Speciation of actinides, and more particularly bioligand-binding ability, influences in vivo behavior. Understanding these interactions is essential for estimation of radiological dose and improvement of decorporation strategies for accidentally contaminated victims. Because the handling of actinides imposes overwhelming difficulties, in vitro assays carried out in physiological conditions are lacking and data regarding such interactions are scarce. In this study, we used a bi-compartmental and dynamic assay, providing physiological conditions (presence of inorganic ions, pH, temperature) to e

This Perspective article reviews the current knowledge regarding the interaction of actinide elements with biomolecules, particularly amino acids, peptides, and proteins. We assess the significance of these interactions, especially in connection to nuclear waste disposal and the potential role of these interactions in the origin of life. Actinides have been observed to form stable complexes with carboxylate groups, resulting in oligomerization and affecting their environmental mobility. The text discusses the complex coordination chemistry of actinides, including the prevalence of hexanuclear

Filter swipe tests are used for routine analyses of actinides in nuclear industrial, research, and weapon facilities as well as following accidental release. Actinide physicochemical properties will determine in part bioavailability and internal contamination levels. The aim of this work was to develop and validate a new approach to predict actinide bioavailability recovered by filter swipe tests. As proof of concept and to simulate a routine or an accidental situation, filter swipes were obtained from a nuclear research facility glove box. A recently-developed biomimetic assay for prediction

Original α-aminobisphosphonate-based copolymers were synthesized and successfully used for actinide complexation. For this purpose, poly(α-chloro-ε-caprolactone-co-ε-caprolactone)-b-poly(ethylene glycol)-b-poly(α-chloro-ε-caprolactone-co-ε-caprolactone) copolymers were first prepared by ring-opening copolymerization of ε-caprolactone (εCL) and α-chloro-ε-caprolactone using poly(ethylene glycol) (PEG) as a macro-initiator and tin(II) octanoate as a catalyst. The chloride functions were then converted to azide moieties by chemical modification, and finally α-aminobisphosphonate alkyne ligand (Tz

Assessing the fate of contaminants in the environment requires a deep understanding of intrinsic adsorption mechanisms on natural minerals such as Fe-oxyhydroxides. In this study, we proposed an innovative approach to probe site heterogeneities on the goethite surface by comparing the adsorption behavior of rare earth elements (REEs, including Sc, Y, and all lanthanides; Ln) except Pm, as well as Th and U. A surface loading-dependent adsorption of Ln and Y was observed, with a shift from (i) preferential middle to heavy REE adsorption and (ii) limited to substantial fractionation between Y and

Chelation is considered the best method for detoxification by promoting excretion of actinides (Am, Np, Pu, Th, U) from the human body after internal contamination. Chemical agents that possess carboxylic acid or hydroxypyridinonate groups play a vital role in actinide decorporation. In this review article, we provide considerable background details on the chelation chemistry of actinides with an aim to formulate better decorporation agents. Nanocarriers for pulmonary delivery represent an exciting prospect in the development of novel therapies for actinide decorporation that both reduce toxic

In case of an incident in the nuclear industry or an act of war or terrorism, the dissemination of plutonium could contaminate the environment and, hence, humans. Human contamination mainly occurs via inhalation and/or wounding (and, less likely, ingestion). In such cases, plutonium, if soluble, reaches circulation, whereas the poorly soluble fraction (such as small colloids) is trapped in alveolar macrophages or remains at the site of wounding. Once in the blood, the plutonium is delivered to the liver and/or to the bone, particularly into its mineral part, mostly composed of hydroxyapatite.

This review is focused on recent developments on hydroxypyri(mi)dines, as aluminum and actinide chelating agents to combat the toxicity due to accumulations of these metal ions in human body resulting from excessive metal exposure. After a brief update revision of the most common processes of aluminum (Al) exposure, as well as the associated toxicities and pathologies, we will focus on the current available Al chelators and future perspective as potential antidotes of Al toxicity. Due to the similarity between Al and Fe, a major emphasis is given to the hydroxypyridinone and hydroxypyrimidinon

OBJECTIVES: To describe the process of the development of evidence-based guidelines on the assessment and clinical management of internal contamination with transuranic actinides (specifically plutonium, americium, and curium) in incidents where workers, emergency responders, and the public might uptake these radionuclides internally through inhalation, ingestion, or wound contamination.
METHODS: The World Health Organization (WHO) set up a guidelines development group (GDG) that follows the protocol required for producing evidence-based recommendations as described elsewhere. The GRADE® appr

Complexation of trivalent actinides with DTPA (diethylenetriamine pentaacetic acid) was studied as a function of pcH and temperature in (Na,H)Cl medium of 0.1 M ionic strength. Formation constants of both complexes AnHDTPA(-) and AnDTPA(2-) (where An stands for Am, Cm, and Cf) were determined by TRLFS, CE-ICP-MS, spectrophotometry, and solvent extraction. The values of formation constants obtained from the different techniques are coherent and consistent with reinterpreted literature data, showing a higher stability of Cf complexes than Am and Cm complexes. The effect of temperature indicates

