Plasma concentrations of 5-methoxytryptamine, 5-methoxytryptophol and melatonin after 5-methoxytryptamine administration of golden hamsters: physiological implications.

5-Methoxytryptamine (5-MT), 5-methoxytryptophol (5-ML) and melatonin (Mel) were measured in the plasma after 2, 5, and 8 weeks administration of 25 micrograms 5-MT to golden hamsters kept under long photoperiod. 5-MT showed a one compartment kinetic profile in the plasma with half lives of 14.8 min after 2 weeks, 15 min after 5 weeks and 19.1 min after 8 weeks. A rapid metabolism of 5-MT was shown, Mel and 5-ML being detected in the plasma following 5-MT administration. However it was also shown that the gonadal atrophy observed after 5-MT administration cannot be due to its metabolism into th

Structural pharmacology and therapeutic potential of 5-methoxytryptamines.

Psychedelic substances such as lysergic acid diethylamide (LSD) and psilocybin show potential for the treatment of various neuropsychiatric disorders1-3. These compounds are thought to mediate their hallucinogenic and therapeutic effects through the serotonin (5-hydroxytryptamine (5-HT)) receptor 5-HT2A (ref. 4). However, 5-HT1A also plays a part in the behavioural effects of tryptamine hallucinogens5, particularly 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT), a psychedelic found in the toxin of Colorado River toads6. Although 5-HT1A is a validated therapeutic target7,8, little is known about

Identification and diurnal studies of pineal and serum 5-methoxytryptamine in the rat and quail.

Picomole to femtomole per gland/ml levels of 5-methoxytryptamine (5MT) in the pineal gland and serum of the rat and quail were determined, in the absence of a monoamine oxidase inhibitor, by using the ultra-sensitive technique of capillary column gas chromatography/electron-capture negative ion chemical ionization mass spectrometry. Diurnal rhythms of pineal 5MT with high levels at mid-light and low levels at mid-dark were found in both species, while serum 5MT levels showed less or no diurnal variations. The definitive presence of 5MT in the pineal gland and blood circulation provides further

5-Methoxytryptamine is metabolized by monoamine oxidase A in the pineal gland and plasma of golden hamsters.

Pineal concentrations of 5-methoxytryptamine, 5-methoxytryptophol, 5-methoxyindole acetic acid and melatonin were determined using high performance liquid chromatography following administration of different monoamine oxidase (MAO) inhibitors (clorgyline, pargyline and deprenyl) to golden hamsters. Plasma concentrations of 5-methoxytryptamine, 5-methoxytryptophol and melatonin were also measured following 5-methoxytryptamine administration to hamsters pretreated with MAO inhibitors. A significant increase in pineal and plasma 5-methoxytryptamine together with a decrease in 5-methoxytryptophol

ICS 205-930 reduces 5-methoxytryptamine-induced short-circuit current in stripped pig jejunum.

The effect of ICS 205-930 on serotonin (5-hydroxytryptamine, 5-HT) and 5-methoxytryptamine (5-MeOT) induced short-circuit current (SCC) was studied in muscle-stripped pig jejunum in vitro. The selective 5-HT4 receptor agonist 5-MeOT and 5-HT both induced a concentration-dependent increase in SCC. The maximal efficacy of 5-HT was twice that of 5-MeOT. ICS 205-930 (Tropisetron) induced a small increase in SCC. Furthermore, ICS 205-930 reduced the 5-MeOT but not the 5-HT induced increase in SCC, except at the 100 microM ICS 205-930 concentration. Pretreatment with bumetanide reduced 5-HT, 5-MeOT,

Biochemotherapy with immunomodulating pineal hormones other than melatonin: 5-methoxytryptamine as a new oncostatic pineal agent.

Several experiments have demonstrated that pineal gland plays a physiological anticancer role. Melatonin (MLT), its most investigated hormone, is a natural anticancer agent. However, MLT would not be the only endocrine molecule responsible for the anticancer property of the pineal gland. In fact, another pineal indole hormone, the 5-methoxytryptamine (5-MTT), has appeared to exert in vitro an antitumour activity superior to that of MLT itself. Previous studies have already shown the therapeutic anticancer action of MLT in association with chemotherapy also in human neoplasms. This study was pe

Secretion of the methoxyindoles melatonin, 5-methoxytryptophol, 5-methoxyindoleacetic acid, and 5-methoxytryptamine from trout pineal organs in superfusion culture: effects of light intensity.

The teleost pineal organ is a photoreceptive endocrine organ that synthesizes the hormone melatonin through specialized intrapineal photoreceptor cells in a light-dependent manner. The present study investigated whether the methoxyindoles 5-methoxytryptophol (5-MTOL), 5-methoxyindoleacetic acid (5-MIAA), and 5 methoxytryptamine (5-MT) correspond to melatonin secretion synthesized and released from explanted, superfused pineal organs in response to direct light stimulation and whether their release is correlated with the level of dark adaptation. Melatonin release in superfusion cultures using

Reduction of cisplatin-induced anemia by the pineal indole 5-methoxytryptamine in metastatic lung cancer patients.

