C and 180 °C. The degradation of stigmasterol and fatty acids was considered, as was derivative formation. The results show that the liposomes fulfilled their intended function. The ST liposomes were smallest, while the ME liposomes were similar in size to the OE liposomes. The degree of degradatio

Effect of sterol structure on molecular interactions and lateral domain formation in monolayers containing dipalmitoyl phosphatidylcholine.

Molecular associations between different sterols and dipalmitoyl phosphatidylcholine (DPPC) were examined in monolayers at the air/water interface. The sterols examined included cholesterol, 5-cholesten-3- one, 4-cholesten-3 beta-ol, 4-cholesten-3-one, cholesteryl acetate, and cholesteryl methyl-and ethyl ether. Information about the long-range order in pure sterol monolayers, as well as lateral domain-formation in mixed sterol/DPPC monolayers was obtained from the lateral miscibility or distribution of NBD-cholesterol (present at 0.5 mol%), as determined by monolayer epifluorescence microscop

Revealing the Morphology of Small Multilamellar Lipid Nanoparticles (SMLPs) made by In-Vial Homogenization.

Previous research shows that thermosensitive small multilamellar lipid nanoparticles (tSMLPs) offer promising features for temperature-triggered cytostatic drug delivery, remaining completely stable at body temperature (37°C) and releasing their payload under mild hyperthermia conditions (42°C). A distinguishing characteristic of tSMLPs is their unique particle morphology - multiple tightly packed bilayers with progressively decreasing intermembrane spacing toward the particle core. In this study, we shift the focus from their thermosensitivity to an in-depth exploration of the particle's morp

Interaction of Myrsinoic acid a with biomembrane models: Differential effects on DPPC and DPPS properties revealed by surface rheology and vibrational spectroscopy.

This study investigates the interactions of Myrsinoic acid A, a natural compound with reported anti-inflammatory and antitumor properties, with lipid monolayers composed of dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylserine (DPPS). Utilizing tensiometry, polarization-modulation infrared reflection-absorption spectroscopy (PM-IRRAS), Brewster Angle Microscopy (BAM), and surface rheology, we analyzed how Myrsinoic acid A affects the structural and mechanical properties of these lipid systems. The PM-IRRAS spectra revealed that Myrsinoic acid A induced disorder in the DPPC mo

Effect of Ester Moiety on Structural Properties of Binary Mixed Monolayers of Alpha-Tocopherol Derivatives with DPPC.

Phospholipid membranes are ubiquitous components of cells involved in physiological processes; thus, knowledge regarding their interactions with other molecules, including tocopherol ester derivatives, is of great importance. The surface pressure-area isotherms of pure α-tocopherol (Toc) and its derivatives (oxalate (OT), malonate (MT), succinate (ST), and carbo analog (CT)) were studied in Langmuir monolayers in order to evaluate phase formation, compressibility, packing, and ordering. The isotherms and compressibility results indicate that, under pressure, the ester derivatives and CT are ab

Curcumin Accelerates the Lateral Motion of DPPC Membranes.

Curcumin, the main ingredient in turmeric, has attracted attention due to its potential anti-inflammatory, anticancer, wound-healing, and antioxidant properties. Though curcumin efficacy is related to its interaction with biomembranes, there are few reports on the effects of curcumin on the lateral motion of lipids, a fundamental process in the cell membrane. Employing the quasielastic neutron scattering technique, we explore the effects of curcumin on the lateral diffusion of the dipalmotylphosphatidylcholine (DPPC) membrane. Our investigation is also supported by Fourier transform infrared s

Hanatoxin inserts into phospholipid membranes without pore formation.

Hanatoxin (HaTx), a 35-residue polypeptide from spider venom, functions as an inhibitor of Kv2.1 channels by interacting with phospholipids prior to affecting the voltage-sensor. However, how this water-soluble peptide modifies the gating remains poorly understood, as the voltage-sensor is deeply embedded within the bilayer. To determine how HaTx interacts with phospholipid bilayers, in this study, we examined the toxin-induced partitioning of liposomal membranes. HPLC-results from high-speed spin-down vesicles with HaTx demonstrated direct binding. Dynamic light scattering (DLS) and leakage a

Nanodomain Formation of Ganglioside GM1 in Lipid Membrane: Effects of Cholera Toxin-Mediated Cross-Linking.

