[Seasonal variations of vitamin D metabolites in man].

Studies concerning the influence of seasons and ultraviolet radiations on the metabolism of 1,25-dihydrocholecalciferol (1,25 (OH)2D), the active form of vitamin D, have given conflicting results. In the present study serum concentrations of the 3 main vitamin D metabolites--25-hydroxyvitamin D, 24,25-dihydroxyvitamin D and 1,25-dihydroxyvitamin D--were measured by radiocompetitive assay (a) monthly during one year in 7 normal subjects, and (b) before, during and after 4 weeks of "whole body" exposure to ultraviolet radiations in 11 other subjects. In study (a) parallel changes in 25-hydroxyvi

The good and the bad of vitamin D inactivation.

While disorders of impaired vitamin D activation and action have long been appreciated, the consequences of abnormalities in pathways leading to the inactivation of vitamin D metabolites have only recently been identified. Two recent articles have shed new light on this area of vitamin D biology. The report by Martineau et al., published in the JCI, describes a pathway in which binding of the vitamin D metabolite 24R,25(OH)2D3 to its effector molecule FAM57B2 plays an important role in endochondral ossification during bone repair. This work follows, and adds to, another recent JCI publication

[Hypervitaminosis D].

Excessive vitamin D causes marked and prolonged hypercalcemia by accelerating intestinal calcium absorption and bone resorption. Vitamin D induced hypercalcemia includes the toxic ingestion of excessive amount of vitamin D preparations, granulomatous diseases and lymphoproliferative malignancies. In vitamin D toxicity, the clinical courses vary depending on the vitamin D preparation responsible for the hypercalcemia. Hypercalcemia state continues for several months when D2 or D3 are responsible for the toxicity whereas the hypercalcemia would subside in a week when 1 alpha(OH) D3 or 1,25 (OH)2

Circulating levels of vitamin D metabolites after renal transplantation.

We investigated the change in the serum levels of vitamin D metabolites after renal transplantation in 5 uremic children. The serum 1,25-(OH)2D level showed a transient increase on the 1st or 2nd day after transplantation in all the patients. In 3 cases, it decreased after the initial peak and increased gradually to the normal level again within 2-4 weeks although, in the other 2 cases, it remained within the normal range after a slight decline. Serum 25-OHD and 24,25-(OH)2D level remained low for 4-8 weeks. Hypophosphatemia and/or hyperparathyroidism, which still remained after transplantatio

Healing of rachitic lesions in chicks by 24R,25-dihydroxycholecalciferol administered locally into bone.

In an attempt to further define the nature of the active metabolite in bone formation, a series of experiments were conducted whereby vitamin D metabolites were administered locally in vivo into the proximal epiphyseal growth plate of the tibiae of rachitic chicks. Local administration of 3 micrograms of 24,25(OH)2D3 in vivo to D-deficient chicks resulted in disappearance of the rachitic lesions in the same leg. Administration of 1 microgram 1,25(OH)2D3 in a similar manner failed to show any sign of healing. Injection of 5 micrograms 25(OH)D3 was followed by recovery from rickets in both the i

Metabolism and pharmacokinetics of vitamin D in patients with cystic fibrosis.

Patients with cystic fibrosis (CF) commonly have lower circulating concentrations of 25-hydroxyvitamin D (25(OH)D) than healthy populations. We comprehensively compared measures of vitamin D metabolism among individuals with CF and healthy control subjects. In a cross-sectional study, serum from participants with CF (N = 83) and frequency-matched healthy control subjects by age and race (N = 82) were analyzed for: 25(OH)D2 and 25(OH)D3, 1α,25-dihydroxyvitamins D2 and D3 (1α,25(OH)2D2 and 1α,25(OH)2D3), 24,25-dihydroxyvitamin D3 (24,25(OH)2D3), 4β,25-dihydroxyvitamin D3 (4β,25(OH)2D3), 25-hydro

Effects of age and estrogen on renal vitamin D metabolism in the female rat.

Young and adult, intact and ovariectomized female rats were used to study the effects of increasing age and estrogen loss on the conversion of 25-hydroxycholecalciferol [25(OH)D] to 1,25-dihydroxycholecalciferol [1,25(OH)2D] and 24,25-dihydroxycholecalciferol [24,25(OH)2D]. The recovery level of radioactivity in plasma as 1,25(OH)2D after [3H]25(OH)D administration was lower in older animals; ovariectomy reduced [3H]1,25(OH)2D recovery in both young and adult animals. 1,25(OH)2D synthesis by kidney slices was reduced in all adult animals and in ovariectomized young animals. Increase of bone volume in vitamin D-repleted rats by massive administration of 24R,25(OH)2D3.

A large dose of 24R,25(OH)2D3 was administered to the vitamin D-repleted rat to examine its effect on the bone. Male Wistar rats were fed a diet containing 0, 0.025, 1.25, 4.0, and 12.5 ppm 24R, 25(OH)2D3 for 2 years starting at age 6 weeks. The estimated amounts of daily intake of 24R,25(OH)2D3 were 0, 93, 4640, 14680, and 49580 ng/100 g body weight, respectively. No notable difference was found in either the weight or the death rate of the animal. The long-term administration of massive doses of 24R,25(OH)2D3 did not lead to hypercalcemia nor did it affect the blood phosphorus, alkaline-phos

Field rickets in turkey poults: biochemical findings.

Two clinical cases of field rickets and one of nutritional rickets in turkey poults were studied. Plasma levels of calcium, phosphorus, alkaline phosphatase, and vitamin D metabolites were determined. Concentrations of calcium, phosphorus, and alkaline phosphatase in affected poults were typical of nutritional rickets. The mean plasma calcium concentration in rachitic poults was not significantly different from that in controls. Mean values for plasma phosphorus were 1.5 to 2.4 mg/dl lower in the rachitic poults, and values for alkaline phosphatase activity were 1.3-1.7 times greater. In assay

Association between serum vitamin D metabolite levels and cognitive function in community-dwelling older adults: A cross-sectional study.

