Vitamin K-dependent gamma-carboxyglutamic acid protein 1 promotes pancreatic ductal adenocarcinoma progression through stabilizing oncoprotein KRAS and tyrosine kinase receptor EGFR.

BACKGROUND: Vitamin K-dependent γ-glutamic acid carboxylation (Gla) proteins are calcium-binding and membrane-associated, participating in coagulation, bone turnover, and cancer biology. The molecular function of transmembrane proline-rich Gla proteins (PRRGs) remains unexplored. METHODS: Analysis of pancreatic ductal adenocarcinoma (PDAC) datasets, including transcription profiles, clinical data, and tissue microarrays, was conducted to evaluate PRRG1 expression and its clinical relevance. PDAC cell lines with overexpressed, knockdown, and mutated PRRG1 were developed to study biological fun

Mechanism of the calcium-dependent self-association of bovine prothrombin. Use of a covalent cross-linking reagent to study the reaction.

The present study has made use of a covalent cross-linking agent, dithiobis(succinimidylpropionate), to study the self-association of prothrombin and has demonstrated that the covalent dimerization reaction involves the gamma-carboxyglutamic acid region of prothrombin (1-42 of 582). An essential role for the gamma-carboxyglutamic acid residues of prothrombin in the association reaction was demonstrated by experiments that converted gamma-carboxyglutamic acid residues to gamma-methylene glutamic acid or glutamic acid and resulted in a prothrombin species that was inactive in our cross-linking a

Characteristics and culture of osteoblasts derived from avian long bone.

A method is presented for isolating primary osteoblasts from the periosteal surface of chick tibia. The culture system identified supports both cell proliferation and phenotype retention. Cell numbers increased 8-fold in Week 1 and 20-fold over a total of 12 days. Well-established osteoblast markers, alkaline phosphatase staining, gamma-carboxyglutamic acid, osteocalcin, type I collagen, and parathyroid hormone binding were detected. Osteocalcin, gamma-carboxyglutamic acid, and type I collagen were present on culture Day 4, and were increased in amount by Day 8, but were similar to the earlier

Gamma-carboxyglutamate in normal and pathological human middle ear bones.

The presence of the amino acid gamma-carboxyglutamate (GLA) was established in human middle ear bones. Proteins containing GLA have been described as being associated with normal as well as pathological calcifications. The GLA content of human incus, malleus, and stapes with 1-2 nmol/mg bone is in the range previously reported for a variety of bone. A limited number of samples with middle ear pathology, including otosclerosis, did not show altered GLA levels.

[Vitamin K and bone metabolism - on the effect of vitamin K deficiency and gamma-carboxyglutamic acid (author's transl)].

Vitamin K was thought to be primarily involved in the production of clotting factors. The recent findings of gamma-carboxyglutamic acid (Gla) - vitamin K-dependent calcium binding amino acid - in various calcified tissues, however, have prompted suggestions that vitamin K may play an important role in calcification mechanism. The present study was designed to investigate the relation between vitamin K and calcification. Morphological, biomechanical and biochemical analyses were undertaken on bones from vitamin K deficient rats raised on vitamin K deficient diet combined with administration of

The role of the Gla domain in the activation of bovine coagulation factor X by the snake venom protein XCP.

The activation by XCP of coagulation factor X and a factor X species lacking the Gla-domain was studied in the presence and absence of Ca2+. Both proteins could be activated at low rates in the absence of Ca2+. The activation of the unmodified factor X was stimulated by the addition of Ca2+, whereas GD factor X activation was insensitive to Ca2+. The stimulatory effect of Ca2+ seen with the unmodified factor X correlated strongly with a calcium-dependent change in intrinsic protein fluorescence. This conformational change required the Gla-domain as the fluorescence emission of GD factor X was

[Mechanisms of non-specific prothrombin activation].

