Unexpected 16S rRNA heterogeneity in 'Acetobacterium dehalogenans' and reclassification as Acetobacterium malicum subsp. dehalogenans subsp. nov.

Strain MCT is a strictly anaerobic, homoacetogenic bacterium with the ability to utilize methyl chloride as the sole energy source. It was tentatively assigned to the genus Acetobacterium as 'Acetobacterium dehalogenans'. Due to sequence ambiguities, it was not possible to determine the 16S rRNA gene sequence of this strain by direct sequencing of a PCR-amplified DNA segment. Whole-genome sequencing revealed significant heterogeneity amongst the five rRNA operons detected in this strain, with maximum sequence differences between the individual 16S rRNA genes exceeding 1.4%, compared to <0.8% i

Biotransformation and biodefluorination of chlorinated polyfluorocarboxylic acids by Acetobacterium species.

Chlorinated per- and polyfluoroalkyl substances (Cl-PFAS) represent an important subgroup in replacement chemicals for legacy PFAS, and they have been detected in various environments including water bodies, soil, as well as air particles. However, the biodegradability of these chemicals is largely unknown. A recent study reported dechlorination triggered anaerobic biodefluorination of Cl-PFCAs by activated sludge communities. In this study, we further investigated the biodefluorination of Cl-PFCAs by pure cultures. Six of ten selected Cl-PFCAs displayed significant defluorination by Acetobact

Engineering Acetobacterium woodii for the production of isopropanol and acetone from carbon dioxide and hydrogen.

The capability of four genetically modified Acetobacterium woodii strains for improved production of acetone from CO2 and hydrogen was tested. The acetone biosynthesis pathway was constructed by combining genes from Clostridium acetobutylicum and Clostridium aceticum. Expression of acetone production genes was demonstrated in all strains. In bioreactors with continuous gas supply, all produced acetic acid, acetone, and, surprisingly, isopropanol. The production of isopropanol was caused by an endogenous secondary alcohol dehydrogenase (SADH) activity at low gas-feeding rate. Although high amou

Enhanced biotransformation of carbon tetrachloride by Acetobacterium woodii upon addition of hydroxocobalamin and fructose.

The objective of this study was to evaluate the effect of hydroxocobalamin (OH-Cbl) on transformation of high concentrations of carbon tetrachloride (CT) by Acetobacterium woodii (ATCC 29683). Complete transformation of 470 microM (72 mg/liter [aqueous]) CT was achieved by A. woodii within 2.5 days, when 10 microM OH-Cbl was added along with 25.2 mM fructose. This was approximately 30 times faster than A. woodii cultures (live or autoclaved) and medium that did not receive OH-Cbl and 5 times faster than those controls that did receive OH-Cbl, but either live A. woodii or fructose was missing.

A characterized promoter suite enables metabolic rewiring of the acetogen Acetobacterium woodii toward ethanol production.

Acetogenic bacteria are promising platforms for converting gaseous and liquid one-carbon (C1) substrates such as carbon monoxide, carbon dioxide (CO2), formate and methanol into fuels and chemicals, but their application is limited by sparse genetic tools and poor control over metabolic fluxes. Here, we developed a modular plasmid assembly platform and a suite of characterized native, heterologous and methanol-inducible promoters for the model acetogen Acetobacterium woodii, enabling rapid and tunable control of gene expression together with CRISPR-based genome editing. Using an anaerobe-compa

Redirecting electron flow in Acetobacterium woodii enables growth on CO and improves growth on formate.

Anaerobic, acetogenic bacteria are well known for their ability to convert various one-carbon compounds, promising feedstocks for a future, sustainable biotechnology, to products such as acetate and biofuels. The model acetogen Acetobacterium woodii can grow on CO2, formate or methanol, but not on carbon monoxide, an important industrial waste product. Since hydrogenases are targets of CO inhibition, here, we genetically delete the two [FeFe] hydrogenases HydA2 and HydBA in A. woodii. We show that the ∆hydBA/hydA2 mutant indeed grows on CO and produces acetate, but only after a long adaptation

[Medium optimization and fermentation process regulation strategies for enhancing CO2 conversion to acetic acid by Acetobacterium woodii].

