Renewable acrylonitrile production.

Acrylonitrile (ACN) is a petroleum-derived compound used in resins, polymers, acrylics, and carbon fiber. We present a process for renewable ACN production using 3-hydroxypropionic acid (3-HP), which can be produced microbially from sugars. The process achieves ACN molar yields exceeding 90% from ethyl 3-hydroxypropanoate (ethyl 3-HP) via dehydration and nitrilation with ammonia over an inexpensive titanium dioxide solid acid catalyst. We further describe an integrated process modeled at scale that is based on this chemistry and achieves near-quantitative ACN yields (98 ± 2%) from ethyl acryla

Clinical toxicology of acrylonitrile.

Acrylonitrile monomer is used in the production of artificial fibres and resins. It has been used as a fumigant. Acrylonitrile has acute toxic effects for men and animals on over-exposure by inhalation of the vapor, dermal absorption of the liquid and oral intake. Symptoms in men are non-specific and predominantly related to the central nervous system, the respiratory tract, the skin and to the gastrointestinal tract. Severe acrylonitrile intoxication is followed by loss of consciousness, convulsions, respiratory arrest and death. The detailed investigation of a patient with complaints after c

Acrylonitrile metabolism in the rat.

Acrylonitrile (AN) is metabolized by an oxidative pathway via glycidonitrile and glycolaldehyde cyanohydrin to cyanide (CN-), which is transformed to thiocyanate. The major route of AN metabolism (more than 2/3), however, proceeds via cyanoethylation of glutathione, to N-acetyl-S-(2-cyanoethyl)cysteine (AN-mercapturic acid) as a final product.

Mechanisms of immunotoxic effects of acrylonitrile.

Cholinesterase reactivator dipyroxime and hydrogen cyanide antidote anticyan partially restore cell and humoral immune response in mice after acute acrylonitrile poisoning (0.5 LD(50)). When used in combination these drugs virtually completely prevented the suppression of immune reactions. The main mechanism of immunotoxic effect of acrylonitrile is mediated through inhibition of T lymphocyte esterases and a(3) component of cytochrome c oxidase of immunocyte mitochondrial respiration enzymes, which is important for prevention and treatment of immune disturbances caused by this toxin.

Acrylonitrile induces autolysis Bacillus subtilis.

Acrylonitrile (AN) is an industrial chemical used in the manufacture of plastics and other polymers. AN has been reported to be an acute toxin and is a known carcinogen in rodents. When AN was mixed with suspensions of Bacillus subtilis, the bacteria began autolysis. It was determined that AN is partially converted to cyanide, a strong protonophore in B. subtilis. Autolytic enzymes in B. subtilis become active when the protonmotive force is dissipated. The amount of cyanide produced from AN, however, was not enough to promote autolysis in exponential B. subtilis. This is the first report showi

Two routes of acrylonitrile metabolism.

Acrylonitrile (AN) is metabolized in two different routes. The minor route of AN metabolism is its conversion to cyanide. In vitro experiments confirmed that this biotransformation proceeds via glycidonitrile (catalyzed by hepatic monooxygenases) and glycolaldehyde cyanohydrin (catalyzed by epoxide hydrase). Cyanohydrin is spontaneously decomposed to cyanide, which is then metabolized in vivo to thiocyanate by rhodanese. A major route of AN-metabolism (more than 2/3), however, proceeds via its conjugation with glutathione catalyzed by glutathione S-alkenetransferases, to N-acetyl-S-(2-cyanoeth

Possible functional immunotoxicity of acrylonitrile (VCN).

Acrylonitrile (vinyl cyanide, VCN), an environmental pollutant, has been shown to be an animal and human carcinogen particularly for the GIT. In a previous work done in our laboratory, VCN induced immunosuppressive effects as indicated by a decrease in plaque forming cell (PFC) response to SRBCs (sheep red blood cell) immunization, a marked depletion of spleen lymphocyte subsets by flow cytometric analysis as well as bacterial translocation of the normal flora leading to brachial lymph node abscess. This work was carried out to evaluate the systemic and/or local immunotoxic potential of VCN. A

Occupational contact dermatitis due to acrylonitrile.

Within DSM Chemicals BV, a producer of acrylonitrile, skin complaints are frequent. The majority of these are of an irritant nature, while a smaller portion is based on acquired allergies. Allergological examination revealed 5 employees with an allergy to acrylonitrile. 1 of these subjects also developed paraesthesiae in the skin sites affected, a finding not previously described for acrylonitrile. In the guinea pig maximization test (GPMT), acrylonitrile showed strong allergenic potential. For prevention and treatment of contact allergologic disorders, close cooperation between occupational h

Gas-phase photocatalytic oxidation of acrylonitrile.

Photocatalytic oxidation (PCO) of acrylonitrile (AN) on titanium dioxide in the gaseous phase was studied. AN readily undergoes photocatalytic degradation in a gas-solid system by using TiO(2) Degussa P25. The AN PCO volatile products, visible in the infrared spectra, included nitrogen dioxide, nitrous oxide, carbon dioxide, water, hydrogen cyanide and carbon monoxide. Longer contact time resulted in deeper oxidation of AN with decreasing hydrogen cyanide and increasing nitrogen dioxide content. The effect of temperature increasing from 60 to 130 degrees C was observed to be slightly negative

Acrylonitrile: in vivo metabolism in rats and mice.

