Testicular Architecture of Men with 46,XX Testicular Disorders of Sex Development.

BACKGROUND: A subtype of disorders of sex development (DSD) in individuals with a 46,XX karyotype who are phenotypically male is classified as testicular DSD (46,XX TDSD). These individuals develop testes but are infertile due to germ cell loss. However, little is known about their testicular architecture. METHODS: We analyzed biopsies of four SRY positive 46,XX TDSD men for testicular architecture, Sertoli (SCs) and Leydig cells (LCs). These were compared with biopsies of men with normal spermatogenesis (NS, n = 4), men with Klinefelter syndrome, 47 XXY (KS, n = 4), and men with AZF deletion

Possible misdiagnosis of 46,XX testicular disorders of sex development in infertile males.

Objectives: The 46,XX disorders of sex development (DSD) is a rare genetic cause of male infertility and possible misdiagnosis of this condition has never been reported. We aim to investigate clinical characteristics and laboratory results of infertile males with possibly misdiagnosed 46,XX DSD. Methods: Between January 2008 and December 2017, a retrospective case series study was performed involving sixteen 46,XX DSD males without azoospermia factor (AZF) deletion. Demographics, clinical features, laboratory results and assisted reproductive technology (ART) outcomes of these patients were de

46,XX T testicular disorder of sex development. Case report.

OBJECTIVE: We present a case of X-Y translocation with male phenotype (46,XX testicular disorder of sex development) and review the literature. METHODS: Disorders of sex development with mismatch of genetic, gonadal and phenotypic sex are quite rare, and some are due to genetic or chromosomal abnormalities. The karyotype was investigated by a cytogenetic study of peripheral blood (phytohemagglutinin-timulated lymphocyte culture over 72 hours). G-banding analysis of 25 metaphases showed a 46,XX chromosome constitution (46 chromosomes with XX sexual composition). Fluorescence in situ hybridizat

Two males with SRY-positive 46,XX testicular disorder of sex development.

The 46,XX testicular disorder of sex development (46,XX testicular DSD) is a rare phenotype associated with disorder of the sex chromosomes. We describe the clinical, molecular, and cytogenetic findings of a 16- and a 30-year-old male patient with sex-determining region Y (SRY)-positive 46,XX testicular DSD. Chromosomal analysis revealed 46,XX karyotype. Fluorescence in situ hybridization (FISH) showed the SRY region translocated to the short arm of the X chromosome. The presence of the SRY gene was also confirmed by polymerase chain reaction (PCR). The X chromosome inactivation (XCI) assay sh

46,XX Testicular Disorder of Sex Development (DSD) Presenting With Male Hypogonadism.

Disorders of sex development are genetically complex, and phenotypes range from hypospadias to completely masculinized or feminized genitalia with a discordant karyotype. They arise as a result of alterations in gonad formation (sex determination stage) or function (sex differentiation stage). Reaching a specific diagnosis with the aid of molecular technologies is important for individualized management, genetic counseling, and prognostic prediction for fertility and risk of tumor development. This case report describes a young adult male who was referred initially with concern for Klinefelter

Prenatal sex discordance revealing SRY-positive 46,XX testicular disorder of sex development.

46,XX testicular disorder of sex development (DSD) is a rare condition usually diagnosed during adolescence or adulthood due to infertility or hypogonadism. Early diagnosis in phenotypically normal infants is uncommon. We report a male infant identified following discordant prenatal sex assessment, with noninvasive prenatal testing suggesting a female fetus and prenatal ultrasonography demonstrating male genitalia. Postnatal chromosomal analysis revealed 46,X,add(X) (p22.3) with SRY positivity. Physical examination showed normal male external genitalia with bilaterally descended testes and no

Atypical presentation of potter sequence associated with 46, XX testicular disorder of sex development.

Prenatal diagnosis of congenital anomalies is primarily based on ultrasound assessment. This may be limited in settings with structural abnormalities and diagnoses that rely heavily on genetic testing. A 33-year-old white woman, gravida 1, para 0, at 17 weeks of gestation, presented to the outpatient clinic the day after being involved in a motor vehicle collision, without any obstetric complaints. Upon physical examination, minimal tenderness was noted in the left lateral back. Serological markers were unremarkable. Ultrasound revealed an absence of fetal urinary bladder and bilateral renal a

SRY-positive 46,XX testicular disorder of sex development in adult monozygotic twins.

46,XX testicular disorder of sex development (DSD), also known as De la Chapelle syndrome, is a rare condition characterized by a male phenotype in individuals with a 46,XX karyotype, most commonly caused by translocation of the sex-determining region Y (SRY) gene. Diagnosis is frequently delayed until adulthood because external genitalia are typically unambiguous. We report adult monozygotic twins referred for infertility evaluation who were found to have azoospermia and markedly reduced testicular volume. Cytogenetic analysis revealed a 46,XX karyotype in both individuals, and molecular test

Disorders of Sex Development with Testicular Differentiation in SRY-Negative 46,XX Individuals: Clinical and Genetic Aspects.

Virilisation of the XX foetus is the result of androgen excess, resulting most frequently from congenital adrenal hyperplasia in individuals with typical ovarian differentiation. In rare cases, 46,XX gonads may differentiate into testes, a condition known as 46,XX testicular disorders of sex development (DSD), or give rise to the coexistence of ovarian and testicular tissue, a condition known as 46,XX ovotesticular DSD. Testicular tissue differentiation may be due to the translocation of SRY to the X chromosome or an autosome. In the absence of SRY, overexpression of other pro-testis genes, e.

INTRODUCTION: The gonadal genetics and germ cell tumor (GCT) risk in SRY-negative 46,XX testicular and ovotesticular disorders of sex development (DSD) are poorly understood and debated. This study aimed to investigate the gonadal genetic etiology and evaluate GCT risk in an independent cohort. METHODS: We conducted a retrospective analysis of SRY-negative 46,XX testicular and ovotesticular DSD patients who underwent gonadal biopsy or gonadectomy. Routine next-generation sequencing was performed on peripheral blood samples. In gonadal tissues, quantitative PCR was utilized for SRY gene detect

A 46,XX testicular disorder of sex development caused by a Wilms' tumour Factor-1 (WT1) pathogenic variant.

Molecular diagnosis is rarely established in 46,XX testicular (T) disorder of sex development (DSD) individuals with atypical genitalia. The Wilms' tumour factor-1 (WT1) gene is involved in early gonadal development in both sexes. Classically, WT1 deleterious variants are associated with 46,XY disorders of sex development (DSD) because of gonadal dysgenesis. We report a novel frameshift WT1 variant identified in an SRY-negative 46,XX testicular DSD girl born with atypical genitalia. Target massively parallel sequencing involving DSD-related genes identified a novel heterozygous WT1 c.1453_1456