To meet the demand for CO2 resource utilization under the "dual-carbon" goals, the application of medium optimization combined with fermentation process regulation to enhance the acetate production and gas utilization capacity of Acetobacterium woodii represents an effective process optimization strategy. In this study, a multi-strategy approach combining medium optimization and fermentation process regulation was adopted to enhance the acetate yield of an A. woodii SL70 strain obtained via adaptive laboratory evolution (ALE). Firstly, through single-factor experiments and response surface met

Sodium limitation stimulates Acetobacterium woodii enrichment and boosts CO2 conversion to acetate in self-controlled microbial electrosynthesis systems.

Microbial electrosynthesis system (MES) enables the conversion of CO2 into acetate with low energy consumption. However, the accumulation of acetate brings inevitable survival troubles to the model microorganisms, the coulombic efficiency of MES is far from the theoretical value. Establishing a self-controlled system and providing optimized living conditions for model microorganisms can be regarded as a strategy to improve the coulombic efficiency. In this study, a flux-pH adaptive MES coupled with Acetobacterium woodii was constructed and the Na + -dependent characteristics of A. Woodii was f

Improving autotrophic D-lactate production by heterologous expression of a pyruvate formate lyase in an LDHD-expressing Acetobacterium woodii strain.

The anaerobic acetogen Acetobacterium woodii is a promising candidate producing chemicals while converting the greenhouse gas CO2. In this study, A. woodii was genetically engineered to express a lactate dehydrogenase (LDHD) from Leuconostoc mesenteroides, and a pyruvate formate lyase (PFL) as well as its activating enzyme (PFL-AE) originated from Clostridium pasteurianum to autotrophically produce D-lactate next to acetate, using H2 + CO2. In this study, an improved D-lactate production was observed compared to the control strain without PFL and PFL-AE. Especially, during anabolism, positive

Metabolic Reconstruction and Modeling Microbial Electrosynthesis.

Microbial electrosynthesis is a renewable energy and chemical production platform that relies on microbial cells to capture electrons from a cathode and fix carbon. Yet despite the promise of this technology, the metabolic capacity of the microbes that inhabit the electrode surface and catalyze electron transfer in these systems remains largely unknown. We assembled thirteen draft genomes from a microbial electrosynthesis system producing primarily acetate from carbon dioxide, and their transcriptional activity was mapped to genomes from cells on the electrode surface and in the supernatant. Transformation of tetrachloroethylene to trichloroethylene by homoacetogenic bacteria.

Eight homoacetogenic strains of the genera Acetobacterium, Clostridium and Sporomusa were tested for their ability to dechlorinate tetrachloroethylene (perchloroethene, PCE). Of the organisms tested only Sporomusa ovata was able to reductively dechlorinate PCE with methanol as an electron donor. Resting cells of S. ovata reductively dechlorinated PCE at a rate of 9.8 nmol h-1 (mg protein)-1 to trichloroethylene (TCE) as the sole product. The dechlorination activity depended on concomitant acetogenesis from methanol and CO2. Cell-free extracts of S. ovata, Clostridium formicoaceticum, Acetobact

Fe@NC-hybrid electrode enhancing acetate synthesis from CO2via microenvironment regulation in microbial electrochemical systems.

The practical implementation of CO2 valorization via microbial electrochemical systems (MES) is constrained by its relatively suboptimal production rate. Rational design of electrode-localized microenvironments offers a promising strategy to overcome this bottleneck. This study provided a regulation strategy with metal-microbial hybrid electrode which synergistically enhanced CO2 conversion efficiency and microbial activity in MES. The engineered Fe@NC-hybrid electrode demonstrated exceptional performance in modulating the electrode-suspended sludge interface microenvironment, exhibiting outst

Sustained Dechlorination of Vinyl Chloride to Ethene in Dehalococcoides-Enriched Cultures Grown without Addition of Exogenous Vitamins and at Low pH.

Trichloroethene (TCE) bioremediation has been demonstrated at field sites using microbial cultures harboring TCE-respiring Dehalococcoides whose growth is cobalamin (vitamin B12)-dependent. Bioaugmentation cultures grown ex situ with ample exogenous vitamins and at neutral pH may become vitamin-limited or inhibited by acidic pH once injected into field sites, resulting in incomplete TCE dechlorination and accumulation of vinyl chloride (VC). Here, we report growth of the Dehalococcoides-containing bioaugmentation culture KB-1 in a TCE-amended mineral medium devoid of vitamins and in a VC-amend

Synthetic Microbial Cocultivation for Targeted Production of Odd-Chain Carboxylates and Alcohols from Carbon Monoxide.