The review of different mechanisms of non-physiological non-specific prothrombin activation is given as compared with the specific activation by the factor Xa. The use of snake venom enzymes or staphylocoagulase makes possible the thrombin generation from pathological forms of prothrombin lacking of gamma-carboxyglutamic acid and incapable of complete activation into thrombin by the specific activator, factor Xa. This fact stimulated the use of non-specific activators in medicine. Investigation of non-specific prothrombin activators made possible to reveal and to trace pathways of the formatio

Discrimination of normal and abnormal prothrombin and protein C in plasma using a calcium ion-inhibited monoclonal antibody to a common epitope on several vitamin K-dependent proteins.

Vitamin K deficiency or administration of vitamin K antagonists results in the biosynthesis of abnormal des-gamma-carboxy forms of the vitamin K-dependent proteins. Monoclonal antibody H-11 binds several vitamin K-dependent proteins at a determinant that includes the first two residues of gamma-carboxyglutamic acid. Antibody H-11 binds fully carboxylated prothrombin and protein C in the presence of EDTA but binding is inhibited by the divalent metal ions, calcium, magnesium, and manganese. By contrast, des-gamma-carboxy prothrombin and protein C bind antibody H-11 the same in the presence of E

Vitamin K-dependent carboxylase.

Vitamin K-dependent carboxylase is found in the liver, where it is involved in the synthesis of four blood coagulation factors and protein C. The hepatic enzyme has partly been purified and several mechanisms have been postulated for the vitamin K-dependent carboxylation reaction. Recently the enzyme has also been detected in other tissues including the lung, kidney, spleen, testis, bone and arterial vessel wall. The proteins produced by these non-hepatic carboxylases are now being characterized, but in most cases their function is still unknown. This paper is meant to review our present knowl

gamma-Carboxyglutamic acid, a component in human pediatric bladder stones containing calcium salts.

The amino acid gamma-carboxyglutamic acid (Gla) has been previously detected in the vitamin K-requiring blood clotting factors, proteins of calcified vertebrate tissue, renal tissue, plasma protein C, ectopic calcifications, and calcium-containing renal calculi. This paper reports the presence of Gla in the EDTA-soluble, non-dialyzable material recovered from human pediatric bladder stones containing calcium salts. In bladder stones composed of calcium oxalate, uric acid and ammonium acid urate, 73 Gla residues per 1,000 amino acid residues were detected. Bladder stones composed of calcium oxa

Long non-coding RNA Linc01133 promotes osteogenic differentiation of human periodontal ligament stem cells via microRNA-30c / bone gamma-carboxyglutamate protein axis.

Periodontitis is a chronic inflammation caused by the deposition of dental plaque on the tooth surface. Human periodontal ligament stem cells (hPDLSCs) have the potential of osteogenic differentiation. Long non-coding RNAs (lncRNAs) are collectively involved in periodontitis. This study was designed to explore the roles of Linc01133 in osteogenic differentiation of hPDLSCs. hPDLSCs obtained from the periodontal ligament (PDL) of patients with periodontitis were used to collect Linc01133, microRNA-30c (miR-30c), and bone gamma-carboxyglutamate protein (BGLAP) expression data, and their expressi

Postoophorectomy bone loss is associated with reduced bone Gla protein serum levels: a possible effect of osteoblastic insufficiency.

To further investigate the relationship between oophorectomy (OF) and mineral bone loss, 15 women who underwent total hysterectomy and bilateral oophorectomy were studied for 12 months after surgery. Mineralometric and metabolic data were obtained before and after 3, 6, and 12 months. The women lost bone mineral content (measured by single photon absorptiometry) at the same rate they lost cortical and trabecular bone, suggesting that bone loss after OF is a generalized phenomenon. Our data also show that an increase in bone resorption takes place only in the first period after OF; the persiste

Immunolocalization of gamma-carboxyglutamic acid-containing proteins in developing molar tooth germs of the rat.

The localization of dentin gamma-carboxyglutamic acid containing proteins (DGP's) was examined in rat molar tooth germs at various stages of development by immunostaining with the peroxidase-antiperoxidase method. Odontoblasts and their processes stained positive, whereas all other cells were negative. No staining was found in predentin. Dentin stained only after a mild demineralization. The results support the concept that DGP's are deposited by odontoblasts directly into the mineralizing dentin.

