Ethanol is detoxified in the liver, and its intake causes hepatic lipid accumulation. The liver receptor homolog-1 (LRH-1) regulates lipid and bile acid metabolism, but its role in ethanol metabolism remains unclear. This study aimed to explore the relationship between ethanol-induced lipid accumulation and LRH-1. To investigate the role of LRH-1 in hepatic ethanol metabolism, LRH-1f/f and liver-specific LRH-1f/cre+ mice were fed a Lieber-DeCarli diet for 3 weeks. The results showed that ethanol-fed LRH-1f/cre+ mice exhibited increased neutral fat, total cholesterol, liver damage markers, and

Aldehyde Dehydrogenase 1, Family Member A2 (ALDH1A2) is essential for the synthesis of retinoic acid from vitamin A. Studies in model organisms demonstrate a critical role for ALDH1A2 in embryonic development, yet few pathogenic variants are linked to congenital anomalies in humans. We present three siblings with multiple congenital anomaly syndrome linked to biallelic sequence variants in ALDH1A2. The major congenital malformations affecting these children include tetralogy of Fallot, absent thymus, diaphragmatic eventration, and talipes equinovarus. Upper airway anomalies, hypocalcemia, and

Unlike the well-documented effects of aldehyde dehydrogenase 2 (ALDH2) hypoactive variants, the impact of hypoactive ALDH1B1 has yet to be elucidated. Using the Global Human Genome Aggregation Database, we identified 38 ALDH1B1 variants, common (>0.1% allele frequency) to different human genetic ancestral groups. Some missense ALDH1B1 human variants are notably less active and less stable, and three variants represent a complete loss of function (LoF) due to early stop codon. Since these inactive variants are common among humans, we studied the impact of carrying both ALDH1B1 LoF and ALDH2 ina

Disulfiram is an FDA-approved drug used to treat chronic alcoholism. This drug works by blocking the second step of ethanol metabolism by inhibiting aldehyde dehydrogenase-2 (ALDH2), the enzyme responsible for acetaldehyde oxidation into acetic acid. This leads to the accumulation of acetaldehyde in the blood following alcohol ingestion and to highly unpleasant symptoms known as acetaldehyde syndrome. Disulfiram also inhibits ALDH1a1, another member of the aldehyde dehydrogenases that catalyzes the oxidation of retinal into retinoic acid. ALDH1a1 represents a key therapeutic target for the tre

Autosomal recessive cutis laxa (ARCL) is a connective tissue disorder characterized by wrinkled, inelastic skin, frequently associated with a neurologic involvement and multisystem disease. Next generation sequencing was performed in genetically unsolved patients with progeroid features, neurological and eye involvement to assess the underlying etiology. We describe an 6 month old child, diagnosed with a novel, homozygous nonsense mutation c.2339T>C in exon 18 of the ALDH18A1 gene, and reviewed all reported P5CS patients. So far 10 patients were described with mutations in ALDH18A1. Features o

Cyclophosphamide (CP) is one of the most widely used anticancer drugs for various malignancies. However, its long-term use leads to ALDH1A1-mediated inactivation and subsequent resistance which necessitates the development of potential ALDH1A1 inhibitors. Currently, ALDH1A1 inhibitors from different chemical classes have been reported, but these failed to reach the market due to safety and efficacy problems. Developing a new treatment from the ground requires a huge amount of time, effort, and money, therefore it is worthwhile to improve CP efficacy by proposing better adjuvants as ALDH1A1 inh

While numerous studies have examined the subjective response to alcohol as an intermediate phenotype to understand its variability, heritability, and predictive capacity for alcohol-related disorders, in-depth analyses linking alcohol reactivity indicators to genetic factors within a large cohort have been absent. Our study aimed to quantify the exact contribution of each genetic variant relevant to the alcohol metabolism to the variability in alcohol response. Specifically, we focused on two primary genes involved in alcohol metabolism (ALDH2 and ADH1B) and three additional loci (ALDH1B1, ALD

Air pollution is a serious environmental threat to public health; however, the molecular basis underlying its detrimental effects on respiratory fitness remains poorly understood. Here, we showed that exposure to particulate matter ≤ 2.5 μm (PM2.5), a substantial fraction of air pollutants, induced the generation of reactive aldehyde species in the airway. We identified aldehyde dehydrogenase 1A1 (ALDH1A1), which was selectively expressed in airway epithelium, as an enzyme responsible for detoxifying these reactive aldehyde species. Loss of ALDH1A1 function resulted in the accumulation of alde

Cheng et al. identify aldehyde dehydrogenase 1A1 as a novel risk factor for autosomal dominant polycystic kidney disease, by both activating proliferative pathways and acting as a transcription factor. Targeting aldehyde dehydrogenase 1A1 with disulfiram delays cyst growth in autosomal dominant polycystic kidney disease mice. Combining low-dose disulfiram with anti-programmed death ligand 1 antibody synergistically attenuates cyst progression and improves the immune microenvironment. Although aldehyde dehydrogenase 1A1 upregulation occurs in other kidney diseases and its transcriptional mechan

Disruption of the retinoid signaling pathway by environmental chemicals is currently not directly assessed in regulatory testing. Although in vitro and in silico methods are available to assess disruption of this modality, more data on the selectivity and in vivo predictivity of specific assays are needed. To address this, we tested twenty-one pesticides and pharmaceuticals for their ability to agonize or antagonize the retinoic acid receptor alpha (RARα) or to inhibit the activity of aldehyde dehydrogenases (ALDH1As). Experimental data was generated using a RARα reporter gene assay and a cell