Acrylonitrile (VCN) is metabolized to cyanide in rats and mice. Cyanide levels following oral administration of an LD50 of VCN or KCN were determined in blood and organs of treated rats and mice. After administration of VCN, cyanide levels were significantly lower than those following treatment with KCN in rats, whereas in mice the difference was not significant. Differences in VCN toxicity signs were observed in rats and mice. In rats, early VCN toxicity signs were cholinomimetic such as: salivation, diarrhea, peripheral vasodilatation, and excessive gastric secretion. These signs differed fr

Metabolism of acrylonitrile to cyanide. In vitro studies.

In liver fractions from Sprague-Dawley rats, the metabolism of acrylonitrile (VCN) to cyanide (CN-) was localized the microsomal fraction and required NADPH and O2 for maximal activity. The biotransformation of VCN to CN- was characterized with respect to time, microsomal protein concentration, pH, and temperature. Metabolism of VCN was increased in microsomes obtained from phenobarbital-, Aroclor 1254-, or 3-methylcholanthrene-treated rats (479%, 414%, and 142% of control, respectively) and decreased with CoCl2 treatment (54% of control). Acrylonitrile-induced oxidative DNA damage in rat astrocytes.

Chronic administration of acrylonitrile results in a dose-related increase in astrocytomas in rat brain, but the mechanism of acrylonitrile carcinogenicity is not fully understood. The potential of acrylonitrile or its metabolites to induce direct DNA damage as a mechanism for acrylonitrile carcinogenicity has been questioned, and recent studies indicate that the mechanism involves the induction of oxidative stress in rat brain. The present study examined the ability of acrylonitrile to induce DNA damage in the DI TNC1 rat astrocyte cell line using the alkaline Comet assay. Oxidized DNA damage

Lactoperoxidase catalyzes in vitro activation of acrylonitrile to cyanide.

Acrylonitrile (ACN) is a widely used industrial chemical. Although it is a well reported animal carcinogen, its current designation to humans is "possibly carcinogenic". The present study aimed at investigating the ability of LPO enzyme system to oxidize ACN to cyanide (CN(-)) in vitro. Detection of CN(-) served as a marker for the possible generation of free radical intermediates implicated in ACN induced toxicity in the activation process. Optimum conditions for the oxidation of ACN to CN(-) were characterized with respect to pH, temperature and time of incubation as well as ACN, LPO and H(2

Assessment of the acute acrylonitrile-induced neurotoxicity in rats.

Acrylonitrile (VCN) is an aliphatic nitrile which is used extensively in manufacturing of synthetic fibers, plastics, and rubber. Although the neurotoxicity of VCN is recognized, no thorough characterization of this effect has been reported. Current studies were designed to quantitatively characterize the acute phase of VCN-induced cholinomimetic neurotoxicity, and to determine the effects of dose, route of administration, and atropine on such toxicity. Administration of a single gavage or subcutaneous doses of 20, 40, or 80 mg VCN/kg to male Sprague-Dawley rats causes two distinctive phases o

[N-Acetylcysteine as an antidote in accidental acrylonitrile poisoning].

Acute acrylonitrile intoxications are followed by loss of consciousness, convulsions, respiratory arrest, and may end fatally. Common cyanide antidotes have not proved to be effective. In previous animal experiments we could demonstrate that the cyanide antidotes show some protective effect only after oral acrylonitrile application. Only a minimal amount of inhaled acrylonitrile will be transformed into cyanide, in contrast to pharmacokinetics after oral intake. After acrylonitrile inhalation the toxic effect of the whole molecule ( cyanethylation ) is important, and sulfhydryl compounds show

Removal of cyanide from acrylonitrile wastewater using gas membrane.

Acrylonitrile wastewater is one of the most refractory industrial wastewaters as it contains cyanide at a high concentration. This study introduced a safe, effective and economic strategy, that is, use of the gas membrane to acrylonitrile wastewater treatment. Due to the complicated constituents of acrylonitrile wastewater, cyanide removal rate by gas membrane is very low. In order to enhance HCN removal, the operational conditions were optimized; pre-treatment strategies for fouling mitigation were also proposed and tested for acrylonitrile wastewater. The optimal operational parameters were

Acute nephrotoxic potential of acrylonitrile in Fischer-344 rats.

In view of its very limited information, we have evaluated the acute nephrotoxic potential of acrylonitrile (ACN) in male Fischer-344 rats. ACN was given i.p. in saline to groups of 6 rats at doses 0, 10, 20, 40, 60 and 80 mg/kg. Urines were collected for 48 h and the animals were sacrificed at 24 and 48 h after the exposure. Significant increased levels of urinary volumes, glucose as well as increased levels of N-acetyl-beta-D-glucosaminidase (NAG), were observed at higher dose levels of ACN at 24 h. Light microscopy showed an increase in size and number of renal lysosomes or dense bodies at
