Alpha-adrenergic antagonists: correlation of the effect on intraocular pressure and on alpha 2-adrenergic receptor binding specificity in the rabbit eye.

Six alpha-adrenergic antagonists, which have a range of selectivities for alpha 1- and alpha 2-adrenoreceptor subtypes, were compared with respect to their ability to reduce intraocular pressure (IOP) after topical application to the rabbit eye, and their affinity and selectivity for alpha 2-adrenoreceptors, as determined by binding to membranes prepared from rabbit iris-ciliary body. A routine assay for alpha 2-adrenoreceptors using [3H]-rauwolscine was developed for this purpose. ICB contained 200-300 fmol (mg protein)-1 alpha 2-adrenoreceptors which represents approximately two-thirds of th

Effect of alpha 2-adrenergic receptor antagonist (midaglizole) on gastrointestinal motility in conscious dogs.

To clarify the physiological role of the mechanism that adrenergic nerve inhibits Ach release from intramural cholinergic nerve endings, the influence of Midaglizole, alpha 2-adrenergic receptor antagonist, to postprandial gastrointestinal motilities in conscious dogs was investigated. Postprandial motilities of gastric antrum, duodenum, ileum, and colon were significantly enhanced by Midaglizole (3.0-5.0 mg/kg body weight, i.v.). These excitatory responses were abolished by atropine (0.05-0.1 mg/kg body weight, i.v.). On the other hand, in most cases (29 cases out of 32), when Midaglizole was

Adrenergic alpha-2 receptor antagonists cease augmented oxidation of plasma proteins and anxiety of rats caused by chronic noise exposure.

BACKGROUND: Noise is one of the environmental factors, which is considered as a powerful stressor for the organism. Generally, the acoustic stress affects the behavior and physiological state of humans and animals. AIMS: The goal of this study is to investigate the relationship between chronic noise exposure and the effects of adrenergic alpha-2 receptor antagonists, beditin and mesedin, on the anxiety and oxidation of plasma proteins and fibrinogen in rats. METHODS: The experiments were carried out on non-linear albino male rats, divided into four groups (six animals in each): 1. Healthy co

Identification and characterization of alpha 2D-adrenergic receptors in bovine pineal gland.

The vertebrate mechanisms for generating circadian rhythms, regulating the synthesis of melatonin, and modulating the activities of pineal indole-synthesizing enzymes differ significantly among species. The synthesis of melatonin in mammalian pineal glands is stimulated by beta 1-adrenergic receptor agonists and is potentiated by alpha 1-adrenergic receptor agonists, whereas in the avian pineal gland alpha 2-adrenergic receptor agonists inhibit its synthesis. By using [3H]rauwolscine, a selective alpha 2-adrenergic receptor antagonist, we have identified for the first time alpha 2-adrenergic r

Activation of alpha-1 adrenergic receptors modulates the control of left/right sidedness in rat embryos.

Presomite stage rat embryos were cultured for 45-49 hr with medium containing various adrenergic agonists and antagonists. L-Norepinephrine but not D-norepinephrine (several orders of magnitude less potent than the L-isomer at alpha-1 adrenergic receptors) resulted in a dose-dependent increase of situs inversus similar to that found for phenylephrine, an alpha-1 adrenergic agonist. Prazosin, an alpha-1 adrenergic antagonist, inhibited phenylephrine-induced situs inversus in a dose-dependent manner. Neither dexmedetomidine, an alpha-2 adrenergic agonist, nor isoproterenol, a beta adrenergic ago

Binding and functional characterization of alpha-2 adrenergic receptor subtypes on pig vascular endothelium.

Alpha-2 adrenergic receptor subtypes were characterized in membranes of pig vascular endothelium using [3H]rauwolscine. Alpha-2 adrenergic receptor subtypes that mediate endothelium-dependent vascular relaxation were studied in vitro by using ring segments of pig epicardial coronary arteries. Specific [3H]rauwolscine binding in endothelial membranes was saturable and to a single class of high-affinity sites with a mean KD of 0.217 +/- 0.05 nM and Bmax of 156 +/- 28 fmol/mg of protein. Nonlinear regression analysis indicated that competition binding curves for drugs that distinguish the alpha-2

Facilitation of estrous behavior by vaginal cervical stimulation in female rats involves alpha1-adrenergic receptor activation of the nitric oxide pathway.