Cross-linking of specific lipid components by proteins mediates transmembrane signaling and material transport. In this work, we conducted coarse-grained simulation to investigate the interactions of binding units of chorela toxin (CTB) with mixed ganglioside GM1 and dipalmitoylphosphatidylcholine (DPPC) lipid bilayer membrane. We determine that the binding of CTB pentamers cross-links GM1 molecules into protein-sized nanodomains that have distinct lipid order compared with the bulk. The toxin in the nanodomain partially penetrates into the membrane. The local disordering can also transmit acr

Effect of high pressure on fully hydrated DPPC and POPC bilayers.

Enhanced hydrostatic pressure can induce phase transitions in hydrated lipid bilayers especially those composed of saturated phospholipids. In this work, the phase behavior of fully hydrated DPPC (1,2-dipalmitoyl-sn-glycero-3-phosphocholine) and POPC (2-oleoyl-1-palmitoyl-sn-glycero-3-phosphocholine) bilayers as a function of pressure up to 3000 atm has been examined in atomic detail on time scales of up to 1.0 μs, using the molecular dynamics simulation technique. DPPC bilayers formed a rippled gel-like phase comprising a minor disordered fluid-like region and a major ordered gel-like region

[Evaluation of lung surfactant function in terms of lipid monolayer micromechanics].

The analysis of compression isotherms of lipid monolayers and thermodynamical estimation of the conditions of the formation of layered systems underlies the explanation of some details of lung surfactant work. When the chemical potential of the monolayer during compression reaches the value of the chemical potential of the liquid bilayer membrane free of the mechanical stress, the conversion of monolayer into bilayer phase commences by means of squeezing out of monolayer in subphase creating bilayer ripples. During inspiration when alveolar surface expands, ripples turn back. Hence, in the cou

Interfacial Organization and Structural Changes in Model Lung Surfactants Induced by Methylxanthines.

This study investigates the interfacial interactions between the selected methylxanthines, theophylline (Theo) and its derivative theophylline-7-acetic acid (TheoAcid), and model pulmonary surfactants. Chronic obstructive pulmonary disease (COPD) and asthma treatments utilizing these drugs are often limited by a narrow therapeutic window and systemic toxicity. We explore the biophysical feasibility of localized inhalation delivery by analyzing physicochemical drug effects on two-dimensional (2D) monolayers of DPPC, DPPG, and their binary mixture (8:2 molar ratio) at the air-water interface. Understanding the Molecular Mechanism of Anesthesia: Effect of General Anesthetics and Structurally Similar Non-Anesthetics on the Properties of Lipid Membranes.

General anesthesia can be caused by various, chemically very different molecules, while several other molecules, many of which are structurally rather similar to them, do not exhibit anesthetic effects at all. To understand the origin of this difference and shed some light on the molecular mechanism of general anesthesia, we report here molecular dynamics simulations of the neat dipalmitoylphosphatidylcholine (DPPC) membrane as well as DPPC membranes containing the anesthetics diethyl ether and chloroform and the structurally similar non-anesthetics n-pentane and carbon tetrachloride, respecti

Modulation of membrane physical properties by natural insecticidal ketones.

The insecticidal activity of Mentha oil and its main components has been tested and established for various insects/pests. Several mint ketones have demonstrated to act on GABAA receptors (GABAA-R), a transmembrane channel target of several important insecticides whose activity can be modulated by surface-active compounds and by changes in the physical properties of the lipid membrane. In the present work, we analyze the capacity of monoterpenic ketones most commonly found in Mentha species, pulegone and menthone, to interact with DPPC membranes by molecular dynamics (MD) simulations and Langm

A coarse-grained molecular dynamics study of nanoscale phospholipid-cholesterol vesicles: curvature effects on cholesterol distribution and vesicle structure.

