Characterization of the soluble allergenic proteins of cashew nut (Anacardium occidentale L.).

The allergens associated with cashew food allergy have not been well-characterized. We sought to identify the major allergens in cashew nut by performing IgE immunoblots to dissociated and reduced or nonreduced cashew protein extracts, followed by sequencing of the peptides of interest. Sera from 15 subjects with life-threatening reactions to cashews and 8 subjects who tolerate cashews but have life-threatening reactions to other tree nuts were compared. An aqueous cashew protein extract containing albumin/globulin was separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS

Antimicrobial nano-zinc oxide-2S albumin protein formulation significantly inhibits growth of "Candidatus Liberibacter asiaticus" in planta.

Huanglongbing (HLB, also known as citrus greening) is considered to be the most devastating disease that has significantly damaged the citrus industry globally. HLB is caused by the Candidatus Liberibacter asiaticus (CLas), the fastidious phloem-restricted gram-negative bacterium, vectored by the asian citrus psyllid. To date, there is no effective control available against CLas. To alleviate the effects of HLB on the industry and protect citrus farmers, there is an urgent need to identify or develop inhibitor molecules to suppress or eradicate CLas from infected citrus plant. In this paper, w

The quest for ingested peanut protein in human serum.

BACKGROUND: There is mounting evidence that systemic uptake of food allergens is key to triggering anaphylaxis. However, direct proof for this theory is still lacking. The purpose of this study was to quantify the absorption and to determine the absorption kinetics of immunoreactive peanut protein in relation to the allergic response in human. METHODS: Quantitative protein assays including mass spectrometry, dot blots and Western blotting were developed to determine the level of Ara h 2 absorption in human serum. The double monoclonal sandwich ELISA was applied to quantify absorbed Ara h 2 an

Comparative proteomics of amaranth and quinoa seeds reveals species-specific solubility traits of 11S globulins after Osborne and polarity-based extractions.

Amaranth and quinoa are nutritious grains rich in essential amino acids, vitamins, and phytochemicals. The use of these emergent functional foods is still limited and their proteins are poorly characterized. Here, we compared amaranth and quinoa seeds by profiling their proteins using Osborne and polarity-based extraction methods and evaluating their relative protein content. The Osborne fractions and the two fractions generated by the polarity-based method (hydrophilic and hydrophobic) were quantified and analyzed by 1D-SDS-PAGE in the absence and presence of a reducing agent, as well as by d

Ranking primary food allergens by specific IgE-binding and clinical relevance in Dutch adults to identify reference protein sets for allergenicity assessment.

The increasing introduction of novel protein sources globally requires more robust allergenicity evaluations to predict their sensitising capacity. This study ranked plant-derived and animal-derived food allergens to establish reference proteins that may support the future development of predictive tools for de novo sensitisation. Sera from 307 Dutch adult patients presenting with suspected food allergy at an allergology outpatient clinic were analysed with Allergy Xplorer2 multiplex immunoassay. Food components were ranked based on specific IgE-binding frequencies, intensities, and clinical r

[Progress of researches on the allergens Ara h 1, Ara h 2 and Ara h 3 from peanut].

Peanut is one of the most popular foods in the world due to its high nutrition; however, it contains multiple seed storage proteins which are identified as allergens and hence are the most common cause of life-threatening, IgE-mediated anaphylaxis among the hypersensitive individuals. Three peanut proteins, Arachis hypogaea allergy 1, 2, 3 (Ara h 1, Ara h 2 and Ara h 3), which have the common biochemical characteristics like resistance to proteases and heat, are considered as the major allergens because they are recognized by serum IgE from a peanut-allergic patient population. The linear IgE-

Variable IgE cross-reactivity between peanut 2S-albumins: The case for measuring IgE to both Ara h 2 and Ara h 6.

BACKGROUND: 2S-albumins Ara h 2 and Ara h 6 are the most potent peanut allergens and levels of specific immunoglobulin E (IgE) towards these proteins are good predictors of clinical reactivity. Because of structural homologies, Ara h 6 is generally considered to cross-react extensively with Ara h 2. OBJECTIVE: We aimed to quantify the IgE cross-reactivity between Ara h 2 and Ara h 6. METHODS: Peanut 2S-albumins were purified from raw peanuts. The IgE cross-reactivity between Ara h 2 and Ara h 6 was evaluated with 32 sera from French and US peanut-allergic patients by measuring the residual I

Detection of Circulating Peanut Components in Serum after Ingestion.