In estrogen-primed female rats, vaginal cervical stimulation (VCS) provided by male intromissions or by an experimenter enhances estrous behaviors exhibited by females during subsequent mating with a male. We tested the hypothesis that alpha(1)-adrenergic receptors, acting via the nitric oxide-cGMP-protein kinase G pathway, mediate VCS-induced facilitation of female reproductive behaviors. Ovariectomized, estradiol-primed rats received intracerebroventricular (icv) infusions of vehicle or pharmacological antagonists 15 or 60min before VCS. Estrous behaviors (lordosis and proceptivity) in the p

Converging adrenergic and cholinergic mechanisms in the inhibition of Cl secretion in fish opercular epithelium.

The rate of Cl secretion (Isc) by the opercular epithelium of Fundulus heteroclitus is stimulated by elevations in cyclic AMP (cAMP) levels elicited via beta 1-adrenergic receptor activation, and inhibited by both alpha 2-adrenergic and muscarinic cholinergic receptor activation via mechanisms presently unknown. A comparison of these two inhibitory responses was made using clonidine, an alpha 2-adrenergic agonist, and acetylcholine (ACh), a cholinergic agonist. The dose required for maximum inhibition was 100 times greater for ACh, but in all other respects the responses elicited by both agoni

Blockade of alpha-2 adrenergic receptors in the rostral ventrolateral medulla attenuates the sympathoinhibitory response to cocaine.

The purpose of this study was to determine whether neurons in the rostral ventrolateral medulla play a role in the sympathoinhibitory response elicited by i.v. administration of cocaine and, if so, to identify the type(s) of receptors involved. Adrenergic antagonists were microinjected bilaterally into the rostral ventrolateral medulla in pentobarbital-anesthetized rats in an attempt to block the decrease in sympathetic nerve discharge (SND) elicited by cocaine (1 mg/kg i.v.). After the bilateral microinjection of saline, cocaine elicited a -56 +/- 5% (mean +/- S.E.) decrease in SND lasting 36

[3H]Dihydroergocryptine binding to alpha-adrenergic receptors of human platelets. A reassessment using the selective radioligands [3H]prazosin, [3H]yohimbine, and [3H]rauwolscine.

Which subtype(s) of the alpha-adrenergic receptor occurs on human platelets? Studies of platelet responsiveness to adrenergic compounds and indirect radioligand binding studies addressing this question have yielded contradictory conclusions. These bindings studies employed the ligand [3H]dihydroergocryptine ( [3H]DHE), an alpha-adrenergic antagonist that does not select between alpha 1- and alpha 2-adrenergic receptors and that also binds to other receptor types in some tissues. To determine the subtype of the platelet alpha-adrenergic receptor, we have examined the binding to intact human pla

Cathine, an amphetamine-related compound, acts on mammalian spermatozoa via beta1- and alpha2A-adrenergic receptors in a capacitation state-dependent manner.

BACKGROUND: Mammalian spermatozoa have been shown to have beta(1,2,3)- and alpha(2A)-adrenergic receptors, the former functioning only in uncapacitated spermatozoa and the latter only in capacitated cells. Cathine, an amphetamine-related metabolite of a compound found in Catha edulis leaves, accelerates capacitation and inhibits spontaneous acrosome loss by regulating cAMP production. This study tested the hypothesis that adrenergic receptors are involved in these responses. METHODS: Uncapacitated and capacitated mouse sperm suspensions were incubated with cathine +/- specific antagonists for

Inhibitory effects of clonidine and dopamine on adenylate cyclase of rabbit ciliary processes.