Nanoscale lipid bilayer vesicles are widely used as model systems for exosomes and synaptic vesicles, released by cells for intercellular communication, in which strong membrane curvature couples molecular packing to asymmetry between the two lipid-bilayer monolayers. Here, we investigate DPPC, POPC or DLiPC phospholipid (PC)/cholesterol bilayer vesicles using coarse-grained molecular dynamics simulations, systematically varying phospholipid chain unsaturation, cholesterol content (0-50 mol%), and vesicle diameter (10-30 nm for the DPPC system). Cholesterol undergoes rapid redistribution on su

The amount of the settled surfactant dipalmitoylphosphatidylcholine (DPPC) in the heart of rats does not correlate with decompression illness.

INTRODUCTION: The lung surfactant dipalmitoylphosphatidylcholine (DPPC) settles on the luminal aspect of blood vessels to form active hydrophobic spots (AHS). The nanobubbles that develop at these spots are gas micronuclei from which decompression bubbles expand upon decompression. Variability between "bubblers" and "non-bubblers" is derived from the variability in the AHS. We expected that the level of DPPC in the heart would correlate with the severity of decompression illness (DCI). METHODS: Thirty-six rats were exposed to 8 bar for 32 minutes. RESULTS: After decompression, they were exam

Effect of ipratropium bromide on the interfacial organization of model lung surfactant membranes.

The interactions of inhaled drugs with lung surfactant are important for understanding their behavior at the first biological barrier of the respiratory tract. In this work, we investigated the effect of the anticholinergic bronchodilator, ipratropium bromide (IPB), on model lung surfactant membranes composed of dipalmitoylphosphatidylcholine (DPPC), dipalmitoylphosphatidylglycerol (DPPG), and their 8:2 molar mixture. Langmuir monolayer studies showed that ipratropium bromide has little influence on zwitterionic DPPC, while it interacts strongly with negatively charged DPPG, leading to concent

Advanced Spectroscopic Studies of the AIE-Enhanced ESIPT Effect in a Selected 1,3,4-Thiadiazole Derivative in Liposomal Systems with DPPC.

Liposomal systems are advanced carriers of active substances which, thanks to their ability to encapsulate these substances, significantly improve their pharmacokinetics, bioavailability, and selectivity. This article presents the results of spectroscopic studies for a selected compound from the 1,3,4-thiadiazole group, namely 4-[5-(naphthalen-1-ylmethyl)-1,3,4-thiadiazol-2-yl]benzene-1,3-diol (NTBD, see below in the text), in selected liposomal systems formed from the phospholipid 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC). Effects of Nanoplastics on Lipid Membranes and Vice Versa: Insights from All-Atom Molecular Dynamics Simulations.

We compute the potential of mean force (PMF) between semicrystalline polyethylene (PE) nanoplastics (NPLs) and model POPC and DPPC bilayers, which approximate in vivo membranes, using atomistic simulations. Our work shows that atomistic resolution is required to characterize the NPL and lipid interactions. By analyzing the PMF, we demonstrate that the mechanical properties of membranes, rather than NPL semicrystalline morphologies, govern NPL-membrane interactions. Resistance to NPL penetration arises from the elastic energy of the membrane deformation. The flexible POPC membranes resist NPL t

Molecular-Level Insights into the Interactions of Nicotine and Cotinine with DPPC Phospholipid Bilayers.

Small alkaloids such as nicotine and its major metabolite, cotinine, are widely present in biological and environmental systems, yet their membrane interaction mechanisms and related cytotoxic effects remain poorly quantified. Here, molecular dynamics simulations combined with umbrella sampling were used to investigate the adsorption, distribution, orientation, and transmembrane behavior of nicotine and cotinine in DPPC bilayers. Both molecules rapidly migrated from bulk water to the water-lipid interface within 8 ns and showed strong interfacial enrichment, with residence probabilities of 85%
