Nine groups of four 18- to 24-month-old rams were inoculated with Actinobacillus seminis by the following routes: intraconjunctival, intranasal, oral, intravenous, intramuscular, intraepididymal, vas deferens, intraurethral or intrapreputial. Eight similar rams were left uninoculated as controls. Systemic clinical signs were minimal and were confined primarily to the inoculation sites and the scrotal contents. Mild to severe epididymitis resulted from all the routes of inoculation except intraconjunctival and intranasal. Direct inoculation into the genital tract, especially into the cauda epid

Actinobacillus seminis is a gram-negative bacterium of the Pasteurellaceae family that is involved in ovine epididymitis. Looking for a protein specific to this species, we determined the protein profile of subcellular fractions of A. seminis (American Type Culture Collection number 15768): proteins from the outer membrane (OMPs), inner membrane (IMPs), and cytoplasm (CPs). These profiles provide the first data, to our knowledge, regarding subcellular fractions of A. seminis. In the OMP fraction, we identified a protein with a molecular mass of 75 kDa that proved to be immunogenic and apparent

A survey was conducted to establish the prevalence of Actinobacillus seminis in ovine semen and the possible importance of the organism as a cause of ram infertility. A seminis was isolated from three of 16 infertile rams and the infected animals had lesions of the genital tract and produced poor quality semen. A seminis was also isolated from two of 96 fertile rams used as donors of semen for artificial insemination programmes. The strains of A seminis could be identified on the basis of their API ZYM profiles. The results suggest that in the United Kingdom A seminis should be considered as a

Twenty-four isolates of Actinobacillus seminis were typed by PCR ribotyping, repetitive extragenic palindromic element (REP)-based PCR, and enterobacterial repetitive intergenic consensus (ERIC)-based PCR. Five types were distinguished by REP-PCR, and nine types were distinguished by ERIC-PCR. PCR ribotyping produced the simplest pattern and could be useful for identification of A. seminis and for its differentiation from related species. REP- and ERIC-PCR could be used for strain differentiation in epidemiological studies of A. seminis.

AIM: To investigate and assess the sequential pathological changes in the epididymis and testis of young rams injected intra-epididymally with Actinobacillus seminis.
METHODS: Twenty yearling Suffolk and Suffolk-cross rams were randomly divided into two groups comprising 16 test and four control animals. Each test ram received 2.3 x 109 cfu/ml of A. seminis injected intra-epididymally. Every 24 h post-inoculation (p.i.), two test rams were randomly selected, euthanised, and necropsied, until the end of the experiment at 192 h p.i. One control animal was euthanised at 24 h, 48 h, 96 h and 144

The objective of this study was to experimentally evaluate the pathogenicity of an Actinobacillus seminis isolate named SAAS01 in goats. Animals were challenged with 2 mL of a suspension containing 1,5 × 108 CFU/mL of A. seminis (SAAS01 isolate) through the intrapreputial, epididymis tail, and conjunctival routes. Epididymis and testicular fragments were submitted to histopathological exam, and semen samples underwent microbiological and molecular diagnoses. Clinically, a unilateral increase in firm consistency was observed in the epididymis and testicles of two animals inoculated in epididymi

Infectious epididymitis is considered a major cause of economic losses for the sheep industry worldwide. This study aimed to investigate clinical and pathological changes associated with experimental infections with A. seminis and H. somni in rams. Twenty rams of age 18 to 24 months were infected by intraepididymal inoculation of A. seminis (n = 10) and H. somni (n = 10). Rams were weekly examined and biological samples were collected during six weeks. All rams inoculated with A. seminis and 80% inoculated with H. somni became infected. The recovery of bacteria was possible in semen and urine

Actinobacillus seminis causes epididymitis in ruminants and spontaneous abortion. A. seminis virulence factors remain unknown; however, it has been shown that testosterone increased bacterial growth, adhesins expression, and protease expression, as well as biofilm production, suggesting the presence of a bacterial receptor for this hormone. The present study reported the isolation of two potential testosterone receptors from A. seminis grown in the presence of testosterone. A 75 kDa and a 55 kDa proteins were isolated from bacterial lysate with triazine orange-Sepharose CL6B column or with imm

Ovine epididymitis, caused by Actinobacilus seminis, is an infectious disease that produces atrophy of the testis, low fertility, and sterility in infected animals. Iron is a microelement necessary for different vital functions in all organisms and most microorganisms. A. seminis iron acquisition mechanisms are undiscovered. For this reason, this work aimed to know the mechanisms this bacterium possesses to respond when grown in an iron restriction culture medium. A. seminis up-expressed three proteins, including a transferrin binding protein, and down-expressed seven (enzymes and putative adh

BACKGROUND: Infectious ovine epididymitis results in substantial economic losses worldwide due to reproductive failure and culling of breeders. The most common causative agents of these infections are Brucella ovis, Actinobacillus seminis, and Histophilus somni. The aim of this study was to develop a multiplex PCR assay for simultaneous detection of Brucella ovis, Actinobacillus seminis, and Histophilus somni with species-specific primers applied to biological samples for molecular diagnosis of these infections.
RESULTS: The multiplex assay was capable of detecting B. ovis, A. seminis, and H.

Actinobacillus seminis infection in rams constitutes a spectrum of pathological changes in various genital organs, with a predilection for the cauda epididymis. There is a need to understand the disease, as it represents a significant factor contributing to infertility and sterility. Here, we aim to provide a comprehensive overview of the biological characteristics of A. seminis, modes of transmission, epidemiology and pathogenesis, clinical signs and pathological changes of the disease, the laboratory techniques that have been used in diagnosis, differential diagnosis, treatment and preventio

Objective of the present paper is to review microbial diseases of the genital system of male small ruminants. The paper reviews the infections and the diseases by taking an organ approach within the genital system, whilst relevant health management actions are also discussed. Diseases of the genital organs of male small ruminants include orchitis, of bacterial or viral aetiology, epididymitis, primarily caused by Brucella ovis, by other bacteria as well (e.g., Actinobacillus seminis, Haemophilus somni), infections of the accessory glands, orf, other infections of the penis or prepuce and infec

Seventy-nine suspected false-positive sera, obtained over 1 year from routine submissions for Brucella ovis serological testing, were used in this study. These sera, which exhibited titres in the complement fixation test, but which because of their epidemiological history and their reactions in the enzyme-linked immunosorbent assay and gel diffusion test were suspected to be false positives, were further analysed by immunoblotting. In blots, using B. ovis antigens, rough lipopolysaccharide was identified as the major, immuno-reactive bacterial component. Antibodies against this macromolecule w

To date, there is a lack of research into the vaginal and sperm microbiome and its bearing on artificial insemination (AI) success in the ovine species. Using hypervariable regions V3-V4 of the 16S rRNA, we describe, for the first time, the combined effect of the ovine microbiome of both females (50 ewes belonging to five herds) and males (five AI rams from an AI center) on AI outcome. Differences in microbiota abundance between pregnant and non-pregnant ewes and between ewes carrying progesterone-releasing intravaginal devices (PRID) with or without antibiotic were tested at different taxonom