Skin contamination by alpha-emitting actinides is a risk to workers during nuclear fuel production and reactor decommissioning. Also, the list of items for potential use in radiological dispersal devices includes plutonium and americium. The actinide chemical form is important and solvents such as tributyl phosphate, used to extract plutonium, can influence plutonium behavior. This study investigated skin fixation and efficacy of decontamination products for these actinide forms using viable pig skin in the Franz cell diffusion system. Commonly used or recommended decontamination products such

Studies were conducted to demonstrate the ability to directly extract U from pure and saline waters and then rapidly detect trace (0.02-0.2 mBq/mL) gross alpha particle emissions using centrifugally tensioned metastable fluid detectors (CTMFDs), which enable 4π (∼99 %) intrinsic alpha detection efficiency with keV-level resolution, while also exhibiting insensitivity to gamma and beta radiation. Actinide resin-extractant cartridges were used to extract U (in fresh and saline water) and Pu (in fresh water). A CTMFD-specific protocol was developed to remove the extractant and actinide from the c

In the case of an accidental nuclear event, contamination of human bodies by actinide elements may occur. Such elements have the particularity to exhibit both radiological and chemical toxicities that may induce severe damages at several levels, depending on the biokinetics of the element. In order to eliminate the actinide elements before they are stored in target organs (liver, kidneys, or bone, depending on the element), sequestering agents must be quickly injected. However, to date, there is still no ideal sequestering agent, despite the recent interest in this topic due to contamination c

Worldwide stocks of actinides and lanthanide fission products produced through conventional nuclear spent fuel are increasing continuously, resulting in a growing risk of environmental and human exposure to these toxic radioactive metal ions. Understanding the biomolecular pathways involved in mammalian uptake, transport and storage of these f-elements is crucial to the development of new decontamination strategies and could also be beneficial to the design of new containment and separation processes. To start unraveling these pathways, our approach takes advantage of the unique spectroscopic

The work of the Task Group 5.2 'Research Studies on Biokinetic Models' of the CONRAD project is presented. New biokinetic models have been implemented by several European institutions. Quality assurance procedures included intercomparison of the results as well as quality assurance of model formulation. Additionally, the use of the models was examined leading to proposals of tuning parameters. Stable isotope studies were evaluated with respect to their implications to the new models, and new biokinetic models were proposed on the basis of their results. Furthermore, the development of a biokin

The release of actinides into the environment poses a significant risk of their uncontrolled migration in various physical and chemical forms. A key migration mechanism for all actinides is their pseudocolloidal transport, which occurs through sorption onto suspended particles of soils, clays, iron oxides and hydroxides, and other colloids surface in groundwater. This phenomenon facilitates their potential transport over considerable distances, creating additional environmental risks. The behavior of these pseudocolloidal particles is currently not well understood and is highly dependent on a

A systematic study of interactions of U, Np and Pu ions with water in their all possible oxidation states (OSs) has been performed using DFT with triple zeta basis sets, hybrid functional and D3 dispersion correction. In gaseous phase, the maximum coordination of 9 water molecules is observed for U in both +3 and + 4 OSs and Np in +4 OS, whereas the same of 10 water molecules is obtained in +3 OS of Np and both +3 and + 4 OSs of Pu. However, in +5 and + 6 OSs, the actinide oxy ions are solvated by maximum 5 water molecules for all the three actinides, whereas in +7 state of both Np and Pu, the

The release of actinides into the environment represents a significant potential public health concern. Chelation therapy utilizing diethylenetriamine pentaacetate (DTPA) is a U.S. Food and Drug Administration (FDA)-approved therapy capable of mitigating the deposition of some absorbed actinides in the body. However, the pharmacokinetic profile of DTPA is not ideal for prophylactic applications. In this study, we examine the incorporation of DTPA into a HPMA copolymer (P-DTPA) to investigate if the enhanced blood circulation time can offer superior prophylactic protection and of improving in v

All of the actinides are radioactive. Taken into the body, they damage and induce cancer in bone and liver, and in the lungs if inhaled, and U(VI) is a chemical kidney poison. Containment of radionuclides is fundamental to radiation protection, but if it is breached accidentally or deliberately, decontamination of exposed persons is needed to reduce the consequences of radionuclide intake. The only known way to reduce the health risks of internally deposited actinides is to accelerate their excretion with chelating agents. For more than three decades, the United States Transuranium and Uranium Registries (USTUR) have studied the biokinetics, dosimetry, and biological effects of plutonium, uranium, and americium through voluntary postmortem tissues from persons with known intakes. Radiochemical analyses of tissue obtained at autopsy have shown that plutonium and americium have different biokinetics and an appreciable deposition in the soft tissues of the body as well as the known depots in skeleton and liver. Studies of whole-body Thorotrast donors to the USTUR indicate that commonly accepted risk coefficients fo