INTRODUCTION: Detection of specific food allergens brought into circulation following ingestion is complicated by the minute amounts present in serum. We therefore aimed to assess the utility of selective removal of IgE against peanut components using ImmunoCAP combined with the basophil histamine release (HR) assay (BHRA) to identify the absorbed allergen components after ingestion of peanut. METHODS: Serum from six healthy individuals was drawn before and 1 h after ingestion of 100 g peanut. Serum from two peanut allergic patients was depleted for IgE against single allergen components by i

Changes in Serum Protein-Peptide Patterns in Atopic Children Allergic to Plant Storage Proteins.

Next to cow's milk and eggs, plant foods, i.e., legumes, tree nuts and cereal grains, most often sensitise atopic children. Storage proteins constitutes the most relevant protein fraction of plant foods, causing primary sensitisation. They exhibit strong allergenic properties and immunogenicity. Our goal was to analyse sensitisation to 26 plant storage proteins in a group of 76 children aged 0-5 years with chronic symptoms of atopic dermatitis using Allergy Explorer ALEX2 and to discover changes in serum protein-peptide patterns in allergic patients with the use of MALDI-TOF-MS. We reported th

A dual-functional paper-based analytical device for ultrasensitive detection of peanut allergen-specific IgE.

Increasing attention has been caught by the allergy-related food safety issue. The rapid and sensitive diagnosing approaches are still in high demand for providing clinical reference. Paper-based analytical devices (PADs) are appealing candidates for allergy diagnosis and prediction due to their portability, stability, and operational easiness. However, the sensitivity of PADs needs to be further improved for the targets with low abundance. In addition to the complex signal amplifications, an alternative strategy that requires fewer reagents, steps, and shorter time is anticipated. (82) RESULT

Germline-encoded recognition of peanut underlies development of convergent antibodies in humans.

Humans develop immunoglobulin G (IgG) antibodies to the foods they consume. In the context of food allergy, allergen-specific IgG antibodies can sequentially class-switch to pathogenic IgE. However, the mechanism underlying the antigenicity of food proteins remains uncharacterized. Here, we identified convergent antibodies arising from different antibody gene rearrangements that bind to the immunodominant peanut allergen Ara h 2 and characterized allelic and junctional constraints on germline antibody specificity. Structurally, we found similar epitope-paratope interactions across multiple gen

Fine resolution of the N-terminal IgE-binding epitope of Ara h 2: Discovery of variants with enhanced IgE binding.

BACKGROUND: IgE binding to linear peptides from the N-terminal region of Ara h 2 (epitope 1) is associated with the achievement of sustained unresponsiveness in young children receiving oral immunotherapy and may be important for cross-reactivity between peanuts and tree nuts. This region is also part of the binding site for neutralizing IgG monoclonal antibodies associated with sustained unresponsiveness following oral immunotherapy. OBJECTIVE: We sought to perform alanine scanning of this epitope to determine the importance of individual amino acids and then amino acid scanning to screen fo

Design of an Ara h 2 hypoallergen from conformational epitopes.

INTRODUCTION: Adverse reactions are relatively common during peanut oral immunotherapy. To reduce the risk to the patient, some researchers have proposed modifying the allergen to reduce IgE reactivity, creating a putative hypoallergen. Analysis of recently cloned human IgG from patients treated with peanut immunotherapy suggested that there are three common conformational epitopes for the major peanut allergen Ara h 2. We sought to test if structural information on these epitopes could indicate mutagenesis targets for designing a hypoallergen and evaluated the reduction in IgE binding via imm

Added Diagnostic Value of Peanut Component Testing: A Cross-Sectional Study in Australian Children.