Dopaminergic neurons in the substantia nigra pars compacta (SNpc) demonstrate regionally selective susceptibility in Parkinson's disease (PD) compared to those in the ventral tegmental area (VTA). However, the molecular mechanism for this distinct vulnerability remains unclear. Here, it is shown that Legumain, also known as asparagine endopeptidase (AEP), is activated in a subgroup of SRY-box transcription factor 6 /Aldehyde dehydrogenase 1 family member A1, (Sox6+/ALDH1A1+) neurons in the ventral tier of the SNpc and cleaves Sox6 and ALDH1A1, leading to repression of Special AT-rich sequence

Retinoic acid signaling pathway plays a role in regulating vertebrate development, cell differentiation, and homeostasis. As a key enzyme that catalyzes the oxidation of retinal to retinoic acid, aldehyde dehydrogenase 1 family member A2 (Aldh1a2) is involved in cardiac development, while whether it functions in heart diseases remains to be studied. In this study, we infected primary cardiomyocytes with adenovirus overexpressing Aldh1a2 (Ad-Aldh1a2) to explore the effects of Aldh1a2 overexpression on the biological function of cardiomyocytes. The results showed that the infection with Ad-Aldh1

Adult stem cells are thought to drive the regenerative potential of the endometrium and contribute to the pathogenesis of endometriosis; however, their identity and defining features remain to be characterized. Here, we used in vivo and in vitro approaches to demonstrate that cells with high aldehyde dehydrogenase 1 activity (ALDHHI cells) were long-lived progenitors in the endometrium with a higher organoid formation capacity, long-term passaging potential, and stemness gene signatures. Aldehyde dehydrogenase 1 (ALDH-1) is a marker of stem cells in a variety of diseases, but its role in individuals with chronic inflammatory periapical lesions remains unknown. The aim of this study was to investigate the presence of cells with a stem cell profile based on the immunoexpression of ALDH-1 in periapical granulomas (PGs) and radicular cysts (RCs). A total of 51 cases of periapical lesions (25 PGs and 26 RCs) were subjected to immunohistochemical study. The anti-ALDH-1 antibody was applied using the immunoperoxidase technique. An immunoexpression score (intensity vs. percentage of c

Folic acid (FA) supplementation during pregnancy is the commonly accepted treatment to prevent neural tube defects. The mechanism by which FA prevents neural tube defects (NTDs) remains unclear. FA also prevents other developmental malformations, including alcohol-induced malformations in Fetal Alcohol Syndrome models. We show that FA acts through a metabolic link to retinoic acid (RA) signaling. Using a pax3-knockdown Xenopus model of FA-rescuable NTDs, we show that RA or its precursors equally rescue these defects. Similarly, FA rescues alcohol-induced NTDs in a model previously shown to be

The transcription factor nuclear factor erythroid 2-related factor 2 (NRF2) is a master regulator of the cellular antioxidant response that defends against ferroptosis and facilitates tumour development. Here, by performing an aldehyde dehydrogenase (ALDH) family shRNA screen, our study identifies aldehyde dehydrogenase 3 family member A2 (ALDH3A2) as a potent NRF2 activator that inhibits ferroptosis. Interestingly, ALDH3A2 efficiently activates NRF2 in both wild-type (WT) Kelch-like ECH-associated protein 1 (KEAP1) and mutant KEAP1 tumour cells. Mechanistically, ALDH3A2 inhibits the phosphory

BACKGROUND: Although hypersensitivity reactions following intake of alcoholic drinks are common in Caucasians, the underlying mechanisms and clinical significance are not known. In contrast, in Asians, alcohol-induced asthma and flushing have been shown to be because of a single nucleotide polymorphism (SNP), the acetaldehyde dehydrogenase 2 (ALDH2) 487lys, causing decreased acetaldehyde (the metabolite of ethanol) metabolism and high levels of histamine. However, the ALDH2 487lys is absent in Caucasians.
OBJECTIVES: To investigate the genetic determinants of self-reported alcohol-induced hyp

ALDH1B1 is a mitochondrial aldehyde dehydrogenase required for the survival of stem-like colorectal and pancreatic cancer cells. Isoform-selective guanidinyl antagonists of aldehyde dehydrogenases (IGUANAs) have been developed for ALDH1B1, and here we describe structure-activity relationship (SAR) studies of three scaffold components: the bicyclic guanidine core, a substituted aryl ring that engages the catalytic site, and a biaryl group that interacts with flanking residues. We further show that previously reported IGUANAs are metabolically labile and rapidly effluxed from intestinal epitheli

PA2125 from Pseudomonas aeruginosa PAO1 is annotated as an aldehyde dehydrogenase (ALDH), but its substrate specificity, catalytic properties, and phylogenetic family assignment had not been established. Here, we cloned the PA2125 gene, produced and purified the recombinant PA2125 protein, and combined extensive substrate screening and steady-state kinetic characterisation with molecular dockings, molecular dynamics simulations, and phylogenetic analysis. PA2125 preferred NAD+ over NADP+, and screening of 54 aldehydes revealed a narrow substrate range centred on substituted benzaldehydes, part

Slow-transit constipation (STC) is a disabling motility disorder with unclear smooth-muscle mechanisms. Through spatial proteomics of human colon and functional assays in primary human colonic smooth muscle cells (HCoSMCs), we identified Bridging Integrator 1 (BIN1) and Aldehyde Dehydrogenase 1B1 (ALDH1B1) as key regulators of intestinal motility. Both proteins were markedly reduced in smooth muscle from STC patients and localized to fibrotic areas. Lentiviral knockdown of BIN1 or ALDH1B1 impaired ATP-evoked Ca2 + responses and contraction, disrupted mitochondrial architecture, and increased r