Beta-hydroxyaspartic acid in vitamin K-dependent plasma proteins from scorbutic and warfarin-treated guinea pigs.

beta- Hydroxyaspartic acid is a rare amino acid, present in all vitamin K-dependent plasma proteins except prothrombin, and is formed by a post-translational hydroxylation of aspartic acid. We have now investigated whether this hydroxylation, like that of proline in collagen, is vitamin C-dependent. The vitamin K-dependent plasma proteins were isolated from normal and scorbutic guinea pig plasma by barium citrate adsorption and the beta- hydroxyaspartic acid content was determined. Compared with normal animals, scorbutic animals showed no significant reduction of beta- hydroxyaspartic acid con

Dietary induced subclinical vitamin K deficiency in normal human subjects.

A subclinical vitamin K deficiency was induced in 32 healthy subjects (four groups of eight males and females) aged 20-40 and 60-80 yr residing in the Metabolic Research Unit of the Human Nutrition Research Center on Aging at Tufts University. Volunteers were initially fed (4 d) a baseline-period diet containing the recommended daily allowance for vitamin K which is equivalent to 80 micrograms/d of phylloquinone (vitamin K1). During the baseline period various parameters of vitamin K nutritional status were monitored. The baseline period was followed by a 13-d depletion period during which the

Reduced axial bone mineral content in patients taking an oral anticoagulant.

Recent literature indicates that the vitamin K cycle plays a role in the calcification process, presumably via its intervention on gamma-carboxylation of the noncollagenous proteins of bone osteocalcin and matrix gamma-carboxyglutamic acid protein. The major clinical evidence of this interference is fetal bone defect caused by oral anticoagulants given to the mother during the first trimester of pregnancy. No bone abnormalities have been reported so far in adults receiving oral anticoagulants. We studied 56 women who had had cardiac valve replacement and who were given acenocoumarol as anticoa

[Diagnostic evaluation of plasma free gamma-carboxyglutamic acid in maintenance hemodialytic patients with renal osteodystrophy].

The levels of serum Bone Gla Protein (BGP) and plasma free gamma-carboxyglutamic acid (free gamma-Gla) were measured in 36 patients on maintenance hemodialysis and compared with conventional parameters of renal osteodystrophy (ROD), such as C-PTH, intact PTH, Al-PIII and hydroxyproline levels in serum, and sigma GS/D. Even though the values of BGP were significantly elevated in patients group, 64.5 +/- 1.74 ng/ml, but the low values within normal range could be found in 3 of 36 patients, also. On the other hand, plasma levels of free gamma-Gla were markedly elevated in all cases as high as 10.

Treatment of congenital osteopetrosis with high-dose calcitriol.

We administered high doses of calcitriol (up to 32 micrograms per day) to an infant with malignant osteopetrosis, in an attempt to stimulate bone resorption. The patient was placed on a low-calcium diet to prevent hypercalcemia. Measures of bone turnover increased during calcitriol therapy; hydroxyproline excretion rose from 140 to 1358 micrograms per milligram of creatinine per 24 hours, with parallel increases in the ratio of calcium to creatinine in the urine, urinary gamma-carboxyglutamic acid, serum osteocalcin, and serum alkaline phosphatase. Differences in the metal ion structure between Sr- and Ca-prothrombin fragment 1.

The structure of Sr-prothrombin fragment 1 has been solved and refined by restrained least-squares methods at 2.5-A resolution to a crystallographic R value of 0.167. The protein structure is very similar to that of Ca-fragment 1. A polymeric array of five Sr2+ ions separated by about 4.0 A is buried among six gamma-carboxyglutamic acid (Gla) residues; three other Sr2+ ions interact with other Gla residues and are located further apart. One of these was not found in the Ca-fragment 1 structure. The coordination of the Sr2+ ions resembles that of Ca2+, but there are some significant differences