BACKGROUND: Peanut components are widely used in clinical practice; however, their utility to predict challenge outcome in the Australian children, outside of infants, is not well studied. OBJECTIVE: Can peanut component testing predict outcome of challenge in peanut-allergic children. METHODS: All children attending peanut challenges, regardless of previous allergic reactions to peanut or sensitization (skin prick test or peanut IgE) alone, were recruited. Serum collected before the challenge was analyzed for peanut IgE and Ara h 1, 2, 3, 6, 8, and 9 (ImmunoCap). RESULTS: Of the 222 childr

Anaphylaxis to peanut in a patient predominantly sensitized to Ara h 6.

Diagnosis of peanut allergy has improved thanks to component-resolved diagnostics. Peanut allergen component Ara h 2 is considered to indicate true peanut allergy. The component Ara h 6 is structurally similar to Ara h 2, but the diagnostic value of analyzing IgE antibodies to Ara h 6 is unclear. A boy sensitized (≥0.35 kU(A)/l) to Ara h 8 but not to Ara h 1, Ara h 2 and Ara h 3 was challenged with peanut and developed grade II anaphylaxis. In serum collected at the time of challenge a doubling of IgE to the peanut allergen extract was observed compared to allergy testing 9 months earlier. In

Sunflower Seed-Induced Anaphylaxis With Immunoglobulin E Reactivity to 2S Albumins and Novel 60 kDa Antigen Identified by Immunoblot Analysis.

Sunflower seed allergy is uncommon, and reports of severe systemic reactions, such as anaphylaxis, are limited, particularly in Japan where sunflower seeds are not routinely consumed. A 38-year-old woman developed throat discomfort, dyspnea, and generalized urticaria within minutes of ingesting cookies containing sunflower seeds and was diagnosed with anaphylaxis. Her medical history included bronchial asthma and pollinosis caused by Japanese cedar and cypress. Skin prick testing (SPT) showed a strong positive (4+) reaction to sunflower seeds, while the results were negative for mugwort, cashe

Impact of recombinant expression in Komagataella phaffii on the allergenic properties of the peanut allergen Ara h 2.

INTRODUCTION: Recombinant allergens are an important diagnostic tool for determining the IgE-sensitization profile of patients, assessing the risk of symptom severity and potential clinical cross-reactivity. In this context, the use of different host cells for recombinant expression must be evaluated in terms of IgE reactivity and diagnostic value. Therefore, recombinant Ara h 2, the major peanut allergen, was produced in yeast, structurally characterized and investigated in respect to IgE-binding and allergenic properties. METHODS: Ara h 2 was produced as recombinant protein using yeast and

Increasing the accuracy of peanut allergy diagnosis by using Ara h 2.

BACKGROUND: Measurement of whole peanut-specific IgE (sIgE) is often used to confirm sensitization but does not reliably predict allergy. Ara h 2 is the dominant peanut allergen detected in 90% to 100% of patients with peanut allergy and could help improve diagnosis. OBJECTIVES: We sought to determine whether Ara h 2 testing might improve the accuracy of diagnosing peanut allergy and therefore circumvent the need for an oral food challenge (OFC). METHODS: Infants from the population-based HealthNuts study underwent skin prick tests to determine peanut sensitization and subsequently underwent

Influence of plant lipids on immune responses in mice to the major Brazil nut allergen Ber e 1.

BACKGROUND: Lipids, particularly bacterial lipopolysaccharide, can impact on immune responses to proteins, with low doses enhancing type 2 responses. OBJECTIVE: We have examined the influence of natural plant lipid extracts on antibody responses provoked in mice by recombinant Ber e 1, the major allergen in Brazil nuts. METHODS: BALB/c strain mice were immunized (by intraperitoneal injection) with natural or recombinant Ber e l produced in Pichia pastoris and admixed with various lipid fractions isolated from Brazil nuts. Ara h 2-specific IgE is superior to whole peanut extract-based serology or skin prick test for diagnosis of peanut allergy in infancy.

BACKGROUND: Screening of high-risk infants for peanut allergy (PA) before introduction is now recommended in the United States, but the optimal approach is not clear. OBJECTIVE: We sought to compare the diagnostic test characteristics of peanut skin prick test (SPT), peanut-specific IgE (sIgE), and sIgE to peanut components in a screening population of infants before known peanut exposure.