OBJECTIVE: It has been demonstrated that the hematopoiesis is under a neuroendocrine control, namely mediated by the pineal gland. The pineal indole melatonin (MLT) has appeared to exert thrombopoietic and lymphopoietic activity, whereas it has no relevant effect on red cell differentiation. The present study was performed to evaluate the influence of another pineal indole, the 5-methoxytryptamine (5-MTT) on red cell line and hemoglobin production. MATERIALS & METHODS: The study was carried out in metastatic lung cancer patients who underwent a chemotherapeutic combination containing cisplati

Day-night rhythm of 5-methoxytryptamine biosynthesis in the pineal gland of the golden hamster (Mesocricetus auratus).

5-Methoxytryptamine is a potent agonist of presynaptic 5-hydroxytryptamine autoreceptors modulating serotonin release in the central nervous system. This methoxyindole can be synthesized in the pineal gland, but its presence in vivo is still controversial, probably because of rapid catabolism by monoamine oxidase. An improved high-pressure liquid chromatography method, with coulometric detection, has been developed for the simultaneous measurement of melatonin, 5-methoxytryptamine, 5-methoxytryptophol and 5-methoxyindolacetic acid. We have demonstrated a day-night rhythmicity in the amount of

Effect of different photoperiods on the diurnal rhythm of 5-methoxytryptamine in the pineal gland of golden hamsters (Mesocricetus auratus).

This study tested the photo-dependency of the rhythmic synthesis of 5-methoxytryptamine (5-MT) in the pineal gland of golden hamsters. After pargyline administration, pineal 5-methoxytryptamine and melatonin were measured by HPLC in male golden hamsters kept under short and long photoperiod. In both photoperiodic regimes, a clear 5-MT rhythm was observed which fitted a sinusoidal function with high values occurring during the daytime and low values occurring during the night-time. The duration of the low night-time levels was clearly proportional to the length of the dark phase. A marked rhyth

Effects of pinealectomy and melatonin or 5-methoxytryptamine on testicular LH and PRL receptors in Syrian hamsters (Mesocricetus auratus).

The pineal has been previously shown to be an important factor in the regulation of testicular function in photoperiodic mammals. The effects of lack or increase in pineal hormones on testicular hormonal receptors has, therefore, been examined. Pinealectomy decreased the concentration of testicular LH receptors in hamsters exposed to either a long or short photoperiod but had no effect on the concentration of testicular PRL receptors. In animals exposed to a short photoperiod, pinealectomy prevented testicular regression and the concomitant decreases in total LH and PRL receptor contents. Trea

Modulation of anticancer cytokines IL-2 and IL-12 by melatonin and the other pineal indoles 5-methoxytryptamine and 5-methoxytryptophol in the treatment of human neoplasms.

Lymphocyte number still remains one of the most important immune parameters predicting the prognosis of advanced cancer patients. IL-2 and IL-12 are the main antitumor cytokines in humans, and their effect is modulated by the neuroendocrine system, mainly by the pineal gland through the circadian release of melatonin (MLT) and perhaps that of other indole hormones, such as 5-methoxytryptamine (5-MTT), and 5-methoxytryptophol (5-MTP). MLT has been proven to exert important antitumor immunomodulating effects, whereas the possible immunomodulatory properties of the other pineal indoles are still

Effect of melatonin and 5-methoxytryptamine administration on the testis and pineal gland activity of the fresh-water snake, Natrix piscator.

Effects of melatonin (aMT) and 5-methoxytryptamine (MT) were studied on the testicular activity cycle of the fresh-water snake, Natrix piscator. The subcutaneous implantation and the injections (morning as well as evening) of these two methoxyindoles prevented testicular recrudescence, retarded the testicular active phase, and accelerated the rate of regression of testes, while having no effect on the inactive testes. Contrary to this, these two compounds increased the pineal gland weight during different reproductive phases. These results revealed that both aMT and MT, whether they were conti

5-methoxytryptamine improves hepatic inflammation and insulin resistance in a macrophage C-X-C motif chemokine ligand 14 dependent manner.

Accumulating evidence indicates that tryptophan metabolism plays important roles in insulin resistance, and glucolipid metabolism. Our preliminary study revealed a significant decrease in the serum levels of the tryptophan metabolite 5-methoxytryptamine (5MT), in both high-fat diet (HFD)-fed and db/db mice. Notably, these levels were restored following treatment with the hypoglycemic agent, the dipeptidyl peptidase-4 inhibitor (DPP-4i) sitagliptin, suggesting a potential relationship between 5MT and metabolic homeostasis. In the current study, we demonstrated that 5MT treatment ameliorated ins

Suppression of UV-induced erythema by topical treatment with melatonin (N-acetyl-5-methoxytryptamine). Influence of the application time point.

BACKGROUND: In a previous study, we reported a significant and dose-dependent suppression of UV-induced erythema in human skin by a topically applied melatonin preparation. OBJECTIVE: The present double-blind randomized study was designed to examine the influence of the application time point of topical melatonin on this antierythema effect. METHODS: Defined small areas on the lower back of 20 volunteers were treated with 0.6 mg/cm2 melatonin dissolved in a nanocolloid gel carrier either 15 min before or 1, 30 or 240 min after UV irradiation with twice the individual minimal erythema dose de

Low ambient temperature does not affect the pineal concentrations of either 5-methoxytryptamine or melatonin in golden hamsters kept under short photoperiod.