Microbial fermentation of syngas (CO, H2, CO2) using acetogens is a promising route for the revalorisation of one-carbon feedstocks. However, product diversification from syngas using pure cultures of these microorganisms remains a challenge. Here, we present a synthetic microbial community comprising the acetogen Acetobacterium wieringae JM, the propionigenic bacterium Anaerotignum neopropionicum and the chain elongator Clostridium kluyveri, which collectively produce odd- and even-chain carboxylic acids and higher alcohols from CO/CO2. In batch bioreactors, metabolite cross-feeding within th

Using electron balances and molecular techniques to assess trichoroethene-induced shifts to a dechlorinating microbial community.

This study demonstrated the utility in correlating performance and community structure of a trichloroethene (TCE)-dechlorinating microbial consortium; specifically dechlorinators, fermenters, homoacetogens, and methanogens. Two complementary approaches were applied: predicting trends in the microbial community structure based on an electron balance analysis and experimentally assessing the community structure via pyrosequencing and quantitative polymerase chain reaction (qPCR). Fill-and-draw reactors inoculated with the DehaloR^2 consortium were operated at five TCE-pulsing rates between 14 an

Interspecies interaction and effect of co-contaminants in an anaerobic dichloromethane-degrading culture.

An anaerobic stable mixed culture dominated by bacteria belonging to the genera Dehalobacterium, Acetobacterium, Desulfovibrio, and Wolinella was used as a model to study the microbial interactions during DCM degradation. Physiological studies indicated that DCM was degraded in this mixed culture at least in a three-step process: i) fermentation of DCM to acetate and formate, ii) formate oxidation to CO2 and H2, and iii) H2/CO2 reductive acetogenesis. The 16S rRNA gene sequencing of cultures enriched with formate or H2 showed that Desulfovibrio was the dominant population followed by Acetobact

Metabolic Versatility of acetogens in syngas Fermentation: Responding to varying CO availability.

Syngas fermentation using acetogenic bacteria offers a promising route for sustainable chemical production. However, gas-liquid mass transfer limitations and efficient co-utilization of CO and H2 pose significant challenges. This study investigated the kinetics of syngas conversion to acetate by Acetobacterium wieringae and Clostridium species in batch conditions under varying initial CO partial pressures (19 - 110 kPa). A. wieringae strains, exhibited superior growth in all gas compositions, with a maximum growth rate of 0.104 h-1. The distinct CO, H2, and CO2 consumption patterns revealed me

Defined Electrosynthetic Microbial Consortia Reveal Electron Transfer Modes Governing Acetate Production.

Precise manipulation of interspecies electron transfer (IET) is critical for advancing microbial electrosynthesis (MES) toward efficient CO2 bioconversion. Here, defined synthetic consortia are constructed by pairing Shewanella oneidensis MR-1, a model bidirectional electroactive bacterium, with Clostridium aceticum (electroactive acetogen) or Acetobacterium woodii (nonelectroactive acetogen), mimicking functional guilds commonly observed in natural MES communities. Co-cultivation markedly boosts acetate production by up to 88%, achieving 1.16 ± 0.01 and 1.05 ± 0.01 g L-1, with carbon conversi

Steering Bioelectrochemical CO2 Reduction Toward Methanogens Suppression and Acetogen Bioaugmentation.

Biological strategies for converting carbon dioxide (CO2) into valuable compounds are attractive approaches for a carbon-neutral future. Bioelectrochemical systems (BESs) represent an innovative strategy for the control of microbial metabolism, in which electrochemical techniques are adopted to stimulate reductive and oxidative processes. Acetogenesis and methanogenesis are the two main chemoautotrophic pathways of CO2 reduction usually present in anaerobic environments. Due to the syntrophic and competitive relationship between acetogens and methanogens, methanogenesis inhibition strategies s

Mutualistic interaction between dichloromethane- and chloromethane-degrading bacteria in an anaerobic mixed culture.

The microbial mixed culture RM grows with dichloromethane (DCM) as the sole energy source generating acetate, methane, chloride and biomass as products. Chloromethane (CM) was not an intermediate during DCM utilization consistent with the observation that CM could not replace DCM as a growth substrate. Interestingly, cultures that received DCM and CM together degraded both compounds concomitantly. Transient hydrogen (H2 ) formation reaching a maximum concentration of 205 ± 13 ppmv was observed in cultures growing with DCM, and the addition of exogenous H2 at concentrations exceeding 3000 ppmv