The inhibitory effects of the alpha 2-adrenergic agonist clonidine and that of dopamine were studied on the adenylate cyclase activity in homogenates of ciliary processes. Clonidine inhibited in a dose-dependent manner basal adenylate cyclase activity as well as that stimulated by isoproterenol or forskolin. However, the adenylate cyclase activity stimulated by isoproterenol was sensitive to at least one order lower inhibitory concentrations of clonidine than basal or forskolin-stimulated adenylate cyclase. Dopamine inhibited adenylate cyclase stimulated by isoproterenol considerably less pote

Interaction between alpha 2- and beta 2-adrenergic receptors in rabbit ciliary processes.

The interaction between the alpha 2- and beta 2-adrenergic receptors of ciliary processes has been studied by examining dose-response curves for adrenergic agonist stimulation of cyclic AMP production by intact, excised rabbit ciliary processes. Stimulation of cyclic AMP production by 1-isoproterenol is maximum from 0.1 to 1.0 microM; at higher concentrations stimulation decreases and approaches basal levels. Decreased cyclic AMP production at high concentrations of isoproterenol is blocked by the specific alpha 2-adrenergic antagonist, yohimbine, but not by the alpha 1-adrenergic antagonist,

Antinociception following implantation of mouse B16 melanoma cells in mouse and rat spinal cord.

B16 F1C29 melanoma cells, which are thought to contain and release catecholamines, were implanted in mouse and rat spinal subarachnoid space. B16 F1C29 cell implants augmented the antinociceptive effect of morphine in tail-flick test, and this interaction was blocked by either the alpha 2-adrenergic antagonist idazoxan or the opioid antagonist naloxone. B16 F1C29 cell implants also augmented the antinociceptive effect of the catecholamine re-uptake blocker desipramine. Substance P-induced biting and scratching behaviors were inhibited in mice receiving B16 F1C29 cell implants, and this effect

The role of adrenoreceptors in control of stereotyped oral behavior in restricted-fed fowls.

Effects on environmentally induced oral stereotypies (object pecking and drinker-directed activity) of preferential antagonists and agonists of adrenoreceptor subtypes were examined in individually caged broiler breeder fowls subjected to chronic food restriction. Three drugs in each category were injected intravenously at three doses, and their effects compared with a saline control treatment. With the antagonists, object pecking was suppressed more by prazosin (alpha 1) and propranolol (beta) than by yohimbine (alpha 2), while drinker-directed activity showed delayed stimulation with yohimbi

Effect of various catecholamine antagonists on prolactin secretion in conscious male rabbits.

To clarify physiological roles of catecholaminergic systems in the control of rabbit prolactin (PRL) release, the effect of various catecholamine receptor antagonists on plasma PRL levels was examined in conscious, freely moving male rabbits. An intravenous (iv) injection of yohimbin (2.5 mg/kg body wt), an alpha 2-adrenoreceptor antagonist, but not prazosin (2 mg/kg body wt), an alpha 1-adrenergic receptor antagonist, resulted in a significant elevation of plasma PRL. Physiological role of alpha-adrenoceptors in salivary secretion.

The submaxillary gland (SM) of rat is innervated by both branches of the autonomic nervous system. Secretion is mediated by the activation of both muscarinic-cholinergic and alpha/beta adrenergic receptors. Studies of the relative affinity of pharmacological agonists and antagonists have warranted a subclassification of alpha adrenoceptors into types alpha 1 and alpha 2. Our studies involve an analysis of the physiologic role of both types of alpha adrenoceptors in salivary secretion. Alpha-2 adrenergic activity of bromocriptine and quinpirole in chicken pineal gland. Effects on melatonin synthesis and [3H]rauwolscine binding.

In the pineal gland and retina of chickens, serotonin N-acetyl-transferase (NAT) activity and melatonin content are modulated by different receptors, alpha-2 adrenergic receptors in pineal gland and D2-dopamine receptors in retina. The effect of two D2-dopamine receptor agonists, bromocriptine and quinpirole (LY 171555), on melatonin synthesis in these tissues was investigated. Systemic administrations of bromocriptine and quinpirole decreased nocturnal NAT activity and melatonin content of both pineal gland and retina.
