[Effect of cerebral intraventricular administration of 5,6-dihydroxytryptamine and 5,7-dihydroxytryptamine on the cyclic secretion of corticosterone in the rat].

The effect of two potent neurotoxic agents for 5HT neurons, 5,6-DHT and 5,7-DHT, intraventricularly injected, has been studied on the values and rhythm of plasmatic corticosterone secretion in the male Wistar rat model, along an entire day: 4 a.m., 8 a.m., noon, 5 p.m. and midnight. The dose administered for each drug was 100 micrograms/animal in a single injection. The levels of corticosterone were determined 15 and 30 days post-injection. The effects observed for both drugs were similar at similar times: highly significant decreases in all the studied points which fall under the same line of

[Effect of 5,7-dihydroxytryptamine on pain sensitivity and analgesic activity of morphine].

Electron microscopy of rats ultrathin sections from dorsal and central raphe nucleus and spinal cord after 5,7-dihydroxytryptamine intracisternal microinjection (200 micrograms) has revealed neurones and axonal terminals distruction, which associated with tail-flick hypoalgesia and blood pressure nociceptive reactions diminished. In this condition the morphine (2 mg/kg) analgesia and drug depressive effect on pain hemodynamic manifestations increase significantly.

Recovery of brain noradrenaline after 5,7-dihydroxytryptamine-induced axonal lesions in the rat.

Time-dependent changes in regional CNS noradrenaline (NA) concentration, 3H-NA uptake and fluorescence morphology of CNS NA neurons were analysed in the adult rat up to 6 months after intraventricular injection of 5,7-dihydroxytryptamine (5,7-DHT), and compared with the time-course of changes in brain and spinal cord indolamine neurons. Following a substantial depletion of both amines in all CNS regions (telodiencephalon, brainstem and spinal cord) at 10 days after 150 mug 5,7-DHT, brain NA--but not 5-HT--levels recovered to near-normal values in brainstem and forebrain (35% below the age-matc

5,7-Dihydroxytryptamine induced changes in the postnatal development of central 5-hydroxytryptamine neurons.

Systemic administration of the neurotoxic compound 5,7-dihydroxytryptamine (5,7-HT) to newborn rats led acutely (within 1--2 h) to a marked reduction of the in vitro uptake of [3H]5-hydroxytryptamine (5-HT) in homogenates from the cerebral cortex (75% decrease) and the pons-medulla (60% decrease). When 5,7-HT was administered postnatally we found that between days 5 an7 resistance developed in the cerebral cortex towards the 5,7-HT induced reduction in 3H-5-HT uptake. This was most likely the result of the postnatal development of the blood-brain barrier. These results show that 5,7-HT can pas

Effects of 5,7-dihydroxytryptamine lesion of the dorsal raphe nucleus on ethanol discrimination in the rat.

It has been shown that ethanol produces a complex interoceptive cue in rodents with distinct GABAergic, glutamatergic, and serotonergic (5-hydroxytryptamine, 5-HT) components. The present study aimed to examine the contribution of the 5-HT system originating in the dorsal raphe nucleus (DRN) to the discriminative stimulus effects of ethanol in male Wistar rats. Therefore, selective lesions of 5-HT neurons in the DRN were induced by microinfusions of 5,7-dihydroxytryptamine. The DRN- and sham-lesioned rats were trained to discriminate ethanol (1.0 g/kg) from saline in a standard two-lever drug

Autoradiographic evaluation of the influence of hypothalamic 5,7-dihydroxytryptamine lesion on brain serotonin synthesis.

The influence of a unilateral stereotaxically induced 5,7-dihydroxytryptamine (5,7-DHT) lesion in the dorsolateral hypothalamus on brain serotonin synthesis was evaluated by an autoradiographic method, using labelled alpha-methyl-L-tryptophan (alpha-MTrp). The hypothalamus was selected as the lesion site because it receives well defined and relatively large projections from the raphe nuclei. Data suggest that the unilateral lesion in the dorsolateral hypothalamus had a significant influence (an increase) on the rate of serotonin synthesis in the large majority of ipsilateral brain structures e

Elimination of Serotonergic Neurons by Stereotaxic Injection of 5,7-Dihydroxytryptamine in the Dorsal Raphe Nuclei of Mice.

Stereotaxic injection has been widely used for direct delivery of compounds or viruses to targeted brain areas in rodents. Direct targeting of serotonergic neurons in the dorsal raphe nucleus (DRN) can cause excessive bleeding and animal death, due to its location below the superior sagittal sinus (SSS). This protocol describes the generation of a DRN serotonergic neuron-lesioned mouse model (>90% survival rate) with stable loss of >70% 5-HT-positive cells in the DRN. The lesion is induced by stereotaxic injection of a selective serotonergic neurotoxin 5,7-dihydroxytryptamine (5,7-DHT) into th

Regional central serotonin-2 receptor binding and phosphoinositide turnover in rats with 5,7-dihydroxytryptamine lesions.