BACKGROUND & AIMS: Vitamin D deficiency has been associated with cognitive decline and Alzheimer's disease (AD); however, findings remain inconsistent. Assessing vitamin D status based solely on total 25-hydroxyvitamin D [25(OH)D] may be insufficient, and other metabolites, such as 24,25-dihydroxyvitamin D3 [24,25(OH)2D3] and 3-epimer-25-hydroxyvitamin D3 [3-epi-25(OH)D3], may provide additional insights. This study aimed to examine the association between serum vitamin D metabolite concentrations and cognitive function in older adults. METHODS: A cross-sectional study was conducted involving

Genome-wide analyses of gene expression profile identify key genes and pathways involved in skeletal response to phosphate and 1,25-dihydroxyvitamin D3 in vivo.

Phosphate (P) is an essential element involved in various biological actions, such as bone integrity, energy production, cell signaling and molecular component. P homeostasis is modulated by 4 main tissues; intestine, kidney, bone, and parathyroid gland, where 1,25-dihydroxyvitamin D3 (1,25(OH)2D3), parathyroid hormone and fibroblast growth factor 23 (FGF23) are produced and/or have an influence. In bone, serum P level modulates the production of FGF23 which then controls not only P excretion but also vitamin D metabolism in kidney in an endocrine manner. The hormonally active form of vitamin

Vitamin D metabolites in diabetic patients: decreased serum concentration of 24,25-dihydroxyvitamin D.

In order to elucidate if changes in vitamin D metabolism play a role for diabetic bone loss, the serum concentrations of the major vitamin D metabolites were studied in 26 adult male ambulatory insulin-treated diabetics, selected to have normal renal function and a duration of diabetes below 11 years. The patients were studied during usual metabolic control and exhibited wide ranges of hyperglycaemia and glycosuria. The serum concentrations of the major metabolites of vitamin D, 25-hydroxyvitamin D(2 + 3) (25OHD), 24,25-dihydroxyvitamin D(2 + 3) (24,25(OH)2D), and 1,25-dihydroxyvitamin D(2 + 3

Critical Role for 24-Hydroxylation in Homeostatic Regulation of Vitamin D Metabolism.

CONTEXT: The body has evolved homeostatic mechanisms to maintain free levels of Ca+2 and 1,25-dihydroxyvitamin D (1,25(OH)2D) within narrow physiological ranges. Clinical guidelines emphasize important contributions of parathyroid hormone (PTH) in maintaining this homeostasis. OBJECTIVE: This work aimed to investigate mechanisms of homeostatic regulation of vitamin D (VitD) metabolism and to apply mechanistic insights to improve clinical assessment of VitD status. METHODS: This crossover clinical trial studied community participants before and after VitD3 supplementation. Participants includ

Action of vitamin D metabolites on PTH secretion in man.

We have examined the effects of metabolites of vitamin D [25OHD3, 1,25(OH)2D3, 24,25(OH)2D3, and 25,26(OH)2D3] on serum calcium and iPTH in human deficient-D osteomalacia. The four metabolites decreased iPTH, but only for 1,25(OH)2D3 was a significant correlation between increase of serum calcium and decrease of iPTH observed. The 24,25(OH)2D3 and 25,26(OH)2D3 decreased iPTH despite a decrease of serum calcium at the beginning of treatment. The 25OHD decreased iPTH before increased serum calcium. These results could be interpreted as a direct effect of metabolites of vitamin D on PTH secretion

Effects of protein deficiency on plasma levels of 25-hydroxyvitamin and 24,25-dihydroxyvitamin D in the rat.

The effect of protein deficiency on plasma 25-hydroxyvitamin and 24,25-dihydroxyvitamin D concentrations was determined in weanling rats. The concentrations of these metabolites did not differ in rats fed diet providing either 0.5 or 21% casein for 4 weeks postweaning. Also, no change was observed in plasma levels of these metabolites in rats fed the 21% casein diet in amounts restricted to those consumed by the 0.5% casein animals for the same period of time. The results indicate that the synthesis and transport of 25-hydroxyvitamin and 24,25-dihydroxyvitamin D are not significantly affected

Hypercalcemia, nephrolithiasis, and hypervitaminosis D precipitated by supplementation in a susceptible individual.

Vitamin D supplementation is common among the general public. Although generally considered safe, vitamin D supplement-induced toxicity has been reported, often in association with manufacturing or labeling errors. Additionally, selected patient populations may have a hypersensitivity to vitamin D supplementation, leading to consequences due to supraphysiologic serum 25-hydroxyvitamin D levels. A 58-y-old woman developed hypercalcemia and its sequelae while on vitamin D supplementation. Despite being vitamin D replete, a functional medicine practitioner prescribed vitamin D starting at 8000 IU

[Assessment of Vitamin D Metabolism by LC-MS/MS].

Vitamin D is pro-hormone that has important roles in calcium metabolism in the intestine, kidneys, and bone. Many studies have indicated that vitamin D deficiency causes not only rickets and osteomalacia, but also increases the risk of various diseases such as cancer and autoimmune disease. Of the many vitamin D metabolites, 25-hydroxyvitamin D[25OH-D], 1α,25-dihydroxyvitamin D[1,25(OH)2-D], and 24,25-dihydroxyvitamin D [24,25 (OH) 2-D] are important for assessing vitamin D metabolism. They are circulating, biologically active, and major inactive forms of vitamin D metabolite, respectively. Im