"Denervation supersensitivity" of serotonin (5-HT) receptors has been proposed to explain the behavioral supersensitivity to 5-hydroxytryptophan (5-HTP) which develops after lesions of indoleamine neurons with 5,7-dihydroxytryptamine (5,7-DHT). To examine the possible role of receptor recognition sites and second messenger activity in supersensitivity, we measured regional 5-HT2 receptor ligand binding and 5-HT-stimulated phosphoinositide turnover in adult rats with 5,7-DHT lesions made by intracisternal injection and their saline-treated controls. Effects of lateral ventricle injection of 5,7-dihydroxytryptamine on neurons in the medial prefrontal cortex of rats: An electrophysiology study.

The medial prefrontal cortex (mPFC) is closely associated with various psychopathologies in humans, and its dysfunction is invariably accompanied by abnormalities in the serotonin (5-hydroxytryptamine, 5-HT) system of the brain. In this study, in-vivo extracellular recording techniques were used to investigate changes in the excitability of pyramidal neurons and interneurons in the rat mPFC following injection of 5,7-dihydroxytryptamine (5,7-DHT) into the bilateral lateral ventricles to damage the serotoninergic neurons. The levels of 5-HT in the mPFC and dorsal raphe nucleus of rats were dete

Effect of 5,7-dihydroxytryptamine lesion on mianserin-induced conditioned place aversion and on 5-hydroxytryptamine1C receptors in the rat brain.

The lesion of serotonergic neurons (by an intraventricular injection of 5,7-dihydroxytryptamine) potentiated the conditioned place aversion induced by the 5-hydroxytryptamine1C/5-hydroxytryptamine2 antagonist mianserin in rats. This effect was selective for mianserin as the same lesion suppressed the conditioned place aversion induced by the benzodiazepine inverse agonist FG-7142. Previous results had shown the involvement of the 5-hydroxytryptamine1C receptors in the conditioned place aversion induced by mianserin [Rocha et al. (1993) Behav. Pharmac. 4, 101-106]. It was thus of interest to in

The functional significance of neonatal 5,7-dihydroxytryptamine lesions in the rat: response to selective 5-HT1A and 5-HT2,1C agonists.

To study the involvement of serotonin (5-HT) receptor subtypes in behavioral supersensitivity following neonatal 5,7-dihydroxytryptamine (5,7-DHT) lesions, we measured acute behavioral responses to a single dose of selective 5-HT1A (8-OH-DPAT) or 5-HT2,1C (DOI) agonist compared to 5-hydroxytryptophan (5-HTP) in rats injected with 5,7-DHT intraperitoneally or intracisternally 14 weeks earlier. Only intraperitoneal 5,7-DHT injection resulted in brainstem 5-HT hyperinnervation, but cortical 5-HT depletions were also less. Effects of 5,7-dihydroxytryptamine and 6-hydroxydopamine on head-twitch response induced by serotonin, p-chloroamphetamine, and tryptamine in mice.

Head-twitch response (HTR) in mice was induced by intracerebroventricular injection of tryptamine (TRA) as well as serotonin (5-HT) and p-chloroamphetamine (PCA). Pretreatment with 5,7-dihydroxytryptamine enhanced both the 5-HT-induced and the TRA-induced HTR. The PCA-induced HTR, however, was attenuated by the drug. On the other hand, pretreatment with 6-hydroxydopamine did not alter the 5-HT response but enhanced both the PCA- and the TRA-induced response. These results suggest that 5-HT may directly stimulate the post-synaptic receptors, while the PCA response may be based on the release of

Regeneration of normal terminal innervation patterns by central noradrenergic neurons after 5,7-dihydroxytryptamine-induced axotomy in the adult rat.

The regeneration of central adrenergic axons has been followed between 5 days and 18 months after 5,7-dihydroxytryptamine(5,7-DHT)-induced axotomy in the adult rat, using fluorescence histochemistry in combination with noradrenaline (NA) determinations and [3H]NA uptake measurements. Thyrotropin-releasing hormone reverses the supersensitively depressed monosynaptic transmission by serotonin in 5,7-dihydroxytryptamine-treated neonatal rats in vitro.

The interaction of serotonin and thyrotropin-releasing hormone (TRH) on monosynaptic reflex (MSR) in isolated neonatal spinal cords was examined. Superfusion of serotonin (1-30 microM) in untreated cords, depressed the MSR in a dose-dependent manner. The depression was about 25% at 10 microM of serotonin, in either control or vehicle-treated groups. While for the same concentration of serotonin, the depression was 97 +/- 2.1% of the control in cords from 5,7 dihydroxytryptamine (5,7-DHT)-treated animals. Developmental changes in serotonin and 5-hydroxyindoleacetic acid concentrations and opiate receptor binding in rat spinal cord following neonatal 5,7-dihydroxytryptamine treatment.

Normal levels of serotonin and 5-hydroxyindoleacetic (5-HIAA) were measured in the developing rat spinal cord and brain stem using HPLC. Serotonin, 5-HIAA and 3H-naloxone binding were monitored following neonatal 5,7-dihydroxytryptamine (5,7-DHT) treatment. Serotonin in the spinal cord develops biphasically with peaks at gestation day 21 (E21) and postnatal day 8 (P8) while 5-HIAA peaks at P3 and between P8 and P11. In the brain stem, serotonin gradually rises from E15 to adult levels which are reached by P32 while 5-HIAA rises sharply to P15 and thereafter decreases to adult levels by P32. 5,
